1Perspectives of pluripotent stem cells in livestock显示文摘The recent progress in derivation of pluripotent stem cells(PSCs)from farm animals opens new approaches not only for reproduction,genetic engineering,treatment and conservation of these species,but also for screening novel drugs for their efficacy and toxicity,and modelling of human diseases.Initial attempts to derive PSCs from the inner cell mass of blastocyst stages in farm animals were largely unsuccessful as either the cells survived for only a few passages,or lost their cellular potency;indicating that the protocols which allowed the derivation of murine or human embryonic stem(ES)cells were not sufficient to support the maintenance of ES cells from farm animals.This scenario changed by the innovation of induced pluripotency and by the development of the 3 inhibitor culture conditions to support naïve pluripotency in ES cells from livestock species.However,the long-term culture of livestock PSCs while maintaining the full pluripotency is still challenging,and requires further refinements.Here,we review the current achievements in the derivation of PSCs from farm animals,and discuss the potential application areas.Dharmendra Kumar Thirumala R Talluri Naresh L Selokar Iqbal Hyder Wilfried A Ku 2021World Journal of Stem Cells2021,13,1:4

3Stabilization of expansive soils using chemical additives: A review显示文摘Volume instability of expansive soils due to moisture fluctuations is often disastrous,causing severe damages and distortions in the supported structures.It is,therefore,necessary to adequately improve the performance of such soils that they can favorably fulfil the post-construction stability requirements.This can be achieved through chemical stabilization using additives such as lime,cement and fly ash.In this paper,suitability of such additives under various conditions and their mechanisms are reviewed in detail.It is observed that the stabilization process primarily involves hydration,cation exchange,flocculation and pozzolanic reactions.The degree of stabilization is controlled by several factors such as additive type,additive content,soil type,soil mineralogy,curing period,curing temperature,delay in compaction,pH of soil matrix,and molding water content,including presence of nano-silica,organic matter and sulfate compounds.Provision of nano-silica not only improves soil packing but also accelerates the pozzolanic reaction.However,presence of deleterious compounds such as sulfate or organic matter can turn the treated soils unfavorable at times even worser than the unstabilized ones.Dharmendra Barman Sujit Kumar Dash 2022Journal of Rock Mechanics and Geotechnical Engineering2022,14,4:3

5Fake News Classification Using a Fuzzy Convolutional Recurrent Neural Network显示文摘In recent years,social media platforms have gained immense popularity.As a result,there has been a tremendous increase in content on social media platforms.This content can be related to an individual’s sentiments,thoughts,stories,advertisements,and news,among many other content types.With the recent increase in online content,the importance of identifying fake and real news has increased.Although,there is a lot of work present to detect fake news,a study on Fuzzy CRNN was not explored into this direction.In this work,a system is designed to classify fake and real news using fuzzy logic.The initial feature extraction process is done using a convolutional recurrent neural network(CRNN).After the extraction of features,word indexing is done with high dimensionality.Then,based on the indexing measures,the ranking process identifies whether news is fake or real.The fuzzy CRNN model is trained to yield outstanding resultswith 99.99±0.01%accuracy.This work utilizes three different datasets(LIAR,LIAR-PLUS,and ISOT)to find the most accurate model.Dheeraj Kumar Dixit Amit Bhagat Dharmendra Dangi 2022Computers, Materials & Continua2022,,6:1

9Non-random tissue distribution of human na ve umbilical cord matrix stem cells显示文摘AIM: To determine the tissue and temporal distribution of human umbilical cord matrix stem (hUCMS) cells in severe combined immunodeficiency (SCID) mice. METHODS: For studying the localization of hUCMS cells, tritiated thymidine-labeled hUCMS cells were injected in SCID mice and tissue distribution was quantitatively determined using a liquid scintillation counter at days 1, 3, 7 and 14. Furthermore, an immunofluorescence detection technique was employed in which anti-human mitochondrial antibody was used to identify hUCMS cells in mouse tissues. In order to visualize the distribution of transplanted hUCMS cells in H&E stained tissue sections, India Black ink 4415 was used to label the hUCMS cells. RESULTS: When tritiated thymidine-labeled hUCMS cells were injected systemically (iv) in female SCID mice, the lung was the major site of accumulation at 24 h after transplantation. With time, the cells migrated to other tissues, and on day three, the spleen, stomach, and small and large intestines were the major accumulation sites. On day seven, a relatively large amount of radioactivity was detected in the adrenal gland, uterus, spleen, lung, and digestive tract. In addition, labeled cells had crossed the blood brain barrier by day 1. 17Induced pluripotent stem cells: Mechanisms, achievements and perspectives in farm animals显示文摘Pluripotent stem cells are unspecialized cells withunlimited self-renewal, and they can be triggered to differentiate into desired specialized cell types. These features provide the basis for an unlimited cell source for innovative cell therapies. Pluripotent cells also allow to study developmental pathways, and to employ them or their differentiated cell derivatives in pharmaceutical testing and biotechnological applications. Via blastocyst complementation, pluripotent cells are a favoured tool for the generation of genetically modified mice. The recently established technology to generate an induced pluripotency status by ectopic co-expression of the transcription factors Oct4, Sox2, Klf4 and c-Myc allows to extending these applications to farm animal species, for which the derivation of genuine embryonic stem cells was not successful so far. Most induced pluripotent stem(i PS) cells are generated by retroviral or lentiviral transduction of reprogramming factors. Multiple viral integrations into the genome may cause insertional mutagenesis and may increase the risk of tumour formation. Non-integration methods have been reported to overcome the safety concerns associated with retro and lentiviral-derived i PS cells, such as transient expression of the reprogramming factors using episomal plasmids, and direct delivery of reprogramming m RNAs or proteins. In this review, we focus on the mechanisms of cellular reprogramming and current methods used to induce pluripotency. We also highlight problems associated with the generation of i PS cells. 20Immunochemical characterization of antigens of Brucella canis and their use in seroprevalence study of canine brucellosis显示文摘Objective:To explore immunochemical characterization of antigens of Brucella canis(B. canis),and the use in seroprevalence study of canine brucellosis.Methods:External hot phosphate buffer saline extract(HPBSE) and internal sonicated(SA) antigens were prepared from B.canis strain MEX 51 and imniunochemically characterized.These antigens were used to test 527 serum samples of dogs by 2-mercaptoethanol-tubc agglutination test(2 ME-TAT), agar gel immunodiffusion test(AGID).dot-ELISA and indirect enzyme-linked immunosorbent assay(I-ELISA) to assess the seroprevalence of canine brucellosis.Results:The protein content of HPBSE and SA antigens was 0.387 mg/ml.and 0.195 mg/mL,respectively,whereas carbohydrate content was 0.174 mg/mL and 0.150 mg/mL,respectively.The sodium dodecyl sulfate-polyacrylamide gel electrophoresis(12.5%) of HPBSE and SA,revealed 6 and 8 visible peptide bands ranging from 18-80 kDa and 12-45 kDa,respectively.Western blot analysis showed immunodominant bands of MW 12.28.39 and 45 kl)a for HPBSE and 20-24 kl)a for SA. The AGII) revealed HPBSE as more specific antigen than SA but both I-ELISA and dot-ELISA indicated SA antigen to be more specific and reliable than HPBSE.The seroprevalence of canine brucellosis was 2.27%by 2ME-TAT.1.5%by AGID.3.03%by dot-ELISA and 16.12%by I-ELISA.
