3Heavy Metal Accumulation in Medicinal Plants Collected from Environmentally Different Sites显示文摘Objective To estimate the heavy metal content in soil and selected medicinal plants procured from environmentally different sites of the same city. Methods Soil and plant samples of Abutilon indicum, Calotropis procera, Euphorbia hirta, Peristrophe bycaliculata, and Tinospora cordifolia were collected from 3 environmentally different sites of the city: heavy traffic area (HTA), industrial area (IA), and residential area (RA). Pb, Cd, Cr, and Ni were estimated in soil and plant samples by inductively coupled plasma emission spectrometry and compared. Results The level of heavy metal was higher in soil than in plant parts studied. Accumulation of heavy metals varied from plant to plant. Pb was the highest in Calotropis procera root from HTA site and the lowest in Peristrophe bycaliculata whole plant from IA site. It was also lower in residential area than in heavy traffic area. Conclusion The level of heavy metal content differed in the same medicinal plant collected from environmentally different sites of the same city. 4Potential of plant growth-promoting rhizobacteria-plant interactions in mitigating salt stress for sustainable agriculture:A review显示文摘Soil salinization affecting different crops is one of the serious threats to global food security.Soil salinity affects 20%and 33%of the total cultivated and irrigated agricultural lands,respectively,and has been reported to caused a global crop production loss of 27.3 billion USD.The conventional approaches,such as using salt-tolerant varieties,saline soil scrapping,flushing,leaching,and adding supplements (e.g.,gypsum and lime),often fail to alleviate stress.In this context,developing diverse arrays of microbes enhancing crop productivity under saline soil conditions without harming soil health is necessary.Various advanced omics approaches have enabled gaining new insights into the structure and metabolic functions of plant-associated beneficial microbes.Various genera of salt-tolerating rhizobacteria ameliorating biotic and abiotic stresses have been isolated from different legumes,cereals,vegetables,and oil seeds under extreme alkaline and saline soil conditions.Rapid progress in rhizosphere microbiome research has revived the belief that plants may be more benefited from their association with interacting diverse microbial communities as compared with individual members in a community.In the last decade,several salt-tolerating plant growth-promoting rhizobacteria (PGPR) that improve crop production under salt stress have been exploited for the reclamation of saline agrosystems.This review highlights that the interaction of salt-tolerating microbes with plants improves crop productivity under salinity stress along with potential salt tolerance mechanisms involved and will open new avenues for capitalizing on cultivable diverse microbial communities to strengthen plant salt tolerance and,thus,to refine agricultural practices and production under saline conditions.Kailash Chand KUMAWAT Sharon NAGPAL Poonam SHARMA 2022Pedosphere2022,32,2:5
5Membrane-bound mucins and mucin terminal glycans expression in idiopathic or Helicobacter pylori, NSAID associated peptic ulcers显示文摘AIM:To determine the expression of membrane-bound mucins and glycan side chain sialic acids in Helicobacter pylori(H.pylori)-associated,non-steroidal inflammatory drug(NSAID)-associated and idiopathic-gastric ulcers.METHODS:We studied a cohort of randomly selected patients with H.pylori(group 1,n=30),NSAID(group 2,n=18),combined H.pylori and NSAID associated gastric ulcers(group 3,n=24),and patients with idiopathic gastric ulcers(group 4,n=20).Immunohistochemistry for MUC1,MUC4,MUC17,and staining for Erythrina cristagalli agglutinin and Sambucus nigra agglutinin(SNA)lectins was performed on sections from the ulcer margins.RESULTS:Staining intensity of MUC17 was higher in H.pylori ulcers(group 1)than in idiopathic ulcers(group4),11.05±3.67 vs 6.93±4.00 for foveola cells,and10.29±4.67 vs 8.00±3.48 for gland cells,respectively(P<0.0001).In contrast,MUC1 expression was higher in group 4 compared group 1,9.89±4.17 vs 2.93±5.13 in foveola cells and 7.63±4.60 vs 2.57±4.50 for glands,respectively(P<0.0001).SNA lectin staining was increased in group 4,in parallel to elevated MUC1expression,indicating more abundantα2-6 sialylation in that group.CONCLUSION:Cytoplasmic MUC17 staining was sig-nificantly decreased in the cases with idiopathic ulcer.The opposite was observed for both MUC1 and SNA lectin.This observation may reflect important pathogenic mechanisms,since different mucins with altered sialylation patterns may differ in their protection efficiency against acid and pepsin.Yaron Niv Doron Boltin Marisa Halpern Miriam Cohen Zohar Levi Alex Vilkin Sara Morgenstern Vahig Manugian Erica St Lawrence Pascal Gagneux Sukhwinder Kaur Poonam Sharma Surinder K Batra Samuel B Ho 2014World Journal of Gastroenterology2014,20,40:5
