1Gas Hydrates in the Qilian Mountain Permafrost, Qinghai, Northwest China显示文摘Qilian Mountain permafrost, with area about 10×104 km2, locates in the north of Qinghai-Tibet plateau. It equips with perfect conditions and has great prospecting potential for gas hydrate. The Scientific Drilling Project of Gas Hydrate in Qilian Mountain permafrost, which locates in Juhugeng of Muri Coalfield, Tianjun County, Qinghai Province, has been implemented by China Geological Survey in 2008–2009. Four scientific drilling wells have been completed with a total footage of 2059.13 m. Samples of gas hydrate are collected separately from holes DK-1, DK-2 and DK-3. Gas hydrate is hosted under permafrost zone in the 133–396 m interval. The sample is white crystal and easily burning. Anomaly low temperature has been identified by the infrared camera. The gas hydrate-bearing cores strongly bubble in the water. Gas-bubble and water-drop are emitted from the hydrate-bearing cores and then characteristic of honeycombed structure is left. The typical spectrum curve of gas hydrate is detected using Raman spectrometry. Furthermore, the logging profile also indicates high electrical resistivity and sonic velocity. 3Genome Editing Enables Next-Generation Hybrid Seed Production Technology显示文摘The next-generation hybrid seed technology enables the successful production of sortable hybrid seeds from genic male sterile(GMS)lines and maintainers;however,it requires multiple laborious and complicated steps.Here,we designed a simple next-generation hybrid seed production strategy that takes advantage of the CRISPR/Cas9 technology to create a Manipulated GMS Maintainer(MGM)system via a single transformation.Under this schema,the maize male fertility gene ZmMS26 was nullified by removal of its fifth exon using the CRISPR/Cas9 system on a vector,and a second vector carrying a functional ZmMS26 cDNA was co-transformed to restore fertility.The second vector also contains a male gametophyte inactivation gene(ZmAA1)encoding maizeα-amylase driven by the pollen-specific promoter PG47 and an endosperm fluorescent marker(DsRED)driven by the barley endosperm aleurone-specific promoter Ltp2.The derived single-copy hemizygous MGM lines bore a mutated MS26 gene,leading to complete male sterility but normal vegetative growth and grain yield.The MGM system could prevent genetic transmission of the MGM elements via male gametophytes,providing an efficient method for sorting maintainer seeds labeled by DsRED.This strategy can be extended to any GMS gene and to hybrid crops other than maize.Xiantao Qi Congsheng Zhang Jinjie Zhu Changlin Liu Changling Huang Xinhai Li Chuanxiao Xie 2020Molecular Plant2020,13,9:18
4Cooperative effects of hydrogen sulfide and nitric oxide on delaying softening and decay of strawberry显示文摘Hydrogen sulfide(H2S)and nitric oxide(NO)as two signal molecules play prominent roles in plant response to biotic stress.The combination effects of these two molecules on ripening and decay of postharvest strawberry were investigated.Strawberries were first dipped in distilled water,0.8 mmol/L NaHS solution,5μmmol/L sodium nitroprusside(SNP)solution,and combinative solutions of 0.8 mmol/L NaHS and 5μmmol/L SNP for 10 min,respectively,and then stored at 20°C.Results showed that the effects of the co-treatment of 0.8 mmol/L NaHS solution and 5μmmol/L SNP solution were superior to that of other treatments.The former could suppress the percentage of fruit decay,inhibit respiration rate,maintain the original crust color,and slow down the decrease of firmness and the change of relative conductivity,which preserved the fruit quality.Further investigation showed that co-treatment of hydrogen sulfide and nitric oxide maintained higher activities of chitinase(CHI)and beta-1,3-glucanase(GNS),and lowered activities of pectin methylesterase(PME),polygalacturonase(PG)and endo-β-1,4-glucanase(EGase).The study indicated that the synergistic function of H2S and NO could effectively prolong the shelf life and reduce the decay rate of harvested strawberry.Zhang Chang Shi Jingying Zhu Liqin Li Changle Wang Qingguo 2014International Journal of Agricultural and Biological Engineering2014,7,6:12
