1A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 2A Review of Methods for Studying Microbial Diversity in Soils显示文摘Soil microorganisms play a central role in decomposing organic matter, in determining the release of mineral nutrients, and in nutrient cycling. Recently, extensive studies have focused on soil microbial diversity. However, understanding the diversity of this complex microbial community in the soil environment is a challenging task. Thus, it is important to master and comprehend appropriate methods for studying soil microbial diversity. Concepts of soil microbial diversity and major methods of study are briefly introduced in this paper. Then, the application of biochemical-based and molecular-based techniques in this area, and their advantages and disadvantages are evaluated. 6Correlations Between Foliar Stable Carbon Isotope Composition and Environmental Factors in Desert Plant Reaumuria soongorica (Pall.) Maxim.显示文摘Leaves of 407 individuals of Reaumuria soongorica (Pall.) Maxim. collected from the major distribution areas were measured to investigate the distribution characteristics of the stable carbon isotope in this desert plant, as well as correlations between δ13C values and environmental factors. Results showed that δ13C values in R. soongorica ranged from-22.77‰. to-29.85‰. and that the mean δ13C value (-26.52‰)was higher than a previously reported δ13C value for a different desert ecosystem. This indicates that R.soongorica belongs to the C3 photosynthetic pathway and has higher water use efficiency than other species. The correlations between δ13C values and environmental factors demonstrated that the foliar δ13C values in R. soongorica increased significantly with decreasing mean annual precipitation and mean relative humidity, and decreased with decreasing duration of sunshine and evaporation. The spatial distribution trend of δ13C values in R. soongorica was not obvious and there was no significant correlation between the δ13C values and mean annual temperature. We conclude that different distribution trends in δ13C values for R. soongorica were likely caused by stomatal limitation rather than by nutrient-related changes in photosynthetic efficiency and that precipitation played an important role in the wide distribution range of R.soongorica. 7SHRIMP zircon U-Pb dating for subduction-related gran-itic rocks in the northern part of east Junggar, Xinjiang显示文摘SHRIMP U-Pb zircon dating on the Xileketehalasu granodiorite porphyry and Kalasayi monodiorite porphyry that intrude middle Devonian Beitashan Formation at the north part of east Junggar region shows that they were formed at 381±6 Ma and 376±10 Ma respectively. They are interpreted as subduction-related granitic rocks, which is the first report that the isotopic ages for the granitic rocks range from 350 to 390 Ma. Another determined age for the Kalasayi monodiorite porphyry is 408±9 Ma, representing the age of underlain Lower Devonian volcanic rocks. Thus, the U-Pb dates suggest that the northeastward subduction of Junggar ocean from southwest occurred at 408 to 376 Ma (the real inter- val may be larger). 8Postconditioning of sevoflurane and propofol is associated with mitochondrial permeability transition pore显示文摘Background: Sevoflurane and propofol are effective cardioprotective anaesthetic agents, though the cardioprotection of propofol has not been shown in humans. Their roles and underlying mechanisms in anesthetic postconditioning are unclear. Mitochondrial permeability transition pore (MPTP) opening is a major cause of ischemia-reperfusion injury. Here we investigated sevoflurane- and propofol-induced postconditioning and their relationship with MPTP. Methods: Isolated perfused rat hearts were exposed to 40 min of ischemia followed by 1 h of reperfusion. During the first 15 min of reperfusion, hearts were treated with either control buffer (CTRL group) or buffer containing 20 μmol/L atractyloside (ATR group), 3% (v/v) sevoflurane (SPC group), 50 μmol/L propofol (PPC group), or the combination of atractyloside with respective anesthetics (SPC+ATR and PPC+ATR groups). Infarct size was determined by dividing the total necrotic area of the left ventricle by the total left ventricular slice area (percent necrotic area). Results: Hearts treated with sevoflurane or propofol showed significantly better recovery of coronary flow, end-diastolic pressures, left ventricular developed pressure and derivatives compared with controls. Sevoflurane resulted in more protective