| 7 | Evidence from ^(18)O feeding studies for hydroxyl group donor in the reaction catalyzed by cytidylate hydroxymethylase MilA显示文摘5-Hydroxymethylcytosine (5hmC) was present in T-even phage and mammalian DNA. 5hmC in phage is formed by hydroxymethylation of the cytosine base in deoxycytidylate (dCMP) by deoxycytidylate hydroxymethylase (CH), which uses the solvent water as the hydroxyl group donor. By contrast, 5hmC is formed in mammal zygotes by the oxidation of 5-methylcytosine (5mC). 5hmC was also present in a nucleoside antibiotic mildiomycin and its formation is governed by a cytidylate hydroxymethylase MilA. However, the catalytic mechanism remains unknown. In the present study, we purified His-tagged MilA and fed its in vitro reaction with H218O. The LC-MS analysis of the product revealed that 18O was incorporated into the hydroxymethylated CMP (HmCMP), and the secondary MS result of 18O-labeled HmCMP indicated that 18O was incorporated into the cytosine of HmCMP. The results demonstrate that MilA uses solvent water as the hydroxyl group donor like CH. Moreover, Thr102 of MilA was predicted as potential critical amino acid anchoring one molecule of water for hydroxylation. | 8 | GRB10 regulates β-cell mass by inhibiting β-cell proliferation and stimulating β-cell dedifferentiation显示文摘Decreased functional β-cell mass is the hallmark of diabetes, but the cause of this metabolic defect remains elusive. Here, we show that the levels of the growth factor receptor-bound protein 10(GRB10), a negative regulator of insulin and m TORC1 signaling, are markedly induced in islets of diabetic mice and high glucose-treated insulinoma cell line INS-1 cells. β-cell-specific knockout of Grb10 in mice increased β-cell mass and improved β-cell function. Grb10-deficient β-cells exhibit enhanced m TORC1 signaling and reduced β-cell dedifferentiation, which could be blocked by rapamycin. On the contrary, Grb10 overexpression induced β-cell dedifferentiation in MIN6 cells. | 11 | Impact of age and tumor size on the development of the Kasabach-Merritt phenomenon in patients with kaposiform hemangioendothelioma:a retrospective cohort study显示文摘Introduction The Kasabach–Merritt phenomenon(KMP)is a severe complication of kaposiform hemangioendothelioma(KHE).The risk factors for KMP need further investigation.Methods The medical records of patients with KHE were reviewed.Univariate and multivariate logistic regression models were used for the risk factors for KMP,and the area under the receiver operator characteristic(ROC)curve was used to assess the predictive power of risk factors.Results A total of 338 patients with KHE were enrolled.The incidence of KMP was 45.9%.Age of onset(P<0.001,odds ratio[OR]0.939;95%confidence interval[CI]0.914–0.966),lesion size(P<0.001,OR 1.944;95%CI 1.646–2.296),mixed type(P=0.030,OR 2.428;95%CI 1.092–5.397),deep type(P=0.010,OR 4.006;95%CI 1.389–11.556),and mediastinal or retroperitoneal lesion location(P=0.019,OR 11.864;95%CI 1.497–94.003)were correlated with KMP occurrence through multivariate logistic regression.ROC curve analysis revealed that the optimal cutoffs were 4.75 months for the age of onset(P<0.001,OR 7.206,95%CI 4.073–12.749)and a lesion diameter of 5.35 cm(P<0.001,OR 11.817,95%CI 7.084–19.714).Bounded by a lesion size of 5.35 cm,we found significant differences in tumor morphology,age of onset,treatments,and hematological parameters.Using an onset age of 4.75 months as a cutoff,we found significant differences in tumor morphology,lesion size,hematological parameters,and prognosis.Conclusion For KHE patients with an onset age<4.75 months and/or lesion diameter>5.35 cm,clinicians should be wary of the occurrence of KMP.Active management is recommended to improve the prognosis. | Jiangyuan Zhou Yuru Lan Tong Qiu Xue Gong Zixin Zhang Chunshui He Qiang Peng Fan Hu Xuepeng Zhang Guoyan Lu Liqing Qiu Feiteng Kong Yongbo Zhang Siyuan Chen Yi Ji | 2023 | Precision Clinical Medicine2023,6,2: | 0 |

