2Anaerobic biodegradation of PAHs in mangrove sediment with amendment of NaHCO_3显示文摘Mangrove sediment is unique in chemical and biological properties. Many of them suffer polycyclic aromatic hydrocarbon(PAH) contamination. However, the study on PAH biological remediation for mangrove sediment is deficient. Enriched PAH-degrading microbial consortium and electron acceptor amendment are considered as two effective measures. Compared to other electron acceptors, the study on CO2, which is used by methanogens, is still seldom. This study investigated the effect of Na HCO3 amendment on the anaerobic biodegradation of four mixed PAHs, namely fluorene(Fl), phenanthrene(Phe),fluoranthene(Flua) and pyrene(Pyr), with or without enriched PAH-degrading microbial consortium in mangrove sediment slurry. The trends of various parameters, including PAH concentrations, microbial population size, electron-transport system activities, electron acceptor and anaerobic gas production were monitored. The results revealed that the inoculation of enriched PAH-degrading consortium had a significant effect with half lives shortened by 7–13 days for 3-ring PAHs and 11–24 days for 4-ring PAHs. 3Stem cell-derived exosomes-an emerging tool for myocardial regeneration显示文摘Cardiovascular diseases(CVDs) continue to represent the number one cause of death and disability in industrialized countries. The most severe form of CVD is acute myocardial infarction(AMI), a devastating disease associated with high mortality and disability. In a substantial proportion of patients who survive AMI, loss of functional cardiomyocytes as a result of ischaemic injury leads to ventricular failure, resulting in significant alteration to quality of life and increased mortality. Therefore, many attempts have been made in recent years to identify new tools for the regeneration of functional cardiomyocytes. Regenerative therapy currently represents the ultimate goal for restoring the function of damaged myocardium by stimulating the regeneration of the infarcted tissue or by providing cellsthat can generate new myocardial tissue to replace the damaged tissue. Stem cells(SCs) have been proposed as a viable therapy option in these cases. However, despite the great enthusiasm at the beginning of the SC era, justified by promising initial results, this therapy has failed to demonstrate a significant benefit in large clinical trials. One interesting finding of SC studies is that exosomes released by mesenchymal SCs(MSCs) are able to enhance the viability of cardiomyocytes after ischaemia/reperfusion injury, suggesting that the beneficial effects of MSCs in the recovery of functional myocardium could be related to their capacity to secrete exosomes. Ten years ago, it was discovered that exosomes have the unique property of transferring miRNA between cells, acting as miRNA nanocarriers. Therefore, exosomebased therapy has recently been proposed as an emerging tool for cardiac regeneration as an alternative to SC therapy in the post-infarction period. 4Effects of root morphology and anatomy on cadmium uptake and translocation in rice(Oryza sativa L.)显示文摘A clear description of the certain mechanisms of cadmium(Cd) uptake and translocation in rice(Oryza sativa L.) may help to reduce Cd accumulation in rice grain. Hydroponic experiments were carried out to determine the effects of cultivation conditions(aerated and stagnant) on the uptake, translocation and subcellular distribution of Cd in relation to the morphology and anatomy of roots in two rice genotypes with different Cd accumulations in grains. Marked differences in morphology and anatomy were observed between these two genotypes under different cultivation conditions. Genotypes with low Cd accumulation in grains tended to develop fewer root tips per root surface area, larger root porosity and more mature apoplastic barriers. The stagnant cultivation condition decreased the number of root tips per root surface area but increased root porosity and accelerated apoplastic barrier formation in root tissues.Correlative Cd uptake studies revealed that rice plants with fewer number of root tips per root surface area reduced root Cd uptake ability, while mature apoplastic barriers increased root Cd retention in cell walls and the symplast. Thus, the fewer number of root tips per root surface area and the earlier formation of mature apoplastic barriers led to lower Cd uptake and translocation. The results indicated that the morphology and anatomy of roots could play important roles in Cd uptake and translocation in rice, and could be influenced by both genotype and cultivation conditions. 