6Analyses of Lattice Traffic Flow Model on a Gradient Highway显示文摘The optimal current difference lattice hydrodynamic model is extended to investigate the traffic flow dynamics on a unidirectional single lane gradient highway. The effect of slope on uphill/downhill highway is examined through linear stability analysis and shown that the slope significantly affects the stability region on the phase diagram.Using nonlinear stability analysis, the Burgers, Korteweg-deVries(KdV) and modified Korteweg-deVries(mKdV) equations are derived in stable, metastable and unstable region, respectively. The effect of reaction coefficient is examined and concluded that it plays an important role in suppressing the traffic jams on a gradient highway. 8Anti-mycobacterial screening of five Indian medicinal plants and partial purification of active extracts of Cassia sophera and Urtica dioica显示文摘Objective:To find out the anti-mycobacterial potential of Cassia sophera(C.sophera),Urtica dioica(U.dioica),Momordica dioica,Tribulus terrains and Coccinia indica plants against multidrug resistant(MDR) strain of Mycobacterium tuberculosis(M.tuberculosis).Methods:Plant materials were extracted successively with solvents of increasing polarity.Solvent extracts were screened for anti-mycobacterial activity against fast growing,non-pathogenic mycobacterium strain,Mycobacterium semegmatis,by disk diffusion method.The active extracts were tested against MDR and clinical isolates of M.tuberculosis by absolute concentration and proportion methods.The active extracts were subjected to bio-autoassay on TLC followed by silica column chromatography for isolation of potential drug leads.Results:Hexane extract of U.dioica(HEUD) and methanol extract of C.sophera(MECS) produced inhibition zone of 20 mm in disc diffusion assay and MIC of 250 and 125μg/mL respectively in broth dilution assay against Mycobacterium semegmatis.Semipurified traction F2 from MECS produced 86%inhibition against clinical isolate and 60%inhibition against MDR strain of ilf.tuberculosis.F18 from HEUD produced 81%inhibition against clinical isolate and 60%inhibition against MDR strain of M.tuberculosis.Phytochemical analysis indicated that anti-mycobacterial activity of MECS may be due to presence of alkaloids or flavonoids and that of HEUD due to terpenoids.Condosions:C.sophera and U.dioica plant extracts exhibited promising anti-mycobacterial activity against MDR strain of M.tuberculosis. 9A high methionine,low folate and vitamin B_6/B_(12) containing diet can be associated with memory loss by epigenetic silencing of netrin-1显示文摘Memory-epigenetics which is the loss of memory due to epigenetic modifications can be due to the silencing of genes involved in cognitive functions and this is the basis of the current study.We hypothesize that a diet containing high methionine and low vitamins can lead to memory impairment by increasing global DNA methylation and therefore,silencing the netrin-1 gene,which encodes the glycoprotein involved in neurogenesis,axonal guidance and maintenance of the synaptic plasticity.Wild type(C57 BL/6 J) mice were fed with a diet containing excess methionine(1.2%),low-folate(0.08 mg/kg),vitamin B_6(0.01 mg/kg),and B_(12)(10.4 mg/kg) for 6 weeks.Mice were examined weekly for the long-term memory function,using a passive avoidance test,which determined loss of fear-motivated long-term memory starting from the fourth week of diet.Similarly,an increase in brain %5-methyl cytosine was observed starting from the 4 th week of diet in mice.Mice fed with a high methionine,low folate and vitamins containing diet showed a decrease in netrin-1 protein expression and an increase in netrin-1 gene promotor methylation,as determined by methylation-sensitive restriction enzyme-polymerase chain reaction analysis.The increase in methylation of netrin-1 gene was validated by high-resolution melting and sequencing analysis.Furthermore,the association of netrin-1 with memory was established by administering netrin that considerably restored long-term fear motivated memory.Taken together,these results suggest that a diet rich in methionine and lacking in folate and vitamin B_6/B_(12) can induce defects in learning and memory.Furthermore,the data indicates that decrease in netrin-1 expression due to hyper-methylation of its gene can be associated with memory loss.The animal procedures were approved by the Institutional Animal Care and Use Committee,University of Louisville,USA(No.A3586-01) on February 2,2018.Anuradha Kalani Pankaj Chaturvedi Komal Kalani Pradip K.Kamat Poonam Chaturvedi Neetu Tyagi 2019Neural Regeneration Research2019,14,7:4
