1Chinese deserts and sand fields in Last Glacial Maximum and Holocene Optimum显示文摘The Last Glacial Maximum (LGM, c. 26-16 ka) and the Holocene Optimum (HO, c. 9-5 ka) were characterized by cold-dry and warm-wet climates respectively in the recently geological Earth. How Chinese deserts and sand fields responded to these distinctive climatic changes is still not clear, however. To reconstruct environments of the deserts and sand fields during the LGM and HO is helpful to understand the forcing mechanisms of environment change in this arid region, and to test paleoclimatic modeling results. Through our long-term field and laboratory investigations, 400 optically stimulated luminescence (OSL) ages and more than 100 depositional records in the Chinese deserts and sand fields were obtained; on the basis of these data, we reconstruct spatial distributions of the deserts and sand fields during the LGM and HO. Our results show that the sand fields of Mu Us, Hunshandake, Horqin and Hulun Buir in northern and northeastern China had expanded 25%, 37%, 38% and 270%, respectively, during the LGM; the sand fields of Gonghe in the northeastern Qinghai-Tibetan Plateau had expanded 20%, and the deserts of Badain Jaran, Tengger in central northern China had expanded 39% and 29% separately during the LGM; the deserts of Taklimakan, Gurbantünggüt and Kumtag in northwestern China had expanded 10%-20% respectively, compared to their modern areas. On the other hand, all of the sand fields were nearly completely covered by vegetation during the HO; the deserts in northwestern and central northern China were reduced by around 5%-20% in area during this time. Lakes in this arid region were probably expanded during the HO but this conclusion needs more investigation. Compared with the geological distributions of deserts and sand fields, human activity has clearly changed (expanded) the area of active sand dunes at the present time. 5Screening for Y chromosome microdeletions in idiopathic and nonidiopathic infertile men with varicocele and cryptorchidism显示文摘Background Cytogenetic and molecular studies of azoospermic and oligozoospermic males have suggested thepresence of azoospermia factors (AZF) in the Y chromosome.Deletion in AZF regions has been reported todisrupt spermatogenesis and cause infertility.Several candidate genes responsible for spermatogenesis have beenidentified in this region and some of them are thought to be functional in human spermatogenesis.And wereported clinical and molecular studies of Ychromosome microdeletions in Chinese.This study aimed atassessingthe frequency of microdeletions in Chinese men with idiopathic and nonidiopathic infertility problems anddicussing the clinical significance of the AZF region.Methods In this study, we screened 143 infertile men (62 with idiopathic infertilitas and 81 withnonidiopathic infertilitas), in whom karyotype, sperm count, hormonal parameters and fine needle aspirationcytology were evaluated.Genomic DNA was extracted from the peripheral leukocytes.Molecular analysis wasperformed by two multiplexpolymerase chain reactions (PCR) using a set of a sequence tagged sites (STS) from3 different regions of the Y chromosome: AZFa (sY84, sY86), AZFb (sY127, sY134), AZFc (sY254,sY255).Results Nineteen point four percent of idiopathic males (12/62, 19.4%) had microdeletions of either theAZFa, AZFb, AZFc or AZFb +c region.Significantly, a high frequency of microdeletions (9/81, 11.1%) wasfound in nonidiopathic patients with varicocele and cryptorchidism.No deletions were found in healthy fertilemen.There were no significant differences in the localization and extent of deletions between idiopathic andnonidiopathic patients.Conclusions The knowledge of the presence of these deletions in idiopathic and nonidiopathic cases isimportant to understand the prognosis, better management and counsel these patients accordingly.Furthermore,a more extended screening for Y chromosome microdeletions in idiopathic and nonidiopathic men, particularlycandidates for intracytoplasmic sperm injection, is recommended.SONG Ning-hong WU Hong-fei ZHANG Wei ZHUO Zuo-min QIAN Li-xing HUA Li-xing GUO Lin FENG Ning-han 2005Chinese Medical Journal2005,,17:12