| 13 | Effects of Antimicrobial Peptides on Production,Slaughter Performance and Blood Routine of Tan Sheep显示文摘[Objectives]This study was conducted to analyze effects of antimicrobial peptides added to the diet of Tan sheep on their production, slaughter performance and blood composition. [Methods] Ninety two four-month-old Tan sheep were randomly divided into two treatment groups according to their body weight, 46 in each group. The control check group(CK) was feed with conventional diet, and the experimental group was fed with the addition of antimicrobial peptide on the basis of the conventional diet, with the added amount of 2.5 g/sheep per day. The experimental period was 60 d. [Results] The incidence rate was 75.06% lower in the experimental group than in the CK. The average daily weight gain per sheep was 11.27% higher in the experimental group than in the CK(P<0.05). The feed conversion ratio was 8.45% lower in the experimental group than in the CK(P<0.05). The average daily gross profit per sheep was 12.12% higher in the experimental group than in the CK. For slaughter performance, the data difference of each item was not significant. The PH at 45 min and 24 h after slaughter was within the normal range of fresh mutton. The cooked meat percentage and water loss rate showed no significant differences(P>0.05). The marbling ranged from 2.45 to 2.50, indicating that the fat content was moderate, and the difference between groups was not significant(P>0.05). The flesh color ranged from 3.00 to 3.15, between light red and bright red, belonging to the normal color of mutton, and the difference between groups was not significant(P>0.05). The shear force was between 2.50 and 2.65, without a significant difference between groups(P>0.05). The white blood cells, lymphocytes and platelets in the experimental group were lower than those in the CK(P<0.01). The erythrocytes and hemoglobin in the erythrocyte group were higher than those in the CK(P<0.05). The neutrophils and monocytes in the experimental group were lower than those in the CK(P<0.05). | 14 | Particle Size-Controlled Oxygen Reduction and Evolution Reaction Nanocatalysts Regulate Ru(bpy)_(3)^(2+)’s Dual-potential Electrochemiluminescence for Sandwich Immunoassay显示文摘Multiple signal strategies remarkably improve the accuracy and efficiency of electrochemiluminescence(ECL)immunoassays,but the lack of potential-resolved luminophore pairs and chemical cross talk hinders their development.In this study,we synthesized a series of gold nanoparticles(AuNPs)/reduced graphene oxide(Au/rGO)composites as adjustable oxygen reduction reaction and oxygen evolution reaction catalysts to promote and modulate tris(2,2′-bipyridine)ruthenium(II)(Ru(bpy)_(3)^(2+))’s multisignal luminescence.With the increase in the diameter of AuNPs(3 to 30 nm),their ability to promote Ru(bpy)_(3)^(2+)’s anodic ECL was first impaired and then strengthened,and cathodic ECL was first enhanced and then weakened.Au/rGOs with medium-small and medium-large AuNP diameters remarkably increased Ru(bpy)_(3)^(2+)’s cathodic and anodic luminescence,respectively.Notably,the stimulation effects of Au/rGOs were superior to those of most existing Ru(bpy)_(3)^(2+)co-reactants.Moreover,we proposed a novel ratiometric immunosensor construction strategy using Ru(bpy)_(3)^(2+)’s luminescence promoter rather than luminophores as tags of antibodies to achieve signal resolution.This method avoids signal cross talk between luminophores and their respective co-reactants,which achieved a good linear range of 10−7 to 10−1 ng/ml and a limit of detection of 0.33 fg/ml for detecting carcinoembryonic antigen.This study addresses the previous scarcity of the macromolecular co-reactants of Ru(bpy)_(3)^(2+),broadening its application in biomaterial detection.Furthermore,the systematic clarification of the detailed mechanisms for converting the potential-resolved luminescence of Ru(bpy)_(3)^(2+)could facilitate an in-depth understanding of the ECL process and should inspire new designs of Ru(bpy)_(3)^(2+)luminescence enhancers or applications of Au/rGOs to other luminophores.This work removes some impediments to the development of multisignal ECL biodetection systems and provides vitality into their widespread applications. | Shijun Wang Shu Zhu Ziqi Kang Xiangxiu Wang Zixin Deng Kun Hu Jianjun Hu Xiancheng Liu Guixue Wang Guangchao Zang Yuchan Zhang | 2023 | Research2023,,4: | 0 |
