1Autophagy inhibition enhances apigenin-induced apoptosis in human breast cancer cells显示文摘Apigenin (4',5,7-trihydroxyflavone) is a member of the flavone subclass of flavonoids present in fruits and vegetables. The involvement of autophagy in the apigenin-induced apoptotic death of human breast cancer cells was investigated. Cell proliferation and viability were assessed by 3-(4,5-dimethylthiazol- 2-yl)-2,5-diphenyltetrazolium bromide (MTT) and clonogenic assays. Flow cytometry, fluorescent staining and Western blot analysis were employed to detect apoptosis and autophagy, and the role of autophagy was assessed using autophagy inhibitors. Apigenin dose- and time-dependently repressed the proliferation and clonogenic survival of the human breast cancer T47D and MDA-MB-231 cell lines. The death of T47D and MDA-MB-231 cells was due to apoptosis associated with increased levels of Caspase3, PARP cleavage and Bax/Bcl-2 ratios. The results from flow cytometry and fluorescent staining also verified the occurrence of apoptosis. In addition, the apigenin-treated cells exhibited autophagy, as characterized by the appearance of autophagosomes under fluorescence microscopy and the accumulation of acidic vesicular organelles (AVOs) by flow cytometry. Furthermore, the results of the Western blot analysis revealed that the level of LC3-II, the processed form of LC3-I, was increased. Treatment with the autophagy inhibitor, 3-methyladenine (3-MA), significantly enhanced the apoptosis induced by apigenin, which was accompanied by an increase in the level of PARP cleavage. Similar results were also confirmed by flow cytometry and fluorescence microscopy. These results indicate that apigenin has apoptosis-and autophagy-inducing effects in breast cancer cells. 3Direct observation of ultrafast plasmonic hot electron transfer in the strong coupling regime显示文摘Achieving strong coupling between plasmonic oscillators can significantly modulate their intrinsic optical properties.Here,we report the direct observation of ultrafast plasmonic hot electron transfer from an Au grating array to an MoS_(2) monolayer in the strong coupling regime between localized surface plasmons(LSPs)and surface plasmon polaritons(SPPs).By means of femtosecond pump-probe spectroscopy,the measured hot electron transfer time is approximately 40 fs with a maximum external quantum yield of 1.65%.Our results suggest that strong coupling between LSPs and SPPs has synergetic effects on the generation of plasmonic hot carriers,where SPPs with a unique nonradiative feature can act as an‘energy recycle bin’to reuse the radiative energy of LSPs and contribute to hot carrier generation.Coherent energy exchange between plasmonic modes in the strong coupling regime can further enhance the vertical electric field and promote the transfer of hot electrons between the Au grating and the MoS_(2) monolayer.Our proposed plasmonic strong coupling configuration overcomes the challenge associated with utilizing hot carriers and is instructive in terms of improving the performance of plasmonic opto-electronic devices.Hangyong Shan Ying Yu Xingli Wang Yang Luo Shuai Zu Bowen Du Tianyang Han Bowen Li Yu Li Jiarui Wu Feng Lin Kebin Shi Beng Kang Tay Zheng Liu Xing Zhu Zheyu Fang 2019Light(Science & Applications)2019,8,1:7

5Erythromycin loaded by tetrahedral framework nucleic acids are more antimicrobial sensitive against Escherichia coli(E.coli)显示文摘Erythromycin is a commonly used broad-spectrum antibiotic,but resistance to this antibiotic makes its use less effective.Considerable efforts,beside finding alternatives,are needed to enhance its antimicrobial effect and stability against bacteria.Tetrahedral framework nucleic acids(tFNAs),a novel delivery vehicle with a three-dimensional nanostructure,have been studied as a carrying platform of antineoplastic drugs.In this study,the use of tFNAs in delivering erythromycin into Escherichia coli(E.coli)was investigated for the first time.The tFNAs vehicle increased the bacterial uptake of erythromycin and promoted membrane destabilization.Moreover,it increased the permeability of the bacterial cell wall,and reduced drug resistance by improving the movement of the drug across the membrane.The tFNAs-based delivery system enhanced the effects of erythromycin against E.coli.It may therefore provide an effective delivery vehicle for erythromycin in targeting antibiotic-resistant bacteria with thick cell wall.Yue Sun Yuhao Liu Bowen Zhang Shirong Shi Tao Zhang Dan Zhao Taoran Tian Qirong Li Yunfeng Lin 2021Bioactive Materials2021,6,8:4

