1Soil contamination with antibiotics in a typical peri-urban area in eastern China: Seasonal variation, risk assessment, and microbial responses显示文摘The prevalence and persistence of antibiotics in soils has become an emerging environmental issue and an increasing threat to soil security and global public health. The problem is more severe in areas undergoing rapid urbanization; however, the ecological risks of antibiotics,seasonal variability, and associated soil microbial responses in peri-urban soils have not been well-explored. The seasonal soil sampling campaigns were conducted in a typical peri-urban watershed in eastern China to investigate distribution of antibiotics. The results demonstrated higher mean concentrations of most antibiotic compounds in winter than in summer in periurban soils. The seasonal variations of norfloxacin, enrofloxacin, and ciprofloxacin were more significant than those of other antibiotics, due to their higher migration ability and bioavailability.An ecological risk assessment demonstrated that chlortetracycline, ciprofloxacin, doxycycline,and ofloxacin can pose high risks to soil microorganisms. Furthermore, the coexistence of multiple antibiotics obviously poses higher risks than individual compounds. A redundancy analysis demonstrated that tetracyclines mainly showed negative correlations with Firmicutes and Chloroflexi, and quinolones showed obviously negative correlations with Acidobacteria,Gemmatimonadetes, and Nitrospirae, suggesting potential inhibition from antibiotics on biological activities or biodegradation processes. However, the persistence of antibiotics in soil results in a significant decrease in bacterial diversity and a change in dominant species.Our results provide an overview of the seasonal variability of antibiotics and the associated effects on bacterial communities in peri-urban soils. 3Internet of Things to network smart devices for ecosystem monitoring显示文摘Smart, real-time, low-cost, and distributed ecosystem monitoring is essential for understanding and managing rapidly changing ecosystems. However, new techniques in the big data era have rarely been introduced into operational ecosystem monitoring, particularly for fragile ecosystems in remote areas.We introduce the Internet of Things(IoT) techniques to establish a prototype ecosystem monitoring system by developing innovative smart devices and using IoT technologies for ecosystem monitoring in isolated environments. The developed smart devices include four categories: large-scale and nonintrusive instruments to measure evapotranspiration and soil moisture, in situ observing systems for CO2 and d13 C associated with soil respiration, portable and distributed devices for monitoring vegetation variables, and Bi-CMOS cameras and pressure trigger sensors for terrestrial vertebrate monitoring. These new devices outperform conventional devices and are connected to each other via wireless communication networks. The breakthroughs in the ecosystem monitoring IoT include new data loggers and longdistance wireless sensor network technology that supports the rapid transmission of data from devices to wireless networks. The applicability of this ecosystem monitoring IoT is verified in three fragile ecosystems, including a karst rocky desertification area, the National Park for Amur Tigers, and the oasis-desert ecotone in China. By integrating these devices and technologies with an ecosystem monitoring information system, a seamless data acquisition, transmission, processing, and application IoT is created. 6DNA Extraction Protocol for Biological Ingredient Analysis of Liuwei Dihuang Wan显示文摘Traditional Chinese medicine(TCM) preparations are widely used for healthcare and clinical practice. So far, the methods commonly used for quality evaluation of TCM preparations mainly focused on chemical ingredients. The biological ingredient analysis of TCM preparations is also important because TCM preparations usually contain both plant and animal ingredients,which often include some mis-identified herbal materials, adulterants or even some biological contaminants.For biological ingredient analysis, the efficiency of DNA extraction is an important factor which might affect the accuracy and reliability of identification. The component complexity in TCM preparations is high, and DNA might be destroyed or degraded in different degrees after a series of processing procedures. Therefore, it is necessary to establish an effective protocol for DNA extraction from TCM preparations. In this study, we chose a classical TCM preparation,Liuwei Dihuang Wan(LDW), as an example to develop a TCM-specific DNA extraction method.An optimized cetyl trimethyl ammonium bromide(CTAB) method(TCM-CTAB) and three commonlyused extraction kits were tested for extraction of DNA from LDW samples. Experimental results indicated that