5Genetic, metabolic and environmental factors involved in the development of liver cirrhosis in Mexico显示文摘Liver cirrhosis(LC) is a chronic illness caused by inflammatory responses and progressive fibrosis. Globally, the most common causes of chronic liver disease include persistent alcohol abuse, followed by viral hepatitis infections and nonalcoholic fatty liver disease. However, regardless of the etiological factors, the susceptibility and degree of liver damage may be influenced by genetic polymorphisms that are associated with distinct ethnic and cultural backgrounds. Consequently, metabolic genes are influenced by variable environmental lifestyle factors, such as diet, physical inactivity, and emotional stress, which are associated with regional differences among populations. This Topic Highlight will focus on the genetic and environmental factors that may influence the metabolism of alcohol and nutrients in the setting of distinct etiologies of liver disease. The interaction between genes and environment in the current-day admixed population, Mestizo and Native Mexican, will be described. Additionally, genes involved in immune regulation, insulin sensitivity, oxidative stress and extracellular matrix deposition may modulate the degree of severity. In conclusion, LC is a complex disease. The onset, progression, and clinical outcome of LC among the Mexican population are influenced by specific genetic and environmental factors. Among these are an admixed genome with a heterogenic distribution of European, Amerindian and African ancestry; a high score of alcohol consumption; viral infections; a hepatopathogenic diet; and a high prevalence of obesity. 6Relationship of MTHFR gene polymorphisms with renal and cardiac disease显示文摘AIM: To investigate the effects of different methylenetetrahydrofolate reductase(MTHFR) 677C>T gene polymorphism and hyperhomocysteinemia for the development of renal failure and cardiovascular events, which are controversial.METHODS: We challenged the relationship, if any, of MTHFR 677C>T and MTHFR 1298A>C polymorphisms with renal and heart function. The present article is a reappraisal of these concepts, investigating within a larger population, and including a subgroup of dialysis patients, if the two most common MTHFR polymorphisms, C677 T and A1298 C, as homozygous, heterozygous or with a compound heterozygous state, show different association with chronic renal failure requiring hemodialysis. MTHFR polymorphism could be a favorable evolutionary factor, i.e., a protective factor for many ominous conditions, like cancer and renal failure. A similar finding was reported in fatty liver disease in which it is suggested that MTHFR polymorphisms could have maintained and maintain their persistence by an heterozygosis advantage mechanism. We studied a total of 630 Italian Caucasian subject aged 54.60 ± 16.35 years, addressing to the increased hazard of hemodialysis, if any, according to the studied MTHFR genetic polymorphisms. RESULTS: A favorable association with normal renal function of MTHFR polymorphisms, and notably of MTHFR C677 T is present independently of the negative effects of left ventricular hypertrophy, increased IntraRenal arterial Resistance and hyperparathyroidism. 7Immune dysfunction in cirrhosis显示文摘Innate and adaptive immune dysfunction,also referred to as cirrhosis-associated immune dysfunction syndrome,is a major component of cirrhosis,and plays a pivotal role in the pathogenesis of both the acute and chronic worsening of liver function.During the evolution of the disease,acute decompensation events associated with organ failure(s),so-called acute-on chronic liver failure,and chronic decompensation with progression of liver fibrosis and also development of disease specific complications,comprise distinct clinical entities with different immunopathology mechanisms.Enhanced bacterial translocation associated with systemic endotoxemia and increased occurrence of systemic bacterial infections have substantial impacts on both clinical situations.Acute and chronic exposure to bacteria and/or their products,however,can result in variable clinical consequences.The immune status of patients is not constant during the illness;consequently,alterations of the balance between pro-and anti-in-flammatory processes result in very different dynamic courses.In this review we give a detailed