1Gut microbiota imbalance and colorectal cancer显示文摘The gut microbiota acts as a real organ. The symbiotic interactions between resident micro-organisms and the digestive tract highly contribute to maintain the gut homeostasis. However, alterations to the microbiome caused by environmental changes(e.g., infection, diet and/or lifestyle) can disturb this symbiotic relationship and promote disease, such as inflammatory bowel diseases and cancer. Colorectal cancer is a complex association of tumoral cells, non-neoplastic cells and a large amount of micro-organisms, and the involvement of the microbiota in colorectal carcinogenesis is becoming increasingly clear. Indeed, many changes in the bacterial composition of the gut microbiota have been reported in colorectal cancer, suggesting a major role of dysbiosis in colorectal carcinogenesis. Some bacterial species have been identified and suspected to play a role in colorectal carcinogenesis, such as Streptococcus bovis, Helicobacter pylori, Bacteroides fragilis, Enterococcus faecalis, Clostridium septicum, Fusobacterium spp. and Escherichia coli. The potential pro-carcinogenic effects of these bacteria are now better understood. In this review, we discuss the possible links between the bacterial microbiota and colorectal carcinogenesis, focusing on dysbiosis and the potential pro-carcinogenic properties of bacteria, such as genotoxicity and other virulence factors, inflammation, host defenses modulation, bacterial derived metabolism, oxidative stress and anti-oxidative defenses modulation. 2Colon cancer-associated B2 Escherichia coli colonize gut mucosa and promote cell proliferation显示文摘AIM:To provide further insight into the characterization of mucosa-associated Escherichia coli(E.coli)isolated from the colonic mucosa of cancer patients.METHODS:Phylogroups and the presence of cyclomodulin-encoding genes of mucosa-associated E.coli from colon cancer and diverticulosis specimens weredetermined by PCR.Adhesion and invasion experiments were performed with I-407 intestinal epithelial cells using gentamicin protection assay.Carcinoembryonic antigen-related cell adhesion molecule 6(CEACAM6)expression in T84 intestinal epithelial cells was measured by enzyme-linked immunosorbent assay and by Western Blot.Gut colonization,inflammation and procarcinogenic potential were assessed in a chronic infection model using CEABAC10 transgenic mice.Cell proliferation was analyzed by real-time mRNA quantification of PCNA and immunohistochemistry staining of Ki67.RESULTS:Analysis of mucosa-associated E.coli from colon cancer and diverticulosis specimens showed that whatever the origin of the E.coli strains,86%of cyclomodulin-positive E.coli belonged to B2 phylogroup and most harbored polyketide synthase(pks)island,which encodes colibactin,and/or cytotoxic necrotizing factor(cnf)genes.In vitro assays using I-407 intestinal epithelial cells revealed that mucosa-associated B2 E.coli strains were poorly adherent and invasive.However,mucosa-associated B2 E.coli similarly to Crohn’s disease-associated E.coli are able to induce CEACAM6expression in T84 intestinal epithelial cells.In addition,in vivo experiments using a chronic infection model of CEACAM6 expressing mice showed that B2 E.coli strain11G5 isolated from colon cancer is able to highly persist in the gut,and to induce colon inflammation,epithelial damages and cell proliferation.CONCLUSION:In conclusion,these data bring new insights into the ability of E.coli isolated from patients with colon cancer to establish persistent colonization,exacerbate inflammation and trigger carcinogenesis.Jennifer Raisch Emmanuel Buc Mathilde Bonnet Pierre Sauvanet Emilie Vazeille Amélie de Vallée Pierre Déchelotte Claude Darcha Denis Pezet Richard Bonnet Marie-Agnès Bringer Arlette Darfeuille-Michaud 2014World Journal of Gastroenterology2014,20,21:12