alteration of hemodynamics at most time point of reperfusion than propofol. These improvements were paralleled with the reduction of lactate dehydrogenase release and the decrease of infarct size (SPC vs CTRL: (17.48±2.70)% vs (48.47±6.03)%, P<0.05; PPC vs CTRL: (35.60±2.10)% vs (48.47±6.03)%, P<0.05). SPC group had less infarct size than PPC group (SPC vs PPC: (17.48±2.70)% vs (35.60±2.10)%, P<0.05). Atractyloside coadministration attenuated or completely blocked the cardioprotective effect of postconditioning of sevoflurane and propofol. Conclusion: Postconditioning of sevoflurane and propofol has cardio-protective effect against ischemia-reperfusion injury of heart, which is associated with inhibition of MPTP opening. 14Sevoflurane postconditioning reduces myocardial reperfusion injury in rat isolated hearts via activation of PI3K/Akt signaling and modulation of Bcl-2 family proteins显示文摘Sevoflurane postconditioning reduces myocardial infarct size.The objective of this study was to examine the role of the phosphatidylinositol-3-kinase(PI3K)/Akt pathway in anesthetic postconditioning and to determine whether PI3K/Akt signaling modulates the expression of pro-and antiapoptotic proteins in sevoflurane postconditioning.Isolated and perfused rat hearts were prepared first,and then randomly assigned to the following groups:Sham-operation(Sham),ischemia/reperfusion(Con),sevoflurane postconditioning(SPC),Sham plus 100 nmol/L wortmannin(Sham+Wort),Con+Wort,SPC+Wort,and Con+dimethylsulphoxide(DMSO).Sevoflurane postconditioning was induced by administration of sevoflurane(2.5%,v/v) for 10 min from the onset of reperfusion.Left ventricular developed pressure(LVDP),left ventricular end-diastolic pressure(LVEDP),maximum increase in rate of LVDP(+dP/dt),maximum decrease in rate of LVDP(?dP/dt),heart rate(HR),and coronary flow(CF) were measured at baseline,R30 min(30 min of reperfusion),R60 min,R90 min,and R120 min.Creatine kinase(CK) and lactate dehydrogenase(LDH) were measured after 5 min and 10 min reperfusion.Infarct size was determined by triphenyltetrazolium chloride staining at the end of reperfusion.Total Akt and phosphorylated Akt(phospho-Akt),Bax,Bcl-2,Bad,and phospho-Bad were determined by Western blot analysis.Analysis of variance(ANOVA) and Student-Newman-Keuls' test were used to investigate the significance of differences between groups.The LVDP,±dP/dt,and CF were higher and LVEDP was lower in the SPC group than in the Con group at all points of reperfusion(P<0.05).The SPC group had significantly reduced CK and LDH release and decreased infarct size compared with the Con group [(22.9±8)% vs.(42.4±9.4)%,respectively;P<0.05].The SPC group also had increased the expression of phospho-Akt,Bcl-2,and phospho-Bad,and decreased the expression of Bax.Wortmannin abolished the cardioprotection of sevoflurane postconditioning.Sevoflurane postconditioning may protect the isolated rat heart.Activation of PI3K and modulation of the expression of pro-and antiapoptotic proteins may play an important role in sevoflurane-induced myocardial protection.Li-na YU Jing YU Feng-jiang ZHANG Mei-juan YANG Ting-ting DING Jun-kuan WANG Wei HE Tao FANG Gang CHEN Min YAN 2010Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2010,11,9:36

16Intestinal microbiota pathogenesis and fecal microbiota transplantation for inflammatory bowel disease显示文摘The intestinal microbiota plays an important role in inflammatory bowel disease(IBD).The pathogenesis of IBD involves inappropriate ongoing activation of the mucosal immune system driven by abnormal intestinal microbiota in genetically predisposed individuals.However,there are still no definitive microbial pathogens linked to the onset of IBD.The composition and function of the intestinal microbiota and their metabolites are indeed disturbed in IBD patients.The special alterations of gut microbiota associated with IBD remain to be evaluated.The microbial interactions and hostmicrobe immune interactions are still not clarified.Limitations of present probiotic products in IBD are mainly due to modest clinical efficacy,few available strains and no standardized administration.Fecal microbiota transplantation(FMT)may restore intestinal microbial ho-meostasis,and preliminary data have shown the clinical efficacy of FMT on refractory IBD or IBD combined with Clostridium difficile infection.Additionally,synthetic microbiota transplantation with the defined composition of fecal microbiota is also a promising therapeutic approach for IBD.However,FMT-related barriers,including the mechanism of restoring gut microbiota,standardized donor screening,fecal material preparation and administration,and long-term safety should be resolved.The role of intestinal microbiota and FMT in IBD should be further investigated by metagenomic and metatranscriptomic analyses combined with germfree/human flora-associated animals and chemostat gut models.Zi-Kai Wang Yun-Sheng Yang Ye Chen Jing Yuan Gang Sun Li-Hua Peng 2014World Journal of Gastroenterology2014,20,40:30