DNA with the highest purity and concentration was obtained by using TCM-CTAB. To further evaluate the different extraction methods, amplification of the second internal transcribed spacer(ITS2) and the chloroplast genome trnL intron was carried out.The results have shown that PCR amplification was successful only with template of DNA extracted by using TCM-CTAB. Moreover, we performed high-throughput 454 sequencing using DNA extracted by TCM-CTAB. 7Melatonin lowers edema after spinal cord injury显示文摘Melatonin has been shown to diminish edema in rats. Melatonin can be used to treat spinal cord injury. This study presumed that melatonin could relieve spinal cord edema and examined how it might act. Our experiments found that melatonin(100 mg/kg, i.p.) could reduce the water content of the spinal cord, and suppress the expression of aquaporin-4 and glial fibrillary acidic protein after spinal cord injury. 8The influence of urban park characteristics on bird diversity in Nanjing, China显示文摘Background:Habitat loss,fragmentation and decrease of habitat quality caused by urbanization have led to a dramatic decline in biodiversity worldwide.For highly urbanized areas,parks have become“islands”or habitat fragments for wildlife.As an important indicator group of urban ecosystem health,the response of birds to urbanization has attracted the global attention of ecologists.Understanding the key factors affecting bird diversity in urbanized environment is crucial to the protection of biodiversity in urban ecosystems.Methods:We used the line-transect method to survey birds in 37 urban parks in Nanjing,China.We also measured a number of park characteristics(area,isolation,shape index,environmental noise,distance to city center,and habitat diversity)that are commonly assumed to influence bird diversity.We then used the information-theoretic multi-model inference approach to determine which park characteristics had significant impacts on bird species richness.Results:We found that park area,habitat diversity and the distance to city center were the best positive predictors of bird species richness in Nanjing urban parks.By contrast,park isolation,park shape and environmental noise had little or no influence on bird diversity.Conclusions:Our study highlights the importance of park area,habitat diversity and the distance to city center in determining bird diversity in Nanjing city parks.Therefore,from a conservation viewpoint,we recommend that large parks with complex and diverse habitats far away from the city center should be retained or constructed to increase bird diversity in urban design and planning.Xueru Yang Xinwei Tan Chuanwu Chen Yanping Wang 2020Avian Research2020,11,4:5
9Response of Microbial Community to Petroleum Stress and Phosphate Dosage in Sediments of Jiaozhou Bay, China显示文摘The dynamic microcosms were used to evaluate the effect of oil spills on microbial ecological system in marine sediment and the enhancement of nutrient on the oil removal. The function and structure of microbial community caused by the oil pollution and phosphate dosage were simultaneously monitored by dehydrogenase activity assay and PCR-denaturing gradient gel electrophoresis(DGGE) techniques. The results indicated that the amount of total bacteria in all dynamic microcosms declined rapidly with incubation time. The number of petroleum-degrading bacteria and the activity of sediment dehydrogenase were gradually enhanced by petroleum in the oil-treated microcosms, while they both showed no obvious response to phosphate dosage. In comparison, phosphate spiked heterotrophic bacteria and they showed a significant increase in amount. DGGE profiles indicated that petroleum dosage greatly changed community structure, and the bacteria belonged to class Deltaproteobacteria, and phyla Bacteroidetes and Chlorobi were enriched. This study demonstrated that petroleum input greatly impacted the microbial community structure and consequently the marine sediment petroleum-degrading activity was enhanced. 10PDCD5 regulates iNKT cell terminal maturation and iNKT1 fate decision显示文摘Invariant natural killer T1(iNKT1)cells are characterized by the preferential expression of T-box transcription factor T-bet(encoded by Tbx21)and the production of cytokine IFN-γ,but the relationship between the developmental process and iNKT1 lineage diversification in the thymus remains elusive.We report in the present study a crucial role of programmed cell death 5(PDCD5)in iNKT cell terminal maturation and iNKT1 fate determination.Mice with T cell-specific deletion of PDCD5 had decreased numbers of thymic and peripheral iNKT cells with a predominantly immature phenotype and defects in response toα-galactosylceramide.Loss of PDCD5 also selectively abolished the iNKT1 lineage by reducing T-bet expression in iNKT cells at an early thymic developmental stage(before CD44 upregulation).We further demonstrated that TOX2,one of the high mobility group proteins that was highly expressed in iNKT cells at stage 1 and could be stabilized by PDCD5,promoted the permissive histone H3K4me3 modification in the promoter region of Tbx21.These data indicate a pivotal and unique role of PDCD5/TOX2 in iNKT1 lineage determination.They also suggest that the fate of iNKT1 may be programmed at the developmental stage of iNKT cells in the thymus.Ke Wang Xinwei Zhang Yifan Wang Gaowen Jin Mingyang Li Shusong Zhang Jie Hao Rong Jin Xiaojun Huang Hounan Wu Jun Zhang Yingyu Chen Qing Ge 2019Cellular & Molecular Immunology2019,16,9:4

