2Cardamonin from a medicinal herb protects against LPS-induced septic shock by suppressing NLRP3 inflammasome显示文摘Aberrant activation of NLRP3 inflammasome has been implicated in the pathogenesis of diverse inflammation-related diseases, and pharmacological molecules targeting NLRP3 inflammasome are of considerable value to identifying potential therapeutic interventions. Cardamonin(CDN), the major active ingredient of the traditional Chinese medicinal herb Alpinia katsumadai, has exerted an excellent anti-inflammatory activity, but the mechanism underlying this role is not fully understood. Here, we show that CDN blocks canonical and noncanonical NLRP3 inflammasome activation triggered by multiple stimuli. Moreover, the suppression of CDN on inflammasome activation is specific to NLRP3, not to NLRC4 or AIM2 inflammasome. Besides, the inhibitory effect is not dependent on the expression of NF-κB-mediated inflammasome precursor proteins. We also demonstrate that CDN suppresses the NLRP3 inflammasome through blocking ASC oligomerization and speckle formation in a dose-dependent manner.Importantly, CDN improves the survival of mice suffering from lethal septic shock and attenuates IL-1βproduction induced by LPS in vivo, which is shown to be NLRP3 dependent. 3Research on theory and key technology of trusted computing platform security testing and evaluation显示文摘Trusted computing has become a new trend in the area of international information security, and the products of trusted computing platform begin to be used in application. Users will not use the products of information security, unless it goes through the testing and evaluation. Here we concentrate on the testing and evaluation problem of trusted computing platform, begin with constructing proper formalization model of trusted computing platform for testing, and establish a mathematical chain of trust model based on SPA. Moreover, we give a verification method of composite characteristics and find the potential factors threatening the trusted system in the process of remote attestation through analysis. For trusted software stack, we study the problem of automatic generation of test case and propose an improved method of generating the random test case, to raise the quality of test case. Finally, we give a prototype system of trusted computing platform and the actual test data related. The result demonstrates that there exist some flaws in the architecture of the present TCG computing platform. 4Features of colorectal cancer in China stratified by anatomic sites:A hospital-based study conducted in university-affiliated hospitals from 2014 to 2018显示文摘Objective:The clinical and biological characteristics of colorectal cancer have been found to differ depending on the anatomic site of the cancer.However,for Chinese patients,there is limited information on the proportion of cases at each site and the related features.In this study,we explored the location,distribution and other features of colorectal cancers at each anatomic site in Chinese patients.Methods:We conducted a hospital-based study using hospitalization summary reports from 10 Peking University-affiliated hospitals from 2014 to 2018;the reports covered a total of 2,097,347 hospitalizations.Incident cases were chosen as the study population,and their epidemiological features were further analyzed.Results:A total of 20,739 colorectal cancer patients were identified.Rectum was the most common location(48.3%)of the cancer,whereas the proportions of patients with distal and proximal colon cancer were 24.5%and18.6%,respectively.Patients with rectal cancer were predominantly male and were the youngest for all anatomical sites(each P<0.001).The highest proportion of emergency admissions,the longest hospital stays and the highest hospitalization costs were found in patients with proximal colon cancer(each P<0.001).The proximal colon cancer subgroup included the highest proportions of patients with medical histories of cholecystectomy,cholecystolithiasis and/or gallbladder polyps and appendectomy(P=0.009,P<0.001 and P<0.001,respectively).The distal colon cancer subgroup included the highest proportions of patients with medical histories of diabetes and hypertension(P<0.001,respectively).Conclusions:The patterns of colorectal cancer observed in this study differ from those reported for Western patients and show a significantly higher proportion of patients with rectal cancer.Different epidemiological features were also found based on anatomic sites.Further studies based on tumor location should be conducted to facilitate more accurate screening and treatment.Ruize Qu Yanpeng Ma Liyuan Tao Xiaoyuan Bao Xin Zhou Bingyan Wang Fei Li Siyi Lu Lin Tuo Siyan Zhan Zhipeng Zhang Wei Fu 2021Chinese Journal of Cancer Research2021,33,4:10
