| 1 | Pancreatic ductal adenocarcinoma: Risk factors, screening, and early detection显示文摘Pancreatic cancer is the fourth most common cause of cancer-related deaths in the United States, with over 38000 deaths in 2013. The opportunity to detect pancreatic cancer while it is still curable is dependent on our ability to identify and screen high-risk populations before their symptoms arise. Risk factors for developing pancreatic cancer include multiple genetic syndromes as well as modifiable risk factors. Genetic conditions include hereditary breast and ovarian cancer syndrome, Lynch Syndrome, familial adenomatous polyposis, Peutz-Jeghers Syndrome, familial atypical multiple mole melanoma syndrome, hereditary pancreatitis, cystic fibrosis, and ataxia-telangiectasia; having a genetic predisposition can raise the risk of developing pancreatic cancer up to 132-fold over the general population. Modifiable risk factors, which include tobacco exposure, alcohol use, chronic pancreatitis, diet, obesity, diabetes mellitus, as well as certain abdominal surgeries and infections, have also been shown to increase the risk of pancreatic cancer development. Several largevolume centers have initiated such screening protocols, and consensus-based guidelines for screening high-riskgroups have recently been published. | 2 | Autoantibody to MOG suggests two distinct clinical subtypes of NMOSD显示文摘We characterized a unique group of patients with neuromyelitis optica spectrum disorder(NMOSD) who carried autoantibodies of aquaporin-4(AQP4) and myelin-oligodendrocyte glycoprotein(MOG). Among the 125 NMOSD patients, 10(8.0%) were AQP4- and MOG-ab double positive, and 14(11.2%) were MOG-ab single positive. The double-positive patients had a multiphase disease course with a high annual relapse rate(P=0.0431), and severe residual disability(P<0.0001). Of the double-positive patients, 70% had MS-like brain lesions, more severe edematous, multifocal regions on spinal magnetic resonance imaging(MRI), pronounced decreases of retinal nerve fiber layer thickness and atrophy of optic nerves. In contrast, patients with only MOG-ab had a higher ratio of monophasic disease course and mild residual disability. Spinal cord MRI illustrated multifocal cord lesions with mild edema, and brain MRIs showed more lesions around lateral ventricles. NMOSD patients carrying both autoantibodies to AQP4 and MOG existed and exhibited combined features of prototypic NMO and relapsing-remitting form of MS, whereas NMOSD with antibodies to MOG only exhibited an 'intermediate' phenotype between NMOSD and MS. | 4 | Comprehensive and innovative techniques for livertransplantation in rats: A surgical guide显示文摘AIM: To investigate our learning curves of orthotopic liver transplantation (OLT) in rats and the most important factor for successful surgery. METHODS: We describe the surgical procedures for our rat OLT model, and determined the operator learning curves. The various factors that contributed to successful surgery were determined. The most important surgical factors were evaluated between successful and unsuccessful surgeries.RESULTS: Learning curve data indicated that 50 cases were required for operator training to start a study. Operative time, blood loss, warm ischemic time, anhepatic phase, unstable systemic hemodynamic state, and body temperature after surgery significantly affected surgery success by univariate analysis, while the anhepatic phase was the most critical factor for success by multivariate analysis. CONCLUSION: OLT in rats is the only liver transplantation model that provides clinically relevant and reliable results. | 6 | Responses of an old-field plant community to interacting factors of elevated[CO_(2)],warming,and soil moisture显示文摘Aims The direct effects of atmospheric and climatic change factors—atmospheric[CO_(2)],air temperature and changes in precipitation—can shape plant community composition and alter ecosystem function.It is essential to understand how these factors interact to make better predictions about how ecosystems may respond to change.We investigated the direct and interactive effects of[CO_(2)],warming and altered soil moisture in open-top chambers(OTCs)enclosing a constructed old-field community to test how these factors shape plant communities.Materials and methods The experimental facility in Oak Ridge,TN,USA,made use of 4-m diameter OTCs and rain shelters to manipulate[CO_(2)](ambient,ambient+300 ppm),air temperature(ambient,ambient+3.5C)and soil moisture(wet,dry).The plant communities within the chambers comprised seven common old-field species,including grasses,forbs and legumes.We tracked foliar cover for each species and calculated community richness,evenness and diversity from 2003 to 2005.Important findings This work resulted in three main findings:(1)warming had speciesspecific effects on foliar cover that varied through time and were altered by soil moisture treatments;(2)[CO_(2)]had little effect on individual species or the community;(3)diversity,evenness and richness were influenced most by soil moisture,primarily reflecting the response of one dominant species.We conclude that individualistic species responses to atmospheric and climatic change can alter community composition and that plant populations and communities should be considered as part of analyses of terrestrial ecosystem response to climate change.However,prediction of plant community responses may be difficult given interactions between factors and changes in response through time. | E.Cayenne Engel Jake F.Weltzin Richard J.Norby Aimee T.Classen | 2009 | Journal of Plant Ecology2009,2,1: | 8 |
