| 1 | Dysregulation of the miR-146a-Smad4 axis impairs osteogenesis of bone mesenchymal stem cells under inflammation显示文摘Osteoporosis is a common disease that affects patient quality of life, especially among the elderly population.Although inflammation contributes significantly to osteoporosis, the underlying mechanism is unclear. In this study, we found that tumor necrosis factor(TNF)-α, an inflammatory environment mimic, inhibits osteogenesis of bone mesenchymal stem cells(BMSCs), induces mi R-146 a and decreases Smad4. Moreover,overexpression of mi R-146 a inhibited the osteogenic ability of BMSCs, whereas blocking mi R-146 a partially rescued the osteogenesis deficiency under TNF-α treatment. Molecularly, mi R-146 a decreased Smad4 expression at the protein level by binding to an element located in the Smad4 30-untranslated region, and restoration of Smad4 reversed the inhibitory effects of mi R-146 a on osteogenesis. | 2 | Gold nanorods together with HSP inhibitor-VER-155008 micelles for colon cancer mild-temperature photothermal therapy显示文摘Enhancing the heat-sensitivity of tumor cells provides an alternative solution to maintaining the therapeutic outcome of photothermal therapy(PTT). In this study, we constructed a therapeutic system, which was composed of methoxy-polyethylene-glycol-coated-gold-nanorods(MPEG-AuN R) and VER-155008-micelles, to evaluate the effect of VER-155008 on the sensitivity of tumor cells to heat, and further investigate the therapeutic outcome of MPEG-AuN R mediated PTT combined with VER-155008-micelles. VER-155008-micelles downregulate the expression of heat shock proteins and attenuate the heat-resistance of tumor cell. The survival of HCT116 cells treated with VER-155008-micelles under 45 ℃ is equal to that treated with high temperature hyperthermia(55 ℃) in vitro. Furthermore, we proved either the MPEG-AuN R or VER-155008-micelles can be accumulate in the tumor site by photoacoustic imaging and fluorescent imaging. In vivo anti-cancer evaluation showed that tumor size remarkably decreased(smaller than 100 mm^3 or vanished) when treated with combing 45℃ mild PTT system, which contrasted to the tumor size when treated with individual 45℃ mild PTT(around 500nm^3) or normal saline as control(larger than 2000 nm^3). | 3 | Output Characteristics of a Series Three-port Axial Piston Pump显示文摘Driving a hydraulic cylinder directly by a closed-loop hydraulic pump is currently a key research area in the field of electro-hydraulic control technology,and it is the most direct means to improve the energy efficiency of an electro-hydraulic control system.So far,this technology has been well applied to the pump-controlled symmetric hydraulic cylinder.However,for the differential cylinder that is widely used in hydraulic technology,satisfactory results have not yet been achieved,due to the asymmetric flow constraint.Therefore,based on the principle of the asymmetric valve controlled asymmetric cylinder in valve controlled cylinder technology,an innovative idea for an asymmetric pump controlled asymmetric cylinder is put forward to address this problem.The scheme proposes to transform the oil suction window of the existing axial piston pump into two series windows.When in use,one window is connected to the rod chamber of the hydraulic cylinder and the other is linked with a low-pressure oil tank.This allows the differential cylinders to be directly controlled by changing the displacement or rotation speed of the pumps.Compared with the loop principle of offsetting the area difference of the differential cylinder through hydraulic valve using existing technology,this method may simplify the circuits and increase the energy efficiency of the system.With the software SimulationX,a hydraulic pump simulation model is set up,which examines the movement characteristics of an individual piston and the compressibility of oil,as well as the flow distribution area as it changes with the rotation angle.The pump structure parameters,especially the size of the unloading groove of the valve plate,are determined through digital simulation.All of the components of the series arranged three distribution-window axial piston pump are designed,based on the simulation analysis of the flow pulse characteristics of the pump,and then the prototype pump is made.The basic characteristics,such as the pressure,flow and noise of the pumps under different rotation speeds,are measured on the test bench.The test results verify the correctness of the principle.The proposed research lays a theoretical foundation for the further development of a new pump-controlled cylinder system. | ZHANG Xiaogang QUAN Long YANG Yang WANG Chengbin YAO Liwei | 2012 | Chinese Journal of Mechanical Engineering2012,25,3: | 10 |
