| 1 | Notch signaling controls chondrocyte hypertrophy via indirect regulation of Sox9显示文摘RBPjk-dependent Notch signaling regulates both the onset of chondrocyte hypertrophy and the progression to terminal chondrocyte maturation during endochondral ossification. It has been suggested that Notch signaling can regulate Sox9 transcription, although how this occurs at the molecular level in chondrocytes and whether this transcriptional regulation mediates Notch control of chondrocyte hypertrophy and cartilage development is unknown or controversial. Here we have provided conclusive genetic evidence linking RBPjk-dependent Notch signaling to the regulation of Sox9 expression and chondrocyte hypertrophy by examining tissuespecific Rbpjk mutant(Prx1Cre;Rbpjkf/f), Rbpjk mutant/Sox9 haploinsufficient(Prx1Cre;Rbpjkf/f;Sox9f/1),and control embryos for alterations in SOX9 expression and chondrocyte hypertrophy during cartilage development. These studies demonstrate that Notch signaling regulates the onset of chondrocyte maturation in a SOX9-dependent manner, while Notch-mediated regulation of terminal chondrocyte maturation likely functions independently of SOX9. | 6 | A method to deliver patterned electrical impulses to Schwann cells cultured on an artificial axon显示文摘Information from the brain travels back and forth along peripheral nerves in the form of electrical impulses generated by neurons and these impulses have repetitive patterns. Schwann cells in peripheral nerves receive molecular signals from axons to coordinate the process of myelination. There is evidence, however,that non-molecular signals play an important role in myelination in the form of patterned electrical impulses generated by neuronal activity. The role of patterned electrical impulses has been investigated in the literature using co-cultures of neurons and myelinating cells. The co-culturing method, however, prevents the uncoupling of the direct effect of patterned electrical impulses on myelinating cells from the indirect effect mediated by neurons. To uncouple these effects and focus on the direct response of Schwann cells,we developed an in vitro model where an electroconductive carbon fiber acts as an artificial axon. The fiber provides only the biophysical characteristics of an axon but does not contribute any molecular signaling.In our 'suspended wire model', the carbon fiber is suspended in a liquid media supported by a 3D printed scaffold. Patterned electrical impulses are generated by an Arduino 101 microcontroller. In this study, we describe the technology needed to set-up and eventually replicate this model. We also report on our initial in vitro tests where we were able to document the adherence and ensheath of human Schwann cells to the carbon fiber in the presence of patterned electrical impulses(hSCs were purchased from ScienCell Research Laboratories, Carlsbad, CA, USA; ScienCell fulfills the ethic requirements, including donor's consent). | 12 | POLIII-derived non-coding RNAs acting as scaffolds and decoys显示文摘A large variety of eukaryotic small structured POLIII-derived non-coding RNAs(ncRNAs)have been described in the past.However,for only few,e.g.7SL and H1/MRP families,cellular functions are well understood.For the vast majority of these transcripts,cellular functions remain unknown.Recent findings on the role ofY RNAs and other POLIII-derived ncRNAs suggest an evolutionarily conserved function of these ncRNAs in the assembly and function of ribonucleoprotein complexes(RNPs).These RNPs provide cellular‘machineries’,which are essential for guiding the fate and function of a variety of RNAs.In this review,we summarize current knowledge on the role of POLIII-derived ncRNAs in the assembly and function of RNPs.We propose that these ncRNAs serve as scaffolding factors that‘chaperone’RNA-binding proteins(RBPs)to form functional RNPs.In addition or associated with this role,some small ncRNAs act as molecular decoys impairing the RBP-guided control of RNA fate by competing with other RNA substrates.This suggests that POLIII-derived ncRNAs serve essential and conserved roles in the assembly of larger RNPs and thus the control of gene expression by indirectly guiding the fate of mRNAs and IncRNAs. | HendrikTauber Stefan Huttelmaier Marcel Kohn | 2019 | Journal of Molecular Cell Biology2019，11，10： | 2 |