5Bavachin enhances NLRP3 inflammasome activation induced by ATP or nigericin and causes idiosyncratic hepatotoxicity显示文摘Psoraleae Fructus(PF)is a well-known traditional herbal medicine in China,and it is widely used for osteoporosis,vitiligo,and other diseases in clinical settings.However,liver injury caused by PF and its preparations has been frequently reported in recent years.Our previous studies have demonstrated that PF could cause idiosyncratic drug-induced liver injury(IDILI),but the mechanism underlying its hepatotoxicity remains unclear.This paper reports that bavachin isolated from PF enhances the specific stimuli-induced activation of the NLRP3 inflammasome and leads to hepatotoxicity.Bavachin boosts the secretion of IL-ip and caspase-1 caused by ATP or nigericin but not those induced by poly(I:C),monosodium urate crystal,or intracellular lipopolysaccharide.Bavachin does not affect AIM2 or NLRC4 inflammasome activation.Mechanistically,bavachin specifically increases the production of nigericin-induced mitochondrial reactive oxygen species among the most important upstream events in the activation of the NLRP3 inflammasome.Bavachin increases the levels of aspartate transaminase and alanine aminotransferase in serum and hepatocyte injury accompanied by the secretion of IL-ip via a mouse model of lipopolysaccharide-mediated susceptibility to IDILI.These results suggest that bavachin specifically enhances the ATP-or nigericin-induced activation of the NLRP3 inflammasome.Bavachin also potentially contributes to PF-induced idiosyncratic hepatotoxicity.Moreover,bavachin and PF should be evaded among patients with diseases linked to the ATP-or nigericin-mediated activation of the NLRP3 inflammasome,which may be a dangerous factor for liver injury.Nan Qin Guang Xu Yan Wang Xiaoyan Zhan Yuan Gao Zhilei Wang Shubin Fu Wei Shi Xiaorong Hou Chunyu Wang Ruisheng Li Yan Liu Jiabo Wang Haiping Zhao Xiaohe Xiao Zhaofang Bai 2021Frontiers of Medicine2021,15,4:9

7Simultaneous anaerobic-aerobic biodegradation of halogenated phenolic compound under oxygen-limited conditions显示文摘The successful application of co-immobilized aerobic-anaerobic biomass under limited aeration in wastewater treatment systems would eliminate the problems associated with the intermediates mono-chlorophenol(MCP) and di-chlorophenol(DCP) accumulations. With low initial pentachlorophenol(PCP) concentration, all PCP could be completely removed under oxygen-limited strict anaerobic conditions, and the removal efficiencies with different initial headspace oxygen percentage(IHOP) were not obviously different from each other. While at high initial PCP concentration, under strictly anaerobic conditions PCP and their intermediates were clearly higher than that under other conditions, and produced obvious accumulation, the highest PCP reduction was achieved by the system receiving 30 IHOP, oxygen-limited system also exhibited lower residual TOC concentration and lower concentration of metabolic intermediates MCP and DCP. These results suggested that under strictly anaerobic condition the reductive dechlorination of low chlorinated compounds became rate limiting in the reductive dechlorination pathway, less chlorinated compounds be more amenable to aerobic degradation, and the aerobes of outer layers could function under limited oxygen. 11Analysis of QTLs for yield-related traits in Yuanjiang common wild rice (Oryza rufipogon Griff.)显示文摘Using an accession of common wild rice (Oryza rufipogon Griff.) collected from Yuanjiang County, Yunnan Province, China, as the donor and an elite cultivar 93-11, widely used in two-line indica hybrid rice production in China, as the recurrent parent, an advanced backcross populations were developed. Through genotyping of 187 SSR markers and investigation of six yield-related traits of two gen- erations (BC4F2 and BC4F4), a total of 26 QTLs were detected by employing single point analysis and interval mapping in both genera-tions. Of the 26 QTLs, the alleles of 10 (38.5%) QTLs originating from O. rufipogon had shown a beneficial effect for yield-related traits in the 93-11 genetic background. In addition, five QTLs controlling yield and its components were newly identified, indicating that there are potentially novel alleles in Yuanjiang common wild rice. Three regions underling significant QTLs for several yield-related traits were detected on chromosome 1, 7 and 12. The QTL clusters were founded and corresponding agronomic traits of those QTLs showed highly significant correlation, suggesting the pleiotropism or tight linkage. 12Dependence of creep age formability on initial temper of an Al-Zn-Mg-Cu alloy显示文摘The initial temper of the material may directly affect the whole creep age forming(CAF)process. In terms of creep deformation and stress relaxation, using the constant-stress creep aging and constant-strain stress relaxation aging tests, the relationship between initial temper and CAF formability is investigated for an Al-Zn-Mg-Cu alloy at 165 °C for 18 h. Three tempers are selected as the initial tempers in CAF, viz., solution, retrogression and re-solution. The CAF formability of this alloy with initial temper of retrogression is the best, and the creep strain of the retrogression tempered specimen after creep aging of 18 h is about 1.21 and 1.34 times than that of the solution and the re-solution tempered specimens, respectively. The calculated stress exponents of this alloy with three initial tempers range from 7.3 to 9.5, indicating that the CAF of this alloy is mainly controlled by the dislocation creep. The various formability for three initial tempers are attributed to different inhibitions of the transgranular precipitates on the dislocation movement. For the retrogression temper, the initial fine and uniformly distributed precipitates are seriously coarsened after6 h of CAF, which minimally inhibit the dislocation movement. 