10Screening of Antioxidant Potential of Selected Barks of Indian Medicinal Plants by Multiple in vitro Assays显示文摘Objective To evaluate the antioxidant potential in herbal extract barks of five therapeutically important medicinal plants native to India, i.e. Crataeva nurvala Buch.-Ham., Buchanania lanzan Spreng., Aegle marmelos Corr., Dalbergia sissoo Roxb. ex DC., and Cedrela toona Roxb. Methods Standardized aqueous alcoholic extracts from the selected barks having different target radicals, such as superoxide radical, nitric oxide, ABTS radical, and peroxidative decomposition of phospholipids, were prepared and screened by multiple in vitro assays. These extracts were also tested for total phenolic and tannin content and correlated with antioxidant capacity. Results Total phenolic and tannin contents were found to be the highest in C. nurvala (195 GAE mg/g and 218.3 mg/g CE). SOD mimetic activity was found to be the highest in Crataeva nurvula, although all barks showed activity more than 100 units/mg extract. Lipid peroxidation inhibitory potential was found to be the highest in Crataeva nurvala (83.4% inhibition of MDA formation/10 μg extract), and also showed a comparatively high NO quenching capacity (45.5% per 10 μg extract). The highest NO quenching potential was found in Aegle marmelos (47.3% per 10 μg extract). Cedrela toona showed the lowest LPO inhibitory potential and NO quenching capacity (50.5% and 30.5%, respectively). Buchanania lanzan, a medicinal plant extensively used for inflammatory disorders and Dalbergia sissoo also showed 72.5% and 69.1% LPO inhibitory potential/10 μg extract. Trolox equivalent antioxidant capacity ranged from 0.24 to 0.39 mmol/L TEAC/mg extract, indicating that all the barks tested had ABTS+· radical quenching capacity. 12Histone deacetylase inhibitor pre-treatment enhances the efficacy of DNA-interacting chemotherapeutic drugs in gastric cancer显示文摘BACKGROUND The prognosis of gastric cancer continues to remain poor,and epigenetic drugs like histone deacetylase inhibitors(HDACi)have been envisaged as potential therapeutic agents.Nevertheless,clinical trials are facing issues with toxicity and efficacy against solid tumors,which may be partly due to the lack of patient stratification for effective treatments.To study the need of patient stratification before HDACi treatment,and the efficacy of pre-treatment of HDACi as a chemotherapeutic drug sensitizer.METHODS The expression activity of class 1 HDACs and histone acetylation was examined in human gastric cancer cells and tissues.The potential combinatorial regime of HDACi and chemotherapy drugs was defined on the basis of observed drug binding assays,chromatin remodeling and cell death.RESULTS In the present study,the data suggest that the differential increase in HDAC activity and the expression of class 1 HDACs are associated with hypoacetylation of histone proteins in tumors compared to normal adjacent mucosa tissue samples of gastric cancer.The data highlights for the first time that pretreatment of HDACi results in an increased amount of DNA-bound drugs associated with enhanced histone acetylation,chromatin relaxation and cell cycle arrest.Fraction-affected plots and combination index-based analysis show that pre-HDACi chemo drug combinatorial regimes,including valproic acid with cisplatin or oxaliplatin and trichostatin A with epirubicin,exhibit synergism with maximum cytotoxic potential due to higher cell death at low combined doses in gastric cancer cell lines.CONCLUSION Expression or activity of class 1 HDACs among gastric cancer patients present an effective approach for patient stratification.Furthermore,HDACi therapy in pretreatment regimes is more effective with chemotherapy drugs,and may aid in predicting individual patient prognosis.Ramchandra Vigay Amnekar Shafqat Ali Khan Mudasir Rashid Bharat Khade Rahul Thorat Poonam Gera Shailesh V Shrikhande Duane T Smoot Hassan Ashktorab Sanjay Gupta 2020World Journal of Gastroenterology2020,26,6:3

14Molecular identification of sex in Hippophae rhamnoides L. using isozyme and RAPD markers显示文摘In many dioecious plants, gender affects economic value, breeding schemes and opportunities for commercial harvests. Hippophae rhamnoides L. is a dioecious plant species in which female genotypes are commercially preferred over male genotypes. Its berries have rich medicinal, nutritional and pharmaceutical properties because of their large amounts of vitamins, essential oils, proteins, fatty acids, free amino acids and flavanoids. Primary limitation for breeding H. rhamnoides L. is its dioecious nature, since gender cannot be identified by traditional methods. Therefore, some reliable and quick methods need to be developed. This communication deals with the development of isozyme and RAPD markers for early sex identification in this dioecious tree. The isozyme analysis was conducted with four enzyme systems, viz. peroxidase, esterase, malate dehydrogenase and catalase. The peroxidase enzyme system produced a female specific sex marker, which successfully differentiated between the staminate and pistillate genotypes of H. rhamnoides L. Thirty five random decamer primers were used in our study and one male sex linked marker was identified. OPD-20 (5′-ACTTCGCCAC-3′) displayed a band at 911 bp that expressed polymorphism between male and female genotypes. The staminate and pistillate genotypes could be distinguished using RAPD marker OPD-20911. 