| 2 | Establishment and cryopreservation of liver, heart and muscle cell lines derived from the Chinese alligator (Alligator sinensis)显示文摘The Chinese alligator, Alligator sinensis, is a critically endangered species. A conservation project of gene resources for an endangered species first involves the preservation of organs, tissues, gametes, genomic DNA libraries and cell lines. The present study is the first to establish and cryopreserve cell lines of liver, heart and muscle tissues from the Chinese alligator. The study revealed that there was a large discrepancy in cell migration time in primary cultures among liver (11-12 d), heart (13-14 d) and muscle (17-18 d) tissue pieces. The differences in time in primary cell culture suggested that it was relatively easy to build visceral-derived cell lines for reptiles. Biological analysis showed that the population doubling time for thawed cells was approxi- mately 36 h. Karyotyping revealed that the frequency of Chinese alligator cells showing chromosome number as 2n=32 was 88.6%-93.4%. Chinese alligator cell lines established here provide a vital resource for research and are likely to be useful for protection of this rare and critically endangered species. | 3 | Analysis theory of random energy of train derailment in wind显示文摘Based on the analysis theory of random energy of train derailment, an analysis theory of random energy of train derailment in wind is suggested. Two methods are proposed -the time domain method and the frequency domain method of analysis theory of random energy of train derailment in wind. The curves of σ pw -v under various wind speeds are obtained through the computation. The original curve of σ p -v is expanded, which turns the analysis theory of random energy of train derailment into the all-weather theory. Train derailment condition has been established under wind action. The first and second criterions of train derailment have been proposed in light of wind action. The analysis of train derailment cases at home or abroad is made, in- cluding the first analysis of Xinjiang train derailment case encountered 13-level of gale, which explained the inevitability of train derailment. The analysis theory of random energy of train derailment in wind shows its validity and accuracy. The input energy σ pw of the transverse vibration of train-track(bridge)-wind system is linked to train speed. | 4 | Optimal root system strategies for desert phreatophytic seedlings in the search for groundwater显示文摘Desert phreatophytes are greatly dependent on groundwater, but how their root systems adapt to different groundwater depths is poorly understood. In the present study, shoot and root growths of Alhagi sparsifolia Shap. seedlings were studied across a gradient of groundwater depths. Leaves, stems and roots of different orders were measured after 120 days of different groundwater treatments. Results indicated that the depth of soil wetting front and the vertical distribution of soil water contents were highly controlled by groundwater depths. The shoot growth and biomass of A. sparsifolia decreased, but the root growth and rooting depth increased under deeper groundwater conditions. The higher ratios of root biomass, root/shoot and root length/leaf area under deeper groundwater conditions implied that seedlings of A. sparsifolia economized carbon cost on their shoot growths. The roots of A. sparsifolia distributed evenly around the soil wetting fronts under deeper groundwater conditions. Root diameters and root lengths of all orders were correlated with soil water availabilities both within and among treatments. Seedlings of A. sparsifolia produced finer first- and second-order roots but larger third- and fourth-order roots in dry soils. | 7 | Combining single-cell RNA sequencing and population-based studies reveals hand osteoarthritis-associated chondrocyte subpopulations and pathways显示文摘Hand osteoarthritis is a common heterogeneous joint disorder with unclear molecular mechanisms and no disease-modifying drugs.In this study,we performed single-cell RNA sequencing analysis to compare the cellular composition and subpopulationspecific gene expression between cartilage with macroscopically confirmed osteoarthritis(n=5)and cartilage without osteoarthritis(n=5)from the interphalangeal joints of five donors.Of 105142 cells,we identified 13 subpopulations,including a novel subpopulation with inflammation-modulating potential annotated as inflammatory chondrocytes.Fibrocartilage chondrocytes exhibited extensive alteration of gene expression patterns in osteoarthritic cartilage compared with nonosteoarthritic cartilage.Both inflammatory chondrocytes and fibrocartilage chondrocytes showed a trend toward increased numbers in osteoarthritic cartilage.In these two subpopulations from osteoarthritic cartilage,the ferroptosis pathway was enriched,and expression of iron overload-related genes,e.g.,FTH1,was elevated.To verify these findings,we conducted a Mendelian randomization study using UK Biobank and a population-based cross-sectional study using data collected from Xiangya Osteoarthritis Study.Genetic predisposition toward higher expression of FTH1 mRNA significantly increased the risk of hand osteoarthritis(odds ratio=1.07,95%confidence interval:1.02–1.11)among participants(n=332668)in UK Biobank.High levels of serum ferritin(encoded by FTH1),a biomarker of body iron overload,were significantly associated with a high prevalence of hand osteoarthritis among participants(n=1241)of Xiangya Osteoarthritis Study(P-for-trend=0.037).In conclusion,our findings indicate that inflammatory and fibrocartilage chondrocytes are key subpopulations and that ferroptosis may be a key pathway in hand osteoarthritis,providing new insights into the pathophysiology and potential therapeutic targets of hand osteoarthritis. | Hui Li Xiaofeng Jiang Yongbing Xiao Yuqing Zhang Weiya Zhang Michael Doherty Jacquelyn Nestor Changjun Li Jing Ye Tingting Sha Houchen Lyu Jie Wei Chao Zeng Guanghua Lei | 2023 | Bone Research2023,11,4: | 0 |

| 11 | Evolution of Phenotype and Mitochondrial Genome Reveals Limbless and Body-elongated Squamates may Change Their Energy Basis for Locomotion显示文摘Limb reduction in Squamata present the dramatic characteristic to focus and usually accompanied with particularly morphological modifications, impacting tremendous locomotion changing and might generate different energy requirement. Herein, we combined both morphological and mitochondrial genomic data to explore the evolution of phenotypic transformation and mitochondrial genome of limbless and body-elongated squamates. We collected phenotypic measurements of 503 individuals, representing limbed or limbless taxa across all major lineages in Squama ta to investiga te the morphological correla tions with limb-reduction. Furthermore, we provided the mitochondrial genome of the representative limbless and elongated species Dibamus bourreti(Angel, 1935) to detect selective constraints on limbless clades with published mitogenomes of other squa ma te reptiles. Our results evidenced tha t body elongation had certain negative relationship with limbreduction in Squamata lineage and Lacertilia lineage(R = –0.495, P < 2.2 e-16;R= –0.332, P = 1.1 e-13, respectively), while tail length showed slight correlation in both clades(R = 0.156, P = 4.3 e-04;R= 0.192, P = 2.1 e-05, respectively).
