| 4 | The methylenetetrahydrofolate reductase genotype 677CT and non-alcoholic fatty liver disease have a synergistic effect on the increasing homocysteine levels in subjects from Chongqing,China显示文摘The methylenetetrahydrofolate reductase(MTHFR)genotypes 677CT and 677TT are associated with elevated serum homocysteine(Hcy)levels by means of lowering the activity of MTHFR,and the increase in serum Hcy may be linked to increased susceptibility to nonalcoholic fatty liver disease(NAFLD).However,there are contradictory reports of the relationship among the MTHFR 677CT gene polymorphism,Hcy,and NAFLD.Therefore,the aim of this study was to identify potential associations and interactions of either Hcy levels or the MTHFR 677CT gene polymorphism with the susceptibility to NAFLD in a Chinese population.The association between the MTHFR 677 CT gene polymorphism and Hcy levels was determined in 243 subjects with NAFLD and 388 healthy subjects without NAFLD using polymerase chain reactionrestriction fragment length polymorphism analysis and high-performance liquid chromatography.In subjects with NAFLD,there was no statistical difference in the genotypic and allelic frequencies of the MTHFR 677 CT gene polymorphism,while serum Hcy levels were significantly higher in subjects with NAFLD.Furthermore,these results strongly suggest that the MTHFR 677CT gene polymorphism and NAFLD have a potential synergistic effect on Hcy elevation,although the MTHFR 677CT gene polymorphism was not correlated with NAFLD in a Chinese population. | Xiaolin Wang Yongli Zhou Mingjun Zhang Yonghong Wang Bo Qin | 2019 | Genes & Diseases2019,6,1: | 6 |
| 5 | Estimation of correction coefficients for measured coal bed methane contents显示文摘Improving the accuracy and precision of coal bed methane(CBM) estimates requires correction of older data from older coal exploration surveys to newer standards.Three methods,the depth gradient method,the contour aerial weight method,and the well-point aerial weight method,were used to estimate the correction coefficient required to predict CBM gas content from coal exploration data.The data from the Nos.3 and 15 coal seams provided the coal exploration data while the CBM exploration stages within the X1 well block located in the southern part of the Qinshui Basin provided the data obtained using newer standards.The results show the correction coefficients obtained from the two aerial weight methods are similar in value but lower than the one obtained from the depth gradient method.The three methods provide similar results for the Nos.3 and 15 seams in that the correction factor is lower for the former seam.The results from the depth gradient method taken together with the coal seam burial depth and the coal rank suggest that variations in the correction factor increase linearly along with coal seam burial depth and coal rank.The correlation obtained can be applied to exploration and the evaluation of coal bed gas resources located in coalfields. | Lei Bo Fu Xuehai Zhou Baoyan Shen Jia ZOU Mingjun Feng Qing | 2012 | International Journal of Mining Science and Technology2012,22,4: | 5 |
| 6 | Effects of climate,land use and land cover changes on soil loss in the Three Gorges Reservoir area,China显示文摘Climate,land use and land cover(LULC)changes are among the primary driving forces of soil loss.Decoupling their effects can help in understanding the magnitude and trend of soil loss in response to human activities and ecosystem management.Here,the RUSLE model was applied to estimate the spatial-temporal variations of soil loss rate in the Three Gorges Reservoir(TGR)area during 2001-2015,followed by a scenario design to decouple the effects of climate and LULC changes.The results showed that increasing rainfall generated as much as 2.90×10^(7)t soil loss in the TGR area.However,such effect was offset by changes in LULC particularly afforestation,which retained about 1.10×10^(7)t soil annually.Other human activities such as dam development and urbanization aggravated soil loss by as much as 1.40×10^(6)t annually.Because of land use policies that favor economic development,distinct spatial variances of soil loss were observed in TGR area.Soil loss in some counties located downstream of the TGR area(i.e.,close to the dam)was more influenced by dam development,but soil loss in the other counties was more influenced by urbanization.As climate change(i.e.,increasing rainfall)did not affect plant performance in TGR area,our findings suggested that ecological restoration was more beneficial to curb the amount of soil loss caused by urbanization and dam construction. | Chunbo Huang Zhixiang Zhou Mingjun Teng Changguang Wu Pengcheng Wang | 2020 | Geography and Sustainability2020,1,3: | 3 |
| 7 | The FMEDA Based DC Calculation for Railway Safety Computer显示文摘This paper presents a new algorithm to compute the Diagnostic coverage(DC) for railway safety computer using the Failure modes effects and diagnostic analysis(FMEDA) theory. The importance to work out the DC accurately is stressed. A certain type of railway safety computer's output element is taken as an example to show how the DC is worked out using the FMEDA method. The probability of dangerous failures per hour(PFH) of one certain safety computer is obtained considering the DC.The final results show that the DC is 99.6% and the PFH of the safety is 1.165 fit, which means 1.165 dangerous failures may occur during 1 billion hours' working time,running up to the requirement of the Safety integrity level4(SIL4). | 8 | Synthesis and Tribological Properties of Graphene-Copper Nanoparticle Composites as Lithium Grease Additive显示文摘In this paper, the graphene-copper nanoparticle composites(GN/Cu NPs) were prepared