5GW5-Like, a homolog of GW5, negatively regulates grain width, weight and salt resistance in rice显示文摘Grain size is an important determinant of yield potential in crops. We previously demonstrated that natural mutations in the regulatory sequences of qSW5/GW5 confer grain width diversity in rice. However, the biological function of a GW5 homolog, named GW5-Like(GW5 L), remains unknown. In this study, we report on GW5 L knockout mutants in Kitaake, a japonica cultivar(cv.)considered to have a weak gw5 variant allele that confers shorter and wider grains. GW5 L is evenly expressed in various tissues, and its protein product is localized to the plasma membrane. Biochemical assays verified that GW5 L functions in a similar fashion to GW5. It positively regulates brassinosteroid(BR) signaling through repression of the phosphorylation activity of GSK2. Genetic data show that GW5 L overexpression in either Kitaake or a GW5 knockout line, Kasaorf3(indica cv. Kasalath background), causes more slender, longer grains relative to the wild-type. We also show that GW5 L could confer salt stress resistance through an association with calmodulin protein OsCa M1-1. 6Precise base editing of non-allelic acetolactate synthase genes confers sulfonylurea herbicide resistance in maize显示文摘Single-nucleotide polymorphisms contribute to phenotypic diversity in maize. Creation and functional annotation of point mutations has been limited by the low efficiency of conventional methods based on random mutation. An efficient tool for generating targeted single-base mutations is desirable for both functional genomics and precise genetic improvement. The objective of this study was to test the efficiency of targeted C-to-T base editing of two non-allelic acetolactate synthase(ALS) in generating sulfonylurea herbicide-resistant mutants. A CRISPR/Cas9 nickase-cytidine deaminase fused with uracil DNA glycosylase inhibitor(UGI) was employed to achieve targeted conversion of cytosine to thymine in ZmALS1 and ZmALS2. Both protoplasts and recovered mutant plants showed the activity of the cytosine base editor, with an in vivo efficiency of up to 13.8%. Transgene-free edited plants harboring a homozygous ZmALS1 mutation or a ZmALS1 and ZmALS2 double mutation were tested for their resistance at a dose of up to 15-fold the recommended limit of chlorsulfuron, a sulfonylurea herbicide widely used in agriculture. 7Novel Wx alleles generated by base editing for improvement of rice grain quality显示文摘Amylose content(AC),which is regulated by the Waxy(Wx)gene,is a major indicator of eating and cooking quality(ECQ)in rice(Oryza sativa).Thus far,only a limited number of mutations in the N-terminal domain of Wx were found to have a major impact on the AC of rice grains and no mutations with such effects were reported for other regions of the Wx protein.Here,nucleotide substitutions in the middle region of Wx were generated by adenine and cytosine base editors.The nucleotide substitutions led to changes in 15 amino acid residues of Wx,and a series of novel Wx alleles with ACs of 0.3%-29.43%(wild type with AC of 19.87%)were obtained.Importantly,the waxy~(abe2)allele showed a'soft rice'AC,improved ECQ,favorable appearance,and no undesirable agronomic traits.The transgenes were removed from the waxy~(abe2)progeny,generating a promising breeding material for improving rice grain quality.Xiaorui Huang Fei Su Sheng Huang Fating Mei Xiaomu Niu Changle Ma Hui Zhang Xiaoguo Zhu Jian-Kang Zhu Jinshan Zhang 2021Journal of Integrative Plant Biology2021,63,9:4
8Conversion of a normal maize hybrid into a waxy version using in vivo CRISPR/Cas9 targeted mutation activity显示文摘Waxy maize is a specialty maize that produces mainly amylopectin starch with special food or industrial values. The objective of this study was to overcome the limitations of wx mutant allele acquisition and breeding efficiency by conversion of parental lines from normal to waxy maize. The intended mutation activity was achieved by in vivo CRISPR/Cas9 machinery involving desired-target mutation of the Wx locus in the ZC01 background,abbreviated as ZC01-DTM^(wx). Triple selection was applied to segregants to obtain high genome background recovery with transgene-free wx mutations. The targeted mutation was identified, yielding six types of mutations among progeny crossed with ZC01-DTM^(wx).The amylopectin contents of the endosperm starch in mutant lines and hybrids averaged94.9%, while those of the wild-type controls were significantly(P < 0.01) lower, with an average of 76.9%. Double selection in transgene-free lines was applied using the Bar strip test and Cas9 PCR screening. The genome background recovery ratios of the lines were determined using genome-wide SNP data. That of lines used as male parents was as high as98.19% and that of lines used as female parents was as high as 86.78%. Conversion hybrids and both parental lines showed agronomic performance similar to that of their wild-type counterparts. 