Toxicities and risk assessment of heavy metals in sediments of Taihu Lake, China, based on sediment quality guidelines显示文摘The occurrence, toxicities, and ecological risks of five heavy metals(Pb, Cu, Cd, Zn and Ni) in the sediment of Taihu Lake were investigated in this study. To evaluate the toxicities caused by the heavy metals, the toxicities induced by organic contaminants and ammonia in the sediments were screened out with activated carbon and zeolite. The toxicities of heavy metals in sediments were tested with benthic invertebrates(tubificid and chironomid). The correlations between toxicity of sediment and the sediment quality guidelines(SQGs)derived previously were evaluated. There were significant correlations(p < 0.0001) between the observed toxicities and the total risk quotients of the heavy metals based on SQGs,indicating that threshold effect level(TEL) and probable effect level(PEL) were reliable to predict the toxicities of heavy metals in the sediments of Taihu Lake. By contrast, the method based on acid volatile sulfides(AVS) and simultaneously extracted metals(SEM),such as ∑SEM/AVS and ∑SEM-AVS, did not show correlations with the toxicities. Moreover, the predictive ability of SQGs was confirmed by a total predicting accuracy of 77%. Ecological risk assessment based on TELs and PELs showed that the contaminations of Pb, Cu, Cd and Zn in the sediments of Taihu Lake were at relatively low or medium levels. The risks caused by heavy metals in the sediments of northern bay of the lake, which received more wastewater discharge from upper stream, were higher than other area of the lake. | Yanfeng Zhang Yuwei Han Jinxi Yang Lingyan Zhu Wenjue Zhong | 2017 | Journal of Environmental Sciences2017,29,12: | 16

Single-cell RNA sequencing reveals B cell-T cell interactions in vascular adventitia of hyperhomocysteinemia-accelerated atherosclerosis显示文摘Dear Editor Cardiovascular disease(CVD)is the leading cause of death around the world(Truelsen,et al.,2015).Atherosclerosis,the dominant underlying cause of CVD,is a chronic inflam-matory disease characterized by lipid accumulation and immune cell infiltration in plaques and vessels(Weber,et al.,1950).The immune microenvironment is critical for the development of atherosclerosis.Homocysteine(Hcy)is an intermediate product of methionine metabolism, and its ele-vation in plasma(>15μmol/L),known as hyperhomocys-teinemia(HHcy),is an independent risk factor for atherosclerosis.HHcy is more common in Asia because of genetic factors and dietary habits(Huo,et al.,2015).folic acid supplement is one of the most important way to treat HHcy in clinic.Although HHcy potentiates atherosclerosis mainly through endothelial injury and inflammatory activa-tion(Luo,et al.,2016),a comprehensive understanding of the immune microenvironment and potential mechanisms in HHcy-accelerated atherosclerotic aortas(HHcy-AA)is still lacking. | Xiaolong Ma Jiacheng Deng Lulu Han Yuwei Song Yutong Miao Xing Du Guohui Dang Dongmin Yang Bitao Zhong Changtao Jiang Wei Kong Qingbo Xu Juan Feng Xian Wang | 2022 | Protein & Cell2022,13,7: | 2
B cell-derived anti-beta 2 glycoprotein I antibody mediates hyperhomocysteinemia-aggravated hypertensive glomerular lesions by triggering ferroptosis显示文摘Hyperhomocysteinemia(HHcy)is a risk factor for chronic kidney diseases(CKDs)that affects about 85%CKD patients.HHcy stimulates B cells to secrete pathological antibodies, although it is unknown whether this pathway mediates kidney injury. In HHcytreated 2-kidney,1-clip(2K1C)hypertensive murine model,HHcy-activated B cells secreted anti-beta 2 glycoprotein I(β2GPI)antibodies that deposited in glomerular endothelial cells(GECs),exacerbating glomerulosclerosis and reducing renal function.Mechanistically,HHcy 2K1C mice increased phosphatidylethanolamine(PE)(18:0/20:4,18:0/22:6,16:0/20:4)in kidney tissue,as determined by lipidomics.GECs oxidative lipidomics validated the increase of oxidized phospholipids upon Hcy-activated B cells culture medium(Hcy-B CM)treatment, including PE(18:0/20:4+3[O],PE(18:0a/22:4+1[O],PE(18:0/22:4+2[O]and PE(18:0/22:4+3[O]).PE synthases ethanolamine kinase 2(etnk2)and ethanolamine-phosphate cytidylyltransferase 2(pcyt2)were increased in the kidney GECs of HHcy 2K1C mice and facilitated polyunsaturated PE synthesis to act as lipid peroxidation substrates. In HHcy 2K1C mice and Hcy-B CM-treated GECs,the oxidative environment induced by iron accumulation and the insufficient clearance of lipid peroxides caused by transferrin receptor(TFR)elevation and down-regulation of SLC7A11/glutathione peroxidase 4(GPX4)contributed to GECs ferroptosis of the kidneys. In vivo, pharmacological depletion of B cells or inhibition of ferroptosis mitigated the HHcy-aggravated hypertensive renal injury.Consequently, our findings uncovered a novel mechanism by which B cell-derived pathogenic anti-β2GPI IgG generated by HHcy exacerbated hypertensive kidney damage by inducing GECs ferroptosis.Targeting B cells or ferroptosis may be viable therapeutic strategies for ameliorating lipid peroxidative renal injury in HHcy patients with hypertensive nephropathy. | Xing Du Xiaolong Ma Ying Tan Fangyu Shao Chun Li Yang Zhao Yutong Miao Lulu Han Guohui Dang Yuwei Song Dongmin Yang Zhenling Deng Yue Wang Changtao Jiang Wei Kong Juan Feng Xian Wang | 2023 | Signal Transduction and Targeted Therapy2023,8,4: | 2