12Flexible mica films for high-temperature energy storage显示文摘Dielectrics used for energy storage are highly desired for power electronics and pulse power applications and the polymer capacitors are the main commercial ones available.The development of flexible electronics and wearable devices require the relative materials being flexible.Besides,high temperature resistance is also desired because of the rising demand for high power devices and large electricity under elevated temperature.The polymer dielectrics for polymer capacitors are flexible but with relatively low working temperature normally less than 100℃.Therefore,fabricating a dielectric material to satisfy the flexibility and high working temperature simultaneously is still a great challenge.Here we propose one solution by demonstrating a hand-exfoliated fluorophlogopite film with micrometer scale thickness.Among which,the mica film with a thickness of around 10μm(Mica-10)exhibits the inorganic-like temperature stability even polymer-like flexibility.From 25℃to 200℃,Mica-10 has an energy density of around 11.27 J/cm^(3)with a variation within 2%,accompanied by a charge-discharge efficiency of around 95%at an electric field of 500 MV/m.Meanwhile,the thin thickness makes Mica-10 flexible,enabling its excellent flexibility and durability.This work revives the traditional material,mica,providing a way for high-temperature energy storage applications.Xinwei Xu Wenlong Liu Yi Li Yifei Wang Qibin Yuan Jie Chen Rong Ma Feng Xiang Hong Wang 2018Journal of Materiomics2018,4,3:3
13A Novel Artificial Neuron-Like Gas Sensor Constructed from CuS Quantum Dots/Bi2S3 Nanosheets显示文摘Real-time rapid detection of toxic gases at room temperature is particularly important for public health and environmental monitoring.Gas sensors based on conventional bulk materials often suffer from their poor surface-sensitive sites,leading to a very low gas adsorption ability.Moreover,the charge transportation efficiency is usually inhibited by the low defect density of surface-sensitive area than that in the interior.In this work,a gas sensing structure model based on CuS quantum dots/Bi_(2)S_(3) nanosheets(CuS QDs/Bi_(2)S_(3) NSs)inspired by artificial neuron network is constructed.Simulation analysis by density functional calculation revealed that CuS QDs and Bi_(2)S_(3) NSs can be used as the main adsorption sites and charge transport pathways,respectively.Thus,the high-sensitivity sensing of NO_(2) can be realized by designing the artificial neuron-like sensor.The experimental results showed that the CuS QDs with a size of about 8 nm are highly adsorbable,which can enhance the NO_(2) sensitivity due to the rich sensitive sites and quantum size effect.The Bi_(2)S_(3) NSs can be used as a charge transfer network channel to achieve efficient charge collection and transmission.The neuron-like sensor that simulates biological smell shows a significantly enhanced response value(3.4),excellent responsiveness(18 s)and recovery rate(338 s),low theoretical detection limit of 78 ppb,and excellent selectivity for NO_(2).Furthermore,the developed wearable device can also realize the visual detection of NO2 through real-time signal changes.Xinwei Chen Tao Wang Jia Shi Wen Lv Yutong Han Min Zeng Jianhua Yang Nantao Hu Yanjie Su Hao Wei Zhihua Zhou Zhi Yang Yafei Zhang 2022Nano-Micro Letters2022,14,1:3

16Low phosphorus promotes NSP1–NSP2 heterodimerization to enhance strigolactone biosynthesis and regulate shoot and root architecture in rice显示文摘Phosphorus is an essential macronutrient for plant development and metabolism,and plants have evolved ingenious mechanisms to overcome phosphate(Pi)starvation.However,the molecular mechanisms underlying the regulation of shoot and root architecture by low phosphorus conditions and the coordinated utilization of Pi and nitrogen remain largely unclear.Here,we show that Nodulation Signaling Pathway 1(NSP1)and NSP2 regulate rice tiller number by promoting the biosynthesis of strigolactones(SLs),a class of phytohormones with fundamental effects on plant architecture and environmental responses.We found that NSP1 and NSP2 are induced by Oryza sativa PHOSPHATE STARVATION RESPONSE2(OsPHR2)in response