| 15 | Causal Association of Cardiac Function by Magnetic Resonance Imaging with Frailty Index:A Mendelian Randomization Study显示文摘Owing to the susceptibility of conventional observational studies to confounding factors and reverse causation,the causal association between cardiac function and frailty is unclear.We aimed to investigate whether cardiac function has causal effects on frailty.In this study,a two-sample Mendelian randomization(MR)study was conducted using genetic variants associated with cardiac function assessed by magnetic resonance imaging phenotypes as instrumental variables.Genetic variants asso-ciated with cardiac function by magnetic resonance imaging(including seven cardiac function phenotypes)and the frailty index(FI)were obtained from two large genome-wide association studies.MR estimates from each genetic instrument were combined using inverse variance weighted(IVW),weighted median,and MR‒Egger regression methods.We found that the increase in genetically determined stroke volume(beta-0.13,95%CI-0.16 to-0.10,p=1.39E-6),rather than other cardiac phenotypes,was associated with lower FI in MR analysis of IVW after Bonferroni correction.Sensitivity analyses examining potential bias caused by pleiotropy or reverse causality revealed similar findings(e.g.,intercept[SE],-0.008[0.011],p=0.47 by MR‒Egger intercept test).The leave-one-out analysis indicated that the association was not driven by single nucleotide polymorphisms.No evidence of heterogeneity was found among the genetic variants(e.g.,MR‒Egger:Q statistic=14.4,p=0.156).In conclusion,we provided evidence that improved cardiac function could contribute to reducing FI.These findings support the hypothesis that enhancing cardiac function could be an effective prevention strategy for frailty. | Hui Zhang Meng Hao Zixin Hu Yi Li Xiaoxi Hu Xiaoyan Jiang Zuyun Liu Xuehui Sun Xiaofeng Wang | 2022 | Phenomics2022,2,6: | 0 |

| 18 | A phylogenetic approach identifies patterns of beta diversity and floristic subregions of the Qinghai-Tibet Plateau显示文摘Patterns of taxonomic and phylogenetic beta diversity and their relationships with environmental correlates can help reveal the origin and evolutionary history of regional biota.The Qinghai-Tibet Plateau(QTP)harbors an exceptionally diverse flora,however,a phylogenetic perspective has rarely been used to investigate its beta diversity and floristic regions.In this study,we used a phylogenetic approach to identify patterns of beta diversity and quantitatively delimit floristic regions on the Qinghai-Tibet Plateau.We also examined the relationships between multifaceted beta diversity,geographical distance,and climatic difference,and evaluated the relative importance of various factors(i.e.,climate,topography and history)in shaping patterns of beta diversity.Sørensen dissimilarity indices indicated that patterns of species turnover among sites dominated the QTP.We also found that patterns of both taxonomic and phylogenetic beta diversity were significantly related to geographical distance and climatic difference.The environmental factors that contributed most to these patterns of beta diversity include annual precipitation,mean annual temperature,climatic gradients and climatic instability.Hierarchical dendrograms of dissimilarity and non-metric multidimensional scaling ordination based on phylogenetic beta diversity data identified ten floristic subregions in the QTP.Our results suggest that the contemporary environment and historical climate changes have filtered species composition among sites and eventually determined beta diversity patterns of plants in the QTP. | Haibin Yu Man Yang Zixin Lu Weitao Wang Fangyuan Yu Yonghua Zhang Xue Yin Hongjun Yu Junjie Hu David C.Deane | 2024 | Plant Diversity2024,46,1: | 0 |
