3Verification of Mutagen Function of Zeocin in Nannochloropsis oceanica Through Transcriptome Analysis显示文摘Zeocin can cause double strand breaks of DNA and thus is frequently used as a selective antibiotic of eukaryotic Sh ble transformants. In non-transformation system, Zeocin may function as a mutagen if not totally lethal. To verify such function of Zeocin, we mutated Nannochloropsis oceanica by increasing the concentration of Zeocin in medium gradually, and isolated a N. oceanica strain(single cell culture) which survived Zeocin up to 10.0μg mL^(-1). The Zeocin-tolerant strain entered the exponential growth phase later and grew slower than the wild strain. Transcriptome profiling showed that the Zeocin-tolerant N. oceanica strain survived Zeocin mainly by adapting(heritable), rather than acclimating(plastic) to Zeocin. Hence mutating N. oceanica with Zeocin was approved effective. Meanwhile, the physiological characteristics of this Zeocin-tolerant strain were demonstrated. As we proposed, N. oceanica tolerated Zeocin by strengthening its protein degradation and antioxidation. The genes controlling cell division and cellular response to stimuli may also have played important roles in the reduction of growth and the tolerance to Zeocin. Our findings evidenced that Zeocin can serve as an appropriate mutagen of microalgae. 4Structural Variation Analysis of Mutated Nannochloropsis oceanica Caused by Zeocin Through Genome Re-Sequencing显示文摘Zeocin can cause double strand breaks of DNA and thus may be employed as a mutagen.In this study,two strains of Nannochloropsis oceanica,the wild and the Zeocin-tolerant strains,were re-sequenced to verify such function of Zeocin.The results showed that Zeocin can mutate the N.oceanica genome and cause the structural variation.Zeocin either swept away or selected the alleles of genes functioning in ubiquitin-mediated proteolysis,alpha-linolenic acid metabolism,ascorbate and aldarate metabolism,ribosome biogenesis,and circadian rhythm,indicating that N.oceanica may have adjusted its metabolic performances for protein,carbohydrate,and lipid,and changed its ribosome biosynthesis and living rhythm to survive in Zeocin containing medium.In addition,Zeocin caused mutation may have influenced the expression of a set of transcription factors.It was concluded that Zeocin effectively caused the structural variation of the genome of N.oceanica,and forced the microalgae to select out the alleles of a set of genes around these variations in order to adapt to Zeocin containing medium.Further studies on the genetic basis of the phenotypic adaptation of this haploid and asexual microalga and the application of Zeocin to its genetic improvement are very important.LIN Genmei ZHANG Zhongyi GUO Li DING Haiyan YANG Guanpin 2018Journal of Ocean University of China2018,17,5:3

7Information integration research on cumulative effect of 'Siqi,Wuwei,and Guijing' in Traditional Chinese Medicine显示文摘OBJECTIVE:To study the morphological basis of the role of Siqi(cold as winter,cool as autumn,warm as spring,hot as summer),Wuwei(five flavors:sweet,pungent,salty,sour,and bitter),and Guijing(meridian tropism) through the use of information integration.METHODS:A14C-2-deoxy-glucose autoradiography method was adopted to determine the overall impact of treatment with 39 herbs on functions of various tissues and organs.Data was measured at 4hs after a single dose and following the last treatment of repeated doses for a week.Least-squares estimation was used and fitted for each herb regression effect of organs and tissues after singleand repeated treatment.The slope of the regression line represented the cumulative trend of the effect of the herbs(β),and the standard deviation of the slope(Sβ) was compared with those of the untreated animals(t 'test).All significantly cumulative effect trends were applied with an artificial neural network,which integrated the relationship among Siqi,Wuwei,and Guijing with tissues and organs.RESULTS:There is a certain relationship among the Siqi,Wuwei,Guijing and the anatomy of organs and tissues,but the different scores indicate that influence of Siqi,Wuwei,Guijing to anatomy of organs and tissues was a nonlinear state.CONCLUSION:Results demonstrated that the effects of Siqi,Wuwei,and Guijing have a morphological basis,and each concept was associated with multiple anatomical structures.Yang Xuming Qi Mingyuan Li Qian Chen Li Yu Zhongyi Yang Lin 2016Journal of Traditional Chinese Medicine2016,36,4:2