Modulation of Beta Oscillations for Implicit Motor Timing in Primate Sensorimotor Cortex during Movement Preparation显示文摘Motor timing is an important part of sensorimotor control. Previous studies have shown that beta oscillations embody the process of temporal perception in explicit timing tasks. In contrast, studies focusing on beta oscillations in implicit timing tasks are lacking. In this study, we set up an implicit motor timing task and found a modulation pattern of beta oscillations with temporal perception during movement preparation. We trained two macaques in a repetitive visually-guided reach-to-grasp task with different holding intervals. Spikes and local field potentials were recorded from microelectrode arrays in the primary motor cortex, primary somatosensory cortex, and posterior parietal cortex. We analyzed the association between beta oscillations and temporal interval in fixedduration experiments(500 ms as the Short Group and1500 ms as the Long Group) and random-duration experiments(500 ms to 1500 ms). The results showed that the peak beta frequencies in both experiments ranged from15 Hz to 25 Hz. The beta power was higher during the hold period than the movement(reach and grasp) period.Further, in the fixed-duration experiments, the mean poweras well as the maximum rate of change of beta power in the first 300 ms were higher in the Short Group than in the Long Group when aligned with the Center Hit event. In contrast, in the random-duration experiments, the corresponding values showed no statistical differences among groups. The peak latency of beta power was shorter in the Short Group than in the Long Group in the fixed-duration experiments, while no consistent modulation pattern was found in the random-duration experiments. These results indicate that beta oscillations can modulate with temporal interval in their power mode. The synchronization period of beta power could reflect the cognitive set maintaining working memory of the temporal structure and attention. | Hongji Sun Xuan Ma Liya Tang Jiuqi Han Yuwei Zhao Xuejiao Xu Lubin Wang Peng Zhang Luyao Chen Jin Zhou Changyong Wang | 2019 | Neuroscience Bulletin2019,35,5: | 2
Chronic electrical stimulation induces functional network changes in cortical neuron cultures显示文摘Recent studies demonstrated that a functional brain network could be regarded as a complex network. With the help of network theory, neuroscientists can identify common organizational principles of the functional brain networks. As a consequence, some non-random organizational features, such as'small world'(most of the nodes are not connected directly but can communicate with few intermediate relay steps)and'rich club'(nodes that are rich in connections tend to form strongly interconnected clubs),have been found in functional brain network.Recently, the'small world'organizational feature of neuronal functional networks in vitro was found to be influenced by external applications. However, little is known about the influence of chronic electrical stimulation on functional networks of dissociated cortical cultures during network development. In the present study, cortical cultures were electrically stimulated at a frequency of 0,0.02,and 0.2 Hz,between 7 and 26 days in vitro(DIV).The spontaneous activity of the cortical cultures was recorded using MEAs.Next,a cross-covariance method and graph theory were applied to investigate organizational feature of functional networks. Our results showed that over 3 weeks of stimulation, the network density significantly increased with maturation in the control and 0.02 Hz stimulation groups, but not in 0.2 Hz stimulation groups. Moreover, all the cultures had a small-world topology at 14,18,22,and 26 DIV,free from the effect of chronic electrical stimulation.Besides,we found an asymmetry effect that partial electrical stimulation inhibited the formation of node connections in stimulated areas. This effect was more pronounced at 0.2 Hz than at 0.02 Hz stimulation. Our results suggest that electrical stimulation does not affect the small-world properties of neural cultures.Instead, electrical stimulation modulates connectivity patterns, and neurons within the stimulated area are less connected than neurons outside the stimulated area. | WU SongFang WANG LuBin ZHAO YuWei SUN HongJi LANG YiRan WANG ChangYong HAN JiuQi WANG Yan HAN Yao | 2020 | Science China(Technological Sciences)2020,63,4: | 2