13In-situ synthesis and sintering of mullite glass composites by SPS显示文摘The main subject of this work is an investigation of the effects of heating rate and current on the crystallisation of amorphous precursors in spark plasma sintering(SPS).For this,dry gel of Al_(2)O_(3)-SiO_(2) with a molar ratio of 1:1,was synthesized and sintered in-situ by SPS,and also by hot pressing(HP)for comparison.Phase analysis showed that the only crystalline product in both cases was mullite,whose Al_(2)O_(3) content was lower in the SPS specimens.The microstructures showed a low volume fraction of large mullite fibers in the SPS specimens,whereas a high volume fraction of fine equiaxed grains was present in the HP specimen.The main difference in microstructure between HP and SPS specimens could be explained in terms of the higher heating rate of the SPS specimens.The size of the SPS die also affected the size and aspect ratio of the mullite fibers produced,which might have been due to either the different electrical current required or a difference in specimen temperature profile.Jinyong ZHANG Hai ZHAN Zhengyi FU Richard TODD 2014Journal of Advanced Ceramics2014,3,2:4
14Identification of differential gene expressions in colorectal cancer and polyp by cDNA microarray显示文摘AIM:To screen the differential expressed genes in colorectal cancer and polyp tissue samples.METHODS:Tissue specimens containing 16 cases of colorectal adenocarcinoma and colorectal polyp vs normal mucosae were collected and subjected to cDNA microarray and bioinformatical analyses.Quantitative reverse transcription-polymerase chain reaction(qRTPCR) was used to confirm some of the cDNA microarray data.RESULTS:The experimental data showed that eight genes were differentially expressed,most of which were upregulated in adenomatous polyp lesions.Forty-six genes expressions were altered in colorectal cancers,of which 29 were upregulated and 17 downregulated,as compared to the normal mucosae.In addition,18 genes were similarly altered in both adenomatous polyps and colorectal cancer.qRT-PCR analyses confirmed the cDNA microarray data for four of those 18 genes:MTA1,PDCD4,TSC1 and PDGFRA.CONCLUSION:These differentially expressed genes likely represent biomarkers for early detection of colorectal cancer and may be potential therapeutic targets after confirmed by further studies.Yi-Chen Dai Xiao-San Zhu Qing-Zhen Nan Zhang-Xin Chen Jun-Pei Xie Yu-Ka Fu Yuan-Yuan Lin Qing-Na Lian Qiao-Fang Sang Xiao-Juan Zhan 2012World Journal of Gastroenterology2012,18,6:4
15Rapid Determination of Total Content of Five Major Anthraquinones in Rhei Radix et Rhizoma by NIR Spectroscopy显示文摘Objective To establish a new method with near-infrared(NIR) spectroscopy to determine the total content of emodin, chrysophanol, rhein, aloeemodin, and physcion. NIR was used in this study to provide rapid and nondestructive analysis results. Methods In the first place, HPLC was used to measure the total content of emodin, chrysophanol, rhein, aloe-emodin and physcion in Rhei Radix et Rhizoma(RR) as a reference. In the second place, the spectral regions, regression methods, pretreatment methods, and partial least squares(PLS) factors were compared to increase the feasibility of the model. In the last, the root mean square error of calibration(RMSEC), root mean square error of cross validation(RMSECV), root mean square error of prediction(RMSEP), and correlation coefficient(r) were used as assessment parameters. Results PLS with first derivative pretreatment in the ranges of 4242-5581 cm^(-1), 5885-6233 cm^(-1) and 6394-7011 cm^(-1) provided the best results. The RMSEC and RMSEP obtained were 0.134 and 0.226 respectively. The according determination coefficients of the quantitative model were 0.99 and 0.94. 16Natural variations in grain length 10(GL10)regulate rice grain size显示文摘Grain size is an important determinant of grain weight and yield in rice.Although several genes related to grain size have been identified,natural variations in these genes that affect grain size are poorly characterized.Here,we describe the grain length QTL GL10,encoding MADS56,which positively regulates grain length and grain weight.A natural allelic variation of NIL-gl10,containing an~1.0-kb deletion in the first exon that abolishes its transcription,results in shorter grain length,lower grain weight and delayed flowering in gl10 plants.The knockout of GL10 in the HJX74 background leads to grain phenotypes similar to that of NIL-gl10,while overexpression of GL10 results in increased grain length and weight and earlier heading date.GL10 regulates grain length by promoting greater longitudinal cell growth in the grain glume.Additionally,GL10 participates in the regulation of gibberellic acid(GA)signaling pathway genes in young panicle tissues.Analysis of GL10 haplotypes shows obvious divergence between the japonica and indica lineages.Our findings reveal an allelic variation of GL10 that may explain differences in grain length among modern cultivars and could be used to breed rice varieties with optimized grain shape.Penglin Zhan Shuaipeng Ma Zhili Xiao Fangping Li Xin Wei Shaojun Lin Xiaoling Wang Zhe Ji Yu Fu Jiahao Pan Mi Zhou Yue Liu Zengyuan Chang Lu Li Suhong Bu Zupei Liu Haitao Zhu Guifu Liu Guiquan Zhang Shaokui Wang 2022Journal of Genetics and Genomics2022,49,5:3