9A novel multistage anoxic/aerobic process with sludge regeneration zone (R-MAO) for advanced nitrogen removal from domestic sewage显示文摘To achieve advanced nitrogen removal from actual municipal sewage,a novel multistage anoxic/aerobic process with sludge regeneration zone(R-MAO)was developed.The reactor was used to treat actual domestic sewage and the nitrogen removal capacity of the sludge regeneration zone(R zone)was investigated during the long-term operation.The best performance was obtained at the R zone’s Oxidation-Reduction Potential(ORP)of-50±30 mV and hydraulic residence times(HRT)of 1.2 hr.The average effluent COD,TN,NH_(4)^(+)-N and NO_(3)^(−)-N of the R-MAO process were 18.0±2.3,7.5±0.6,1.0±0.5 and 4.6±0.4 mg/L,respectively,with the corresponding removal efficiency of COD,TN and NH_(4)^(+)-N were 92.9%±1.0%,84.1%±1.5% and 97.5%±1.1%.Compared to the sole MAO system,the TN removal efficiency of the R-MAO increased by 10.1%.Besides,under the optimal conditions,the contribution of the R zone in the R-MAO that removal COD,TN,NH_(4)^(+)-N and NO_(3)^(−)-N were 0.36,0.15,0.032 and 0.82 g/day.High-throughput sequencing results showed that uncultured_bacterium_f_Burkholderiaceae(5.20%),OLB8(1.04%)and Ottowia(1.03%)played an important role in denitrification in the R zone.This study provided effective guidance for the design and operation of the R-MAO process in domestic sewage treatment.Lin Cao Rong Sun Wenyi Dong Hongjie Wang Zhongyi Dai Xue Wang Jin Xie Hua Li 2023Journal of Environmental Sciences2023,,2:1

12A gecko-inspired adhesive robotic end effector for critical-contact manipulation显示文摘It is an emerging challenge for robots to achieve non-destructive pick-and-place manipulation for delicate objects under low normal preload, i.e., critical-contact manipulation. The prominent on-off controllable property of the gecko-inspired microwedge adhesive makes critical-contact manipulation possible for robotic end effectors. However, it is difficult for end effectors to actuate the micron-scale microwedge adhesive and detect the adhesion state. In this paper, a gecko-inspired adhesive robotic end effector for critical-contact manipulation is proposed, which consists of a half-scissor variable-scale actuator, a 3-axis high-sensitivity isotropic flexible capacitive tactile sensor, and the microwedge adhesive. The half-scissor variable-scale actuator is designed to provide pure large shear loading for the microwedge adhesive at micronscale displacement by merely controlling the normal macro-scale displacement of the actuator. Besides, the3-axis high-sensitivity isotropic flexible capacitive tactile sensor is designed for accurate detection of multiaxis contact forces and the adhesion state between the adhesive and objects to ensure the success of the critical-contact manipulation. The sensor can sense the shear and normal forces by detecting variations of the overlap and distance between electrodes, while the design of the finger-like electrodes improves the sensitivity. 14Characterization of Lysine Monomethylome and Methyltransferase in Model Cyanobacterium Synechocystis sp.PCC 6803显示文摘Protein lysine methylation is a prevalent post-translational modification(PTM)and plays critical roles in all domains of life.However,its extent and function in photosynthetic organisms are still largely unknown.Cyanobacteria are a large group of prokaryotes that carry out oxygenic photosynthesis and are applied extensively in studies of photosynthetic mechanisms and environmental adaptation.Here we integrated propionylation of monomethylated proteins,enrichment of the modified peptides,and mass spectrometry(MS)analysis to identify monomethylated proteins in Synechocystis sp.PCC 6803(Synechocystis).Overall,we identified 376 monomethylation sites in270 proteins,with numerous monomethylated proteins participating in photosynthesis and carbon metabolism.We subsequently demonstrated that Cpc M,a previously identified asparagine methyltransferase in Synechocystis,could catalyze lysine monomethylation of the potential aspartate aminotransferase Sll0480 both in vivo and in vitro and regulate the enzyme activity of Sll0480.The loss of Cpc M led to decreases in the maximum quantum yield in primary photosystemⅡ(PSⅡ)and the efficiency of energy transfer during the photosynthetic reaction in Synechocystis.We report the first lysine monomethylome in a photosynthetic organism and present a critical database for functional analyses of monomethylation in cyanobacteria.The large number of monomethylated proteins and the identification of Cpc M as the lysine methyltransferase in cyanobacteria suggest that reversible methylation may influence the metabolic process and photosynthesis in both cyanobacteria and plants.Xiaohuang Lin Mingkun Yang Xin Liu Zhongyi Cheng Feng Ge 2020Genomics, Proteomics & Bioinformatics2020,18,3:1