| 4 | Dynamic property tests of frozen red sandstone using a split hopkinson pressure bar显示文摘In this study, frozen red sandstone specimens were impacted by a Split Hopkinson bar (SHPB), with a velocity of 4.558 ~ 6.823 ms-1. The temperature of the specimens was maintained at -15℃ during the experiment. For comparison purposes, static uniaxial compression tests were conducted in advance using a freezing triaxial test machine. Four stress-strain curves were obtained in diff erent average strain rates. The test results suggested that when the average strain rate is low, the specimen strength changes gradually;but when it is high, its strength changes rapidly. When the average strain rate is 120.73 s-1, the peak value of stress is as high as 82.96 MPa, which is about two times that of the static compressive strength of 44.1 MPa. A constitutive model was established that was composed of the damaged, viscoelastic and spring bodies, and revealed the variations of compressive strength and strain for the frozen red sandstone under diff erent high strain rates. The test results also showed that the failure form was correlated to the average strain rate of the frozen red sandstone. When the average strain rate is low, the damage was only distributed on the specimen’s edges. However, as the average strain rate increases, the damage range extended to the central parts of the specimen. | 5 | Strong ozone production at a rural site in the North China Plain: Mixed effects of urban plumes and biogenic emissions显示文摘Regional ozone(O_3) pollution has drawn increasing attention in China over the recent decade, but the contributions from urban pollution and biogenic emissions have not been clearly elucidated. To better understand the formation of the regional O_3 problem in the North China Plain(NCP), intensive field measurements of O_3 and related parameters were conducted at a rural site downwind of Ji'nan, the capital city of Shandong province, in the summer of 2013. Markedly severe O_3 pollution was recorded, with the O_3 mixing ratios exceeding the Chinese national ambient air quality standard on 28 days(a frequency of 78%) and with a maximum hourly value of 198 ppbv. Extensive regional transport of wellprocessed urban plumes to the site was identified. An observation-constrained chemical box model was deployed to evaluate in situ photochemical O_3 production on two episodes.The results show that the in situ formation accounted for approximately 46% of the observed O_3 accumulation, while the remainder(~54%) was contributed by regional transport of the O_3-laden urban plumes. The in situ ozone production was in a mixed controlled regime that reducing either NOx or VOCs would lead to a reduction of ozone formation. Biogenic VOCs played an important role in the local ozone formation. | 7 | Recent advances in biological detection with magnetic nanoparticles as a useful tool显示文摘Magnetic nanoparticles have emerged as a powerful tool for magnetic resonance imaging, biodetection, drug delivery, and hyperthermia. This review focuses on the biological detection of magnetic nanoparticles as well as their physicochemical properties. Substantial progress in the sensitivity of detection has been made by developing variety of methods. Five applications of magnetic nanoparticles in biological detection are discussed in this review: magnetic separation, magnetic sensing, magnetic manipulation, magnetic catalysis, and signal enhancer for surface plasmon resonance(SPR). | 8 | Robust RGB-D Camera and IMU Fusion-based Cooperative and Relative Close-range Localization for Multiple Turtle-inspired Amphibious Spherical Robots显示文摘In the narrow, submarine, unstructured environment, the present localization approaches, such as GPS measurement, dead?rcckoning, acoustic positioning, artificial landmarks-based method, are hard to be used for multiple small-scale underwater robots. Therefore, this paper proposes a novel RGB-D camera and Inertial Measurement Unit (IMU) fusion-based cooperative and relative close-range localization approach for special environments, such as underwater caves. Owing to the rotation movement with zero-radius, the cooperative localization of Multiple Turtle-inspired Amphibious Spherical Robot (MTASRs) is realized. Firstly, we present an efficient Histogram of Oriented Gradient (HOG) and Color Names (CNs) fusion feature extracted from color images ofTASRs. Then, by training Support Vector Machine (SVM) classifier with this fusion feature, an automatic recognition method of TASRs is developed. Secondly, RGB-D camerabased measurement model is obtained by the depth map In order to realize the cooperative and relative close-range localization of MTASRs, the MTASRs model is established with RGB-D camera and IMU. Finally, the depth measurement in water is corrected and the efficiency of RGB-D camera for underwater application is validated. | 9 | Quantitative analysis of hepatoma-specific α-fetoprotein(HS-AFP) by a new mini-column affinity chromatography and its clinical value in diagnosis of hepatocellular carcinoma显示文摘Objective:To establish a convenient and economic method to determine hepatoma-specific α-fetoprotein(HS-AFP) for diagnosis of hepatocellular carcinoma(HCC). Methods:HS-AFP from serum of HCC patients was separated by a mini-column Lens culinaris agglutinin(LCA)-affinity chromatography. The levels of serum total AFP and separated HS-AFP were detected by radioimmunoassay(RIA). Results:Circulating AFP was separated into