9Refined spatial temporal epigenomic profiling reveals intrinsic connection between PRDM9-mediated H3K4me3 and the fate of double-stranded breaks显示文摘Meiotic recombination is initiated by the formation of double-strand breaks(DSBs),which are repaired as either crossovers(COs)or noncrossovers(NCOs).In most mammals,PRDM9-mediated H3K4me3 controls the nonrandom distribution of DSBs;however,both the timing and mechanism of DSB fate control remain largely undetermined.Here,we generated comprehensive epigenomic profiles of synchronized mouse spermatogenic cells during meiotic prophase I,revealing spatiotemporal and functional relationships between epigenetic factors and meiotic recombination.We find that PRDM9-mediated H3K4me3 at DSB hotspots,coinciding with H3K27ac and H3K36me3,is intimately connected with the fate of the DSB.Our data suggest that the fate decision is likely made at the time of DSB formation:earlier formed DSBs occupy more open chromatins and are much more competent to proceed to a CO fate.Our work highlights an intrinsic connection between PRDM9-mediated H3K4me3 and the fate decision of DSBs,and provides new insight into the control of CO homeostasis.Yao Chen Ruitu Lyu Bowen Rong Yuxuan Zheng Zhen Lin Ruofei Dai Xi Zhang Nannan Xie Siqing Wang Fuchou Tang Fei Lan Ming-Han Tong 2020Cell Research2020,30,3:3

11Procedural complexity independent of P2Y12 reaction unit(PRU)values is associated with acute in situ thrombosis in Pipeline flow diversion of cerebral aneurysms显示文摘background Acute in situ thrombosis is an ischaemic phenomenon during Pipeline embolisation device(PED)procedures with potentially high morbidity and mortality.There is controversy regarding the role of platelet function testing with P2Y12 assay as a predictor of intraprocedural thromboembolic events.There is limited knowledge on whether procedural complexity influences these events.Methods Data were collected retrospectively on 742 consecutive PED cases at a single institution.Patients with intraprocedural acute thrombosis were compared with patients without these events.results A cohort of 37 PED cases with acute in situ thrombosis(mean age 53.8 years,mean aneurysm size 8.4 mm)was matched with a cohort of 705 PED cases without intraprocedural thromboembolic events(mean age 56.4 years,mean aneurysm size 6.9 mm).All patients with in situ thrombosis received intra-arterial and/or intravenous abciximab.The two groups were evenly matched in patient demographics,previous treatment/subarachnoid hemorrhage(SAH)and aneurysm location.There was no statistical difference in postprocedural P2Y12 reaction unit(PRU)values between the two groups,with a mean of 156 in the in situ thrombosis group vs 148 in the control group(p=0.5894).Presence of cervical carotid tortuosity,high cavernous internal carotid artery grade,need for multiple PED and vasospasm were not significantly different between the two groups.The in situ thrombosis group had statistically significant longer fluoroscopy time(60.4 vs 38.4 min,p<0.0001),higher radiation exposure(3476 vs 2160 mGy,p<0.0001),higher rates of adjunctive coiling(24.3% vs 8.37%,p=0.0010)and higher utilisation of balloon angioplasty(37.8% vs 12.2%,p<0.0001).Clinically,the in situ thrombosis cohort had higher incidence of major and minor stroke,intracerebral haemorrhage and length of stay.Conclusions Predictors of procedural complexity(higher radiation exposure,longer fluoroscopy time,adjunctive coiling and need for balloon angioplasty)are associated with acute thrombotic events during PED placement,independent of PRU values.Bowen Jiang Matthew T Bender Erick M Westbroek Jessica K Campos Li-Mei Lin Risheng Xu Rafael J Tamargo Judy Huang Geoffrey P Colby Alexander L Coon 2018Stroke & Vascular Neurology2018,3,3:2