16Surgical left atrial appendage occlusion during cardiac surgery: A systematic review and meta-analysis显示文摘AIM To evaluate the safety and efficacy of surgical left atrial appendage occlusion(s-LAAO) during concomitant cardiac surgery.METHODS We performed a comprehensive literature search through May 31 st 2018 for all eligible studies comparing s-LAAO vs no occlusion in patients undergoing cardiac surgery. Clinical outcomes during follow-up included: embolic events, stroke, all-cause mortality, atrial fibrillation(AF), reoperation for bleeding and postoperative complications. We further stratified the analysis based on propensity matched studies and AF predominance.RESULTS Twelve studies(n = 40107) met the inclusion criteria.s-LAAO was associated with lower risk of embolic events(OR: 0.63, 95%CI: 0.53-0.76; P < 0.001) and stroke(OR: 0.68, 95%CI: 0.57-0.82; P < 0.0001).Stratified analysis demonstrated this association was more prominent in the AF predominant strata. There was no significant difference in the incidence risk of allcause mortality, AF, and reoperation for bleeding and postoperative complications.CONCLUSION Concomitant s-LAAO during cardiac surgery was associated with lower risk of follow-up thromboembolic events and stroke, especially in those with AF without significant increase in adverse events. 18Use of ketamine sedation for the management of displaced paediatric forearm fractures显示文摘AIM To determine if ketamine sedation is a safe and cost effective way of treating displaced paediatric radial and ulna fractures in the emergency department. METHODS Following an agreed interdepartmental protocol, fractures of the radius and ulna(moderately to severely displaced) in children between the age of 2 and 16 years old, presenting within a specified 4 mo period, were manipulated in our paediatric emergency department. Verbal and written consent was obtained prior to procedural sedation to ensure parents were informed and satisfied to have ketamine. A single attempt at manipulation was performed. Pre and postmanipulation radiographs were requested and assessed to ensure adequacy of reduction. Parental satisfaction surveys were collected after the procedure to assess the perceived quality of treatment. After closed reduction and cast immobilisation, patients were then followed-up in the paediatric outpatient fracture clinic and functional outcomes measured prospectively. A cost analysis compared to more formal manipulation under a general anaesthetic was also undertaken.RESULTS During the 4 mo period of study, 10 closed, moderate to severely displaced fractures were identified and treated in the paediatric emergency department using our ketamine sedation protocol. These included fractures of the growth plate(3), fractures of both radius and ulna(6) and a single isolated proximal radius fracture. The mean time from administration of ketamine until completion of the moulded plaster was 20 min. The mean time interval from sedation to full recovery was 74 min. We had no cases of unacceptable fracture reduction and no patients required any further manipulation, either in fracture clinic or under a more formal general anaesthetic. There were no serious adverse events in relation to the use of ketamine. Parents, patients and clinicians reported extremely favourable outcomes using this technique. Furthermore, compared to using a manipulation under general anaesthesia, each case performed under ketamine sedation was associated with a saving of £1470, the overall study saving being £14700. 20Quantum chemical analysis for the formation of glycine in the interstellar medium显示文摘Glycine (C2H5NO2) was the first amino acid to be detected in space by the stardust space probe in Comet Wild2, and is used by living organisms to make proteins. We discuss three different reaction paths for the formation of glycine in interstellar space from some simpler molecules detected in the interstellar medium. The possibility of the formation of glycine in interstellar space is considered by radical-radical and radical-molecule interaction schemes using quantum chemical calculations with density functional theory at the B3LYP/6-31G (d,p) level.
