1Divergence in the transcriptional landscape between low temperature and freeze shock in cultivated grapevine (Vitis vinifera)显示文摘Low-temperature stresses limit the sustainability and productivity of grapevines when early spring frosts damage young grapevine leaves.Spring conditions often expose grapevines to low,but not damaging,chilling temperatures and these temperatures have been shown to increase freeze resistance in other model systems.In this study,we examined whole-transcriptome gene expression patterns of young leaf tissue from cuttings of five different grapevine cultivars,exposed to chill and freeze shock,in order to understand the underlying transcriptional landscape associated with cold stress response.No visible damage was observed when grapevine leaves were exposed to chilling temperatures while freeze temperatures resulted in variable damage in all cultivars.Significant differences in gene expression were observed between warm control conditions and all types of cold stress.Exposure to chill stress(4°C)versus freezing stress(−3°C)resulted in very different patterns of gene expression and enriched pathway responses.Genes from the ethylene signaling,ABA signaling,the AP2/ERF,WRKY,and NAC transcription factor families,and starch/sucrose/galactose pathways were among the most commonly observed to be differentially regulated.Preconditioning leaves to chill temperatures prior to freezing temperatures resulted in slight buffering of gene expression responses,suggesting that differences between chill and freeze shock perception complicates identification of candidate genes for cold resistance in grapevine.Overall,the transcriptional landscape contrasts observed between low temperature and freezing stresses demonstrate very different activation of candidate pathways impacting grapevine cold response.Jason P.Londo Alisson P.Kovaleski Jacquelyn A.Lillis 2018Horticulture Research2018,5,1:2
2Segregation-assisted spinodal and transient spinodal phase separation at grain boundaries显示文摘Segregation to grain boundaries affects their cohesion,corrosion,and embrittlement and plays a critical role in heterogeneous nucleation.In order to quantitatively study segregation and low-dimensional phase separation at grain boundaries,here,we apply a density-based phase-field model.The current model describes the grain-boundary thermodynamic properties based on available bulk thermodynamic data,while the grain-boundary-density profile is obtained using atomistic simulations.To benchmark the performance of the model,Mn grain-boundary segregation in the Fe–Mn system is studied.3D simulation results are compared against atom probe tomography measurements conducted for three alloy compositions.We show that a continuous increase in the alloy composition results in a discontinuous jump in the segregation isotherm.The jump corresponds to a spinodal phase separation at grain boundary.For alloy compositions above the jump,we reveal an interfacial transient spinodal phase separation.The transient spinodal phenomenon opens opportunities for knowledge-based microstructure design through the chemical manipulation of grain boundaries.The proposed density-based model provides a powerful tool to study thermodynamics and kinetics of segregation and phase changes at grain boundaries.Reza Darvishi Kamachali Alisson Kwiatkowski da Silva Eunan McEniry Dirk Ponge Baptiste Gault Jörg Neugebauer Dierk Raabe 2020npj Computational Materials2020,,1:2

16Benfotiamine supplementation prevents oxidative stress in anterior tibialis muscle and heart显示文摘Objective: This study aimed to evaluate the influence of oral supplementation with benfotiamine on oxidative stress in the liver, heart and muscles of endurance-trained mice.Methods: Twenty-five male BALB/c mice were allocated to the following treatment groups: standard diet and sedentary activity(Sta–Sed), benfotiamine–supplemented diet and sedentary activity(Ben–Sed),standard diet and training activity(Sta–Tr) and benfotiamine–supplemented diet and training activity(Ben–Tr). The training comprised 6 weeks of endurance swimming training. The concentration of thiobarbituric acid reactive substances(TBARS), carbonylated proteins, total thiols and non-protein thiols was analyzed in the liver, heart and tibialis anterior muscle.Results: In the muscle, TBARS concentration in the Sta–Sed group was higher than that in other groups;in the heart, TBARS concentration in the Sta–Sed and Ben–Tr groups was higher than that in the Ben–Sed group. The carbonyl content of the muscle tissues was higher in the Sta–Sed group than in both supplemented groups. In liver, the carbonyl content was lower in the Ben–Sed group than in the Sta–Sed group.The level of total thiols was lower in the Ben–Sed group than in the Sta–Tr group. In the heart, the level of total thiols was higher in the Ben–Sed group than in the Ben–Tr group. 