14CAPE promotes the expansion of human umbilical cord blood-derived hematopoietic stem and progenitor cells in vitro显示文摘Due to the low number of collectable stem cells from single umbilical cord blood(UCB)unit,their initial uses were limited to pediatric therapies.Clinical applications of UCB hematopoietic stem and progenitor cells(HSPCs)would become feasible if there were a culture method that can effectively expand HSPCs while maintaining their self-renewal capacity.In recent years,numerous attempts have been made to expand human UCB HSPCs in vitro.In this study,we report that caffeic acid phenethyl ester(CAPE),a small molecule from honeybee extract,can promote in vitro expansion of HSPCs.Treatment with CAPE increased the percentage of HSPCs in cultured mononuclear cells.Importantly,culture of CD34+HSPCs with CAPE resulted in a significant increase in total colony-forming units and high proliferative potential colony-forming units.Burst-forming unit-erythroid was the mostly affected colony type,which increased more than 3.7-fold in 1μg mL 1CAPE treatment group when compared to the controls.CAPE appears to induce HSPC expansion by upregulating the expression of SCF and HIF1-α.Our data suggest that CAPE may become a potent medium supplement for in vitro HSPC expansion.LIU YiMing ZHANG BoWen ZHANG Jing WANG SiHan YAO HaiLei HE LiJuan CHEN Lin YUE Wen LI YanHua PEI XueTao 2014Science China(Life Sciences)2014,57,2:2
15Recent Advances in Two-Dimensional Magnets:Physics and Devices towards Spintronic Applications显示文摘The emergence of low-dimensional nanomaterials has brought revolutionized development of magnetism,as the size effect can significantly influence the spin arrangement.Since the first demonstration of truly two-dimensional magnetic materials(2DMMs)in 2017,a wide variety of magnetic phases and associated properties have been exhibited in these 2DMMs,which offer a new opportunity to manipulate the spin-based devices efficiently in the future.Herein,we focus on the recent progress of 2DMMs and heterostructures in the aspects of their structural characteristics,physical properties,and spintronic applications.Firstly,the microscopy characterization of the spatial arrangement of spins in 2D lattices is reviewed.Afterwards,the optical probes in the light-matter-spin interactions at the 2D scale are discussed.Then,particularly,we systematically summarize the recent work on the electronic and spintronic devices of 2DMMs.In the section of electronic properties,we raise several exciting phenomena in 2DMMs,i.e.,long-distance magnon transport,field-effect transistors,varying magnetoresistance behavior,and(quantum)anomalous Hall effect.In the section of spintronic applications,we highlight spintronic devices based on 2DMMs,e.g.,spin valves,spin-orbit torque,spin field-effect transistors,spin tunneling field-effect transistors,and spin-filter magnetic tunnel junctions.At last,we also provide our perspectives on the current challenges and future expectations in this field,which may be a helpful guide for theorists and experimentalists who are exploring the optical,electronic,and spintronic properties of 2DMMs.Vertikasari PNingrum Bowen Liu Wei Wang Yao Yin Yi Cao Chenyang Zha Hongguang Xie Xiaohong Jiang Yan Sun Sichen Qin Xiaolong Chen Tianshi Qin Chao Zhu Lin Wang Wei Huang 2020Research2020,,1:2