to low-Pi stress and form a complex to directly bind the promoters of SL biosynthesis genes,thus markedly increasing SL biosynthesis in rice.Interestingly,the NSP1/2–SL signaling module represses the expression of CROWN ROOTLESS 1(CRL1),a newly identified early SL-responsive gene in roots,to restrain lateral root density under Pi deficiency.We also demonstrated that GR24^(4DO) treatment under normal conditions inhibits the expression of OsNRTs and OsAMTs to suppress nitrogen absorption but enhances the expression of OsPTs to promote Pi absorption,thus facilitating the balance between nitrogen and phosphorus uptake in rice.Importantly,we found that NSP1p:NSP1 and NSP2p:NSP2 transgenic plants show improved agronomic traits and grain yield under low-and medium-phosphorus conditions.Taken together,these results revealed a novel regulatory mechanism of SL biosynthesis and signaling in response to Pi starvation,providing genetic resources for improving plant architecture and nutrient-use efficiency in low-Pi environments.Kun Yuan Hao Zhang Chaoji Yu Nan Luo Jijun Yan Shuang Zheng Qingliang Hu Dahan Zhang Liquan Kou Xiangbing Meng Yanhui Jing Mingjiang Chen Xinwei Ban Zongyun Yan Zefu Lu Jian Wu Yu Zhao Yan Liang Yonghong Wang Guosheng Xiong Jinfang Chu Ertao Wang Jiayang Li Bing Wang 2023Molecular Plant2023,16,11:2

19Sensitivity analysis of CO_2 sequestration in saline aquifers显示文摘Carbon capture and storage (CCS) technology has been considered as an important method for reducing greenhouse gas emissions and for mitigating global climate change. Three primary options are being considered for large-scale storage of CO 2 in subsurface formations: oil and gas reservoirs, deep saline aquifers, and coal beds. There are very many large saline aquifers around the world, which could make a big contribution to mitigating global warming. However, we have much less understanding of saline aquifers than oil and gas reservoirs. Several mechanisms are involved in the storage of CO 2 in deep saline aquifers, but the ultimate goal of injection of CO 2 into the aquifers containing salt water is to dissolve the CO 2 in the water. So it is important to study the solubility trapping and sensitivity factors of CO 2 in saline aquifers. This paper presents results of modeling CO 2 storage in a saline aquifer using the commercial reservoir simulator ECLIPSE. The objective of this study was to better understand the CO 2 /brine phase behavior (PVT properties) and quantitatively estimate the most important CO 2 storage mechanism in brine-solubility trapping. This would provide a tool by performing theoretical and numerical studies that help to understand the feasibility of CO 2 geological storage. A 3-dimensional, 2-phase (water/gas) conceptional reservoir model used finite, homogenous and isothermal formations into which CO 2 is injected at a constant rate. The effects of main parameters were studied, including the vertical to horizontal permeability ratio k v /k h , salinity, and residual phase saturations. The results show that the vertical to horizontal permeability ratio has a significant effect on CO 2 storage. 20Nuciferine protects against high-fat diet-induced hepatic steatosis and insulin resistance via activating TFEB-mediated autophagy——lysosomal pathway显示文摘Nonalcoholic fatty liver disease(NAFLD) is characterized by hepatic steatosis and insulin resistance and there are currently no approved drugs for its treatment.Hyperactivation of mTOR complex1(mTORCl) and subsequent impairment of the transcription factor EB(TFEB)-mediated autophagy-lysosomal pathway(ALP) are implicated in the development of NAFLD.Accordingly,agents that augment hepatic TFEB transcriptional activity may have therapeutic potential against NAFLD.The objective of this study was to investigate the effects of nuciferine,a major active component from lotus leaf,on NAFLD and its underlying mechanism of action.Here we show that nuciferine activated ALP and alleviated steatosis,insulin resistance in the livers of NAFLD mice and palmitic acid-challenged hepatocytes in a TFEB-dependent manner.Mechanistic investigation revealed that nuciferine interacts with the Ragulator subunit hepatitis B X-interacting protein and impairs the interaction of the Ragulator complex with Rag GTPases,thereby suppressing lysosomal localization and activity of mTORC1,which activates TFEB-mediated ALP and further ameliorates hepatic steatosis and insulin resistance.Our present results indicate that nuciferine may be a potential agent for treating NAFLD and that regulation of the mTORCl-TFEB-ALP axis could represent a novel pharmacological strategy to combat NAFLD.Xiliang Du Chiara Di Malta Zhiyuan Fang Taiyu Shen Xiaodi Niu Meng Chen Bo Jin Hao Yu Lin Lei Wenwen Gao Yuxiang Song Zhe Wang Chuang Xu Zhijun Cao Guowen Liu Xinwei Li 2022Acta Pharmaceutica Sinica B2022,12,6:2