17Uncertainty in sap flow of Brazilian mahogany determined by the heat ratio method显示文摘The tropical arboreal species Brazilian mahogany(Swietenia macrophylla) is very important economically and ecologically,for which understanding ecophysiological variables such as sap flow will improve understanding of the species and its cultivation.This paper aims to measure uncertainties(U) involved in the application of the heat ratio method for determining sap flow in Brazilian mahogany using sets of heating probes and thermometers installed on plants of 18 months of age,cultivated in Yellow Latosol,under a weighing lysimeter and located in a protected environment.The uncertainty in sap flow was calculated as the combination of uncertainty in the thermal diffusivity(U_(k)),conductive section(U_(Sc)) and corrected sap velocity(U_(Vc)).U_(k) had greater weight in determining the flow of sap in Brazilian mahogany,when compared to U_(Sc) and U_(Vc).The thermal diffusivity during the cycle,or period evaluated,must be adjusted to improve the accuracy of the heat ratio method because the sap flow overestimated transpiration by 15.0%.When soil water was optimal In addition,the vapor pressure deficit linearly and indirectly influenced the SF with a difference of 14.6%.Alisson Macendo Amaral Frederico Antonio Loureiro Soares Lucas Melo Vellame Marconi Batista Teixeira 2021Journal of Forestry Research2021,32,4:0
18Surface Finish Evaluation of AISI P100 Steel after Grinding with MQL Technique with Different Flow Rates显示文摘Grinding operation is a finishing process often employed when high precision and narrow geometric tolerances are required.These requirements can be achieved only if cutting conditions are properly selected,especially the cooling-lubrication technique.In general,grinding is performed in presence of cutting fluid,however,due to the environmental impacts and costs of the conventional coolant delivery technique(flow rates from 4 L/min to 300 L/min),alternative cooling-lubrication techniques have been developed on restriction of the coolants use.Among the several techniques,MQL(minimum quantity of lubricant)technique has received special attention from machining users because of its advantages in terms of surface quality of workpiece and drastic reduction in use of coolant.In this context,this paper evaluated the performance of the MQL technique as compared to the flood coolant in peripheral surface grinding of AISI P100(VP100)steel with conventional aluminum oxide grinding wheel in relation to the surface roughness(Ra and Rz).Input parameters tested were equivalent chip thickness(0.09μm,0.18μm and 0.27μm)and flow rate of the cutting fluid(60 mL/h,150 mL/h and 240 mL/h)of the MQL system.Results showed that the grinding with MQL technique provided lower surface roughness values compared to conventional flood cooling,especially when machining under the intermediary cutting conditions.Also,with exception of heq of 0.09 m,the MQL technique resulted in lower values of Rz parameter as compared to the conventional coolant technique,regardless of the flow rate tested.Bruno Souza Abrão Mayara Fernanda Pereira Cleudes Guimarães Lurian Souza Vieira da Silva Rodrigo deSouza Ruzzi Rosemar Batista da Silva Eduardo Carlos Bianchi Alisson Rocha Machado 2020Journal of Mechanics Engineering and Automation2020,10,2:0
19Hydrological variability and long-term floristic-structural modifications in different habitats of a tropical semi-deciduous forest显示文摘Shifts in hydrological regimes alter river flow rates and flood pulses, decrease environmental heterogeneity and the floristic-structural complexity of associated plant communities. We tested the hypothesis that drought events affect plant community composition and structure at a small-scale within a riparian fragment towards a reduction in floristic-structural complexity. The tree community was sampled in three habitats (wet, transitional and dry) and monitored in seven inventories carried out between 1991 and 2018. Hydrological variations were evaluated through annual rainfalls, river flow rates and water level data. The species richness and the detrended correspondence analysis axes were used to characterise the temporal modifications in floristic composition. Community structure was described in terms of biomass: accumulated, growth of survivors, mortality and recruitment. Generalised linear mixed models were fitted to evaluate the effects of time and environment in community. It was concluded that the climate has become drier in recent years due to declining precipitation that has affected flow rates and water levels. The floristic-structural complexity of the study fragment was maintained during the monitoring period.