9Investigation of the interaction between two phenylethanoid glycosides and bovine serum albumin by spectroscopic methods显示文摘Two phenylethanoid glycosides, acteoside and forsythoside B, were first isolated from the traditional Chinese herb Callicarpa peii H.T. Chang. The interaction between the two phenylethanoid glycosides and bovine serum albumin (BSA) was investigated by fluorescence, UV-vis absorbance and circular dichroism (CD). The results showed that the quenching mechanism in the drug-BSA binary systems was a combination of static quenching and non-radiative energy transfer. Displacement experiments confirmed that the drug bound to the site I of BSA. 13Large-conductance Ca^2+-activated K^+ channel involvement in suppression of cerebral ischemia/reperfusion injury after electroacupuncture at Shuigou(GV26) acupoint in rats显示文摘Excess activation and expression of large-conductance Ca^(2+)-activated K^+ channels(BKCa channels) may be an important mechanism for delayed neuronal death after cerebral ischemia/reperfusion injury. Electroacupuncture can regulate BKCa channels after cerebral ischemia/reperfusion injury, but the precise mechanism remains unclear. In this study, we established a rat model of cerebral ischemia/reperfusion injury. Model rats received electroacupuncture of 1 m A and 2 Hz at Shuigou(GV26) for 10 minutes, once every 12 hours for a total of six times in 72 hours. We found that in cerebral ischemia/reperfusion injury rats, ischemic changes in the cerebral cortex were mitigated after electroacupuncture. Moreover, BKCa channel protein and m RNA expression were reduced in the cerebral cortex and neurological function noticeably improved. These changes did not occur after electroacupuncture at a non-acupoint(5 mm lateral to the left side of Shuigou). Thus, our findings indicate that electroacupuncture at Shuigou improves neurological function in rats following cerebral ischemia/reperfusion injury, and may be associated with down-regulation of BKCa channel protein and m RNA expression. 14Effects of lipid-lowering agents on diabetic retinopathy: a Meta-analysis and systematic review显示文摘AIM: To clarify this controversy and to provide evidence for application of lipid lowering agents in treatment of diabetic retinopathy(DR).METHODS: We searched the databases of Pub Med, Embase and Cochrane Library Central Register of Controlled Trials(CENTRAL) and abstracts from main annual meetings up to January 1, 2017. Google scholar and ClinicalT rials.gov were also searched for unpublished relevant studies. We included randomized controlled trials(RCTs) that studied lipid-lowering agents in type 1 or type 2 diabetes in this Meta-analysis. The primary endpoint was the progression of DR, and the secondary endpoints included vision loss, development of diabetic macular edema(DME) and aggravation of hard exudates. The pooled odds ratios(OR)with corresponding 95% confidence intervals(95%CIs) were calculated.RESULTS: After systemic and manual literature search by two independent investigators, we included 8 RCTs from 7 published articles with 13 454 participants in this Meta-analysis. The results revealed that lipid-lowering drugs were associated with reduced risk in DR progression [OR=0.77(95%CI: 0.62, 0.96), P=0.02]. Lipid-lowering agents might have protective effect on DME compared to placebo, although the difference was not statistically significant [OR=0.60(95%CI: 0.34, 1.08), P=0.09]. However, no significant differences in the worsening of vision acuity [OR=0.96(95%CI: 0.81,1.14), P=0.64] and hard exudates [OR=0.50(95%CI:0.15, 1.74), P=0.28] were found between the lipidlowering drugs and the placebo groups.CONCLUSION: In DR patients, lipid-lowering agents show a protective effect on DR progression and might be associated with reduced risk in the development of DME. However, lipid-lowering agents have no effects on vision loss and hard exudates aggravation. 16Effects of dietary supplementation with essential oils and protease on growth performance,antioxidation,inflammation and intestinal function of weaned pigs显示文摘This experiment evaluated the impacts of essential oils(EO)and protease(PRO),independently or in combination,on growth performance,antioxidation,inflammation and intestinal function of weaned pigs.One hundred and sixty weaned pigs(21 d of age,BW of 6.74±0.20 kg)were randomly divided into 4 treatments with 8 replicate pens of 5 pigs per pen.Dietary treatments included the following:1)control diet(CON),2)CON with 300 mg/kg essential oils(EO),3)CON with 500 mg/kg protease(PRO),4)CON with 300 mg/kg essential oil and 500 mg/kg protease(EO + PRO).On d 8,one pig from each penwas selected for sampling.The remaining pigs were fed for an additional week and growth performance was monitored during this period.Dietary treatments had no marked effects(P>0.05)on the growth performance of pigs.However,pigs receiving EO diet had higher(P<0.05)serumglutathione peroxidase(GSH-Px)activity,and tended to decrease(P=0.063)serum concentration of tumor necrosis factor-a(TNF-a).In addition,pigs receiving EO diet had higher(P<0.05)abundances of phylum Actinobacteria,and genera Bifidobacterium,and lower(P<0.05)phylum Bacteroidetes and genera Alloprevotella in colonic digesta.Pigs receiving PRO