| 1 | Gene editing and its applications in biomedicine显示文摘The steady progress in genome editing, especially genome editing based on the use of clustered regularly interspaced short palindromic repeats(CRISPR) and programmable nucleases to make precise modifications to genetic material, has provided enormous opportunities to advance biomedical research and promote human health. The application of these technologies in basic biomedical research has yielded significant advances in identifying and studying key molecular targets relevant to human diseases and their treatment. The clinical translation of genome editing techniques offers unprecedented biomedical engineering capabilities in the diagnosis, prevention, and treatment of disease or disability. Here, we provide a general summary of emerging biomedical applications of genome editing, including open challenges. | 2 | Quantitative Trait Loci Mapping of Flag-leaf Ligule Length in Rice and Alignment with ZmLG1 Gene显示文摘A doubled haploid(DH) population,which consists of 120 lines derived from anther culture of a typical indica and japonica hybrid 'CJ06'/'TN1',was used in this study.Ligule lengths of flag leaf were investigated for quantitative trait loci(QTL) mapping using the DH population.Five QTLs(qLL-2,qLL-4,qLL-6,qLL-10 and qLL-12) controlling the ligule length(LL) were detected on chromosomes 2,4,6,10 and 12,with the variances explained 11.4%,13.6%,27.8%,22.1% and 11.0%,respectively.Using four known genes of ZmGL1,ZmGL2,ZmGL3 and ZmGL4 in maize from the MaizeGDB,their homologs in rice were aligned and integrated into the existing simple sequence repeats linkage map by in silico mapping.A ZmLG1 homolog gene,OsLG1 encoding a squamosa promoter binding protein,was located between the markers RM255 and RM280,which is just identical to the interval of qLL-4 on the long arm of chromosome 4.The results are beneficial to dissection of the ligule molecular mechanism and the study of cereal evolution. | Dali Zeng Jiang Hu Guojun Dong Jian Liu Longjun Zeng Guangheng Zhang Longbiao Guo Yihua Zhou Qian Qian | 2009 | Journal of Integrative Plant Biology2009,51,4: | 6 |
| 3 | Polyoxometalates-engineered hydrogen generation rate and durability of Pt/CNT catalysts from ammonia borane显示文摘Heterogeneously catalyzed hydrolytic dehydrogenation of ammonia borane is a remarkable structure sensitive reaction. In this work, a strategy by using polyoxometalates(POMs) as the ligands is proposed to engineer the surface and electronic properties of Pt/CNT catalysts toward the enhanced hydrogen generation rate and durability. Three kinds of POMs, i.e., silicotungstic acid(STA), phosphotungstic acid(PTA)and molybdophosphoric acid(PMA), are comparatively studied, among which the STA shows positive effects on the catalytic activity and durability. A catalyst structure-performance relationship is established by a combination of kinetic and isotopic analyses with multiple characterization techniques, such as HAADF-STEM, EDS, Raman spectroscopy and XPS. It is shown that the STA compared to the other two POMs can increase the Pt binding energy and thus promote the reaction. | 5 | Inhibition of aberrant Hif1αactivation delays intervertebral disc degeneration in adult mice显示文摘The intervertebral disc(IVD) is the largest avascular tissue. Hypoxia-inducible factors(HIFs) play essential roles in regulating cellular adaptation in the IVD under physiological conditions. Disc degeneration disease(DDD) is one of the leading causes of disability, and current therapies are ineffective. This study sought to explore the role of HIFs in DDD pathogenesis in mice. The findings of this study showed that among HIF family members, Hif1α was significantly upregulated in cartilaginous endplate(EP) and annulus fibrosus(AF) tissues from human DDD patients and two mouse models of DDD compared with controls. Conditional deletion of the E3 ubiquitin ligase Vhl in EP and AF tissues of adult mice resulted in upregulated Hif1α expression and age-dependent IVD degeneration. Aberrant Hif1α activation enhanced glycolytic metabolism and suppressed mitochondrial function. On the other hand, genetic ablation of the Hif1α gene delayed DDD pathogenesis in Vhl-deficient mice. Administration of 2-methoxyestradiol(2ME2), a selective Hif1α inhibitor, attenuated experimental IVD degeneration in mice. | 11 | Extraction Difficulty of Lithium Ions from Various Crystal Planes of Lithium Titanate显示文摘To study the extraction difficulty of lithium ions from various crystal planes of Li_2 TiO_3, according to the first principle, four representative crystal surfaces of Li_2TiO_3(precursor),(-133),(-206),(002) and(-131), were selected to establish a model and to calculate the surface energy, bond length and population using Materials Studio 5.5(MS 5.5). The results demonstrate that there is no direct relationship between the surface energy and the order of disappearance of the four diffraction peaks when lithium titanate is treated with hydrochloric acid, instead, the difficulty of Li^+ extraction from various crystal faces corresponds to the Li-O bond strength. Lithium ion is easy to remove from(-133) and(-206) due to the relatively weak Li-O bond strength. | 16 | Chiral phonon activated spin Seebeck effect in chiral materials显示文摘Efficient generation of spin polarization is very important for spintronics and quantum computation. In chiral materials without magnetic order nor spin-orbit coupling, we find a new spin selectivity effect—chiral phonon activated spin Seebeck(CPASS)effect. Starting with the nonequilibrium distribution of chiral phonons under a temperature gradient, the CPASS coefficients are computed based on the Boltzmann transport theory. With both the phonon-drag and band transport contributions, the spin accumulations generated by the CPASS effect exhibit quadratic dependence on the temperature gradient. The strength of the CPASS