overview of acquired immune dysfunction and its consequences for cirrhosis.We demonstrate the substantial influence of inherited innate immune dysfunction on acute and chronic inflammatory processes in cirrhosis caused by the pre-existing acquired immune dysfunction with limited compensatory mechanisms.Moreover,we highlight the current facts and future perspectives of how the assessment of immune dysfunction can assist clinicians in everyday practical decision-making when establishing treatment and care strategies for the patients with end-stage liver disease.Early and efficient recognition of inappropriate performance of the immune system is essential for overcoming complications,delaying progression and reducing mortality.Nora Sipeki Peter Antal-Szalmas Peter L Lakatos Maria Papp 2014World Journal of Gastroenterology2014,20,10:10
8Characterizing the optimal operation of photocatalytic degradation of BDE-209 by nano-sized TiO_2显示文摘Brominated flame retardants have been widely used in industry.There is a rapid growing public concern for their availabilities in the environment.Advanced oxidation process(AOP) is a promising and efficient technology which may be used to remove emerging chemicals such as brominated flame retardants.This study aims at investigating optimal operational conditions for the removal of BDE-209 using nano-scaled titanium(IV) oxide.The residual PBDE congeners after photocatalytical degradation of BDE-209 by TiO 2 were analysed by gas chromatography-mass spectrometry(GC-MS).It was found that the degradability of BDE-209 by TiO 2 was attributed to its photocatalytic activity but not the small size of the particles.The half-life of removing BDE-209 by TiO 2 was 3.05 days under visible light.Tetra-and penta-BDEs were the major degraded products of BDE-209.Optimum conditions for photocatalytical degradation of BDE-209 was found to be at pH 12(93% ± 1%),5,10,20 mg/L(93.0% ± 1.70%,91.6% ± 3.21%,91.9% ± 0.952%,respectively),respectively of humic acid and in the form of anatase/rutile TiO 2(82% ± 3%).Hence,the efficiency of removing BDE-209 can be maximized while being cost effective at the said operating conditions.Ka Lai Chow Yu Bon Man Jin Shu Zheng Yan Liang Nora Fung Yee Tam Ming Hung Wong 2012Journal of Environmental Sciences2012,24,9:8
9Development of a new connection for precast concrete walls subjected to cyclic loading显示文摘The Industrialized Building System(IBS) was recently introduced to minimize the time and cost of project construction.Accordingly, ensuring the integration of the connection of precast components in IBS structures is an important factor that ensures stability of buildings subjected to dynamic loads from earthquakes, vehicles, and machineries.However, structural engineers still lack knowledge on the proper connection and detailed joints of IBS structure construction.Therefore, this study proposes a special precast concrete wall-to-wall connection system for dynamic loads that resists multidirectional imposed loads and reduces vibration effects(PI2014701723).This system is designed to connect two adjacent precast wall panels by using two steel U-shaped channels(i.e., male and female joints).During casting, each joint is adapted for incorporation into a respective wall panel after considering the following conditions: one side of the steel channel opens into the thickness face of the panel; a U-shaped rubber is implemented between the two channels to dissipate the vibration effect; and bolts and nuts are used to create an extension between the two U-shaped male and female steel channels.The developed finite element model of the precast wall is subjected to cyclic loads to evaluate the performance of the proposed connection during an imposed dynamic load.Connection performance is then compared with conventional connections based on the energy dissipation, stress, deformation, and concrete damage in the plastic range.The proposed precast connection is capable of exceeding the energy absorption of precast walls subjected to dynamic load, thereby improving its resistance behavior in all principal directions.Ramin Vaghei Farzad Hejazi Hafez Taheri Mohd Saleh Jaafar Farah Nora Aznieta Abdul Aziz 2017Earthquake Engineering and Engineering Vibration2017,16,1:8