three peaks(AFP-1,AFP-2,and AFP-3) by LCA-affinity chromatography. During the elution course,the AFP-1 and AFP-2 could be eluted with TE buffer. HS-AFP(AFP-3) from sera of HCC patients was eluted clearly on the LCA-sepharose gel mini-column with a solution containing α-methyl-D-mannoside. It was a part of total AFP and only found in sera of HCC patients. A ratio of more than 15% for HS-AFP to total AFP in serum was considered as a specific marker for HCC diagnosis with higher sensitivity(92.7%) and specificity(88.2%). Conclusion:The new assay for circulating HS-AFP analysis is more sensitive,repeatable,and convenient. | 11 | Abnormal expression of vascular endothelial growth factor (VEGF) and its clinical features in tissues of human lung cancer显示文摘Objective: Angiogennesis, the formation of new blood vessels from the existing vascular bed, is essential step for growth and invasion of primary tumor. Vascular endothelial growth factor (VEGF) is known to play crucial role in tumor angiogenesis. In the present study, we investigate the expression of VEGF and VEGF-mRNA in the angiogennesis, metastasis and prognosis of lung cancer. Methods: The VEGF cellular distributions and expression in 38 specimens of patients with lung cancer were investigated with immunohistochemistry stain technology. The total RNAs in 38 tissues of lung cancer was measured, then the levels of VEGF-mRNA expression were analyzed by a reverse-transcription polymerase chain reaction (RT-PCR) assay. The levels of VEGF in sera of patients with lung cancer, benign lung diseases and healthy controls were detected through Enzyme linked immunosorbent assay (ELISA) method. Results: The VEGF positive stain was 76% in 38 cases of lung cancer specimens. The 89% rate of VEGF stain was found for clinical stage Ⅲ cases and 92% for stage Ⅳ lung cancers. The significantly higher expression of VEGF was evidenced in patients with lymph node metastasis (84%), distant metastasis (90%), and lung cancers with lower histological differentiation (89%), respectively. The expression level of total RNA was significantly higher in patients with lung cancers than that in their paracancerous or distant lung tissues. The VEGF expressions were tightly correlated with total RNA concentration of lung carcinoma (P<0.01). The predominant expressions of VEGF121 and VEGF165 gene fragments were found in lung cancer specimens by RT-PCR analysis. No significant difference of serum VEGF levels was detected between cases with lung cancer and patients with benign diseases. However, the VEGF level of cases with benign diseases was decreased significantly after patients with anti-inflammation medication. | 12 | Characterization of different-Al-content AlGaN/GaN heterostructures on sapphire显示文摘Al x Ga 1-x N/GaN high-electron-mobility transistor (HEMT) structures with Al composition ranging from x = 0.13 to 0.36 are grown on sapphire substrates by low-pressure metalorganic chemical vapor deposition (LP-MOCVD). The effects of Al content on crystal quality, surface morphology, optical and electrical characteristics of the AlGaN/GaN heterostructures have been analyzed. Although high Al-content (36%) heterostructure exhibits a distinguished photoluminescence peak related to recombination between the two-dimensional electron gas and photoexcited holes (2DEG-h), its crystal quality and rough surface morphology are poor. 2DEG mobility increases with the Al content up to 26% and then it apparently decreases for high Al-content (36%) AlGaN/GaN heterostructure. The increase of sheet carrier density with the increase of Al content has been observed. A high mobility at room temperature of 2105 cm 2 /V s with a sheet carrier density of n s = 1.10 × 10 13 cm -2 , for a 26% Al-content AlGaN/GaN heterostructure has been obtained, which is approaching state-of-the-art for HEMT grown on SiC. | 13 | Facile synthesis of hierarchical Na2Fe(SO4)2@rGO/C as high-voltage cathode for energy density-enhanced sodium-ion batteries显示文摘Fe-based sulfates are ideal cathode candidates for sodium-ion batteries(SIBs) owing to their high operating voltage and low cost but suffer from the nature of poor power performance. Herein, a hierarchical porous Na2Fe(SO4)2@reduced graphene oxide/carbon dot(Na2Fe(SO4)2@rGO/C) with low carbon content(4.12 wt%) was synthesized via a facile homogeneous strategy benefiting for engineering application,which delivers excellent sodium storage performance(high voltage plateau of 3.75 V, 85 m Ah g-1 and330 Wh kg-1 at 0.05 C;5805 W kg-1 at 10 C) and high Na+diffusion coefficient(1.19 × 10-12 cm2 s-1).Moreover, the midpoint voltage of assembled full cell could reach 3.0 V. The electron transfer and reaction kinetics are effectively boosted since the nanoscale Na2Fe(SO4)2 is supported by a robust crosslinked carbon matrix with rGO sheets and carbon dots. The slight rGO sheets sufficiently enhance the electron transfer like a current collecter and restrain the aggregation, as well as ensure smooth ion channels. Meanwhile, the carbon dots in the whole space connect with Na2Fe(SO4)2 and help rGO to promote the conductivity of the electrode.