effect and the relative magnitude of both contributions are tunable by the chemical potential modulation. | 17 | Resistance analysis of the rice variety Huaidao 5 against root-knot nematode Meloidogyne graminicola显示文摘Meloidogyne graminicola has emerged as one of the most destructive plant-parasitic nematodes affecting rice(Oryza sativa)production worldwide.Resistance to M.graminicola in rice could be the most effective option for its management.However,sources of germplasm with resistance to M.graminicola in rice remain limited.Here,we describe the root attraction,gall formation and genetic analysis of the resistance to M.graminicola in the rice variety Huidao 5.A nematode attraction assay showed that second-stage juveniles(J2s)of M.graminicola were attracted at the root tip of Huaidao 5 within 8 h without a significant reduction in attraction compared to the susceptible rice variety Nanjing 9108.Microscopic observation of the infection revealed that the J2s invaded root tissues 12 h after inoculation,but their subsequent movement to the root tip was hindered in Huaidao 5,resulting in decreased nematode number compared to Nanjing 9108.Additionally,we used the soil and hydroponic culture systems to simulate upland and flooding conditions in the paddy fields respectively,and found that gall number was significantly reduced,and nematode development was clearly suppressed in Huaidao 5.To investigate the genetic basis of this resistance,cross breeding was performed between the Huaidao 5 and Nanjing 9108 varieties.There was no reduction in the resistance of the F_(1) offspring to M.graminicola in the greenhouse or field trials,suggesting that a dominant gene could control resistance in Huaidao 5.In summary,this study provides a detailed characterization of a novel source of resistance to M.graminicola in rice,which is of great potential for use in crop breeding. | FENG Hui ZHOU Can-rong ZHU Feng LE Xiu-hu JING De-dao Paul DALY ZHOU Dong-mei WEI Li-hui | 2023 | Journal of Integrative Agriculture2023,22,10: | 0 |
| 18 | A de novo missense mutation in MPP2 confers an increased risk of Vogt–Koyanagi–Harada disease as shown by trio-based whole-exome sequencing显示文摘Vogt–Koyanagi–Harada(VKH)disease is a leading cause of blindness in young and middle-aged people.However,the etiology of VKH disease remains unclear.Here,we performed the first trio-based whole-exome sequencing study,which enrolled 25 VKH patients and 50 controls,followed by a study of 2081 VKH patients from a Han Chinese population to uncover detrimental mutations.A total of 15 de novo mutations in VKH patients were identified,with one of the most important being the membrane palmitoylated protein 2(MPP2)p.K315N(MPP2-N315)mutation.The MPP2-N315 mutation was highly deleterious according to bioinformatic predictions.Additionally,this mutation appears rare,being absent from the 1000 Genome Project and Genome Aggregation Database,and it is highly conserved in 10 species,including humans and mice.Subsequent studies showed that pathological phenotypes and retinal vascular leakage were aggravated in MPP2-N315 mutation knock-in or MPP2-N315 adeno-associated virus-treated mice with experimental autoimmune uveitis(EAU).In vitro,we used clustered regularly interspaced short palindromic repeats(CRISPR‒Cas9)gene editing technology to delete intrinsic MPP2 before overexpressing wild-type MPP2 or MPP2-N315.Levels of cytokines,such as IL-1β,IL-17E,and vascular endothelial growth factor A,were increased,and barrier function was destroyed in the MPP2-N315 mutant ARPE19 cells.Mechanistically,the MPP2-N315 mutation had a stronger ability to directly bind to ANXA2 than MPP2-K315,as shown by LC‒MS/MS and Co-IP,and resulted in activation of the ERK3/IL-17E pathway.Overall,our results demonstrated that the MPP2-K315N mutation may increase susceptibility to VKH disease. | Xianyang Liu Jiayu Meng Xingyun Liao Yusen Liu Qian Zhou Zongren Xu Shuming Yin Qingfeng Cao Guannan Su Siyuan He Wanqian Li Xiaotang Wang Guoqing Wang Dali Li Peizeng Yang Shengping Hou | 2023 | Cellular & Molecular Immunology2023,20,11: | 0 |
| 19 | Biocompatibility of Surface-Modified Biphasic Calcium Phosphate/Poly-L-Lactide Biocomposite in vitro and in vivo显示文摘The biocompatibility of surface-modified biphasic calcium phosphate (mBCP)/poly-L-Lactide (PLLA) biocomposite was investigated through a series of experiments in vitro and in vivo. Acute toxicity and short term systemic toxicity experiments revealed no toxicity of the materials. Hemolysis assay indicated the good blood compatibility of the composite. In cytotoxicity assay, L929 mouse fibroblasts could well differentiate and proliferate. Animal experiments in vivo were performed by implanting the materials into rabbits muscle for 8 weeks. The decreasing of inflammatory cells, the building of fibrous tissue layer as well as the growing of blood cells into materials indicated the nontoxicity of the composite. | 20 | Characterization of ZnO Based Varistor Derived from Nano ZnO Powders and Ultrafine Dopants显示文摘Nanosized ZnO powders were prepared with a two-step precipitation method. The average size of ZnO particles was about 80 nm and their size distribution was narrow. Combining with ultrafine additive powders, ZnO base varistor was produced via an oxide mixing route. ZnO varistor derived from normal reagent grade starting materials was investigated for comparison purpose. Outstanding microstructure of the ZnO varistor derived from nanosize ZnO powders and ultrafine dopants was obtained: uniform distribution of fine ZnO grains (less than 3 microns), grain boundary and the dopant position. Higher varistor voltage (U=492 V/mm) and nonlinear coefficient (α=56.2) as well as lower leakage current (IL=1.5μA) were achieved. The better electrical properties were attributed to the uniform microstructure, which in turn led to stable and uniform potential barriers.