by in situ chemical reduction in aqueous solution. The influence of pH value and the concentration of PVP on the particle size and structure of the GN/Cu NPs as well as the element distribution were analyzed via the scanning electron microscopy(SEM), the transmission electron microscopy(TEM), the X-ray diffraction(XRD), and the energy dispersive spectroscopy(EDS). The tribological properties of the grease with GN/Cu NPs composites have been studied by SRV-IV. It was found that compared to the base grease, the GN/Cu NPs composites could help to reduce the wear loss of the disk by 85.5% and the average friction coefficient by 15.5%. | 10 | DYRK1A suppression restrains Mcl-1 expression and sensitizes NSCLC cells to Bd-2 inhibitors显示文摘Objective:Mcl-1 overexpression confers acquired resistance to Bcl-2 inhibitors in non-small cell lung cancer(NSCLC),but no direct Mcl-1 inhibitor is currently available for clinical use.Thus,novel therapeutic strategies are urgently needed to target Mcl-1 and sensitize the anti-NSCLC activity of Bcl-2 inhibitors.Methods:Cell proliferation was measured using sulforhodamine B and colony formation assays,and apoptosis was detected with Annexin V-FITC staining.Gene expression was manipulated using siRNAs and plasmids.Real-time PCR and Western blot were used to measure mRNA and protein levels.Immunoprecipitation and immunofluorescence were used to analyze co-localization o f dual specificity tyrosine-phosphorylation-regulated kinase 1A(DYRK1A)and Mcl-1.Results:Suppression of DYRK1A resulted in reduced Mcl-1 expression in NSCLC cells,whereas overexpression of DYRK1A significantly increased Mcl-1 expression.Suppression of DYRK1A did not alter Mcl-1 mRNA levels,but did result in an accelerated degradation o f Mcl-1 protein in NSCLC cells.Furthermore,DYRK1A mediated proteasome-dependent degradation o f Mcl-1 in NSCLC cells,and DYRK1A co-localized with Mcl-1 in NSCLC cells and was co-expressed with Mcl-1 in tumor samples from lung cancer patients,suggesting that Mcl-1 may be a novel DYRK1A substrate.We showed that combined therapy with harmine and Bcl-2 antagonists significantly inhibited cell proliferation and induced apoptosis in NSCLC cell lines as well as primary NSCLC cells.Conclusions:Mcl-1 is a novel DYRK1A substrate,and the role of DYRK1A in promoting Mcl-1 stability makes it an attractive target for decreasing Bcl-2 inhibitor resistance. | Yangling Li Dongmei Zhou Shuang Xu Mingjun Rao Zuoyan Zhang Linwen Wu Chong Zhang Nengming Lin | 2020 | Cancer Biology & Medicine2020,17,2: | 2 |

| 16 | A nuclear localized protein ZCCHC9 is expressed in cerebral cortex and suppresses the MAPK signal pathway显示文摘The CCHC-type zinc finger motif has numerous biological activities (such as DNA binding and RNA binding) and can also mediate protein-protein interaction. This article gives a primary report about the human ZCCHC9 gene. Protein ZCCHC9 contains four CCHC motifs and is highly conserved in humans, mice, and rats. The whole cDNA sequence of the ZCCHC9 gene has been amplified by PCR and a number of plasmids have been constructed for further study. The results show that ZCCHC9 is localized in the nucleus, and especially concentrated in the nucleolus. It is highly expressed in the brain and testicles of the mouse. This has been confirmed by real-time reverse transcription polymerase chain reaction (RT-PCR). In situ hybridization of the mouse brain indicates that ZCCHC9 is mainly expressed in the cerebral cortex. | 18 | Anti-infection effects of heparin on SARS-CoV-2 in a diabetic mouse model显示文摘Severe acute respiratory syndrome coronavirus 2(SARSCo V-2)infection can result in more severe syndromes and poorer outcomes in patients with diabetes and obesity.However,the precise mechanisms responsible for the combined impact of coronavirus disease 2019(COVID-19)and diabetes have not yet been elucidated,and effective treatment options for SARS-Co V-2-infected diabetic patients remain limited.To investigate the disease pathogenesis,K18-h ACE2 transgenic(h ACE2^(Tg))mice with a leptin receptor deficiency(h ACE2-Lepr^(-/-))and high-fat diet(h ACE2-HFD)background were generated.The two mouse models were intranasally infected with a 5×10^(5) median tissue culture infectious dose(TCID_(50))of SARSCo V-2,with serum and lung tissue samples collected at 3days post-infection.The h ACE2-Lepr^(-/-)mice were then administered a combination of low-molecular-weight heparin(LMWH)(1 mg/kg or 5 mg/kg)and insulin via subcutaneous injection prior to intranasal infection with1×10^(4) TCID_(50)of SARS-Co V-2.Daily drug administration continued until the euthanasia of the mice.Analyses of viral RNA loads,histopathological changes in lung tissue,and inflammation factors were conducted.Results demonstrated similar SARS-Co V-2 susceptibility in h ACE2^(Tg)mice under both lean(chow diet)and obese(HFD)conditions.However,compared to the h ACE2-Lepr^(+/+)mice,h ACE2-Lepr^(-/-)mice exhibited more severe lung injury,enhanced expression of inflammatory cytokines and hypoxia-inducible factor-1α(HIF-1α),and increased apoptosis.Moreover,combined LMWH and insulin treatment effectively reduced disease progression and severity,attenuated lung pathological changes,and mitigated inflammatory responses.In conclusion,preexisting diabetes can lead to more severe lung damage upon SARS-Co V-2 infection,and LMWH may be a valuable therapeutic approach for managing COVID-19patients with diabetes. | Zhongyun Zhang Ning Zhang Xuancheng Lu Min Zhou Xiaoxiang Yan Weiqiong Gu Jingru Yang Qin Zhang Cheng Zhang Yuhuan Gong Mingjun Jia Xiaoyu Zhang Peng Ning Mei Liu Xiaoyan Li Xiaomeng Shi Wenjun Liu George FGao Guang Ning Jiqiu Wang Yuhai Bi | 2023 | Zoological Research2023,44,6: | 1 |
