1High expression of ACE2 receptor of 2019-nCoV on the epithelial cells of oral mucosa显示文摘It has been reported that ACE2 is the main host cell receptor of 2019-nCoV and plays a crucial role in the entry of virus into the cell to cause the final infection. To investigate the potential route of 2019-n Cov infection on the mucosa of oral cavity, bulk RNA-seq profiles from two public databases including The Cancer Genome Atlas(TCGA) and Functional Annotation of The Mammalian Genome Cap Analysis of Gene Expression(FANTOM5 CAGE) dataset were collected. RNA-seq profiling data of 13 organ types with para-carcinoma normal tissues from TCGA and 14 organ types with normal tissues from FANTOM5 CAGE were analyzed in order to explore and validate the expression of ACE2 on the mucosa of oral cavity. Further, single-cell transcriptomes from an independent data generated in-house were used to identify and confirm the ACE2-expressing cell composition and proportion in oral cavity. The results demonstrated that the ACE2 expressed on the mucosa of oral cavity. Interestingly, this receptor was highly enriched in epithelial cells of tongue. 4Single-cell RNA-seq highlights intra-tumoral heterogeneity and malignant progression in pancreatic ductal adenocarcinoma显示文摘Pancreatic ductal adenocarcinoma (PDAC) is the most common type of pancreatic cancer featured with high intra-tumoral heterogeneity and poor prognosis. To comprehensively delineate the PDAC intra-tumoral heterogeneity and the underlying mechanism for PDAC progression, we employed single-cell RNA-seq (scRNA-seq) to acquire the transcriptomic atlas of 57,530 individual pancreatic cells from primary PDAC tumors and control pancreases, and identified diverse malignant and stromal cell types, including two ductal subtypes with abnormal and malignant gene expression profiles respectively, in PDAC. We found that the heterogenous malignant subtype was composed of several subpopulations with differential proliferative and migratory potentials. Cell trajectory analysis revealed that components of multiple tumor-related pathways and transcription factors (TFs) were differentially expressed along PDAC progression. Furthermore, we found a subset of ductal cells with unique proliferative features were associated with an inactivation state in tumor-infiltrating T cells, providing novel markers for the prediction of antitumor immune response. 7New Collection of Crude Drugs in Chinese Pharmacopoeia 2010 Ⅰ.Callicarpa Linn. and Related Items显示文摘Callicarpa Linn. (beautyberry) is one of the major genera in Verbenaceae, about 20 of which are medicinal plants. Beautyberry, called Zizhu in China, is a generic name of those species and largely used as hemostatic medicine. The Chinese Pharmacopoeia 2010 has admitted three new crude drugs from the genus of Callicarpa Linn. including Callicarpae Macrophyllae Folium, Callicarpae Caulis et Folium, and Callicarpae Formosanae Folium for the first time since the 1977 version of the Chinese Pharmacopoeia. In order to better understand these new crude drugs, we systematically described their bibliography, admission reasons, botanical identification, chemistry, and pharmacology. 8MicroRNA-143 suppresses gastric cancer cell growth and induces apoptosis by targeting COX-2显示文摘AIM:To investigate the function of microRNA-143(miR-143)in gastric cancer and explore the target genes of miR-143.METHODS:A quantitative real-time reverse transcription-polymerase chain reaction(RT-PCR)analysis was performed to evaluate miR-143 expression in gastric cancer cell lines.After transfecting gastric cancer cells with miR-143-5p and miR-143-3p precursors,Alamar blue and apoptosis assays were used to measure the respective proliferation and apoptosis rates.Cyclooxygenase-2(COX-2)expression was determined by realtime RT-PCR and Western blot assays after miR-143transfection.Reporter plasmids were constructed,and a luciferase reporter assay was used to identify the miR-143 binding site on COX-2.RESULTS:Both miR-143-5p and miR-143-3p were significantly downregulated in multiple gastric cancer cell lines.Forced miR-143-5p and miR-143-3p expression in gastric cancer cells produced a profound cytotoxic effect.MiR-145-5p transfection into gastric cancer cells resulted in a greater growth inhibitory effect(61.23%±3.16%vs 46.58%±4.28%,P<0.05 in the MKN-1cell