| 1 | Does flagship species tourism benefit conservation? A case study of the golden snub-nosed monkey in Shennongjia National Nature Reserve显示文摘Tourism has been widely used as a conservation strategy that directly compensates local people or management authorities, thereby contributing to development and creating incentives to preserve the landscape. Reserves or parks that are inhabited by flagship species may be able to generate more revenue through tourism than those without such species because flagship species have great potential for raising funds and creating publicity for conservation goals. In this paper, we examine flagship species tourism focused on observing the golden snub-nosed monkey (Rhinopithecus roxellana) in Shennongjia National Nature Reserve (SNNR). After the program was launched in 2006, both the number of tourists and amount of revenue received by SNNR increased as the general trends of tourism revenue in the country also increased. As a result, SNNR significantly increased rein- vestment in tourism and conservation infrastructure. Furthermore, after senior politicians visited the golden snub-nosed monkey, they abruptly changed their attitudes toward biological conservation, as indicated by an increase in both regular and additional funding for the species conservation. We conclude that flagship species tourism may not only generate revenue for conservation and management but also can improve officials' attitudes toward preserving biodiversity. | 4 | Effect of P-selectin monoclonal antibody on metastasis of gastric cancer and immune function显示文摘AIM: To investigate the effect of cell adhesion molecule Pselectin monoclonal antibody (Mab) on metastasis and immune function of mice orthototopically implanted with human gastric cancer tissue.METHODS: SCID mice were implanted orthotopically with SGC-7901 human gastric carcinoma tissue. Starting from day 3 after operation, animals were given intravenously PBS or P-selectin Mab (100 μg/injection) (for both normal mice and tumor-implanted mice with tumors), twice weekly for 3weeks. Two animals in each group were sacrificed randomly at the 1st, 2nd, 4th week and 6th week. While T cell and B cell transformation indices were determined with the 3H TdR infiltration method, the NK cell activity was detected by the LDH release method.RESULTS: The metastatic rate in the P-selectin Mab treated group was lower than that in the PBS treated group (with tumors). | 6 | Fractal characteristics of surface crack evolution in the process of gas-containing coal extrusion显示文摘In this paper, simulated experiment device of coal and gas outburst was employed to perform the experiment on gas-containing coal extrusion. In the experiment, coal surface cracks were observed with a high-speed camera and then the images were processed by sketch. Based on the above description, the paper studied the fractal dimension values from different positions of coal surface as well as their changing laws with time. The results show that there is a growing parabola trend of crack dimension value in the process of coal extrusion. Accordingly, we drew the conclusion that extruded coal crack evolution is a process of fractal dimension value increase. On the basis of fractal dimension values taken from different parts of coal masses, a fractal dimension of the contour map was drawn. Thus, it is clear that the contour map involves different crack fractal dimension values from different positions. To be specific, where there are complicated force and violent movement in coal mass, there are higher fractal dimension values, i.e., the further the middle of observation surface is from the exit of coal mass, and the lower the fractal dimension value is. In line with fractal geometry and energy theory of coal and gas outburst, this study presents the relation between fractal dimension and energy in the process of extruding. | 9 | PGE2 activates EP4 in subchondral bone osteoclasts to regulate osteoarthritis显示文摘Prostaglandin E2(PGE2), a major cyclooxygenase-2(COX-2) product, is highly secreted by the osteoblast lineage in the subchondral bone tissue of osteoarthritis(OA) patients. However, NSAIDs, including COX-2 inhibitors, have severe side effects during OA treatment. Therefore, the identification of novel drug targets of PGE2 signaling in OA progression is urgently needed. Osteoclasts play a critical role in subchondral bone homeostasis and OA-related pain. However, the mechanisms by which PGE2 regulates osteoclast function and subsequently subchondral bone homeostasis are largely unknown. Here, we show that PGE2 acts via EP4 receptors on osteoclasts during the progression of OA and OA-related pain. Our data show that while PGE2 mediates migration and osteoclastogenesis via its EP2 and EP4 receptors, tissue-specific knockout of only the EP4 receptor in osteoclasts(EP4 Lys M) reduced disease progression and osteophyte formation in a murine model of OA. Furthermore, OA-related pain was alleviated in the EP4 Lys M mice, with reduced Netrin-1 secretion and CGRP-positive sensory innervation of the subchondral bone. The expression of plateletderived growth factor-BB(PDGF-BB) was also lower in the EP4 Lys Mmice, which resulted in reduced type H blood vessel formation in subchondral bone. Importantly, we identified a novel potent EP4 antagonist, HL-43, which showed in vitro and in vivo effects consistent with those observed in the EP4 Lys Mmice. Finally, we showed that the Gαs/PI3 