10One fifth of hospitalizations for peptic ulcer-related bleeding are potentially preventable显示文摘AIM:To calculate the proportion of potentially preventable hospitalizations due to peptic ulcer disease(PUD),erosive gastritis(EG)or duodenitis(ED).METHODS:Retrospective cohort study using ICD-10codes to identify all patients with upper gastrointestinal hemorrhage secondary to endoscopically proven PUD,EG or ED during the period from March 2007 to October 2010 in three major metropolitan hospitals in Melbourne,Australia.Patients were divided into'high risk'(those who would benefit from gastroprotection)and'not high risk'groups as defined by established guidelines.Mean Rockall score,transfusion requirement,length of stay,rebleeding rates,need for surgery and in-hospital mortality was compared between'high risk'and'not high risk'groups.Within the'high risk'group,those on gastroprotection and those with no gastroprotection were also compared.RESULTS:Five hundred and seven patients were included for analysis of which 174 were classified as high risk.Median values of complete Rockall Score(5vs 4,P=0.002)and length of stay(5 d vs 4 d,P=0.04)were higher in the high risk group but in-hospital mortality was lower(0.6%vs 3.9%,P=0.03).130out of the 174 patients in the high risk group were not taking recommended gastroprotective therapy prior to hospitalization.Past history of PUD(OR=3.7,P=0.006)and clopidogrel use(OR=3.2,P=0.007)significantly predicted prescription of gastroprotective therapy.Using proton pump inhibitor protection rates of 50%-85%from published studies,an estimation of13%to 22%of the total number of the hospitalizations due to PUD or EG/ED related bleeding may have been preventable.CONCLUSION:Up to one fifth of all hospitalizations for bleeding secondary to PUD or EG/ED are potentially preventable.Ray Boyapati Sim Ye Ong Bei Ye Anuk Kruavit Nora Lee Rhys Vaughan Sanjay Nurkar Peter Gibson Mayur Garg 2014World Journal of Gastroenterology2014,20,30:8

13Stem cell therapy for the treatment of Leydig cell dysfunction in primary hypogonadism显示文摘The production of testosterone occurs within the Leydig cells of the testes. When production fails at this level from either congenital, acquired, or systemic disorders,the result is primary hypogonadism. While numerous testosterone formulations have been developed, none are yet fully capable of replicating the physiological patterns of testosterone secretion. Multiple stem cell therapies to restore androgenic function of the testes are under investigation. Leydig cells derived from bone marrow, adipose tissue, umbilical cord, and the testes have shown promise for future therapy for primary hypogonadism. In particular, the discovery and utilization of a group of progenitor stem cells within the testes, known as stem Leydig cells(SLCs), has led not only to a better understanding of testicular development, but of treatment as well. When combining this with an understanding of the mechanisms that lead to Leydig cell dysfunction, researchers and physicians will be able to develop stem cell therapies that target the specific step in the steroidogenic process that is deficient. The current preclinical studies highlight the complex nature of regenerating this steroidogenic process and the problems remain unresolved. In summary, there appears to be two current directions for stem cell therapy in male primary hypogonadism. The first method involves differentiating adult Leydig cells from stem cells of various origins from bone marrow, adipose, or embryonic sources. The second method involves isolating, identifying, and transplanting stem Leydig cells into testicular tissue. 17Fatty liver disease,an emerging etiology of hepatocellular carcinoma in Argentina显示文摘AIM To investigate any changing trends in the etiologies of hepatocellular carcinoma(HCC) in Argentina during the last years. METHODS A longitudinal cohort study was conducted by 14 regional hospitals starting in 2009 through 2016. All adult patients with newly diagnosed HCC either with pathology or imaging criteria were included. Patients were classified as presenting non-alcoholic fatty liver disease(NAFLD) either by histology or clinically, provided that all other etiologies of liver disease were ruled out, fatty liver was present on abdominal ultrasound and alcohol consumption was excluded. Complete follow-up was assessed in all included subjects since the date of HCC diagnosis until death or last medical visit.RESULTS A total of 708 consecutive adults with HCC were included. Six out of 14 hospitals were liver transplant centers(n = 484). The prevalence of diabetes mellitus was 27.7%. Overall, HCV was the main cause of liver disease related with HCC(37%) including cirrhotic and non-cirrhotic patients, followed by alcoholic liver disease 20.8%, NAFLD 11.4%, cryptogenic 9.6%, HBV 5.4% infection, cholestatic disease and autoimmune hepatitis 2.2%, and other causes 9.9%. A 6-fold increase in the percentage corresponding to NAFLDHCC was detected when the starting year, i.e., 2009 was compared to the last one, i.e., 2015(4.3% vs 25.6%; P < 0.0001). Accordingly, a higher prevalence of diabetes mellitus was present in NAFLD-HCC group 61.7% when compared to other than NAFLD-HCC 23.3%(P < 0.0001). Lower median AFP values at HCC diagnosis were observed between NAFLD-HCC and non-NAFLD groups(6.6 ng/m L vs 26 ng/m L; P = 0.02). 