17Generation maintenance scheduling based on multiple objectives and their relationship analysis显示文摘In a market environment of power systems, each producer pursues its maximal profit while the independent system operator is in charge of the system reliability and the minimization of the total generation cost when generating the generation maintenance scheduling(GMS). Thus, the GMS is inherently a multi-objective optimization problem as its objectives usually conflict with each other. This paper proposes a multi-objective GMS model in a market environment which includes three types of objectives, i.e., each producer's profit, the system reliability, and the total generation cost. The GMS model has been solved by the group search optimizer with multiple producers(GSOMP) on two test systems. The simulation results show that the model is well solved by the GSOMP with a set of evenly distributed Pareto-optimal solutions obtained. 19Cloning,expression,identification and bioinformatics analysis of Rv3265c gene from Mycobacterium tuberculosis in Escherichia coli显示文摘Objective:To clone and express Rv3265c gene of Mycobacterium tuberculosis in Escherichia coli (E.coli) under optimistic conditions,obtain and identify protein expressed,analyze the structure and characteristics of the protein using bioinformatics methods for future applications.Methods: Rv3265c gene from Mycobacterium tuberculosis H37Rv was amplified by polymerase chain reaction,and was cloned into the pET-30a vector after purification and recovery.The recombinant plasmid was sequenced and expressed in E.coli BL21(DE3),and then purified and identified by western blotting.The essential physical-chemical properties of the protein were predicated by bioinformatics tools,including subcellular location,secondary structure,domains,antigenic epitopes,etc.Tertiary structure of the protein based on homology modeling was estabUshed,while multi-sequence homological alignment and phylogenetic analysis were preformed.Results:The recombinant protein was obtained in soluble fraction from expression system in E.coli B121(DE3) carrying pET30- Rv3265c plasmid,and Rv3265c gene was expressed correctly.Bioinformatics analysis showed the protein contained no signal peptide and transmembrane helices,located outside of membrane.Secondary structure analysis revealed it containedα-helix,extended strand and random coil,46.8%,14.6%,38.6%,respectively.Furthermore,it possessed six potential antigenic epitopes,one glycosyl transferase domain.A simple three-dimensional model of this protein was constructed by Swiss-model sever.Both sequences and structures were conservative and especial either in gene or in protein.Conclusions:Rv3265c gene might be a desirable molecular target for anti-tuberculosis drug and vaccine.The purified protein from expression will be utilized to study the kinetics of L-rhamnosyltransferase and to develope an enzyme assay for screening vaccine or drug.Qiang Wu Peng Zhou Shiyun Qian Xi Qin Qiongyao Fu Zhinong Zhan Hua Pei 2011Asian Pacific Journal of Tropical Medicine2011,4,4:2
20Screening compounds against HCV based on MAVS/IFN-β pathway in a replicon mode显示文摘AIM:To develop a sensitive assay for screening compounds against hepatitis C virus (HCV).METHODS:The proteolytic cleavage of NS3/4A on enhanced yellow fluorescent protein (eYFP)-mitochondrial antiviral signaling protein (MAVS) was examined by reporter enzyme secreted placental alkaline phosphatase (SEAP),which enabled us to perform ongoing monitoring of anti-HCV drugs through repeated chemiluminescence.Subcellular localization of eYFP-MAVS was assessed by fluorescence microscopy.Cellular localization and protein levels were examined by Western blotting.RESULTS:HCV NS3/4A protease cleaved eYFP-MAVSfrom mitochondria to block the activation of interferon (IFN)-β promoter,thus resulting in downregulation of SEAP activity.The decrease in SEAP activity was proportional to the dose of active NS3/4A protease.Also this reporter assay was used to detect anti-HCV activity of IFN-α and cyclosporine A.CONCLUSION:Our data show that this reporter system is a sensitive and quantitative reporter of anti-HCV inhibitors.This system will constitute a new tool to allow the efficient screening of HCV inhibitors.Qiu-Xia Fu Li-Cui Wang Shuai-Zheng Jia Bo Gao Yong Zhou Juan Du Ying-Li Wang Xiao-Hui Wang Jian-Chun Peng Lin-Sheng Zhan 2010World Journal of Gastroenterology2010,16,44:2
