| 1 | A Kelch Motif-Containing Serine/Threonine Protein Phosphatase Determines the Large Grain QTL Trait in Rice显示文摘A thorough understanding of the genetic basis of rice grain traits is critical for the improvement of rice (Oryza sativa L.) varieties.In this study,we generated an F 2 population by crossing the large-grain japonica cultivar CW23 with Peiai 64 (PA64),an elite indica small-grain cultivar.Using QTL analysis,17 QTLs for five grain traits were detected on four different chromosomes.Eight of the QTLs were newly-identified in this study.In particular,qGL3-1,a newly-identified grain length QTL with the highest LOD value and largest phenotypic variation,was fine-mapped to the 17 kb region of chromosome 3.A serine/threonine protein phosphatase gene encoding a repeat domain containing two Kelch motifs was identified as the unique candidate gene corresponding to this QTL.A comparison of PA64 and CW23 sequences revealed a single nucleotide substitution (C→A) at position 1092 in exon 10,resulting in replacement of Asp (D) in PA64 with Glu (E) in CW23 for the 364th amino acid.This variation is located at the D position of the conserved sequence motif AVLDT of the Kelch repeat.Genetic analysis of a near-isogenic line (NIL) for qGL3-1 revealed that the allele qGL3-1 from CW23 has an additive or partly dominant effect,and is suitable for use in molecular marker-assisted selection. | Zejun Hu Haohua He Shiyong Zhang Fan Sun Xiaoyun Xin Wenxiang Wang Xi Qian Jingshui Yang Xiaojin Luo | 2012 | Journal of Integrative Plant Biology2012,54,12: | 41 |

| 3 | Nasal delivery of broadly neutralizing antibodies protects mice from lethal challenge with SARS-CoV-2 delta and omicron variants显示文摘Multiple new variants of severe acute respiratory syndrome coronavirus 2(SARS-Co V-2)have constantly emerged,as the delta and omicron variants,which have developed resistance to currently gained neutralizing antibodies.This highlights a critical need to discover new therapeutic agents to overcome the variants mutations.Despite the availability of vaccines against coronavirus disease 2019(COVID-19),the use of broadly neutralizing antibodies has been considered as an alternative way for the prevention or treatment of SARS-Co V-2 variants infection.Here,we show that the nasal delivery of two previously characterized broadly neutralizing antibodies(F61 and H121)protected K18-h ACE2 mice against lethal challenge with SARS-Co V-2 variants.The broadly protective efficacy of the F61 or F61/F121 cocktail antibodies was evaluated by lethal challenge with the wild strain(WIV04)and multiple variants,including beta(B.1.351),delta(B.1.617.2),and omicron(B.1.1.529)at 200or 1000 TCID_(50),and the minimum antibody administration doses(5-1.25 mg/kg body weight)were also evaluated with delta and omicron challenge.Fully prophylactic protections were found in all challenged groups with both F61 and F61/H121 combination at the administration dose of 20 mg/kg body weight,and corresponding mice lung viral RNA showed negative,with almost all alveolar septa and cavities remaining normal.Furthermore,low-dose antibody treatment induced significant prophylactic protection against lethal challenge with delta and omicron variants,whereas the F61/H121 combination showed excellent results against omicron infection.Our findings indicated the potential use of broadly neutralizing monoclonal antibodies as prophylactic and therapeutic agent for protection of current emerged SARS-Co V-2 variants infection. | Jia Lu Qiangling Yin Rongjuan Pei Qiu Zhang Yuanyuan Qu Yongbing Pan Lina Sun Ding Gao Cuiqin Liang Jingwen Yang Wei Wu Jiandong Li Zongqiang Cui Zejun Wang Xinguo Li Dexin Li Shiwen Wang Kai Duan Wuxiang Guan Mifang Liang Xiaoming Yang | 2022 | Virologica Sinica2022,37,2: | 4 |

| 6 | Scattered seeding of CAR T cells in solid tumors augments anticancer efficacy显示文摘Chimeric antigen receptor T cell(CAR T) therapy was a milestone in the treatment of relapsed and refractory B cell malignancies. However, beneficial effects of CAR T cells have not been obtained in solid tumors yet. Herein, we implement a porous microneedle patch that accommodates CAR T cells and allows in situ penetration-mediated seeding of CAR T cells when implanted in the tumor bed or in the post-surgical resection cavity. CAR T cells loaded in the pores of the microneedle tips were readily escorted to the tumor in an evenly scattered manner without losing their activity. Such microneedle-mediated local delivery enhanced infiltration and immunostimulation of CAR T cells as compared to direct intratumoral injection. | 7 | Subcellular Localization of Large Yellow Croaker(Larimichthys crocea) TLR21 and Expression Profiling of Its Gene in Immune Response显示文摘Toll-like receptor 21(TLR21) is a non-mammalian type TLR, and plays an important role in innate immune response in fish. In this paper, the full-length cDNA sequence of TLR21 gene was identified and characterized from large yellow croaker, Larimichthys crocea and was termed as LcTLR21. It consists of 3365 bp, including a 5'-terminal untranslated region(UTR) of 97 bp, a 3'-terminal UTR of 331 bp, and an open reading frame(ORF) of 2937 bp encoding a polypeptide of 978 amino acid residues. The deduced LcTLR21 contains a signal peptide domain at N-terminal, 12 