| 1 | The adult human testis transcriptional cell atlas显示文摘Human adult spermatogenesis balances spermatogonial stem cell (SSC) self-renewal and differentiation,alongside complex germ cell-niche interactions,to ensure long-term fertility and faithful genome propagation. Here,we performed single-cell RNA sequencing of -6500 testicular cells from young adults. We found five niche/somatic cell types (Leydig,myoid,Sertoli,endothelial,macrophage),and observed germline-niche interactions and key human-mouse differences. Spermatogenesis,including meiosis,was reconstructed computationally,revealing sequential coding,non-coding,and repeat-element transcriptional signatures. Interestingly,we identified five discrete transcriptional/developmental spermatogonial states,including a novel early SSC state,termed State 0. Epigenetic features and nascent transcription analyses suggested developmental plasticity within spermatogonial States. To understand the or丨gin of State 0,we profiled testicular cells from infants,and identified distinct similarities between adult State 0 and infant SSCs. | 2 | Role of MGMT as biomarker in colorectal cancer显示文摘O6-methylguanine DNA methyltransferase(MGMT) gene promoter methylation plays an important role in colorectal carcinogenesis, occurring in about 30%-40% of metastatic colorectal cancer. Its prognostic role has not been defined yet, but loss of expression of MGMT, which is secondary to gene promoter methylation, results in an interesting high response to alkylating agents such as dacarbazine and temozolomide. In a phase 2 study on heavily pre-treated patients with MGMT methylated metastatic colorectal cancer, temozolomide achieved about 30% of disease control rate. Activating mutations of RAS or BRAF genes as well as mismatch repair deficiency may represent mechanisms of resistance to alkylating agents, but a dose-dense schedule of temozolomide may potentially restore sensitivity in RAS-mutant patients. The aim of this article is to present a new commercialized calcium silicate-based material named Biodentine with physical improved properties compared to MTA in a clinical application. Two endodontic microsurgeries were performed by using specific armamentarium(microsurgical instrumentation, ultrasonic tips) under high-power magnification with an operatory microscope. Biodentine was used as a root-end filling in order to seal the root canal system. The two cases were considered completely healed at 1 year and were followed for one more year. | 16 | Cretaceous tectonic evolution of the Neo-Tethys in Central Iran:Evidence from petrology and age of the Nain-Ashin ophiolitic basalts显示文摘The Nain and Ashin ophiolites consist of Mesozoic melange units that were emplaced in the Late Cretaceous onto the continental basement of the Central-East Iran microcontinent(CEIM).They largely consist of serpentinized peridotites slices;nonetheless,minor tectonic slices of sheeted dykes and pillow lavas-locally stratigraphically associated with radiolarian cherts-can be found in these ophiolitic melanges.Based on their whole rock geochemistry and mineral chemistry,these rocks can be divided into two geochemical groups.The sheeted dykes and most of the pillow lavas show island arc tholeiitic(IAT)affinity,whereas a few pillow lavas from the Nain ophiolites show calc-alkaline(CA)affinity.Petrogenetic modeling based on trace elements composition indicates that both IAT and CA rocks derived from partial melting of depleted mantle sources that underwent enrichment in subduction-derived components prior to melting.Petrogenetic modeling shows that these components were represented by pure aqueous fluids,or sediment melts,or a combination of both,suggesting that the studied rocks were formed in an arc-forearc tectonic setting.Our new biostratigraphic data indicate this arc-forearc setting was active in the Early Cretaceous.Previous tectonic interpretations suggested that the Nain ophiolites formed,in a Late Cretaceous backarc basin located in the south of the CEIM(the so-called Nain-Baft basin).However,recent studies showed that the CEIM underwent a counter-clockwise rotation in the Cenozoic,which displaced the Nain and Ashin ophiolites in their present day position from an original northeastward location.This evidence combined with our new data and a comparison of the chemical features of volcanic rocks from different ophiolites around the CEIM allow us to suggest that the Nain-Ashin volcanic rocks and dykes were formed in a volcanic arc that developed on the northern margin of the CEIM during the Early Cretaceous in association with the subduction,below the CEIM,of a Neo-Tethys oceanic branch that was existing between the CEIM and the southern margin of Eurasia.As a major conclusion of this paper,a new geodynamic model for the Cretaceous evolution of the CEIM and surrounding Neo-Tethyan oceanic basins is proposed. | Tahmineh Pirnia Emilio Saccani Ghodrat Torabi Marco Chiari Spela Gorican Edoardo Barbero | 2020 | Geoscience Frontiers2020,11,1: | 2 |

| 19 | Matched or nonmatched interventions based on the transtheoretical model to promote physical activity.A meta-analysis of randomized controlled trials显示文摘Purpose: The aim of this study was to examine whether the efficacy of transtheoretical model(TTM)-based interventions on physical activity(PA)varied according to the following criteria:(1) interventions targeted the actual stages of change(SOCs) or did not;(2) participants were selected according to their SOC or were not; and(3) its theoretical constructs(decisional balance, temptation, self-efficacy, processes of change).Methods: Thirty-three randomized controlled trials assessing TTM-based interventions promoting PA in adults were systematically identified.Results: The between-group heterogeneity statistic(Qb) did not reveal any differential efficacy either in interventions targeting the actual SOC compared with those that did not(Qb = 1.48, p = 0.22) or in interventions selecting participants according to their SOC compared with those that did not(Qb = 0.01, p = 0.91). TTM-based interventions enhanced PA behavior whether they targeted the actual SOC(Cohen's d = 0.36; 95%confidence interval(CI): 0.22–0.49) or not(d = 0.23; 95%CI: 0.09–0.38) and whether they selected their participants according to their SOC(d = 0.33; 95%CI: 0.13–0.53) or not(d = 0.32; 95%CI: 0.19–0.44).
