| 1 | Peptide aptamers:The versatile role of specific protein function inhibitors in plant biotechnology显示文摘In recent years,peptide aptamers have emerged as novel molecular tools that have attracted the attention of researchers in various fields of basic and applied science,ranging from medicine to analytical chemistry.These artificial short peptides are able to specifically bind,track,and inhibit a given target molecule with high affinity,even molecules with poor immunogenicity or high toxicity,and represent a remarkable alternative to antibodies in many different applications.Their use is on the rise,driven mainly by the medical and pharmaceutical sector.Here we discuss the enormous potential of peptide aptamers in both basic and applied aspects of plant biotechnology and food safety.The different peptide aptamer selection methods available both in vivo and in vitro are introduced,and the most important possible applications in plant biotechnology are illustrated.In particular,we discuss the generation of broadbased virus resistance in crops, 'reverse genetics' and aptasensors in bioassays for detecting contaminations in food and feed.Furthermore,we suggest an alternative to the transfer of peptide aptamers into plant cells via genetic transformation,based on the use of cell-penetrating peptides that overcome the limits imposed by both crop transformation and Genetically Modified Organism commercialization. | Monica Colombo Chiara Mizzotti Simona Masiero Martin M.Kater Paolo Pesaresi | 2015 | Journal of Integrative Plant Biology2015,57,11: | 4 |
| 2 | Cytoskeleton reorganization and ultrastructural damage induced by gliadin in a three-dimensional in vitro model显示文摘AIM: To evaluate the interplay between gliadin and LoVo cells and the direct effect of gliadin on cytoskeletal patterns.METHODS: We treated LoVo multicellular spheroids with digested bread wheat gliadin in order to investigate their morphology and ultrastructure (by means of light microscopy and scanning electron microscopy), and the effect of gliadin on actin (phalloidin fluorescence)and the tight-junction protein occludin and zonula occluden-1.RESULTS: The treated spheroids had deep holes and surface blebs, whereas the controls were smoothly surfaced ovoids. The incubation of LoVo spheroids with gliadin decreased the number of intracellular actin filaments, impaired and disassembled the integrity of the tight-junction system.CONCLUSION: Our data obtained from an 'in vivolike' polarized culture system confirm the direct noxious effect of gliadin on the cytoskeleton and tight junctions of epithelial cells. | 9 | Orthotopic induction of CH157MN convexity and skull base meningiomas into nude mice using stereotactic surgery and MRI characterization显示文摘Meningioma in vivo research is hampered by the difficulty of establishing an easy and reproducible orthotopic model able to mimic the characteristics of a human meningioma. Moreover, leptomeningeal dissemination and high mortality are often associated with such orthotopical models, making them useless for clinical translation studies. An optimized method for inducing meningiomas in nude mice at two different sites is described in this paper and the high reproducibility and low mortality of the models are demonstrated. Skull base meningiomas were induced in the auditory meatus and convexity meningiomas were induced on the brain surface of 23 and 24 nude mice, respectively. Both models led to the development of a mass easily observable by imaging methods. Dynamic contrast enhanced MRI was used as a tool to monitor and characterize the pathology onset and progression.
