2雷公藤多苷对寻常型银屑病患者外周血Th17/Treg 相关细胞因子的影响显示文摘目的观察雷公藤多昔治疗前后寻常型银屑病患者血清辅助性T细胞17(Thl7)/调节性T细胞(Treg)相关细胞因子白介素-17(1L-17),白介素-23(IL-23)和白介素-10(IL-10)的含量,探讨雷公藤多昔治疗寻常型银屑病的作用机制。方法对蚌埠医学院第一附属医院皮肤科2016年12月—2017年6月就诊的41例寻常型银屑病患者采用口服雷公藤多昔片治疗,每日3次,每次20mg,疗程8周,以银屑病皮损面积和严重程度(PASI)评分评价疗效;采用酶联免疫吸附法(ELISA)检测正常对照组及患者治疗前后血清中IL-17、L-23和IL-10水平变化。结果寻常型银屑病患者外周血Thl7相关细胞因子IL-17和IL-23水平显著高于正常对照组(均P<0.01);而Treg相关细胞因子IL-10水平显著低于正常对照组。雷公藤多昔治疗后IL-17和IL-23水平明显降低,与治疗前相比差异有统计学意义(均P<0.01);而IL-10水平明显升高,与治疗前相比差异也有统计学意义(P<0.01)。雷公藤多昔治疗后PASI评分与治疗前相比显著下降,差异有统计学意义(P<0.01).患者雷公藤多昔治疗前后PASI评分与IL-17和IL-23水平呈正相关性,而与IL-10呈负相关性(均P<0.01).结论雷公藤多昔治疗寻常型银屑病疗效显著,可能通过调节Thl7/Treg相关细胞因子水平发挥其治疗作用。TUEKAM TUEKAM Erna Gaelle 江从军 2019中华全科医学2019,17,8:18
3Intestinal alkaline phosphatase in the colonic mucosa of children with inflammatory bowel disease显示文摘AIM:To investigate intestinal alkaline phosphatase(iAP) in the intestinal mucosa of children with inflammatory bowel disease(IBD).METHODS:Colonic biopsy samples were taken from 15 newly diagnosed IBD patients and from 10 healthy controls.In IBD patients,specimens were obtainedboth from inflamed and non-inflamed areas.The iAP mRNA and protein expression was determined by reverse transcription-polymerase chain reaction and Western blotting analysis,respectively.Tissue localization of iAP and Toll-like receptor(TLR) 4 was investigated by immunofluorescent staining.RESULTS:The iAP protein level in the inflamed mucosa of children with Crohn's disease(CD) and ulcerative colitis(UC) was significantly decreased when compared with controls(both P < 0.05).Similarly,we found a significantly decreased level of iAP protein in the inflamed mucosa in CD compared with non-inflamed mucosa in CD(P < 0.05).In addition,the iAP protein level in inflamed colonic mucosa in patients with UC was decreased compared with non-inflamed mucosa in patients with CD(P < 0.05).iAP protein levels in the non-inflamed mucosa of patients with CD were similar to controls.iAP mRNA expression in inflamed colonic mucosa of children with CD and UC was not significantly different from that in non-inflamed colonic mucosa with CD.Expression of iAP mRNA in patients with noninflamed mucosa and in controls were similar.Co-localization of iAP with TLR4 showed intense staining with a dotted-like pattern.iAP was present in the inflamed and non-inflamed mucosa of patients with CD,UC,and in control biopsy specimens,irrespective of whether it was present in the terminal ileum or in the colon.However,the fluorescent signal of TLR4 was more pronounced in the colon compared with the terminal ileum in all groups studied.CONCLUSION:Lower than normal iAP protein levels in inflamed mucosa of IBD patients may indicate a role for iAP in inflammatory lesions in IBD.Based on our results,administration of exogenous iAP enzyme to patients with the active form of IBD may be a therapeutic option.Kriszta Molnár dám Vannay Beáta Szebeni Nóra Fanni Bánki Erna Sziksz ron Cseh Hajnalka Gyrffy Péter László Lakatos Mária Papp András Arató Gábor Veres 2012World Journal of Gastroenterology2012,18,25:5
4Qualitative analysis of catechins from green tea GMB-4 clone using HPLC and LC-MS/MS显示文摘Objective:To identify the bioactive compounds in catechins isolation and its components from green tea GMB-4 clone.Methods:Green tea GMB-4 clones were extracted with distilled water at 90C.Samples were eluted into the column with 10%ethanol.Subsequently,the column was eluted with95%ethanol and evaporated separately.Green tea extract was identified by thin layer chromatography.Catechins were separated by the stationary phase in column chromatography using polyamide with 10%ethanol eluent and 95%ethanol.The results of isolations were analyzed by high performance liquid chromatographic(HPLC)and LCMS/MS.Analysis of catechins by HPLC was done by external standard.Results:Fraction from 10%ethanol showed that four major peaks at retention time of1.663,2.367,2.950 and 4.890,indicated the presence of four catechins components including catechin,epicatechins,gallocatechin and epigallocatechin.Whereas,fraction from 95%ethanol showed two main peaks at retention time of 5.167 and 9.82,which indicated the presence of epigallocatechin gallate(EGCG)and epicatechin gallate(ECG).EGCG(m/z 459),epigallocatechin(m/z 307),ECG(m/z 443),and epicatechin(m/z 291)were isolated and separated successfully using HPLC and LC-MS/MS.Conclusions:The HPLC and LC-MS/MS methods were successfully tuned for the qualitative analysis of green tea extract with EGCG and ECG.Four major catechins were separated and identified by LC-MS/MS,such as EGCG,epigallocatechin,ECG and epicatechin.The result of HPLC analysis showed that EGCG and ECG were main components from catechins isolation of green tea GMB-4 clone.Erna Susanti Ciptati Retty Ratnawati Aulanni'am Achmad Rudijanto 2015Asian Pacific Journal of Tropical Biomedicine2015,5,12:3