17Detection and classification of breast lesions using multiple information on contrast-enhanced mammography by a multiprocess deep-learning system: A multicenter study显示文摘Objective: Accurate detection and classification of breast lesions in early stage is crucial to timely formulate effective treatments for patients. We aim to develop a fully automatic system to detect and classify breast lesions using multiple contrast-enhanced mammography(CEM) images.Methods: In this study, a total of 1,903 females who underwent CEM examination from three hospitals were enrolled as the training set, internal testing set, pooled external testing set and prospective testing set. Here we developed a CEM-based multiprocess detection and classification system(MDCS) to perform the task of detection and classification of breast lesions. In this system, we introduced an innovative auxiliary feature fusion(AFF)algorithm that could intelligently incorporates multiple types of information from CEM images. The average freeresponse receiver operating characteristic score(AFROC-Score) was presented to validate system’s detection performance, and the performance of classification was evaluated by area under the receiver operating characteristic curve(AUC). Furthermore, we assessed the diagnostic value of MDCS through visual analysis of disputed cases,comparing its performance and efficiency with that of radiologists and exploring whether it could augment radiologists’ performance.Results: On the pooled external and prospective testing sets, MDCS always maintained a high standalone performance, with AFROC-Scores of 0.953 and 0.963 for detection task, and AUCs for classification were 0.909[95% confidence interval(95% CI): 0.822-0.996] and 0.912(95% CI: 0.840-0.985), respectively. It also achieved higher sensitivity than all senior radiologists and higher specificity than all junior radiologists on pooled external and prospective testing sets. Moreover, MDCS performed superior diagnostic efficiency with an average reading time of 5 seconds, compared to the radiologists’ average reading time of 3.2 min. 18Clinical application of Aconitum carmichaelii Debx. (Fu Zi in Chinese) by traditional Chinese medicine physicians–A cross-sectional questionnaire survey in Beijing显示文摘Objective:To investigate clinical application of aconite root(Aconitum carmichaelii Debx.;hereinafter“aconite”)by traditional Chinese medicine(TCM)physicians in TCM hospitals in Beijing and explore the specific use of aconite and the effect of different doses on decoction time.Methods:This was a cross-sectional study using two-stage sampling.Questionnaires were administered among TCM physicians from TCM hospitals in Beijing between September 2020 and August 2021.The questionnaire mainly included general demographics,clinical backgrounds of TCM physicians,and clinical application of aconite.The data analyses were performed using SPSS 20.0.Results:A total of 387 questionnaires were collected,of which 385 were valid.Of the 385 participating TCM physicians,331(85.97%)used aconite in clinical practice.The top three medical diagnoses treated with aconite were heart failure,diarrhea without cause,and chronic gastritis.The TCM pattern treated most frequently with a high dose of aconite was spleen-kidney yang deficiency,and the TCM pattern treated most frequently with a low dose was spleen yang deficiency,while spleen-kidney yang deficiency was treated most frequently with a common dose of aconite.The top three common formulas were Sini decoction,Mahuang Xixin Fuzi decoction,and Zhenwu decoction.Hei Shun Pian was the most common herbal pieces of aconite.The highest frequency of the maximum dose of aconite was 15 g,that of the minimum dose was 3 g,and that of the common dose was 10 g.There was a certain correlation between decoction time and the dose of aconite.Most TCM physicians advised their patients to take medicine twice a day.Conclusion:In this study,we refined and quantified the use of aconite and enriched the exploration of the clinical application of aconite besides its provisions in the Pharmacopoeia of the People's Republic of China.Xiaopeng Zhao Shenglou Ni Ning Liang Qiuchen Huang Zhongyi Pan Lin Zhang Jia Song Yanling Fu 2021Journal of Traditional Chinese Medical Sciences2021,8,4:0