line)and a higher apoptosis rate(28.74%±1.93%vs 22.13%±3.31%,P<0.05 in the MKN-1 cell line)than miR-143-3p transfection.Further analysis indicated that COX-2 expression was potently suppressed by miR-143-5p but not by miR-143-3p.The activity of a luciferase reporter construct that contained the 3’-untranslated region(UTR)of COX-2 was downregulated by miR-143-5p(43.6%±4.86%,P<0.01)but not by miR-143-3p.A mutation in the miR-145-5p binding site completely ablated the regulatory effect on luciferase activity,which suggests that there is a direct miR-145-5p binding site in the 3’-UTR of COX-2.CONCLUSION:Both miR-143-5p and miR-143-3p function as anti-oncomirs in gastric cancer.However,miR-143-5p alone directly targets COX-2,and it exhibits a stronger tumor suppressive effect than miR-143-3p.Xiao-Li Wu Bin Cheng Pei-Yuan Li Huan-Jun Huang Qiu Zhao Zi-Li Dan Dean Tian Peng Zhang 2013World Journal of Gastroenterology2013,19,43:15

11Typical atmospheric haze during crop harvest season in northeastern China:A case in the Changchun region显示文摘This study presents the mass concentrations of PM_(2.5),O_3,SO_2 and NOxat one urban,one suburban and two rural locations in the Changchun region from September 25 to October 272013. Major chemical components of PM_(2.5)at the four sites were daily sampled and analyzed. Most of daily concentrations of SO_2(7–82 μg/m^3),O_3(27–171 μg/m^3) and NOx(14–213 μg/m^3) were below the limits of the National Ambient Air Quality Standard(NAAQS)in China. However,PM_(2.5)concentrations(143–168 μg/m^3) were 2-fold higher than NAAQS.Higher PM_(2.5)concentrations(~ 150 μg/m^3) were measured during the pre-harvest and harvest at the urban site,while PM_(2.5)concentrations significantly increased from 250 to400 μg m^(-3) at suburban and rural sites with widespread biomass burning. At all sites,PM_(2.5)components were dominated by organic carbon(OC) and followed by soluble component sulfate(SO_4^(2-)),ammonium(NH_4^+) and nitrate(NO_3^-). Compared with rural sites,urban site had a higher mineral contribution and lower potassium(K^+and K) contribution to PM_(2.5).Severe atmospheric haze events that occurred from October 21 to 23 were attributed to strong source emissions(e.g.,biomass burning) and unfavorable air diffusion conditions.Furthermore,coal burning originating from winter heating supply beginning on October 18 increased the atmospheric pollutant emissions. 12Association of NOD1 and NOD2 genes polymorphisms with Helicobacter pylori related gastric cancer in a Chinese population显示文摘AIM:To investigate the association between the tag single nucleotide polymorphisms(TagSNPs) of NOD1 and NOD2 and the risk of developing gastric cancer.METHODS:We conducted a hospital-based case-control study including 296 incident gastric cancer patients and 160 gastritis controls.Eight TagSNPs in the NOD1 and NOD2 genes were selected from the Hapmap database using the haploview software and genotyped by the Sequenom MassArray system.The serum levels of anti-Helicobacter pylori(H.pylori) IgG were measured by enzyme-linked immunosorbent assay to indicate H.pylori infection.The odds ratios(OR) and 95% confidence intervals(CI) were calculated by unconditional logistic regression,including sex and age as confounding factors.RESULTS:The NOD1 rs2907749 GG genotype showed a decreased risk for gastric cancer(OR 0.50,95% CI:0.26-0.95,P = 0.04) while the rs7789045 TT genotype showed an increased risk(OR 2.14,95% CI:1.20-3.82,P = 0.01).An elevated susceptibility to gastric cancer was observed in the subjects with H.pylori infection and the NaOD1 rs7789045 TT genotype(OR 2.05,95% CI:1.07-3.94,P = 0.03) or the NOD2 rs7205423 GC genotype(OR 2.52,95% CI:1.05-6.04,P = 0.04).Haplotype analysis suggested that the distribution of AGT(rs2907749,rs2075820 and rs7789045) in NOD1 between the cases and control groups was significantly different(P corrected:0.04),and the diplotype AGT/AGT was associated with an elevated gastric cancer risk(OR 1.98,95% CI:1.04-3.79,P = 0.04).The association of the NOD1 rs7789045 TT genotype and the diplotype AGT/AGT was significant with H.pylori-related diffuse-type gastric cancer(OR 3.00,95% CI:1.38-6.53,P = 0.01;OR 4.02,95% CI:1.61-10.05,P < 0.01,respectively).CONCLUSION:Genetic polymorphisms in NOD1 and NOD2 may interact with H.pylori infection and may play important roles in promoting the development of gastric cancer in the Chinese population.Peng Wang Li Zhang Jian-Ming Jiang Dan Ma Hao-Xia Tao Sheng-Ling Yuan Yan-Chun Wang Ling-Chun Wang Hao Liang Zhao-Shan Zhang Chun-Jie Liu 2012World Journal of Gastroenterology2012,18,17:9