leucine-rich repeats(LRRs) at the extracellular region, a transmembrane domain and a cytoplasmic toll-interleukin-1 receptor(TIR) domain at the C-terminal. Subcellular localization analysis revealed that the LcTLR21-GFP was constitutively expressed in cytoplasm. Tissue expression analysis indicated that LcTLR21 gene broadly expressed in most of the examined tissues, with the most predominant abundance in spleen, followed by head-kidney and liver, while the weakest expression was detected in brain. The expression level of LcTLR21 after LPS, poly I:C and Vibrio parahaemolyticus challenges was investigated in spleen, head-kidney and liver. | 9 | Temporal and spatial stability of the EM/PM molecular subtypes in adult diffuse glioma显示文摘Detailed characterizations of genomic alterations have not identified subtype-specific vulnerabilities in adult gliomas. Mapping gliomas into developmental programs may uncover new vulnerabilities that are not strictly related to genomic alterations. After identifying conserved gene modules co-expressed with EGFR or PDGFRA (EM or PM), we recently proposed an EM/PM classification scheme for adult gliomas in a histological subtype- and grade-independent manner. By using cohorts of bulk samples, paired primary and recurrent samples, multi-region samples from the same glioma, single-cell RNA-seq samples, and clinical samples, we here demonstrate the temporal and spatial stability of the EM and PM subtypes. The EM and PM subtypes, which progress in a subtype-specific mode, are robustly maintained in paired longitudinal samples. Elevated activities of cell proliferation, genomic instability and microenvironment, rather than subtype switching, mark recurrent gliomas. Within individual gliomas, the EM/PM subtype was preserved across regions and single cells. Malignant cells in the EM and PM gliomas were correlated to neural stem cell and oligodendrocyte progenitor cell compartment, respectively. Thus, while genetic makeup may change during progression and/or within different tumor areas, adult gliomas evolve within a neurodevelopmental framework of the EM and PM molecular subtypes. | 12 | Involvement of triadimefon induced early ABA-dependent H2O2 accumulation in soybean against water stress显示文摘The present study showed that pretreatment of triadimefon(TDM),a triazole compound,could improve tolerance of soybean seedlings to subsequent water stress.TDM pretreatment resulted in early and late rise in superoxide dismutase(SOD)and catalase(CAT)activities,and upregulation of ascorbate(As A)content in non-stressed and water-stressed seedlings,leading to late increase in net photosynthetic rate(Pn),late decrease in hydrogen peroxide(H2O2)and electrolyte leakage in stressed ones.These TDM-induced changes were blocked by application of abscisic acid(ABA)biosynthesis inhibitor tungstate,which inhibited early rise of ABA and H2O2contents in non-stressed and stressed seedlings.However,ABA pretreatment overcomed the effects of this inhibitor.Application of NADPH oxidase inhibitor diphenyleneiodonium(DPI),polyamine oxidase(PAO)inhibitor 2-hydroxyethylhydrazine(2-HEH)and H2O2scavenger dimethylthiourea(DMTU)prevented early TDM-induced rise of H2O2content.DPI,2-HEH and DMTU also decreased SOD,CAT and As A levels,but did not affect ABA content during early and late phases in both seedlings pretreated with TDM.In addition,these chemicals decreased Pn,and increased H2O2content and electrolyte leakage during late phase in TDM-pretreated stressed seedlings.Overall,these results indicated that TDM pretreatment alleviated adverse effects of water stress on soybean seedlings,which was at least in part,due to increase of antioxidant capacity and decrease of oxidative damage induced by early ABA-dependent H2O2generation. | Zejun Xu Fei Tong Yujun Qi Donglei Sun Nengfei Bian Xing Wang | 2020 | Oil Crop Science2020,5,1: | 0 |
| 13 | TATA-box binding protein-associated factor 2 regulates grain size in rice显示文摘Grain size,characterized by a combination of grain length,width,and thickness,is one of the major determinants of yield in rice.The present study identified TATA-box binding protein-associated factor 2(TAF2)as an essential component regulating transcription and determining grain size in rice.Map-based cloning showed that a G/T substitution in TAF2 resulted in a naturally occurring mutant called reduced grain size and plant height 1(rgh1).The mutants,with weak edited rgh1 alleles,exhibited a small grain phenotype with reduced grain length and width,while the severe knockout mutant(rgh1-2s)was dwarf and completely sterile.Allelic test performed between rgh1 and several edited alleles confirmed that the mutation in TAF2 caused the rgh1 phenotype.GUS staining showed that TAF2 was mainly expressed in the vascular bundles of roots,stems,leaves,and grains.The cytological analysis revealed that reduced cell division in the glumes resulted in the small grain phenotype of rgh1.Further RNA-sequencing detected altered expression of genes involved in the basic biological processes in rgh1 mutant.These findings provide novel insights into the TAF2-mediated genetic mechanism regulating grain size in rice. | Ling Jiang Ning Jiang Zejun Hu Xuejun Sun Xian Xiang Yahui Liu Mingwei Wu Chunming Liu Xiaojin Luo | 2023 | The Crop Journal2023,11,2: | 0 |
