2Tree ring based streamflow reconstruction for the Upper Yellow River over the past 1234 years显示文摘We established a Juniperus przewalski tree ring width chronology, based on tree ring cores collected from the A’nyêmaqên Mountains. Statistical analysis showed that the chronology was highly correlated with instrumental streamflow records from previous August to current July from the Tangnaihai Station in the upper reaches of the Yellow River, with a correlation coefficient of 0.656. Streamflow for the upper reaches of the Yellow River was reconstructed for the past 1234 years. Low flow periods for the 11-year averaged streamflow reconstruction were definite as lower than mean plus 1 standard deviation, and high flow periods were higher than mean minus 1 standard deviation. Over the past 1234 years, high flows occurred 18 times, and low flows occurred 12 times. The main low flow periods were identified as AD 1140–1156, AD 1295–1309, AD 1473–1500, and AD 1820–1847, and the main high flow periods were identified as AD 846–873, and AD 1375–1400. Extremely low streamflow over the reconstruction period was seen during the late 15th century, coinciding with a widespread drought phenomenon, which took place in the northeastern Tibetan Plateau over the same period. Reconstructed streamflow shows significant low-frequency variability, which is in line with drought variability of neighboring regions, as inferred from tree rings and other proxies. 4A 457-year reconstruction of precipitation in the southeastern Qinghai-Tibet Plateau,China using tree-ring records显示文摘This paper presents a 457-year reconstruction of precipitation in the southeastern Qinghai-Tibet Plateau using tree-ring records.Tree-ring samples were collected from the Hengduan Mountains in the southeastern part of the Qinghai-Tibet Plateau,China.A nearly 500-year chronology was developed using tree-ring width records.Correlation analysis shows moisture is the main factor limiting tree growth in this region.Ring-widths were significantly positively correlated with the Palmer Drought Severity Index(PDSI) and precipitation in many months.The highest correlation coefficient was found between the annual growth of trees and precipitation from the previous September to the current June(0.738).Based on this relationship,we reconstructed the precipitation history from 1509 to 2006.The reconstruction explains 54.4%(Radj2=53.5%,N=49,F=56.12) of the actual precipitation variation during the calibration period(1958-2006).During the reliable period of the reconstruction(1549-2006),some low-frequency climate signals are included,indicating this region has been getting wetter in the last 20 years.The reconstruction documents six apparently dry and five pluvial periods and the 17th century dry period lasted longer than any other.When compared with other recent studies,this study and these earlier reconstructions show a similar trend in the variation of drought and pluvial.Further spatial correlation analysis confirms that the reconstructed precipitation adequately represents the rainfall history of the entire Hengduan Mountain area.The Multi-taper method,a type of spectral analysis,reveals that precipitation in this area had significant(P<0.01) spectral peaks at 3-5 a,60 a and 79-85 a.GOU XiaoHua YANG Tao GAO LinLin DENG Yang YANG MeiXue CHEN FaHu 2013Chinese Science Bulletin2013,58,10:16
5Ubiquitination of Rheb governs growth factor-induced mTORC1 activation显示文摘Mechanistic target of rapamycin mTOR complex 1 (mTORC1)plays a key role in the integration of various environmental signals to regulate cell growth and metabolism,mTORC1 is recruited to the lysosome where it is activated by its interaction with GTP-bound Rheb GTPase.However,the regulatory mechanism of Rheb activity remains largely unknown.Here,we show that ubiquitination governs the nucleotide-bound status of Rheb.Lysosome-anchored E3 ligase RNF152 catalyzes Rheb ubiquitination and promotes its binding to the TSC complex.EGF enhances the deubiquitination of Rheb through AKT-dependent USP4 phosphorylation,leading to the release of Rheb from the TSC complex.Functionally,ubiquitination of Rheb is linked to mTORC1-mediated signaling and consequently regulates tumor growth.Thus,we propose a mechanistic model whereby Rheb-mediated mTORC1 activation is dictated by a dynamic opposing act between Rheb ubiquitination and deubiquitination that are catalyzed by RNF152 and USP4 respectively.Lu Deng Lei Chen Linlin Zhao Yan Xu Xiaoping Peng Xinbo Wang Lin Ding Jiali Jin Hongqi Teng Yanming Wang Weijuan Pan Fei Yu Lujian Liao Li Li Xin Ge Ping Wang 2019Cell Research2019,29,2:16