14Clue to a thorough understanding of terahertz pulse generation by femtosecond laser filamentation显示文摘In this work, it has been demonstrated that in order to fully understand the terahertz(THz) pulse generation process during femtosecond laser filamentation, the interaction between THz wave and air plasma has to be taken into account. This interaction is mainly associated with the spatial confinement of the THz pulse by the plasma column, which could be described by the one-dimensional negative dielectric(1DND) waveguide model. By combining the 1 DND model with the conventional four-wave mixing(4WM) and photocurrent(PC) models,the variation of THz spectral amplitude and width obtained in experiments could be better understood. 15In vivo and in vitro effects of QHF combined with chemotherapy on hepatocellular carcinoma显示文摘Objective:To investigate the synergistic anti-tumor effect of QHF (a Chinese medicine formula with antitumor active ingredients, including 800 mg/kg Cinobufotalin, 14 mg/kg Ginsenoside Rg3, 5.5 mg/kg Notoginseng and 100 mg/kg Lentinan) when combined with the chemotherapy drug cisplatin (DDP). Methods: Hepatocellular carcinoma H22 cells were implanted into mice and after the transplants were successfully established the animals were divided into four groups, namely a normal saline(NS) control group, QHF group, DDP group and QHF+DDP group. The tumor growth was monitored and the survival time determined. In vitro studies employing H22 cells used the first three groups, and determined the effects of QHF and DDP on tumor cell cycle distribution, apoptosis and morphologic changes in vitro. Results: QHF significantly inhibited the growth of tumors and prolonged the survival time of mice with hepatocellular carcinomas. QHF combined with DDP could attenuate DDP-induced leucopenia, spleen and thymus atrophy and other indicators of toxicity. The inhibition rate of tumor growth reached 82.54% with QHF+DDP, and QHF prolonged the life span of DDP-treated mice by 66.83%. In the in vitro experiments tumor cells showed morphological changes characteristic of apoptosis by both light and transmission electron microscopy in the QHF group, and the apoptosis rate was 33.85%. Moreover, the proportion of cells in the G0/G1 phase was increased and those in the S-phase decreased. 16Cross-correlation matrix Root-MUSIC algorithm for bistatic multiple-input multiple-output radar显示文摘A novel Root-MUSIC algorithm based on cross-correlation matrix is proposed to estimate the direction of arrival(DOA) and direction of departure(DOD) for the bistatic multiple-input multiple-output(MIMO)radar. In the algorithm, the cross-correlation matrix is first constructed by using the received data which are arranged according to the transmit array and the receive array, respectively. And the DOAs and DODs are estimated independently employing cross-correlation Root-MUSIC algorithms without eigendecomposition,where the null spaces are constructed with a linear operation of the cross-correlation matrices. Secondly the pair-matching of DOAs and DODs is accomplished by solving an angle estimate optimization. 17RNA-binding protein LIN28B inhibits apoptosis through regulation of the AKT2/FOXO3A/BIM axis in ovarian cancer cells显示文摘LIN28B is an evolutionarily conserved RNA-binding protein that regulates mRNA translation and miRNA let-7 maturation in embryonic stem cells and developing tissues.Increasing evidence demonstrates that LIN28B is activated in cancer and serves as a critical oncogene.However,the underlying molecular mechanisms of LIN28B function in tumorigenesis are still largely unknown.Here we report that LIN28B was expressed in over half of the patients with epithelial ovarian cancer who were examined(n=584).Functional experiments demonstrated that LIN28B inhibited ovarian cancer cell apoptosis.Furthermore,we showed that the proapoptotic factor BIM played an essential role in the antiapoptotic function of LIN28B.RNA-IP microarray analysis suggested that LIN28B binds to mRNAs that are associated with the DNA damage pathway,such as AKT2,in ovarian cancer cells.By binding to AKT2 mRNA and enhancing its protein expression,LIN28B regulated FOXO3A protein phosphorylation and decreased the transcriptional level of BIM,which antagonized the antiapoptosis activity of LIN28B.Taken together,these results mechanistically linked LIN28B and the AKT2/FOXO3A/BIM axis to the apoptosis pathway.The findings may have important implications in the diagnosis and therapeutics of ovarian cancer.Xiaojuan Lin Jianfeng Shen Dan Peng Xinhong He Congjian Xu Xiaojun Chen Janos LTanyi Kathleen Montone Yi Fan Qihong Huang Lin Zhang Xiaomin Zhong 2018Signal Transduction and Targeted Therapy2018,3,1:8
