1Function of lncRNAs and approaches to lncRNA-protein interactions显示文摘Long non-coding RNAs(lncRNAs),which represent a new frontier in molecular biology,play important roles in regulating gene expression at epigenetic,transcriptional and post-transcriptional levels.More and more lncRNAs have been found to play important roles in normal cell physiological activities,and participate in the development of varieties of tumors and other diseases.Previously,we have only been able to determine the function of lncRNAs through multiple mechanisms,including genetic imprinting,chromatin remodeling,splicing regulation,mRNA decay,and translational regulation.Application of technological advances to research into the function of lncRNAs is extremely important.The major tools for exploring lncRNAs include microarrays,RNA sequencing(RNA-seq),Northern blotting,real-time quantitative reverse transcription-polymerase chain reaction(qRT-PCR),fluorescence in situ hybridization(FISH),RNA interference(RNAi),RNA-binding protein immunoprecipitation(RIP),chromatin isolation by RNA purification(ChIRP),crosslinking-immunopurification(CLIP),and bioinformatic prediction.In this review,we highlight the functions of lncRNAs,and advanced methods to research lncRNA-protein interactions.ZHU JuanJuan FU HanJiang WU YongGe ZHENG XiaoFei 2013Science China(Life Sciences)2013,56,10:54
2Optimal configuration of a class of endoreversible heat engines for maximum efficiency with radiative heat transfer law显示文摘Optimal configuration of a class of endoreversible heat engines with fixed duration,input energy and radiative heat transfer law (q∝Δ(T4)) is determined. The optimal cycle that maximizes the efficiency of the heat engine is obtained by using opti-mal-control theory,and the differential equations are solved by the Taylor series expansion. It is shown that the optimal cycle has eight branches including two isothermal branches,four maximum-efficiency branches,and two adiabatic branches. The interval of each branch is obtained,as well as the solutions of the temperatures of the heat reservoirs and the working fluid. A numerical example is given. 4The trend of the APRGP velocity field and plates movement derived from GPS data显示文摘High-precision data processing theories and methods for the wide area GPS network–––Asia Pacific Regional Geodesy Project (APRGP), are systematically discussed in this article. With the software bernese5.0 update, the annual results of APRGP coordinate, velocity field and the trend of plate movement are reestimated, and the corresponding accuracy of adjustment with different constraints, also the velocity field of APRGP, are presented and analyzed. 5Virus-Mediated Overexpression of ETS-1 in the Ventral Hippocampus Counteracts Depression-Like Behaviors in Rats显示文摘ETS-1 is a transcription factor that is a member of the E26 transformation-specific(ETS) family. Galanin receptor 2(Gal R2), a subtype of receptors of the neuropeptide galanin, has been shown to have an antidepressant-like effect after activation in rodents. Our previous study has shown that overexpression of ETS-1 increases the expression of Gal R2 in PC12 phaeochromocytoma cells. However, whether ETS-1 has an antidepressant-like effect is still unclear. In this study, we found that chronic mild stress(CMS) decreased the expression of both ETS-1 and Gal R2 in the ventral hippocampus of rats. Meanwhile,we demonstrated that overexpression of ETS-1 increased the expression of Gal R2 in primary hippocampal neurons.Importantly, we showed that overexpression of ETS-1 in the ventral hippocampus counteracted the depression-like behaviors of CMS rats. Furthermore, we found that overexpression of ETS-1 increased the level of downstream phosphorylated extracellular signal-regulated protein kinases 1 and 2(p-ERK1/2) of Gal R2 in the ventral hippocampus of CMS rats. 6Effect of Nanocomposites Coatings on Exterior Performance of Bamboo during Aging显示文摘Bamboo deterioration caused by light becomes a significant problem as application of bamboo extends from indoor to outdoor.The color and roughness of bamboo are two important exterior performances.This study focused on UV resistant nano composites coatings for exterior use and what aimed to find a formulation which could enhance the durability of bamboo.Four kinds of film-forming materials and four kinds of nano particles were used to improve bamboo exterior performance.The color and roughness of nano composites coatings were measured during accelerated weathering.Artificial aging time was 174 h at the temperature of 50 ℃.The experiment showed nanocomposites coatings had significant improvement in UV-shielding and nearly all the samples presented a darkening tendency in ΔL* and increasing tendency in Δa*,ΔE* and ΔRa.The orthogonal experiment obtained the least change in color and Ra which could be achieved with nano composites coating made by melamine resin and TiO2 when a* was considered as an important index.Film-forming material was the only factor which had significant influence on color and roughness when a* was not considered as important index,then melamine resin was the best choice.LI Neng CHEN Yuhe BAO Yongjie ZOU Yijia CAI Hanjiang 2012Chinese Forestry Science and Technology2012,11,Z1:4