6Meningeal lymphatic vessels regulate brain tumor drainage and immunity显示文摘Recent studies have shown that meningeal lymphatic vessels(MLVs),which are located both dorsally and basally beneath the skull,provide a route for draining macromolecules and trafficking immune cells from the central nervous system(CNS)into cervical lymph nodes(CLNs),and thus represent a potential therapeutic target for treating neurodegenerative and neuroinflammatory diseases.However,the roles of MLVs in brain tumor drainage and immunity remain unexplored.Here we show that dorsal MLVs undergo extensive remodeling in mice with intracranial gliomas or metastatic melanomas.RNA-seq analysis of MLV endothelial cells revealed changes in the gene sets involved in lymphatic remodeling,fluid drainage,as well as inflammatory and immunological responses.Disruption of dorsal MLVs alone impaired intratumor fluid drainage and the dissemination of brain tumor cells to deep CLNs(dCLNs).Notably,the dendritic cell(DC)trafficking from intracranial tumor tissues to dCLNs decreased in mice with defective dorsal MLVs,and increased in mice with enhanced dorsal meningeal lymphangiogenesis.Strikingly,disruption of dorsal MLVs alone,without affecting basal MLVs or nasal LVs,significantly reduced the efficacy of combined anti-PD-1/CTLA-4 checkpoint therapy in striatal tumor models.Furthermore,mice bearing tumors overexpressing VEGF-C displayed a better response to anti-PD-1/CTLA-4 combination therapy,and this was abolished by CCL21/CCR7 blockade,suggesting that VEGF-C potentiates checkpoint therapy via the CCL21/CCR7 pathway.Together,the results of our study not only demonstrate the functional aspects of MLVs as classic lymphatic vasculature,but also highlight that they are essential in generating an efficient immune response against brain tumors.Xueting Hu Qiuping Deng Lu Ma Qingqing Li Yidong Chen Yuhan Liao Fan Zhou Chen Zhang Linlin Shao Jun Feng Tubao He Weihai Ning Yan Kong Yingqing Huo Aibin He Bing Liu Jingjing Zhang Ralf Adams Yulong He Fuchou Tang Xiuwu Bian Jincai Luo 2020Cell Research2020,30,3:11
7Development and characterization of a clinical strain of Coxsackievirus A16 and an eGFP infectious clone显示文摘Coxsackievirus A16(CA16) is one of the major causes of hand, foot, and mouth disease(HFMD) worldwide, which is a common illness that affects children. The frequent occurrence of HFMD outbreaks has become a serious public health problem in Asia. Therefore, it is important to understand the pathogenesis and replication of CA16. In this study, a stable infectious c DNA clone of an epidemic strain of Coxsackievirus A16(CA16) was assembled, and subsequently a reporter virus(e GFP-CA16) was constructed by inserting the e GFP gene between the 5'-UTR and the N-terminus of VP4, with the addition of a 2A protease cleavage site(ITTLG) at its C-terminus. This was transfected into Vero cells to generate infectious recombinant viruses. The growth characteristics and plaque morphology, in vitro, in mammalian cells were found to be indistinguishable between the parental and recombinant viruses. Although the e GFP-CA16 showed smaller plaque size as compared to recombinant CA16, both were found to exhibit similar growth trends and EC50 of NITD008. In summary, this stable infectious c DNA clone should provide a valuable experimental system to study CA16 infection and host response. 8Controllable synthesis of highly crystallized mesoporous TiO2/WO3 heterojunctions for acetone gas sensing显示文摘Mesoporous semiconducting metal oxides(SMOs)heterojunctions are appealing sensors for gas detecting.However,due to the different hydrolysis and condensation mechanism of every metal precursor and the contradiction between high crystallinity and high surface area,the synthesis of mesoporous SMOs heterojunctions with highly o rdered mesostructures,highly crystallized frameworks,and high surface area remains a huge challenge.In this work,we develop a novel'acid-base pair'adjusted solvent evaporation induced self-assembly(EISA)strategy to prepare highly crystallized ordered mesoporous TiO2/WO3(OM-TiO2/WO3)heterojunctions.The WCl6 and titanium isopropoxide(TIPO)are used as the precursors,respectively,which function as the'acid-base pair',enabling the coassembly with the structure directing agent(PEO-b-PS)into highly ordered meso structures.In addition,PEO-b-PS can be converted to rigid carbon which can protect the meso structures from collapse during the crystallization process.The resultant OM-TiO2/WO3 heterojunctions possess primitive cubic mesostructures,large pore size(~21.1 nm),highly crystalline frameworks and surface area(~98 m2/g).As a sensor for acetone,the obtained OM-TiO2/WO3 show excellent re sponse/recovery perfo rmance(3 s/5 s),good linear dependence,repeatability,selectivity,and long-term stability(35 days).Changyao Wang Yuhui Li Pengpeng Qiu Linlin Duan Wei Bi Yan Chen Dingyi Guo Yupu Liu Wei Luo Yonghui Deng 2020Chinese Chemical Letters2020,31,5:4