K/AKT/MAPK signaling pathway is downstream of EP4 activation via PGE2 in osteoclasts. Initiated by observations of different endoplasmic reticulum(ER) responses of host to seasonal H1N1 and highly pathogenic avian influenza(HPAI) A H5N1 infections, we identified an alternative antiviral role of tauroursodeoxycholic acid(TUDCA), a clinically available ER stress inhibitor, both in vitro and in vivo. Rather than modulating ER stress in host cells, TUDCA abolished the proton conductivity of viral M2 by disrupting its oligomeric states, which induces inefficient viral infection. We also showed that M2 penetrated cells, whose intracellular uptake depended on its proton channel activity,an effect observed in both TUDCA and M2 inhibitor amantadine. | 14 | Establishment Method of a Mixture Model and Its Practical Application for Transmission Gears in an Engineering Vehicle显示文摘Highly versatile machines, such as wheel loaders, forklifts, and mining haulers, are subject to many kinds of working conditions, as well as indefinite factors that lead to the complexity of the load. The load probability distribution function (PDF) of transmission gears has many distributions centers; thus, its PDF cannot be well represented by just a single-peak function. For the purpose of representing the distribution characteristics of the complicated phenomenon accurately, this paper proposes a novel method to establish a mixture model. Based on linear regression models and correlation coefficients, the proposed method can be used to automatically select the best-fitting function in the mixture model. Coefficient of determination, the mean square error, and the maximum deviation are chosen and then used as judging criteria to describe the fitting precision between the theoretical distribution and the corresponding histogram of the available load data. The applicability of this modeling method is illustrated by the field testing data of a wheel loader. Meanwhile, the load spectra based on the mixture model are compiled. The comparison results show that the mixture model is more suitable for the description of the load-distribution characteristics. | 18 | Efficient transformation and expression of gfp gene in the edible mushroom Pleurotus nebrodensis显示文摘An efficient transformation method mediated by PEG-protoplasts was developed for the newly commercial edible mushroom Pleu- rotus nebrodensis. Two plasmids were used to co-transform protoplasts of P. nebrodensis. One plasmid is pAN7-1 containing a positive selectable marker gene hph conferring hygromycin B resistance. Another plasmid is pBlue-GFP containing a reporter gene gfp conferring green fluorescent protein. PCR and Southern blot analysis showed that hph gene or/and gfp gene were integrated into the genome of P. nebrodensis transformants. The transformation efficiency of the positive selectable marker gene hph was 3 transformants per microgram of plasmid pAN7-1 DNA, which was about 30 times higher than that previously reported in thoroughly studied Pleurotus species such as Pleurotus ostreatus. The transformation efficiency of the reporter gene gfp was 9 transformants per microgram of plasmid pBlu-GFP DNA. The co-transformation efficiency was 23.68%. | 19 | The role of GPCRs in bone diseases and dysfunctions显示文摘The superfamily of G protein-coupled receptors (GPCRs) contains immense structural and functional diversity and mediates a myriad of biological processes upon activation by various extracellular signals.Critical roles of GPCRs have been established in bone development,remodeling,and disease.Multiple human GPCR mutations impair bone development or metabolism,resulting in osteopathologies.Here we summarize the disease phenotypes and dysfunctions caused by GPCR gene mutations in humans as well as by deletion in animals.To date,92 receptors (5 glutamate family,67 rhodopsin family,5 adhesion,4 frizzled/taste2 family,5 secretin family,and 6 other 7TM receptors) have been associated with bone diseases and dysfunctions (36 in humans and 72 in animals).By analyzing data from these 92 GPCRs,we found that mutation or deletion of different individual GPCRs could induce similar bone diseases or dysfunctions,and the same individual GPCR mutation or deletion could induce different bone diseases or dysfunctions in different populations or animal models.Data from human diseases or dysfunctions identified 19 genes whose mutation was associated with human BMD:9 genes each for human height and osteoporosis;4 genes each for human osteoarthritis (OA) and fracture risk;and 2 genes each for adolescent idiopathic scoliosis (AIS),periodontitis,osteosarcoma growth,and tooth development.Reports from gene knockout animals found 40 GPCRs whose deficiency reduced bone mass,while deficiency of 22 GPCRs increased bone mass and BMD;deficiency of 8 GPCRs reduced body length,while 5 mice had reduced femur size upon GPCR deletion.Furthermore,deficiency in 6 GPCRs induced osteoporosis;4 induced osteoarthritis;3 delayed fracture healing;3 reduced arthritis severity;and reduced bone strength,increased bone strength,and increased cortical thickness were each observed in 2 GPCR-deficiency models.The ever-expanding number of GPCR mutation-associated diseases warrants accelerated molecular analysis,population studies,and investigation of phenotype correlation with SNPs to elucidate GPCR function in human diseases. | Jian Luo Peng Sun Stefan Siwko Mingyao Liu Jianru Xiao | 2019 | Bone Research2019,7,2: | 5 |