diet decreased(P<0.05)the serum concentration of malondialdehyde(MDA)and diamine oxidase activity,increased(P<0.05)the villus height and the ratio of villus height to crypt depth in duodenum,increased sucrase activity in jejunal mucosa,and also increased the abundance of phylum Actinobacteria in colonic digesta.Furthermore,the synergistic effects of EO and PROwas observed(P<0.05)for pigs with decreasing serum TNF-a concentration and increasing serum GSH-Px activity.Collectively,the results indicated that dietary supplementation of EO and PRO had no significant effects on growth performance of weaned pigs.EO diet appeared to improve antioxidant activity and intestinal microbiota,while PRO diet improved intestinal morphology and digestive enzyme activity,and there was a synergistic effect of EO and PRO on reducing inflammatory parameters in weaned pigs.Xie Peng Qiang Zhou Cheng Wu Jian Zhao Quan Tan Ying He Liang Hu Zhengfeng Fang Yan Lin Shengyu Xu Bin Feng Jian Li Yong Zhuo Chris Van Ginneken Xuemei Jiang De Wu Lianqiang Che 2022Animal Nutrition2022,,2:8
17Construct hepatic analog by cell-matrix controlled assembly technology显示文摘A mixture of hepatic cells and chito- san/gelatin solution was deposited to construct a hepatic analog by way of layer-by-layer deposition technique using a home-made devise. The size and cell concentration of the analogs can be controlled freely. Approximately 90% of the hepatic cells re- mained viable under 0.2 Mpa extrusion pressure. Cultured in vitro 8 weeks before animal test, hepatic cells in structure maintained their phenotype and kept proliferating, and albumin and other secretion of the cells increased. Cords and hepaton-like structures were observed after culture for 20 d. These results indicate that hepatic cells could be assembled di- rectly into a 3D viable structure and expanded to form a hepatic organoid. 18A Quantitative Evaluation of Shale Gas Content in Different Occurrence States of the Longmaxi Formation: A New Insight from Well JY-A in the Fuling Shale Gas Field,Sichuan Basin显示文摘Comprehensive quantitative evaluation of shale gas content and the controlling factors in different occurrence states is of great significance for accurately assessing gas-bearing capacity and providing effective well-production strategies. A total of 122 core samples from well JY-A in the Fuling shale gas field were studied to reveal the characteristics of S_1 l shale,15 of which were selected to further predict the shale gas content in different occurrence states, which are dependent on geological factors in the thermal evolution process. Geological parameters were researched by a number of laboratory programs, and the factors influential in controlling shale gas content were extracted by both PCA and GRA methods and prediction models were confirmed by the BE method using SPSS software. Results reveal that the adsorbed gas content is mainly controlled by TOC, Ro, SSA, PD and pyrite content, and the free gas content is mainly controlled by S_2, quartz content, gas saturation and formation pressure for S_1 l in well JY-A. Three methods, including the on-site gas desorption method, the empirical formula method, and the multiple regression analysis method were used in combination to evaluate the shale gas capacity of well JY-A, all of which show that the overall shale gas content of well JY-A is in the range of 2.0–5.0 m^3/t and that the free gas ratio is about 50%, lower than that of well JY-1. 20Over-expression of the AtGA2ox8 gene decreases the biomass accumulation and lignification in rapeseed (Brassica napus L.)显示文摘Gibberellin 2-oxidase (GA 2-oxidase) plays very important roles in plant growth and development. In this study, the AtGA2ox8 gene, derived from Arabidopsis (Arabidopsis thaliana), was transformed and over-expressed in rapeseed (Brassica napus L.) to assess the role of AtGA2ox8 in biomass accumulation and lignification in plants. The transgenic plants, identified by resistant selection, polymerase chain reaction (PCR) and reverse-transcription PCR (RT-PCR) analyses, and green fluorescence examination, showed growth retardation, flowering delay, and dwarf stature. The fresh weight and dry weight in transgenic lines were about 21% and 29% lower than those in wild type (WT), respectively, and the fresh to dry weight ratios were higher than that of WT. Quantitative measurements demonstrated that the lignin content in transgenic lines decreased by 10%-20%, and histochemical staining results also showed reduced lignification in transgenic lines. Quantitative real-time PCR analysis indicated that the transcript levels of lignin biosynthetic genes in transgenic lines were markedly decreased and were consistent with the reduced lignification.