7Clinical and pathological features of miR-10b and RHOC gene expression in hepatocellular carcinoma显示文摘Aberrant expression of microRNAs(miRNAs)was reported frequently in different human cancers.The major role of miRNA is targeting 30-UTR of coding gene and causing translational repression or mRNA degradation.miR-10b overexpression was reported to promote breast cancer metastasis by up-regulating RHOC expression.But its expression in hepatocellular carcinoma(HCC)remains unclear.Our study indicated that the expression of miR-10b was different in HCC and adjacent tissue samples,and reduced expression of miR-10b in HCC was related tovein invasion.High-level expression of RHOC was also related to vein invasion in HCC.But no correlation was found between miR-10b and RHOC expression.These results suggest that miR-10b and RHOC are independent predictors of HCC invasion and metastasis.Jingli Du Yulan Wang Hanjiang Fu Xiaofei Zheng Minggui Lin Mingzhou Guo Lixin Wei 2014Chinese Science Bulletin2014,59,19:4

9Treatment of discontinuities inside Earth models:Effects on computed coseismic deformations显示文摘In this paper,we study how coseismic deformations calculated in 1066 Earth models are affected by how the models treat Earth discontinuities.From the results of applying models 1066A(continuous)and 1066B(discontinuous),we find that the difference in Love numbers of strike-slip and horizontal tensile sources are bigger than dip-slip and vertical tensile sources.Taken collectively,discontinuities have major effects on Green’s functions of four independent sources.For the near-field coseismic deformations of the 2013 Okhotsk earthquake(Mw 8.3),the overall differences between theoretical calculations in vertical displacement,geoid,and gravity changes caused by discontinuities are 10.52 percent,9.07 percent and 6.19 percent,with RMS errors of 0.624 mm,0.029 mm,and 0.063μGal,respectively.The difference in far-field displacements is small,compared with GPS data,and we can neglect this effect.For the shallow earthquake,2011 Tohoku-Oki earthquake(Mw 9.0),the differences in near-field displacements are 0.030 m(N-S),0.093 m(E-W),and 0.025 m(up-down)in our study area with the ARIA slip model,which gives results closer to GPS data than those from the USGS model.The difference in vertical displacements and gravity changes on the Earth’s surface caused by discontinuities are larger than 10 percent.The difference in the theoretical gravity changes at spatially fixed points truncated to degrees 60,as required by GRACE data,is 0.0016μGal and the discrepancy is 11 percent,with the theoretical spatial gravity changes from 1066B closer to observations than from 1066A.The results show that an Earth model with discontinuities in the medium has a large effect on the calculated coseismic deformations.Jie Dong Gabriele Cambiotti HanJiang Wen Roberto Sabadini WenKe Sun 2021Earth and Planetary Physics2021,5,1:2

12Reducing relapse and accelerating osteogenesis in rapid maxillary expansion using an injectable mesoporous bioactive glass/fibrin glue composite hydrogel显示文摘Rapid maxillary expansion(RME),as a common treatment for craniomaxillofacial deformity,faces the challenge of high relapse rates and unsatisfactory therapeutic effects.In this study,a standardized Sprague-Dawley(SD)rat RME model was first established with a modified expander as well as retainer design and optimized anterior maxillary expanding force of 100 g which exerted the most synchronized mobility of mid-palatal suture and incisors.Via the standardized model,the high relapse rate was proven to be attributed to insufficient osteogenesis in expanded suture,requiring long-term retainer wearing in clinical situations.To reduce the relapse rate,mesoporous bioactive glass/fibrin glue(MBG/FG)composite hydrogels were developed for an in situ minimal invasive injection that enhance osteogenesis in the expanded palate.The component of 1 wt%MBG was adopted for