9Creation of aromatic maize by CRISPR/Cas显示文摘Aroma is an important quality parameter for breeding in rice(Oryza sativa).For example,the aromatic rice varieties basmati and jasmine rice,with a popcorn-like scent,are popular worldwide and routinely command a price premium.2-acetyl-1-pyrroline(2AP)is a key flavor compound among over 200 volatiles identified in fragrant rice.A naturally fragrant germplasm exists in multiple plant species besides rice,which all exhibit lower activity of BETAINE ALDEHYDE DEHYDROGENASE 2(BADH2).However,no equivalent aromatic germplasm has been described in maize(Zea mays).Here,we characterized the two maize BADH2 homologs,ZmBADH2a and ZmBADH2b.We generated zmbadh2a and zmbadh2b single mutants and the zmbadh2a-zmbadh2b double mutant by CRISPR/Cas in four inbred lines.A popcorn-like scent was only noticeable in seeds from the double mutant,but not from either single mutant or in wild type.In agreement,we only detected 2AP in fresh kernels and dried mature seeds from the double mutant,which accumulated between 0.028 and 0.723 mg/kg 2AP.These results suggest that ZmBADH2a and ZmBADH2b redundantly participate in 2AP biosynthesis in maize,and represent the creation of the world's first aromatic maize by simultaneous genome editing of the two BADH2 genes.Yanxiao Wang Xiaoqin Liu Xiuxiu Zheng Wenxia Wang Xunqing Yin Haifeng Liu Changle Ma Xiaomu Niu Jian-Kang Zhu Fei Wang 2021Journal of Integrative Plant Biology2021,63,9:3
10Baicalin inhibits inflammation of lipopolysaccharide-induced acute lung injury via toll like receptor-4/myeloid differentiation primary response 88/nuclear factor-kappa B signaling pathway显示文摘OBJECTIVE:To explore the effect and mechanism of baicalin in the treatment of acute lung injury(ALI)by in vivo and in vitro experiments.METHODS:ALI was induced by instilling 10 mg/m L lipopolysaccharide(LPS)into the airway of rats.Different doses of baicalin(50 and 100 mg·kg^(-1)·d^(-1 ))were administered by gavage one day before modeling.RESULTS:Baicalin significantly reduced the permeability of the alveolocapillary membrane,alleviated tissue injury and inflammatory infiltration,and inhibited the secretion of inflammatory factors and the infiltration of neutrophils.The decline in these inflammations was related to the inhibition of the toll like receptor-4(TLR4)/myeloid differentiation factor 88(My D88)/nuclear factor-kappa B(NF-κB)/nod-like receptor pyrin containing 3(NLRP3)signaling pathway and the mitogen-activated protein kinase(MAPK)signaling pathway.CONCLUSIONS:Baicalin inhibits the secretion of inflammatory factors by inhibiting the TLR4-My D88-NF-κB/NLRP3 pathway and the MAPK signaling pathway.Thus,it reduces lung bronchial epithelial layer,alveolar damage,and pulmonary edema as detected in the in vivo and in vitro experiments.Therefore,baicalin may be a potential preventive and therapeutic drug for ALI.ZHU Changle FENG Cuiling FENG Feng Yao Xiaoqin WANG Guishu SHI Liangtian ZHENG Jiakun 2022Journal of Traditional Chinese Medicine2022,42,2:3
11Effect of pulsatile perfusion during cardiopulmonary bypass in terms of radial artery sphygmogram显示文摘OBJECTIVE: To investigate a quantitative method for using radial artery pulse waveforms to assess the effect of pulsatile flow during cardiopulmonary bypass(CPB).METHODS: A total of 34 adults with heart disease who underwent open-heart surgery between April2010 and January 2011 were randomized into a pulsatile perfusion group(n=17) and a non-pulsatile perfusion group(n=17). Radial arterial pulse waveforms of pulsatile and non-pulsatile perfusion patients were observed and compared before and during CPB.RESULTS: No pulse waveform could be detected at patients' radial artery in both groups when the aorta was cross-clamped. Pulse waveforms could be detected at pulsatile perfusion patients' radial artery, but could not be detected at non-pulsatile perfusion patients' radial artery during CPB. 12Ribonuclease H-like gene SMALL GRAIN2 regulates grain size in rice through brassinosteroid signaling pathway显示文摘Grain size is a key agronomic trait that determines the yield in plants.Regulation of grain size by brassinosteroids(BRs)in rice has been widely reported.However,the relationship between the BR signaling pathway and grain size still requires further study.Here,we isolated a rice mutant,named small grain2(sg2),which displayed smaller grain and a semi-dwarf phenotype.The decreased grain size was caused by repressed cell expansion in spikelet hulls of the sg2 mutant.Using map-based cloning combined with a Mut Map approach,we cloned SG2,which encodes a plant-specific protein with a ribonuclease H-like domain.SG2 is a positive regulator downstream of GLYCOGEN SYNTHASE KINASE2(GSK2)in response to BR signaling,and its mutation causes insensitivity to exogenous BR treatment.Genetical and biochemical analysis showed that GSK2 interacts with and phosphorylates SG2.We further found that BRs enhance the accumulation of SG2 in the nucleus,and subcellular distribution of SG2 is regulated by GSK2 kinase activity.In addition,Oryza sativa OVATE family protein 19(OsOFP19),a negative regulator of grain shape,interacts with SG2 and plays an antagonistic role with SG2 in controlling gene expression and grain size.Our results indicated that SG2 is a new component of GSK2-related BR signaling response and regulates grain size by interacting with Os OFP19.Yunshuai Huang Hui Dong Changling Mou Ping Wang Qixian Hao Min Zhang Hongmin Wu Fulin Zhang Tengfei Ma Rong Miao Kai Fu Yaping Chen Ziyan Zhu Cheng Chen Qikai Tong Zhuoran Wang Shirong Zhou Xi Liu Shijia Liu Yunlu Tian Ling Jiang Jianmin Wan 2022Journal of Integrative Plant Biology2022,64,10:1