5Increased duodenal expression of mi R-146a and-155 in pediatric Crohn's disease显示文摘AIM: To evaluate the role of micro RNA(mi R)-146 a,-155 and-122 in the duodenal mucosa of pediatric patients with Crohn's disease(CD) and the effect of transforming growth factor-β(TGF-β) on these mi Rs in duodenal epithelial and fibroblast cells.METHODS: Formalin-fixed, paraffin-embedded biopsies derived from the macroscopically inflamed(CD inflamed: n = 10) and intact(CD intact: n = 10) duodenal mucosa of pediatric CD patients and control children(C: n = 10) were examined. Expression of mi R-146 a,-155 and-122 was determined by realtime polymerase-chain reaction(PCR). The expression of the above mi Rs was investigated in recombinant human TGF-β(1 nmol/L, 24 h) or vehicle treated small intestinal epithelial cells(CCL-241) and primary duodenal fibroblast cells derived from healthy children as well.RESULTS: Expression of mi R-146 a was significantly higher in the inflamed duodenal mucosa compared to the intact duodenal mucosa of children with CD(CD inflamed: 3.21 ± 0.50 vs CD intact: 0.62 ± 0.26, p ≤ 0.01) and to the control group(CD inflamed: 3.21 ± 0.50 vs C: 1.00 ± 0.33, p ≤ 0.05). The expression of mi R-155 was significantly increased in the inflamed region of the duodenum compared to the control group(CD inflamed: 4.87 ± 1.02 vs Control: 1.00 ± 0.40, p ≤ 0.001). The expression of mi R-122 was unchanged in the inflamed or intact mucosa of CD patients compared to controls. TGF-β treatment significantly decreased the expression of mi R-155 in small intestinal epithelial cells(TGF-β: 0.7 ± 0.083 vs Control: 1 ± 0.09, p ≤ 0.05) and also the expression of mi R-146a(TGF-β: 0.67 ± 0.04 vs Control: 1 ± 0.15, p ≤ 0.01) and mi R-155(TGF-β: 0.72 ± 0.09 vs Control: 1 ± 0.06, p ≤ 0.05) in primary duodenal fibroblasts compared to corresponding vehicle treated controls. TGF-β treatment did not influence the expression of mi R-122.CONCLUSION: The elevated expression of mi R-146 a and-155 in the inflamed duodenal mucosa of CD patients suggests the role of these mi Rs in the pathomechanism of inflammatory bowel disease. 6Hepatoprotective and antioxidant activity of pentagamavunon-0 against carbon tetrachloride-induced hepatic injury in rats显示文摘Objective:To investigate the hepatoproteetivc ami antioxidant activity of pentagamavunon-0(PGV-0) against CCl-4-induced hepatic injury in rats.Methods:The groups of animals were administered with PGV-0 at die doses 2.5.5,10,and 20 mg/kg b.w.,p.o.once in a day for 6 days and at day 7 the animals were administrated with carbon tetrachloride(CClj)(20%,2 ml/kg b.w.in liquid paraffin dp.).The effect of PGV-0 on serum transaminase(SGPT),alkaline phosphates(ALP and total bilirubin were determined in CCl-4-indueed hepatotoxicity in rats.Further,the effects of PGV-0 on glutathione(GSU) content,cutalase(CAT) and NO free radical scavenging activity also were investigated.Results:The results demonstrated that PCV-0 significantly reduced the activity of SGPT,serum ALP and total bilirubin in CCl-4 induced rat hepatotoxicity.PGV-0 has effect on the antioxidant and free radical defense system.It prevented the depletion level of GSH and decrease activity of CAT in CCl-4-induced liver injury in rats.PCV-0 also demonstrated the free radical scavenger effects on NO free radical scavenging activity with ES value of 32.32μM. 8Morpho-Physiological Response of Oryza glaberrima to Gradual Soil Drying显示文摘Soil drought occurrence during dry season has been the main constraint, besides prolonged flooding during rainy season, in increasing cropping intensity and rice productivity in tropical riparian wetland. Use of drought tolerant rice genotype might be a suitable option for overcoming such problem. This study focused on the effects of gradual soil drying during early vegetative growth stage on morphological and physiological traits of five Oryza glaberrima genotypes, namely RAM12, RAM14, RAM59, RAM97 and RAM101, and two Oryza sativa subsp japonica genotypes, i.e. Koshihikari and Minamihatamochi. The plants were subjected to 6 d of gradual soil drying condition from 15 days after transplanting(DAT) to 20 DAT, and were allowed to recover until 22 DAT. Gradual soil drying reduced plant growth as indicated by dry mass accumulation. Drought reduced stomatal conductance and increased leaf rolling score of all the genotypes. All the genotypes showed comparable response on stomatal conductance, but O. glaberrima genotypes performed higher in leaf rolling recovery. Meanwhile, O. sativa genotypes decreased total leaf area and specific leaf area, but increased specific leaf weight in order to avoid further damages due to drought stress.