10Integrated histopathological,lipidomic,and metabolomic profiles reveal mink is a useful animal model to mimic the pathogenicity of severe COVID-19 patients显示文摘Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)is transmitted on mink farms between minks and humans in many countries.However,the systemic pathological features of SARS-CoV-2-infected minks are mostly unknown.Here,we demonstrated that minks were largely permissive to SARS-CoV-2,characterized by severe and diffuse alveolar damage,and lasted at least 14 days post inoculation(dpi).We first reported that infected minks displayed multiple organ-system lesions accompanied by an increased inflammatory response and widespread viral distribution in the cardiovascular,hepatobiliary,urinary,endocrine,digestive,and immune systems.The viral protein partially co-localized with activated Mac-2+macrophages throughout the body.Moreover,we first found that the alterations in lipids and metabolites were correlated with the histological lesions in infected minks,especially at 6 dpi,and were similar to that of patients with severe and fatal COVID-19.Particularly,altered metabolic pathways,abnormal digestion,and absorption of vitamins,lipids,cholesterol,steroids,amino acids,and proteins,consistent with hepatic dysfunction,highlight metabolic and immune dysregulation.Enriched kynurenine in infected minks contributed to significant activation of the kynurenine pathway and was related to macrophage activation.Melatonin,which has significant anti-inflammatory and immunomodulating effects,was significantly downregulated at 6 dpi and displayed potential as a targeted medicine.Our data first illustrate systematic analyses of infected minks to recapitulate those observations in severe and fetal COVID-19 patients,delineating a useful animal model to mimic SARS-CoV-2-induced systematic and severe pathophysiological features and provide a reliable tool for the development of effective and targeted treatment strategies,vaccine research,and potential biomarkers.Zhiqi Song Linlin Bao Wei Deng Jiangning Liu Erjun Ren Qi Lv Mingya Liu Feifei Qi Ting Chen Ran Deng Fengdi Li Yunpeng Liu Qiang Wei Hong Gao Pin Yu Yunlin Han Wenjie Zhao Junjun Zheng Xujian Liang Fuhe Yang Chuan Qin 2022Signal Transduction and Targeted Therapy2022,7,2:2

19The self-assembly and structural regulation of a hydrogen-bonded dimeric building block formed by two N-H…O hydrogen bonds on HOPG显示文摘The self-assembled behavior of an unsymmetric molecule(BCDTDA)with one imidazole group as center and benzoic acid group as functional group is studied,and the regulatory behaviors of coronene(COR)and three bipyridine derivatives(named BP,PEBP-C4 and PEBP-C8)on BCDTDA self-assembly structures are also investigated.Based on highly oriented pyrolytic graphite(HOPG)substrate,scanning tunneling microscopy(STM)is used to observe the variation of assembled behaviors at the solid-liquid interface.Because of the concentration effect,BCDTDA molecules can assemble into grids and Kagomés structures in the form of N–H…O hydrogen bonded dimers.BCDTDA molecules still maintain dimeric structures in the regulation of COR and BP molecules to BCDTDA self-assembly.However,PEBP-C4 and PEBP-C8 destroy the structure of the dimers,and form a variety of co-assembled structures with BCDTDA.Different guest molecules coordinate the host molecules differently,which makes the experiment more meaningful.Combined with density functional theory(DFT)calculation,the discovery of molecular interactions provides a promising strategy for the construction of functional nanostructures and devices.Peng Lei Lin Ma Siqi Zhang Jianqiao Li Linlin Gan Ke Deng Wubiao Duan Wei Li Qingdao Zeng 2023Chinese Chemical Letters2023,34,8:1
20Development and Characterization of West Nile Virus Replicon Expressing Secreted Gaussia Luciferase显示文摘We developed a Gaussia luciferase (Gluc) reporter replicon of West Nile virus (WNV) and used it to quantify viral translation and RNA replication. The advantage of the Gluc replicon is that Gaussia luciferase is secreted into the culture medium from cells transfected with Gluc replicon RNA, and the medium can be assayed directly for luciferase activity. Using a known Flavivirus inhibitor (NITD008), we demonstrated that the Gluc-WNV replicon could be used for antiviral screening.