enhanced mechanical strength,matched degradation rate and ion dissolution,excellent in vitro biocompatibility and osteoinductivity.Effects of 1%MBG/FG composite hydrogel on osteogenesis in expanded mid-palatal sutures with/without retention were evaluated in the standardized model.The results demonstrated that injection of 1%MBG/FG composite hydrogel significantly promoted bone formation within the expanded mid-palatal suture,inhibited osteoclastogenesis and benefited the balance of bone remodeling towards osteogenesis.Combination of retainer and injectable biomaterial was demonstrated as a promising treatment to reduce relapse rate and enhance osteogenesis after RME.The model establishment and the composite hydrogel development in this article might provide new insight to other craniomaxillofacial deformity treatment and design of bone-repairing biomaterials with higher regenerative efficiency.Hanjiang Zhao Xiangyu Wang Anting Jin Minjiao Wang Zeying Wang Xingtai Huang Jiewen Dai Xudong Wang Dan Lin Steve GF.Shen 2022Bioactive Materials2022,7,12:2

18Effect of Rongchang capsule (茸菖胶囊) on seizure behavior,cognitive impairment,and hippocampal DNA damage and inflammatory factors in epileptic rats显示文摘OBJECTIVE: To observe the behavioral changes and changes in DNA fragments and related inflammatory factors in the hippocampus of epileptic rats pretreated with Rongchang capsule(茸菖胶囊).METHODS: Eighty Sprague-Dawley rats were randomly divided into the normal group(NG), model group(MG), sodium valproate group(VG), and Rongchang capsule group(RG)(n = 20 in each group). Pentylentetrazol was administered to the MG, VG, and RG to induce epilepsy. The VG and RG were pretreated with 1/2 the therapeutic dose of sodium valproate and Rongchang capsule, respectively. Changes in convulsion behavior and water maze learning were observed. Single cell gel electrophoresis was used to detect changes in the DNA in the hippocampus. The tail length(TL) and Olive tail moment(OTM) of cells were analyzed by GASP software. The expression of interleukin-1β(IL-1β),high mobility group box 1(HMGB1), transforming growth factor-β(TGF-β), and CCL4 in the hippocampus was determined by Western blotting.RESULTS: Rongchang capsule had a weaker effect on convulsive latency than sodium valproate, but significantly reduced seizure susceptibility. The spatial learning ability of the RG was better than that of the VG(P ≤ 0.01). The TL and OTM were significantly higher in the MG than the NG(P < 0.01). The RG had a better TL and OTM than the VG(P < 0.01).Combined with the microscopy results, DNA damage was most pronounced in the MG. Drug intervention decreased the DNA damage in the VG and RG. The expressions of IL-1β, CCL4, and HMGB1 in the hippocampus were significantly greater in the MG than the NG(P < 0.01), and were significantly reduced in the RG and VG compared with the MG(P < 0.01);however, there was no intergroup difference in the expression of TGF-β. The average values for the expression of inflammatory factors in the hippocampus were higher in the RG than in the VG;thus, Rongchang capsule may have a weaker effect on reducing the expression of inflammatory factors in the hippocampus than sodium valproate.CONCLUSION: Pretreatment with Rongchang capsule prevents or delays cognitive impairment in rats with induced epilepsy, reduces hippocampal DNA damage, and decreases the hippocampal expressions of IL-1β, CCL4, and HMGB1.PIAO Xiang WANG Wei CHEN Hanjiang YAN Rong LI Mengquan 2021Journal of Traditional Chinese Medicine2021,41,3:1

20A novel method for ionizing radiation-induced RNA damage detection by poly(A)-tailing RT-PCR显示文摘Ionizing radiation (IR) causes severe cellular damage both directly and indirectly and disrupts RNA integrity. RNA strand breaks are the most frequent type of damage caused by IR. RNA damage is involved in the development of degenerative diseases, including Alzheimer’s disease and Parkinson’s disease. However, the mechanism of mRNA damage and any resulting pathophysiological outcomes are poorly understood. This is partly because there is a lack of sensitive tools to monitor damage randomly occurring in RNA, especially RNA strand break damage in a given RNA. In this work, a method using the reverse transcription polymerase chain reaction (RT-PCR) after poly(A) addition to 3′-end of RNA to determine RNA strand break damage in a specific RNA by poly(A) polymerase has been developed. The levels of damage in specific mRNAs, including ABL1, TP53, GADD45A and ATR from IR-treated HeLa cells were examined. Strand breaks were detected in all mRNAs examined.