3脊髓灰质炎灭活疫苗基础免疫效果观察显示文摘目的考察灭活脊髓灰质炎疫苗(inactivated poliomyelitis vaccine,IPV)在中国婴儿中的免疫效果,并与目前常规使用的口服脊髓灰质炎减毒活疫苗(oral poliomyelitis vaccine,OPV)进行比较。方法对2个月龄婴儿采用组群随机法分为2个组,每组208名,分别接种IPV和OPV,并采集免疫前后血清。采用微量中和方法,对血清中抗脊髓灰质炎病毒3个型的中和抗体进行测定,对于抗体保护率比较采用χ^2检验进行统计学处理。抗体滴度进行对数转换后采用Z检验进行比较,所有统计学检验以P〈0.05来确定差异是否具有统计学意义。结果婴儿经初次免疫后,IPV组I、Ⅱ、Ⅲ型病毒中和抗体保护率分别为100.0%(186/186)、97.3%(181/186)、98.9%(184/186),几何平均滴度(GMT)分别为151.2、86.7、211.3,OPV组I、Ⅱ、Ⅲ型病毒中和抗体保护率分别为97.4%(188/193)、100.0%(193/193)、95.3%(184/193),GMT分别为1089.5、538.2、203.7。两组中I、Ⅱ型的保护率差异没有统计学意义(I、Ⅱ型分别为χ^2I=2.991,P=0.084;χ^2Ⅱ=3.512,P=0.061),但Ⅲ型中差异有统计学意义(χ^2Ⅲ=4.143,P=0.042)。IPV组I、Ⅱ型抗体几何平均滴度低于OPV疫苗,差异有统计学意义(ZI=12.537,P=0.000;ZⅡ=13.415,P=0.000),而Ⅲ型抗体几何平均滴度差异没有统计学意义(ZⅢ=0.067,P=0.947)。结论经基础免疫后IPV在婴儿中免疫效果良好,和OPV相比,IPV组I、Ⅱ型保护率与OPV相当,Ⅲ型高于OPV组。IPV组I、Ⅱ型抗体几何平均滴度低于OPV疫苗,而Ⅲ型抗体几何平均滴度与OPV组相当。李长贵 英志芳 王剑锋 方捍华 李艳萍 李荣成 Marie-Claude Bonnet 张燕平 2009中华预防医学杂志2009,43,6:11
4Association of colorectal cancer with pathogenic Escherichia coli: Focus on mechanisms using optical imaging显示文摘AIM: To investigate the molecular or cellular mechanisms related to the infection of epithelial colonic mucosa by pks-positive Escherichia coli(E. coli) using optical imaging.METHODS: We choose to evaluate the tumor metabolic activity using a fluorodeoxyglucose analogue as 2-deoxyglucosone fluorescent probes and to correlate it with tumoral volume(mm^3). Inflammation measuring myeloperoxidase(MPO) activity and reactive oxygen species production was monitored by a bioluminescent(BLI) inflammation probe and related to histological examination and MPO levels by enzyme-linked immunosorbent assay(ELISA) on tumor specimens. The detection and quantitation of these two signals were validated on a xenograft model of human colon adenocarcinoma epithelial cells(HCT116) in nude mice infected with a pks-positive E. coli. The inflammatory BLI signal was validated intra-digestively in the colitisCEABAC10 DSS models, which mimicked Crohn's disease. RESULTS: Using a 2-deoxyglucosone fluorescent probe, we observed a high and specific HCT116 tumor uptake in correlation with tumoral volume(P = 0.0036). Using the inflammation probe targeting MPO, we detected a rapid systemic elimination and a significant increase of the BLI signal in the pks-positive E. coli-infected HCT116 xenograft group(P < 0.005). ELISA confirmed that MPO levels were significantly higher(1556 ± 313.6 vs 234.6 ± 121.6 ng/m L P = 0.001) in xenografts infected with the pathogenic E. coli strain. Moreover, histological examination of tumor samples confirmed massive infiltration of pks-positive E. coli-infected HCT116 tumors by inflammatory cells compared to the uninfected group. These data showed that infection with the pathogenic E. coli strain enhanced inflammation and ROS production in tumors before tumor growth. 