18A deep learning-integrated micro-CT image analysis pipeline for quantifying rice lodging resistance-related traits显示文摘Lodging is a common problemin rice,reducing its yield andmechanical harvesting efficiency.Rice architecture is a key aspect of its domestication and a major factor that limits its high productivity.The ideal rice culm structure,includingmajor_axis_culm,minor axis_culm,andwall thickness_culm,is critical for improving lodging resistance.However,the traditionalmethod ofmeasuring rice culms is destructive,time consuming,and labor intensive.In this study,we used a high-throughput micro-CT-RGB imaging system and deep learning(SegNet)todevelopa high-throughputmicro-CTimageanalysis pipelinethatcanextract 24 riceculmmorphological traits and lodging resistance-related traits.When manual and automatic measurements were compared at themature stage,the mean absolute percentage errors for major_axis_culm,minor_axis_culm,andwall_thickness_culmin 104 indica rice accessionswere 6.03%,5.60%,and 9.85%,respectively,and the R^(2) valueswere 0.799,0.818,and 0.623.We also builtmodels of bending stress using culmtraits at the mature and tillering stages,and the R^(2) values were 0.722 and 0.544,respectively.The modeling results indicated that this method can quantify lodging resistance nondestructively,even at an early growth stage.In addition,we also evaluated the relationships of bending stress toshoot dryweight,culm density,and drought-related traits and found that plants with greater resistance to bending stress had slightly higher biomass,culm density,and culm area but poorer drought resistance.In conclusion,we developed a deep learning-integrated micro-CT image analysis pipeline to accurately quantify the phenotypic traits of rice culms in4.6 min per plant;this pipeline will assist in future high-throughput screening of large rice populations for lodging resistance.Di Wu Dan Wu Hui Feng Lingfeng Duan Guoxing Dai Xiao Liu Kang Wang Peng Yang Guoxing Chen Alan P.Gay John H.Doonan Zhiyou Niu Lizhong Xiong Wanneng Yang 2021Plant Communications2021,2,2:7
19Tumor microenvironments self-activated nanoscale metal-organic frameworks for ferroptosis based cancer chemodynamic/photothermal/chemo therapy显示文摘Ferroptosis,as a newly discovered cell death form,has become an attractive target for precision cancer therapy.Several ferroptosis therapy strategies based on nanotechnology have been reported by either increasing intracellular iron levels or by inhibition of glutathione(GSH)-dependent lipid hydroperoxidase glutathione peroxidase 4(GPX4).However,the strategy by simultaneous iron delivery and GPX4 inhibition has rarely been reported.Herein,novel tumor microenvironments(TME)-activated metal-organic frameworks involving Fe&Cu ions bridged by disulfide bonds with PEGylation(FCSP MOFs)were developed,which would be degraded specifically under the redox TME,simultaneously achieving GSH-depletion induced GPX4 inactivation and releasing Fe ions to produce ROS via Fenton reaction,therefore causing ferroptosis.More ROS could be generated by the acceleration of Fenton reaction due to the released Cu ions and the intrinsic photothermal capability of FCSP MOFs.The overexpressed GSH and H2O2 in TME could ensure the specific TME self-activated therapy.Better tumor therapeutic efficiency could be achieved by doxorubicin(DOX)loading since it can not only cause apoptosis,but also indirectly produce H2O2 to amplify Fenton reaction.Remarkable anti-tumor effect of obtained FCSP@DOX MOFs was verified via both in vitro and in vivo assays.Yu Liang Li Zhang Chao Peng Shiyu Zhang Siwen Chen Xin Qian Wanxian Luo Qing Dan Yongyan Ren Yingjia Li Bingxia Zhao 2021Acta Pharmaceutica Sinica B2021,11,10:7