17Optimization of the OCO-2 Cloud Screening Algorithm and Evaluation against MODIS and TCCON Measurements over Land Surfaces in Europe and Japan显示文摘A method to tighten the cloud screening thresholds based on local conditions is used to provide more stringent schemes for Orbiting Carbon Observatory-2(OCO-2)cloud screening algorithms.Cloud screening strategies are essential to remove scenes with significant cloud and/or aerosol contamination from OCO-2 observations,which helps to save on the data processing cost and ensure high quality retrievals of the column-averaged CO2 dry air mole fraction(XCO2).Based on the radiance measurements in the 0.76μm O2A band,1.61μm(weak),and 2.06μm(strong)CO2 bands,the current combination of the A-Band Preprocessor(ABP)algorithm and Iterative Maximum A Posteriori(IMAP)Differential Optical Absorption Spectroscopy(DOAS)Preprocessor(IDP)algorithm passes around 20%-25%of all soundings,which means that some contaminated scenes also pass the screening process.In this work,three independent pairs of threshold parameters used in the ABP and IDP algorithms are sufficiently tuned until the overall pass rate is close to the monthly clear-sky fraction from the MODIS cloud mask.The tightened thresholds are applied to observations over land surfaces in Europe and Japan in 2016.The results show improvement of agreement and positive predictive value compared to the collocated MODIS cloud mask,especially in summer and fall.In addition,analysis indicates that XCO2 retrievals with more stringent thresholds are in closer agreement with measurements from collocated Total Carbon Column Observing Network(TCCON)sites.Sijie CHEN Shuaibo WANG Lin SU Changzhe DONG Ju KE Zhuofan ZHENG Chonghui CHENG Bowen TONG Dong LIU 2020Advances in Atmospheric Sciences2020,37,4:1
18Effect of High Electric Field on Lifetime Performance of Metalized Film Capacitor in Pulsed-power Applications显示文摘High energy density capacitor is a key device to power supply source for electromagnetic gun (EMG) system, and extending its lifetime is important for increasing the reliability of the power source. Working in high electric field could affect the capacitor lifetime, and this effect on metallized polypropylene film capacitors (MPPFCs) in pulsed-power applications is studied and presented. Experimental re- sults show that the lifetime of MPPFCs decreases with the increasing peak value of charged electric field, and this decrease could be described by function (L/L0) ∝ (E/E0)–m, where, m=7.32. The lifetime of MPPFCs also decreases with the increase of the reversal coeffi- cients in underdamped circuits, which could be described by (L/L0) ∝ (ln(1/K0)/(ln(1/K))–b, where, b=0.7. 19Correlates of Sexual Lifestyles Among Older Adults Living With HIV in China: Findings From the Sexual Well-being (SWELL) Study显示文摘Sexual lifestyles are closely related to overall human health and well-being.Few studies have focused on sexual lifestyles among older adults living with HIV(OALHIV),especially in low-and middle-income countries.This study is a part of the sexual well-being among older adults in China(SWELL)study,which is a multicenter cross-sectional study focused on sexual health among older adults aged 50 years and older.Participants were 680 OALHIV(≥50 years old)from the SWELL study conducted from June 2020 to December 2022.Data were collected through one-on-one interviews.We used logistic regression to assess the correlates of sexual activity and sexual satis-faction.Among all participants,37.1%were sexually active.Being older,male,in a steady relationship and employed were associated with being sexually active.The prevalence of sexual satisfaction was 69.8%among sexually active OALHIV.Being homosexual and reporting a better general health status were associated with sexual satisfaction.The OALHIV who have depressive symptoms were less likely to report sexual satisfaction.To support holistic healthy aging among OALHIV,it is important for healthcare providers to be educated about the importance of enquiring about sexual activity,satisfaction and problems and addressing concerns while conveying sex-positive attitudes during clinical reviews,as these are still critical aspects of their health and well-being.Bingyi Wang Xin Peng Leiwen Fu Xinyi Li Tongxin Shi Bowen Liang Tian Tian Thomas Fitzpatrick Zhen Lu Weijie Zhang JunyeBian Xin Xiao YingWang Yunlong Ao Lin Ouyang YongCai Maohe Yu Guohui Wu Linghua Li Xiaojun Meng Huachun Zou 2023Infectious Microbes & Diseases2023,5,3:1