5Microbial markers in colorectal cancer detection and/or prognosis显示文摘Colorectal cancer(CRC) is the second leading cause of cancer worldwide. CRC is still associated with a poor prognosis among patients with advanced disease. On the contrary, due to its slow progression from detectable precancerous lesions, the prognosis for patients with early stages of CRC is encouraging. While most robust methods are invasive and costly, actual patient-friendly screening methods for CRC suffer of lack of sensitivity and specificity. Therefore, the development of sensitive, non-invasive and cost-effective methods for CRC detection and prognosis are necessary for increasing the chances of a cure. Beyond its beneficial functions for the host, increasing evidence suggests that the intestinal microbiota is a key factor associated with carcinogenesis. Many clinical studies have reported a disruption in the gut microbiota balance and an alteration in the faecal metabolome of CRC patients, suggesting the potential use of a microbialbased test as a non-invasive diagnostic and/or prognostic tool for CRC screening. This review aims to discuss the microbial signatures associated with CRC known to date, including dysbiosis and faecal metabolome alterations, andthe potential use of microbial variation markers for noninvasive early diagnosis and/or prognostic assessment of CRC and advanced adenomas. 10Presence of three dengue serotypes in Ouagadougou(Burkina Faso):research and public health implications显示文摘Background:The significant malaria burden in Africa has often eclipsed other febrile illnesses.Burkina Faso’s first dengue epidemic occurred in 1925 and the most recent in 2013.Yet there is still very little known about dengue prevalence,its vector proliferation,and its poverty and equity impacts.Methods:An exploratory cross-sectional survey was performed from December 2013 to January 2014.Six primary healthcare centers in Ouagadougou were selected based on previously reported presence of Flavivirus.All patients consulting with fever or having had fever within the previous week and with a negative rapid diagnostic test(RDT)for malaria were invited to participate.Sociodemographic data,healthcare use and expenses,mobility,health-related status,and vector control practices were captured using a questionnaire.Blood samples of every eligible subject were obtained through finger pricks during the survey for dengue RDT using SD BIOLINE Dengue Duo(NS1Ag and IgG/IgM)®and to obtain blood spots for reverse transcription polymerase chain reaction(RT-PCR)analysis.In a sample of randomly selected yards and those of patients,potential Aedes breeding sites were found and described.Larvae were collected and brought to the laboratory to monitor the emergence of adults and identify the species.Results:Of the 379 subjects,8.7%(33/379)had positive RDTs for dengue.Following the 2009 WHO classification,38.3%(145/379)had presumptive,probable,or confirmed dengue,based on either clinical symptoms or laboratory testing.Of 60 samples tested by RT-PCR(33 from the positive tests and 27 from the subsample of negatives),15 were positive.The serotypes observed were DENV2,DENV3,and DENV4.Odds of dengue infection in 15-to-20-year-olds and persons over 50 years were 4.0(CI 95%:1.0–15.6)and 7.7(CI 95%:1.6–37.1)times higher,respectively,than in children under five.Average total spending for a dengue episode was 13771 FCFA[1300–67300 FCFA](1$US=478 FCFA).On average,2.6 breeding sites were found per yard.Potential Aedes breeding sites were found near 71.4%(21/28)of patients,but no adult Aedes were found.The most frequently identified potential breeding sites were water storage containers(45.2%).Most specimens collected in yards were Culex(97.9%).Conclusions:The scientific community,public health authorities,and health workers should consider dengue as a possible cause of febrile illness in Burkina Faso.Valéry Ridde Isabelle Agier Emmanuel Bonnet Mabel Carabali Kounbobr Roch Dabiré Florence Fournet Antarou Ly Ivlabèhiré Bertrand Meda Beatriz Parra 2016Infectious Diseases of Poverty2016,5,1:2