17Spatio-temporal dynamics of vegetation and its relationship with climate factors Mongolia, China coverage in Inner显示文摘The vegetation coverage dynamics and its relationship with climate factors on different spatial and temporal scales in Inner Mongolia during 2001-2010 were analyzed based on MODIS-NDVI data and climate data.The results indicated that vegetation coverage in Inner Mongolia showed obvious longitudinal zonality,increasing from west to east across the region with a change rate of 0.2/10°N.During 2001-2010,the mean vegetation coverage was 0.57,0.4 and 0.16 in forest,grassland and desert biome,respectively,exhibiting evident spatial heterogeneities.Totally,vegetation coverage had a slight increasing trend during the study period.Across Inner Mongolia,the area of which the vegetation coverage showed extremely significant and significant increase accounted for 11.25% and 29.13% of the area of whole region,respectively,while the area of which the vegetation coverage showed extremely significant and significant decrease accounted for 7.65% and 26.61%,respectively.On inter-annual time scale,precipitation was the dominant driving force of vegetation coverage for the whole region.On inter-monthly scale,the change of vegetation coverage was consistent with both the change of temperature and precipitation,implying that the vegetation growth within a year is more sensitive to the combined effects of water and heat rather than either single climate factor.The vegetation coverage in forest biome was mainly driven by temperature on both inter-annual and inter-monthly scales,while that in desert biome was mainly influenced by precipitation on both the two temporal scales.In grassland biome,the yearly vegetation coverage had a better correlation with precipitation,while the monthly vegetation coverage was influenced by both temperature and precipitation.In grassland biome,the impacts of precipitation on monthly vegetation coverage showed time-delay effects.MU Shaojie YANG Hongfei LI Jianlong CHEN Yizhao GANG Chengcheng ZHOU Wei JU Weimin 2013Journal of Geographical Sciences2013,23,2:28

19Effects of molasses on the fermentation characteristics of mixed silage prepared with rice straw, local vegetable by-products and alfalfa in Southeast China显示文摘This experiment was conducted to study the effect of molasses on the fermentation characteristics of mixed silage ensiled rice straw and vegetable by-products with alfalfa.Mixture(202 g kg^(–1) dry matter(DM))consisting of rice straw,broccoli residue and alfalfa at the ratio of 5:4:1 was ensiled with three experimental treatments:(1)no additives(control);(2)molasses at 2.5%(M1);(3)molasses at 5%(M2)on a fresh matter basis of mixture,respectively.All treatments were packed into laboratory-scale silos,and three silos per treatment were sampled on days 1,3,5,14 and 30.The result showed that the p H value of all mixed silages decreased gradually with the time of ensiling except for the control silage,in which a significant increase(P<0.05)on day 30 occurred.The lactic acid content increased gradually with the time of ensiling and reached the highest value on day 14,and a marked decrease(P<0.05)was found in the control silage on day 30.The changes of acetic acid content showed similar pattern with lactic acid content.A trace amount of propionic and butyric acid contents were found in the three mixed silages during the fermentation period.Comparing to the control,M1 and M2 treatments improved the fermentation quality of mixed silages as indicated by higher(P<0.05)lactic acid contents and lower(P<0.05)p H and ammonia-N contents.The Flieg points also showed that M1 and M2 silages were well preserved,whereas the control silage had a bad quality.Overall,the findings of this study suggested that adding molasses could improve fermentation quality of mixed silage,and M1 was more suitable for practical application.WANG Jian CHEN Lei YUAN Xian-jun GUO Gang LI Jun-feng BAI Yun-feng SHAO Tao 2017Journal of Integrative Agriculture2017,16,3:24