19Coordination-driven structure reconstruction in polymer of intrinsic microporosity membranes for efficient propylene/propane separation显示文摘Polymers of intrinsic microporosity(PIMs),integrating unique microporous structure and solution-processability,are one class of the most promising membrane materials for energy-efficient gas separations.However,the micropores generated from inefficient chain packing often exhibit wide pore size distribution,making it very challenging to achieve efficient olefin/paraffin separations.Here,we propose a coordination-driven reconstruction(CDR)strategy,where metal ions are incorporated into amidoxime-functionalized PIM-1(AO-PIM)to in situ generate coordination crosslinking networks.By varying the type and content of metal ions,the resulting crosslinking structures can be optimized,and the molecular sieving capability of PIM membranes can be dramatically enhanced.Particularly,the introduction of alkali or alkaline earth metals renders more precise micropores contributing to superior C3H6/C3H8 separation performance.K+incorporated AO-PIM membranes exhibit a high ideal C3H6/C3H8 selectivity of 50,surpassing almost all the reported polymer membranes.Moreover,the coordination crosslinking structure significantly improves the membrane stability under higher pressure as well as the plasticization resistant performance.We envision that this straightforward and generic CDR strategy could potentially unlock the potentials of PIMs for olefin/paraffin separations and many other challenging gas separations.Yanxiong Ren Boyang Chong Wei Xu Zhengqing Zhang Lin Liu Yingzhen Wu Yutao Liu Haifei Jiang Xu Liang Hong Wu Hongjun Zhang Bangjiao Ye Chongli Zhong Guangwei He Zhongyi Jiang 2022The Innovation2022,3,6:0
20Disentangling land model uncertainty via Matrix-based Ensemble Model Inter-comparison Platform(MEMIP)显示文摘Background:Large uncertainty in modeling land carbon(C)uptake heavily impedes the accurate prediction of the global C budget.Identifying the uncertainty sources among models is crucial for model improvement yet has been difficult due to multiple feedbacks within Earth System Models(ESMs).Here we present a Matrix-based Ensemble Model Inter-comparison Platform(MEMIP)under a unified model traceability framework to evaluate multiple soil organic carbon(SOC)models.Using the MEMIP,we analyzed how the vertically resolved soil biogeochemistry structure influences SOC prediction in two soil organic matter(SOM)models.By comparing the model outputs from the C-only and CN modes,the SOC differences contributed by individual processes and N feedback between vegetation and soil were explicitly disentangled.Results:Results showed that the multi-layer models with a vertically resolved structure predicted significantly higher SOC than the single layer models over the historical simulation(1900–2000).The SOC difference between the multi-layer models was remarkably higher than between the single-layer models.Traceability analysis indicated that over 80%of the SOC increase in the multi-layer models was contributed by the incorporation of depth-related processes,while SOC differences were similarly contributed by the processes and N feedback between models with the same soil depth representation.Conclusions:The output suggested that feedback is a non-negligible contributor to the inter-model difference of SOC prediction,especially between models with similar process representation.Further analysis with TRENDY v7 and more extensive MEMIP outputs illustrated the potential important role of multi-layer structure to enlarge the current ensemble spread and the necessity of more detail model decomposition to fully disentangle inter-model differences.We stressed the importance of analyzing ensemble outputs from the fundamental model structures,and holding a holistic view in understanding the ensemble uncertainty.Cuijuan Liao Yizhao Chen Jingmeng Wang Yishuang Liang Yansong Huang Zhongyi Lin Xingjie Lu Yuanyuan Huang Feng Tao Danica Lombardozzi Almut Arneth Daniel SGoll Atul Jain Stephen Sitch Yanluan Lin Wei Xue Xiaomeng Huang Yiqi Luo 2022Ecological Processes2022,11,1:0