Single cell analysis reveals inhibition of angiogenesis attenuates the progression of heterotopic ossification in Mkx^(−/−)mice显示文摘Tendon heterotopic ossification(HO)is characterized by bone formation inside tendon tissue, which severely debilitates people in their daily life.Current therapies fail to promote functional tissue repair largely due to our limited understanding of HO pathogenesis. Here,we investigate the pathological mechanism and propose a potential treatment method for HO.Immunofluorescence assays showed that the Mohawk(MKX)expression level was decreased in human tendon HO tissue, coinciding with spontaneous HO and the upregulated expression of osteochondrogenic and angiogenic genes in the tendons of Mkx^(−/−)mice.Single-cell RNA sequencing analyses of wild-type and Mkx^(−/−)tendons identified three cell types and revealed the excessive activation of osteochondrogenic genes during the tenogenesis of Mkx^(−/−)tendon cells.Single-cell analysis revealed that the gene expression program of angiogenesis, which is strongly associated with bone formation, was activated in all cell types during HO.Moreover, inhibition of angiogenesis by the small-molecule inhibitor BIBF1120 attenuated bone formation and angiogenesis in the Achilles tendons of both Mkx mutant mice and a rat traumatic model of HO.These findings provide new insights into the cellular mechanisms of tendon HO and highlight the inhibition of angiogenesis with BIBF1120 as a potential treatment strategy for HO. | Junxin Lin Yuwei Yang Wenyan Zhou Chao Dai Xiao Chen Yuanhao Xie Shan Han Huanhuan Liu Yejun Hu Chenqi Tang Varitsara Bunpetch Dandan Zhang Yishan Chen Xiaohui Zou Di Chen Wanlu Liu Hongwei Ouyang | 2022 | Bone Research2022,10,4: | 1

Thirteen Dipterocarpoideae genomes provide insights into their evolution and borneol biosynthesis显示文摘Dipterocarpoideae, the largest subfamily of the Dipterocarpaceae,is a dominant component of Southeast Asian rainforests and is widely used as a source of wood, damar resin, medicine, and essential oil. However, many Dipterocarpoideae species are currently on the IUCN Red List owing to severe degradation of their habitats under global climate change and human disturbance.Genetic information regarding these taxa has only recently been reported with the sequencing of four Dipterocarp genomes, providing clues to the function and evolution of these species. Here,we report on 13 high-quality Dipterocarpoideae genome assemblies, ranging in size from 302.6 to 494.8 Mb and representing the five most species-rich genera in Dipterocarpoideae.Molecular dating analyses support the Western Gondwanaland origin of Dipterocarpaceae.Based on evolutionary analysis,we propose a three-step chromosome evolution scenario to describe the karyotypic evolution from an ancestor with six chromosomes to present-day species with 11 and 7 chromosomes. We discovered an expansion of genes encoding cellulose synthase(CesA),which is essential for cellulose biosynthesis and secondary cell-wall formation. We functionally identified five bornyl diphosphate synthase(BPPS)genes, which specifically catalyze the biosynthesis of borneol,a natural medicinal compound extracted from damar resin and oils, thus providing a basis for large-scale production of natural borneol in vitro. | Zunzhe Tian Peng Zeng Xiaoyun Lu Tinggan Zhou Yuwei Han Yingmei Peng Yunxue Xiao Botong Zhou Xue Liu Yongting Zhang Yang Yu Qiong Li Hang Zong Feining Zhang Huifeng Jiang Juan He Jing Cai | 2022 | Plant Communications2022,3,6: | 1
Photoluminescence Properties of Nanocrystalline 3C-SiC Films显示文摘Nanocrystalline (nc) 3C-SiC films on the Si substrate were prepared by the helicon wave plasma enhanced chemical vapor deposi-tion (HW-PECVD) technique. With the SiH4-CH4 gas flow ratio changing, the films exhibit different photoluminescence (PL) character-istics. Under the stoichiometric condition, the PL peak redshift from 470 nm to 515 nm is detected with the increase of excitation wave-length, which can be attributed to the quantum confinement effect radiation of 3C-SiC nanocrystals of different sizes. However, the ap-pearance of an additional PL band at 436 nm in Si-rich film might be sourced back to the excess of Si defect centers in it. This is also the case for C-rich film for its PL band lying at 570 nm. The results above quoted indicate an important influence of gas flow ratio on the PL properties of the SiC films providing an effective guidance for analyzing the luminescence mechanism and exploring the high-efficiency light emission of the SiC films. | YUWei LU Xue-qin LU Wan-bing HAN Li FU Guang-sheng | 2006 | Chinese Journal of Aeronautics2006,19,B12: | 0