18Anti-irritable Bowel Syndrome Syrup Improves Constipation-Predominant Irritable Bowel Syndrome:A Randomized,Placebo-Controlled Trial显示文摘Objective To evaluate the efficacy and safety of administration of the formulated Persian herbal syrup on improving the symptoms of patients with constipation-predominant irritable bowel syndrome(IBS-C).Methods This study was conducted in 70 patients with IBS-C,who were recruited from 3 medical centers in Mashhad,Iran,from November 2017 to August 2018.Seventy patients were randomly assigned to 2 groups including treatment and placebo groups by block randomization,35 cases in each group.Patients in the treatment group received 15 mL of anti-IBS syrup,thrice daily for 6 weeks and followed up for 4 weeks.Placebo syrup was also prepared through similar instruction,BP syrup without plant extract was used.Primary outcome induding IBS Symptom Severity Scale(IBS-SSS)questionnaire and secondary outcomes in terms of Hospital Anxiety and Depression(HADS)questionnaires,the Bristol Stool Form Scale(BSFS)were completed and evaluated at weeks 6 and 10,respectively.Safety indices were collected at the end of the treatment and Common Terminology Criteria for Adverse Events v4.0(CTCAE)was used to evaluate the adverse events.Results The response to treatment was 84.4%(27/32)in the treatment group and 46.4%(13/28)in the placebo group,respectively(P=0.002).Compared with pre-treatment,a significant decrease was found on the IBS-SSS and BSFS scores after 6-week intervention in both groups(P<0.001).Moreover,IBS-SSS and BSFS scores in the treatment group were lower than the placebo group after the intervention(P=0.041).There was no significant difference in the anxiety and depression scores after treatment in both groups(P>0.05).Side effects reported in the treatment group included 2 cases of headache during the first week of the onset of the treatment,1 case of drowsiness,1 case of increase in menstrual bleeding,which did not result in discontinuation of the treatment.In the placebo group,1 case of exacerbation of the disease was reported.Conclusions Anti-IBS syrup significantly reduced the severity of IBS symptoms compared to placebo.However,there was a need for further investigation regarding the anxiety and depression scores.(Registration No.IRCT2017061034446N1).Hamide Khorram Pazhouh Seyyd Musa al-Reza Hosseini Ali Taghipour Shokouhsadat Hamedi Mohammadreza Noras 2020Chinese Journal of Integrative Medicine2020,26,10:4

20Aminoguanidine impedes human pancreatic tumor growth and metastasis development in nude mice显示文摘AIM:To study the action of aminoguanidine on pancreatic cancer xenografts in relation to cell proliferation,apoptosis,redox status and vascularization.METHODS:Xenografts of PANC-1 cells were developed in nude mice. The animals were separated into two groups:control and aminoguanidine treated. Tumor growth,survival and appearance of metastases were determined in vivo in both groups. Tumors were excised and ex vivo histochemical studies were performed. Cell growth was assessed by Ki-67 expression. Apoptosis was studied by intratumoral expression of B cell lymphoma-2 protein (Bcl-2) family proteins and Terminal deoxynucleotidyl transferase biotin-dUTP Nick End Labeling (Tunel). Redox status was evaluated by the expression of endothelial nitric oxide synthase (eNOS),catalase,copper-zinc superoxide dismutase (CuZnSOD),manganese superoxide dismutase (MnSOD) and glutathione peroxidase (GPx). Finally,vascularization was determined by Massons trichromic staining,and by VEGF and CD34 expression.RESULTS:Tumor volumes after 32 d of treatment by aminoguanidine (AG) were significantly lower than in control mice (P < 0.01). Median survival of AG mice was significantly greater than control animals (P < 0.01). The appearance of both homolateral and contralateral palpable metastases was significantly delayed in AG group.
