2Correlation of IL-8 with induction, progression and metastatic potential of colorectal cancer显示文摘AIM: To investigate the expression profile of IL-8 in inflammatory and malignant colorectal diseases to evaluate its potential role in the regulation of colorectal cancer (CRC) and the development of colorectal liver metastases (CRLM). METHODS: IL-8 expression was assessed by quantitative real-time PCR (Q-RT-PCR) and the enzyme-linked immunosorbent assay (ELISA) in resected specimens from patients with ulcerative colitis (UC, n = 6) colorectal adenomas (CRA, n = 8), different stages of colorectal cancer (n = 48) as well as synchronous and metachronous CRLM along with their corresponding primary colorectal tumors (n = 16). RESULTS: IL-8 mRNA and protein expression was significantly up-regulated in all pathological colorectal entities investigated compared with the corresponding neighboring tissues. However, in the CRC specimens IL-8 revealed a significantly more pronounced overexpression in relation to the CRA and UC tissues with an average 30-fold IL-8 protein up-regulation in the CRC specimens in comparison to the CRA tissues. Moreover, IL-8 expression revealed a close correlation with tumor grading. Most interestingly, IL-8 up-regulation was most enhanced in synchronous and metachronous CRLM, if compared with the corresponding primary CRC tissues. Herein, an up to 80-fold IL-8 overexpression in individual metachronous metastases compared to normal tumor neighbor tissues was found. 7High-resolution ex vivo analysis of the degradation and osseointegration of Mg-xGd implant screws in 3D显示文摘Biodegradable magnesium(Mg)alloys can revolutionize osteosynthesis,because they have mechanical properties similar to those of the bone,and degrade over time,avoiding the need of removal surgery.However,they are not yet routinely applied because their degradation behavior is not fully understood.In this study we have investigated and quantified the degradation and osseointegration behavior of two biodegradable Mg alloys based on gadolinium(Gd)at high resolution.Mg-5Gd and Mg-10Gd screws were inserted in rat tibia for 4,8 and 12 weeks.Afterward,the degradation rate and degradation homogeneity,as well as bone-to-implant interface,were studied with synchrotron radiation micro computed tomography and histology.Titanium(Ti)and polyether ether ketone(PEEK)were used as controls material to evaluate osseointegration.Our results showed that Mg-5Gd degraded faster and less homogeneously than Mg-10Gd.Both alloys gradually form a stable degradation layer at the interface and were surrounded by new bone tissue.The results were correlated to in vitro data obtained from the same material and shape.The average bone-to-implant contact of the Mg-xGd implants was comparable to that of Ti and higher than for PEEK.The results suggest that both Mg-xGd alloys are suitable as materials for bone implants.Diana Krüger Silvia Galli Berit Zeller-Plumhoff D.C.Florian Wieland Niccolo Peruzzi Bjorn Wiese Philipp Heuser Julian Moosmann Ann Wennerberg Regine Willumeit-Romer 2022Bioactive Materials2022,7,7:2

19Age-related histomorphologic changes in the canine gastrointestinal tract:A histologic and immunohistologic study显示文摘AIM: To examine the changes in the histomorphology of the gastric, jejunal and colonic wall of dogs due to physiological aging. METHODS: Full thickness biopsies were taken from the gastrointestinal tracts of 28 dogs of different ages. The thickness of the different layers of the wall was measured and the numbers of proliferating cells as indicated by immunohistochemical detection of Ki67 were counted. RESULTS: In the three excision sites, the thickness of all subepithelial layers increased with rising age. The strongest correlation between age and thickness of the intestinal wall was found in the first 10 years of life and in the jejunum (r = 0.6-0.71 for the deep lamina propria mucosa, the muscularis mucosa, and the circular layer of the tunica muscularis). The number of proliferating cells decreased during aging, with the strongest correlation in the lamina propria mucosa and lamina muscularis mucosa of the jejunum and in the colonic submucosa (r = -0.61 to -0.71). Epithelial proliferation was only weakly correlated to the age. CONCLUSION: The morphology of the deeper layers and the proliferation of mesenchymal cells of the intestinal wall of healthy dogs are correlated with age.
