| 1 | DYRK1A suppression restrains Mcl-1 expression and sensitizes NSCLC cells to Bd-2 inhibitors显示文摘Objective:Mcl-1 overexpression confers acquired resistance to Bcl-2 inhibitors in non-small cell lung cancer(NSCLC),but no direct Mcl-1 inhibitor is currently available for clinical use.Thus,novel therapeutic strategies are urgently needed to target Mcl-1 and sensitize the anti-NSCLC activity of Bcl-2 inhibitors.Methods:Cell proliferation was measured using sulforhodamine B and colony formation assays,and apoptosis was detected with Annexin V-FITC staining.Gene expression was manipulated using siRNAs and plasmids.Real-time PCR and Western blot were used to measure mRNA and protein levels.Immunoprecipitation and immunofluorescence were used to analyze co-localization o f dual specificity tyrosine-phosphorylation-regulated kinase 1A(DYRK1A)and Mcl-1.Results:Suppression of DYRK1A resulted in reduced Mcl-1 expression in NSCLC cells,whereas overexpression of DYRK1A significantly increased Mcl-1 expression.Suppression of DYRK1A did not alter Mcl-1 mRNA levels,but did result in an accelerated degradation o f Mcl-1 protein in NSCLC cells.Furthermore,DYRK1A mediated proteasome-dependent degradation o f Mcl-1 in NSCLC cells,and DYRK1A co-localized with Mcl-1 in NSCLC cells and was co-expressed with Mcl-1 in tumor samples from lung cancer patients,suggesting that Mcl-1 may be a novel DYRK1A substrate.We showed that combined therapy with harmine and Bcl-2 antagonists significantly inhibited cell proliferation and induced apoptosis in NSCLC cell lines as well as primary NSCLC cells.Conclusions:Mcl-1 is a novel DYRK1A substrate,and the role of DYRK1A in promoting Mcl-1 stability makes it an attractive target for decreasing Bcl-2 inhibitor resistance. | Yangling Li Dongmei Zhou Shuang Xu Mingjun Rao Zuoyan Zhang Linwen Wu Chong Zhang Nengming Lin | 2020 | Cancer Biology & Medicine2020,17,2: | 2 |

| 11 | Electromagnetic wave absorption properties of cobalt-zinc ferrite nanoparticles doped with rare earth elements显示文摘Traditional ferrites are of poor electromagnetic wave(EMW) absorption while doping rare earth elements(REEs) can greatly enhance their permeability to improve the EMW loss performance.In this study,Co-Zn ferrite nanoparticles doped with various amounts of REEs(Gd^(3+),Nd^(3+)and Pr^(3+)) were synthesized by a hydrothermal method,and their particle morphology and an EMW absorption performance were characterized by using transmission electron microscopy(TEM) and a Vector network analyzer(VNA).The results show that the initial spherical Co-Zn ferrite nanoparticles present an irregular quadrilateral structure after Gd^(3+)doping,and the average particle size of Co_(0.5)Zn_(0.5-x)Gd_(x)Fe_(2) O_(4) increases from 26 to 50 nm with x increasing from 0 to 0.35.At x of 0.25,the reflectivity absorbance achieves-27.94 dB at 18 GHz with the effective absorption bandwidth(EAB) of 4.08 GHz at a sample thickness of 2.5 mm.When Nd^(3+)doping amount reaches x=0.3,the minimum reflection loss(RL)is-25.63 dB at 18 GHz and EAB is 3.91 GHz.Doping Pr^(3+)(x=0.25) in the sample broadens EAB,and the minimum RL is-16.1 dB at 16.81 GHz and EAB is 7.31 GHz.This study shows that the magnetic moment produced by doping REES can form magnetic domains,which affects the incident EMW and improves the magnetic loss.It is expected that REEs-doped Co-Zn ferrite nanoparticles can be used as efficient electromagnetic shielding materials in aerospace. | Changye Mang Zhijun Ma Jun Luo Mingjun Rao Xin Zhang Zhiwei Peng | 2021 | Journal of Rare Earths2021,39,11: | 0 |
| 12 | Enrichment of Nb and Ti from carbonatite pyrochlore ore via calcining-slaking followed by gravity separation显示文摘In this work,a novel process consisting of calcining-slaking followed by gravity separation for the enrichment of niobium(Nb)and titanium(Ti)from carbonatite pyrochlore ore was proposed,validated and compared with the current mainstream flotation method.During calcining of the pyrochlore ore,within which the carbonates were transformed into lime.Subsequently,when the calcined ore was slaked,lime was transformed into hydroxide with fine particles which were amenable to gravity separation.After calcining at 900℃for 60 min,slaking at 90℃for 10 min with a liquid–solid ratio of 3:1(mL/g),approximately 40%of tailings can be removed by gravity separation,the recoveries of Nb and Ti were 94.7%and 91.0%,and the enrichment ratios of Nb and Ti were 1.61 and 1.43,respectively.The new approach exhibits high separation efficiency of carbonate gangue minerals and valuable minerals,satisfactory recoveries of niobium as well as titanium can be achieved. | Shuhui Zhang Mingjun Rao Rendong Xiao Jinxiang You Guanghui Li Tao Jiang | 2022 | International Journal of Mining Science and Technology2022,32,3: | 0 |
