| 3 | Intervertebral disc development and disease-related genetic polymorphisms显示文摘The intervertebral disc(IVD)comprises a gelatinous inner core(nucleus pulposus;NP)and concentric rings(annulus fibrosus;AF).The NP,an important structure for shock absorption in the vertebrate spinal motion segment,can be traced back to the notochord in ontogenetic lineage.In vertebrates,the notochord undergoes mucinoid changes,and had been considered vestigial until recently.However,observed correlations between IVD degeneration and back pain in humans have renewed interest in the IVD in biomedical fields.Beyond its mechanical contribution to development,the notochord is also an essential signaling center,which coordinates formation of the neural tube and somites.The pertinent signaling molecules,particularly TGF-b and bone morphogenetic proteins(BMPs),continue to play roles in the adult tissues and have been utilized for tissue regeneration.Genetic factors are major determinants of who will develop IVD degeneration and related back pain,and seem to correlate better with disc degeneration and back pain than do external forces on the spine.In summary,the spinal column is a landmark development in evolution.Genes directing the development of the IVD may also contribute to its maintenance,degeneration,and regeneration.Likewise,structural genes as well as genes responsible for maintenance of the structure are related to IVD degeneration.Finally,genes responsible for inflammation may play a dual role in exacerbating degeneration or facilitating repair responses depending on the context. | Jason W.Ashley Motomi Enomoto-Iwamoto Lachlan J.Smith Robert L.Mauck Danny Chan Joseph Lee Martin F.Heyworth Howard An Yejia Zhang | 2016 | Genes & Diseases2016,3,3: | 3 |
| 4 | Clinicopathological survey of 23 patients with lacrimal fossa lesions in Hong Kong显示文摘AIM: To survey the clinicopathological correlations of lacrimal fossa lesions in the Hong Kong population.METHODS: A total of 23 patients with lacrimal fossa lesions were identified radiologically.The mean age at the time of diagnosis was 52 year-old.The medical records of patients diagnosed with lacrimal fossa lesions and subsequently underwent radiological investigations were collected and reviewed retrospectively throughout a six years period from 2000 to 2006 from three regional hospitals (Tung Wah Eastern Hospital,Pamela Youde Nethersole Eastern Hospital and Queen Mary Hospital) belonging to the Hong Kong Mega Cluster.Patient demographics,presenting signs and symptoms,imaging modalities,pathological reports and subsequent management were recorded.RESULTS: The most common initial presenting symptom was upper lid swelling (83%) followed by proptosis (13%).19 patients (83%) had benign lacrimal fossa lesion and 4 patients (17%) had malignancy.Three of the malignancies were lymphomas and one was adenoid cystic carcinoma.CONCLUSION: Age is an important factor in considering the differential diagnosis of lacrimal fossa lesion.Lymphoproliferative diseases have the highest incidence in patients over 60 years of age while inflammatory lesions are more common in younger patients in Hong Kong. | Danny Siu-Chun Ng Edwin Chan Kin Yau Clement Wai-Nang Chan | 2011 | International Journal of Ophthalmology(English edition)2011,4,3: | 3 |

| 6 | Mitochondrial neuronal uncoupling proteins:a target for potential disease-modification in Parkinson’s disease Philip显示文摘This review gives a brief insight into the role of mitochondrial dysfunction and oxidative stress in the converging pathogenic processes involved in Parkinson’s disease(PD).Mitochondria provide cellular energy in the form of ATP via oxidative phosphorylation,but as an integral part of this process,superoxides and other reactive oxygen species are also produced.Excessive free radical production contributes to oxidative stress.Cells have evolved to handle such stress via various endogenous anti-oxidant proteins.One such family of proteins is the mitochondrial uncoupling proteins(UCPs),which are anion carriers located in the mitochondrial inner membrane.There are five known homologues(UCP1 to 5),of which UCP4 and 5 are predominantly expressed in neural cells.In a series of previous publications,we have shown how these neuronal UCPs respond to 1-methyl-4-phenylpyridinium(MPP+;toxic metabolite of MPTP)and dopamine-induced toxicity to alleviate neuronal cell death by preserving ATP levels and mitochondrial membrane potential,and reducing oxidative stress.We also showed how their expression can be influenced by nuclear factor kappa-B(NF-B)signaling pathway specifically in UCP4.Furthermore,we previously reported an interesting link between PD and metabolic processes through the protective effects of leptin(hormone produced by adipocytes)acting via UCP2 against MPP+-induced toxicity.There is increasing evidence that these endogenous neuronal UCPs can play a vital role to protect neurons against various pathogenic stresses including those associated with PD.Their expression,which can be induced,may well be a potential therapeutic target for various drugs to alleviate the harmful effects of pathogenic processes in PD and hence modify the progression of this disease. | WL Ho Jessica WM Ho Hui-Fang Liu Danny HF So Zero HM Tse Koon-Ho Chan David B Ramsden Shu-Leong Ho | 2012 | Translational Neurodegeneration2012,1,1: | 3 |

| 18 | Bevacizumab for neovascular age-related macular degeneration in Chinese patients in a clinical setting显示文摘AIM:To determine the outcome of non-investigational treatment with intravitreal bevacizumab(IVB) in neovascular age-related macular degeneration(AMD)patients.· METHODS:Retrospective chart review of 81 eyes with neovascular AMD followed-up for at least 12 mo and received 3-monthly loading IVB injections.Re-treat was based upon the individual clinician's judgment.Bestcorrected visual acuity(BCVA) and optical coherence tomography measurements of central foveal thickness outcomes were evaluated at 12,24 mo.· RESULTS:Eighty-one eyes(of 75 patients) completed12 mo of follow-up and 44 eyes(of 41 patients) completed24 mo of follow-up.The mean baseline logMAR BCVA significantly improved from 0.94±0.69 to 0.85±0.68 at12mo(P<0.001) and from 0.91±0.65 to 0.85±0.60(P=0.004)at 24 mo.The proportion of eyes that lost <15 logMAR letters at 12 mo was 90.1%and at 24 mo was 81.8%.IVB was effective in improving visual acuity in both treatment naive and previous photodynamic therapy(PDT)-treated subgroups.Treatment naive patients required significantly fewer injections than patients with prior PDT.Multiple regression analysis identified that poorer baseline visual acuity was associated with greater improvement in visual acuity(P=0.015).· CONCLUSION:Fewer injections in clinical practice may result in suboptimal visual outcomes compared with clinical trials of IVB in neovascular AMD patients.Poor baseline visual acuity and prior PDT treatment may also improve vision after IVB.The safety and durability of effect was maintained at 24 mo. | Danny Siu-Chun Ng Alvin Kwan-Ho Kwok Justin Man-Kit Tong Clement Wai-Nang Chan Walton Wai-Tat Li | 2016 | International Journal of Ophthalmology(English edition)2016,9,3: | 1 |