17Effect of frequency and intensity of rotating magneticfield on the microstructures of Pb-Sn alloys显示文摘The solidification microstructures of Pb-45% Sn hypoeutectic and Pb-85%Sn hypereutectic alloys were studied in rotating magnetic field (RMF). A transition of primary phase from dendrite to spherical growth was caused by the RMF, which simultaneously fractured and fined grains. The fracture and refinement increased first and then decreased with the increase in RMF intensity. When magnetic intensity exceeded a critical value, the size of the primary phase became bigger on the contrary. Therefore, there existed an optimum value of magnetic intensity in fracture and refinement of grain. Moreover, the rotating frequency determined the skin depth of magnetic field, and further affected the homogenization of temperature and solute. The rotating frequency and magnetic intensity were key factors affect- ing refinement and uniformity of solidification microstructures. 18Effect of spleen-invigorating, Qi-replenishing and blood-arresting formula on zebrafish models with simvastatin-induced hemorrhage caused by spleen failing to control blood, in terms of theory of Traditional Chinese Medicine显示文摘OBJECTIVE: To investigate the hemostasis effect of spleen-invigorating, Qi-replenishing and blood-arresting formula, on a zebrafish models with simvastatin-induced hemorrhage, and with symptom pattern caused by spleen failing to control blood, in terms of theory of Traditional Chinese Medicine(TCM).METHODS: In the first experiment, 60 AB strain wild type zebrafishes were randomly assigned into two groups: normal group and model group. The model group was treated with 50 μM simvastatinfor 24 h. The second experiment: The melanin allele mutated Albino strain zebrafishes were divided into normal, model, A group and B group. The observational parameters were as follows: blood flow, velocity of movement, hemorrhage ratio and improvement ratio of hemorrhage.RESULTS: Hemorrhage ratio: in the first experiment, brain hemorrhage ratio was 75%. In the second experiment, heart hemorrhage ratio was 65%.Blood flow: compared with the normal group,there was a significantly decrease in the model group(P < 0.001). Velocity of movement: in the first experimental, compared with the normal group,there was a significantly decrease in the model group(P < 0.001). Improvement ratio of hemorrhage: agents A had little effect in heart hemorrhage of the zebrafish; agents B could reduce heart hemorrhage ratio of the zebrafish, and increase the improvement ratio of hemorrhage.CONCLUSION: The manifestation of zebrafish model with simvastatin-induced hemorrhage is basically similar to that of the clinical symptom pattern caused by spleen's failure to control blood. 20Barocaloric Material with High Thermal Conductivity for Room-Temperature Refrigeration显示文摘Barocaloric refrigeration technology,one of the caloric-effect refrigeration technologies,is drawing more and more attention.Neopentyl glycol(NPG)was reported to have a giant barocaloric effect,making it a potential barocaloric material.However,the high solid-solid(S-S)phase transition temperature and low thermal conductivity hinder the application of NPG in barocaloric refrigeration.This work lowers the S-S phase transition temperature and improves the thermal conductivity of the NPG-based barocaloric material.An NPG/TMP(TMP:Trimethylolpropane)binary system with an S-S phase transition temperature of 283.15 K is prepared,in which the mass ratio of TMP is 20%.Graphene is then added to the binary system to enhance thermal conductivity,and the optimal mass ratio of graphene was determined to be 5%.The thermal conductivity of this composite is 0.4 W/(m·K),increased by 110%compared to the binary system.To predict the effect of enhanced thermal conductivity on the cold-extraction process of the barocaloric refrigeration cycle,a numerical model is developed.The results show that the cold-extraction time of the barocaloric refrigeration cycle utilizing the composite with 5%graphene as the refrigerant is shortened by 50%compared with that using the binary system.ZHU Liutao DAI Zhaofeng GAO Yuanzhi WU Dongxu WANG Changling ZHAO Dongliang SHE Xiaohui DING Yulong ZHANG Xiaosong 2023Journal of Thermal Science2023,32,6:0
