| 1 | Auxin-induced AUXIN RESPONSE FACTOR4 activates APETALA1 and FRUITFULL to promote flowering in woodland strawberry显示文摘Flowering time is known to be regulated by numerous pathways,such as the autonomous,gibberellin,aging,photoperiod-mediated,and vernalization pathways.These regulatory mechanisms involve both environmental triggers and endogenous hormonal cues.Additional flowering control mechanisms mediated by other phytohormones,such as auxin,are less well understood.We found that in cultivated strawberry(Fragaria×ananassa),the expression of auxin response factor4(FaARF4)was higher in the flowering stage than in the vegetative stage.Overexpression of FaARF4 in Arabidopsis thaliana and woodland strawberry(Fragaria vesca)resulted in transgenic plants flowering earlier than control plants.In addition,FveARF4-silenced strawberry plants showed delayed flowering compared to control plants,indicating that FaARF4 and FveARF4 function similarly in regulating flowering.Further studies showed that ARF4 can bind to the promoters of the floral meristem identity genes APETALA1(AP1)and FRUITFULL(FUL),inducing their expression and,consequently,flowering in woodland strawberry.Our studies reveal an auxin-mediated flowering pathway in strawberry involving the induction of ARF4 expression. | Xiangxiang Dong Yanjun Li Yuhan Guan Shaoxi Wang He Luo Xiaoming Li He Li Zhihong Zhang | 2021 | Horticulture Research2021,8,1: | 4 |

| 18 | Computational investigation of contraction behavior in a liquid-solid fluidized bed显示文摘Contraction behavior of a liquid-solid fluidized bed has been investigated numerically. Based on a simple hydrodynamic model proposed by Brandani and Zhang (2006), a case study for solid particles with a density of 3,000 kg/m3 and a diameter of 2.5× 10 -3 m is simulated in a two-dimensional fluidized bed (0.50 m height and 0.10 m width). Due to the continuity of numerical computation, there is a transition region between two zones of different solid holdups when the liquid velocity is suddenly changed. The top, middle and bottom interfaces are explored to obtain a reasonable interface height. The simulated results show that the steady time of the middle interface is more close to Gibilaro's theory and suitable for describing the contraction process of a phase interface. | 20 | Hepatitis B Virus Induces Microtubule Stabilization to Promote Productive Infection through Upregulating Microtubule-associated Protein 1S显示文摘Background and Aims:Continuous release and transmission of hepatitis B virus(HBV)is one of the main factors leading to chronic hepatitis B(CHB)infection.However,the mechanism of HBV-host interaction for optimal viral transport is unclear.Hence,we aimed to explore how HBV manipulates microtubule-associated protein 1S(MAP1S)and microtubule(MT)to facilitate its transport and release.Methods:The expression of MAP1S or acetylated MT was investigated by immunofluorescence,RT-PCR,immunoblotting,and plasmid transfection.MAP1S overexpression or knockdown was performed by lentiviral infection or shRNA transfection,respectively.HBV DNA was quantified using q-PCR.Results:Significantly higher level of MAP1S in HepG2215 cells compared with HepG2 cells was detected using RT-PCR(p<0.01)and immunoblotting(p<0.001).Notably,stronger MAP1S expression was observed in the liver tissues of patients with CHB than in healthy controls.MAP1S overexpression or knockdown demonstrated that MAP1S promoted MT acetylation and reduced the ratio of HBV DNA copies inside to outside cells.Further,transfection with the hepatitis B virus X protein(HBx)-expressing plasmids induced significantly higher level of MAP1S than that in controls(p<0.0001),whereas HBVX−mutant-encoding HBV proteins(surface antigen,core protein,and viral DNA polymerase)hardly affected its expression.Conclusions:These results demonstrate that HBx induces the forma tion of stable MTs to promote the release of HBV particles through upregulating MAP1S.Thus,our studies delineate a unique molecular pathway through which HBV manipulates the cytoskeleton to facilitate its own transportation,and indicate the possibility of targeting MAP1S pathway for treatment of patients with CHB. | Yuanyue Guan Bin Sun Shijie Zhang Yuan Zhuang Yanxiang Huang Minghua Lin Rongling Zheng Dexi Chen Ying Shi Yanjun Wang | 2022 | Journal of Clinical and Translational Hepatology2022,10,3: | 0 |