| 7 | Novel interactions of mitochondria and reactive oxygen/nitrogen species in alcohol mediated liver disease显示文摘Mitochondrial dysfunction is known to be a contributing factor to a number of diseases including chronic alcohol induced liver injury. While there is a detailed understanding of the metabolic pathways and proteins of the liver mitochondrion, little is known regarding how changes in the mitochondrial proteome may contribute to the development of hepatic pathologies. Emerging evidence indicates that reactive oxygen and nitrogen species disrupt mitochondrial function through post-translational modifications to the mitochondrial proteome. Indeed, various new affinity labeling reagents are available to test the hypothesis that post-translational modification of proteins by reactive species contributes to mitochondrial dysfunction and alcoholic fatty liver disease. Specialized proteomic techniques are also now available, which allow for identification of defects in the assembly of multi-protein complexes in mitochondria and the resolution of the highly hydrophobic proteins of the inner membrane. | 8 | The effects of plant density of Melastoma malabathricum on the erosion rate of slope soil at different slope orientations显示文摘Malaysia's cut slopes,especially for road lines accommodation,are prone to erosions and landslides.These problems mainly occur due to lack of vegetation cover and strong erosive forces.In addition,the topography factors have also become a major factor affecting soil degradation.Thus,this study is aimed at determining the effects of planting density of a selected species,namely Melastoma malabathricum;one,two,and three seedlings per box,on the erosion rate at selected slopes of different orientation(morning and evening sun) at the Guthrie Corridor Expressway,Selangor.In six months of observation,treatment with three seedlings/box on the morning sun slope showed a lower erosion rate by 69.2%than those with the same treatment on the evening sun slope.In addition,the treatment of the three seedlings recorded at month six(final observation) had the highest reduction of soil saturation level(STL),by 23.6%.Furthermore,the physiological values of the species studied,grown on the morning sun slope,were higher in terms of stomatal conductance and photosynthetic rate by 12.1%and 15.8%(three seedlings/box),respectively.The overall results showed that plant density was inversely related to the STL and erosion rate on the slope.In conclusion,a planting density of three seedlings/box and morning sun orientation gave positive effects on the plant's physiological performance of the slope,reducing the STL,as well as alleviating the erosion rate of slope soils. | Aimee Halim Osman Normaniza | 2015 | International Journal of Sediment Research2015,30,2: | 6 |
| 9 | MicroRNA in pancreatic ductal adenocarcinoma and its precursor lesions显示文摘Pancreatic ductal adenocarcinoma(PDAC) is the 4^(th) deadliest cancer in the United States, due to its aggressive nature, late detection, and resistance to chemotherapy. The majority of PDAC develops from 3 precursor lesions, pancreatic intraepithelial lesions(PanIN), intraductual papillary mucinous neoplasm(IPMN), and mucinous cystic neoplasm. Early detection and surgical resection can increase PDAC 5-year survival rate from 6% for Stage Ⅳ to 50% for Stage Ⅰ. To date, there are no reliable biomarkers that can detect PDAC. MicroRNAs(miRNA) are small noncoding RNAs(18-25 nucleotides) that regulate gene expression by affecting translation of messenger RNA(mRNA). A large body of evidence suggests that miRNAs are dysregulated in various types of cancers. MiRNA has been profiled as a potential biomarker in pancreatic tumor tissue, blood, cyst fluid, stool, and saliva. Four mi RNA biomarkers(miR-21, miR-155, miR-196, and miR-210) have been consistently dysregulated in PDAC. MiR-21, miR-155, and miR-196 have also been dysregulated in IPMN and PanIN lesions suggesting their use as early biomarkers of this disease. | 10 | Macrophages are the primary effector cells in IL-7-induced arthritis显示文摘Synovial macrophages are crucial in the development of joint inflammation and bone damage;however, the pathways that controlmacrophage remodeling in inflammatory M1 cells or bone-eroding osteoclasts are not fully understood. We determined thatelevated IL-7R/CD127 expression is the hallmark of rheumatoid arthritis (RA) M1 macrophages and that these cells are highlyresponsive to interleukin-7 (IL-7)-driven osteoclastogenesis. We established that lipopolysaccharide (LPS), interferon-γ (IFNγ), andtumor necrosis factor-α (TNFα), the classic M1 macrophage mediators, enhance IL-7R expression in RA and murine macrophages.The local expression of IL-7 provokes arthritis, predominantly through escalating the number of F480^(+)iNOS^(+) cells rather than CD3^(+)T cells. Ectopic LPS injection stabilizes IL-7-induced arthritis by increasing myeloid IL-7R expression, in part via IFNγ induction.Hence, in RAG−/− mice, IL-7-mediated arthritis is suppressed because of the reduction in myeloid IL-7R expression due to the lackof IFNγ. Moreover, the amelioration of IL-7-induced arthritis by anti-TNF therapy is due to a decrease in the number of cells in theunique F480^(+)iNOS^(+)IL-7R^(+)CCL5^(+) subset, with no impact on the F480^(+)Arginase^(+) cell or CD3^(+) T cell frequency. Consistent with thepreclinical findings, the findings of a phase 4 study performed with RA patients following 6 months of anti-TNF therapy revealedthat IL-7R expression was reduced without affecting the levels of IL-7. | 11 | Evidence based review of type 2 diabetes prevention and management in low and middle income countries显示文摘AIM:To identify the newest approaches to type 2diabetes(T2DM)prevention and control in the developingworld context.METHODS:We conducted a systematic review of published studies of diabetes prevention and control programs in low and middle-income countries,as defined by the World Bank.We searched Pub Med using Medical Subject Headings terms.Studies needed to satisfy four criteria:(1)Must be experimental;(2)Must include patients with T2DM or focusing on prevention of T2DM;(3)Must have a lifestyle intervention component;(4)Must be written in English;and(5)Must have measurable outcomes related to diabetes.RESULTS:A total of 66 studies from 20 developing countries were gathered with publication dates through September 2014.India contributed the largest number of trials(11/66).Of the total 66 studies reviewed,all but 3 studies reported evidence of favorable outcomes in the prevention and control of type 2 diabetes.The overwhelming majority of studies reported on diabetes management(56/66),and among these more than half were structured lifestyle education programs.The evidence suggests that lifestyle education led by allied health professionals(nurses,pharmacists)were as effective as those led by physicians or a team of clinicians.The remaining diabetes management interventions focused on diet or exercise,but the evidence to recommend one approach over another was weak.CONCLUSION:Large experimental diabetes prevention/control studies of dietary and exercise interventions are lacking particularly those that consider quality rather than quantity of carbohydrates and alternative exercise. | Aimee Afable Nidhi Shree Karingula | 2016 | World Journal of Diabetes2016,7,10: | 3 |

| 13 | Strategies for PET imaging of the receptor for advanced glycation endproducts(RAGE)显示文摘The implication of the receptor for advanced glycation end-products(RAGE)in numerous diseases and neurodegenerative disorders makes it interesting both as a therapeutic target and as an inflammatory biomarker.In the context of investigating RAGE as a biomarker,there is interest in developing radiotracers that will enable quantification of RAGE using positron emission tomography(PET)imaging.We have synthesized potential small molecule radiotracers for both the intracellular([18F]InRAGER)and extracellular([18F]RAGER)domains of RAGE.Herein we report preclinical evaluation of both using in vitro(lead panel screens)and in vivo(rodent and nonhuman primate PET imaging)methods.Both radiotracers have high affinity for RAGE and show good brain uptake,but suffer from off-target binding.The source of the off-target PET signal is not attributable to binding to melatonin receptors,but remains unexplained.We have also investigated use of lipopolysaccharide(LPS)-treated mice as a possible animal model with upregulated RAGE for evaluation of new imaging agents.Immunoreactivity of the mouse brain sections revealed increases in RAGE in the male cohorts,but no difference in the female groups.However,it proves challenging to quantify the changes in RAGE due to off-target binding of the radiotracers.Nevertheless,they are appropriate lead scaffolds for future development of 2nd generation RAGE PET radiotracers because of their high affinity for the receptor and good CNS penetration. | Lindsey R.Drake Allen F.Brooks Jenelle Stauff Phillip S.Sherman Janna Arteaga Robert A.Koeppe Aimee Reed Timothy J.Montavon Marc B.Skaddan Peter J.H.Scott | 2020 | Journal of Pharmaceutical Analysis2020,10,5: | 2 |

Fe K-edge X-ray absorption spectroscopy(XAS) was employed to study the local structure of Fe atoms for samples before and after doping Dy, Tb or Gd. It is found that the bond lengths and coordination numbers are changed. Thus, the exchange interaction between Fe atoms increases with Dy, Tb or Gd doping, resulting in the improvement of Curie temperature of NdFeB permanent magnets. The doping effect is proven by experimental measurement of the magnetic properties.