Ecological impacts of the expansion of offshore wind farms on trophic level species of marine food chain显示文摘The global demand for renewable energy has resulted in a rapid expansion of offshore wind farms(OWFs)and increased attention to the ecological impacts of OWFs on the marine ecosystem.Previous reviews mainly focused on the OWFs’impacts on individual species like birds, bats,or mammals. This review collected numerous field-measured data and simulated results to summarize the ecological impacts on phytoplankton, zooplankton, zoobenthos, fishes, and mammals from each trophic level and also analyze their interactions in the marine food chain.Phytoplankton and zooplankton are positively or adversely affected by the‘wave effect’,‘shading effect’,oxygen depletion and predation pressure, leading to a ±10% fluctuation of primary production.Although zoobenthos are threatened transiently by habitat destruction with a reduction of around 60% in biomass in the construction stage, their abundance exhibited an over 90% increase, dominated by sessile species, due to the‘reef effect’in the operation stage.Marine fishes and mammals are to endure the interferences of noise and electromagnetic, but they are also aggregated around OWFs by the‘reef effect’and‘reserve effect’.Furthermore, the complexity of marine ecosystem would increase with a promotion of the total system biomass by 40% through trophic cascade effects strengthen and resource partitioning alternation triggered by the proliferation of filter-feeders.The suitable site selection,long-term monitoring, and life-cycle-assessment of ecological impacts of OWFs that are lacking in current literature have been described in this review,as well as the carbon emission and deposition. | Lijing Wang Bangguo Wang Wenxi Cen Rui Xu Yuwei Huang Xin Zhang Yinghui Han Yuanxun Zhang | 2024 | Journal of Environmental Sciences2024,,5: | 0
Study on the Problem of Multistage Vaccine Production and Allocation with Capacity Constraints显示文摘Vaccination is the best way to build an immune barrier to control a new infectious disease.Considering the limited production capacity at the initial stage of vaccine production, the problem of multistage vaccine production and allocation is studied under the policy of free treatment for infected patients by the government.Given the population and infection rates of different regions and the vaccine production capacity of each stage,an integer programming model is established to minimize the sum of production-and-vaccination cost and treatment cost of infected patients, which is solved by the GUROBI solver. The correctness of the model is verified by simulation, and the necessity of considering the treatment cost of infected patients in the objective function is further analyzed, which confirmed the correctness of the free treatment policy for patients in China.A heuristic algorithm is designed to solve the large-scale problem and numerical experiments are conducted to verify the efficiency of this algorithm.Furthermore, based on the sensitivity analysis results of parameters, the optimal vaccine production and allocation strategy are proposed, which provide the decision basis for the government departments to make reasonable vaccine production and vaccination plan. | HAN Qianqian ZHANG Yuwei LI Zhenping FANG Yong | 2023 | Journal of Systems Science & Complexity2023,36,5: | 0
Generalized assembly of sandwich-like 0D/2D/0D heterostructures with highly exposed surfaces toward superior electrochemical performances显示文摘Heterostructures composed of two-dimensional(2D)nanosheets and zero-dimensional(0D)nanoparticles(NPs)have attracted increasing attention because of the synergy arising from the coupling interactions between the two mixed-dimensional components.Despite recent advances,it remains a challenge to fabricate 2D/0D heterostructures with clean and accessible surfaces, which is highly desirable for the diversity of catalytic, sensing, and energy storage applications.Herein,we report a generalized methodology that enables the facile assembly of sandwich-like 0D/2D/0D heterostructures with facilitated mass-transport channels and exposed surface active sites.A ligand-exchange strategy with HBF4 is employed to strip off the surface-coating ligands of colloidal NPs,rendering them positively charged and dispersible in polar solvents. This allows subsequent electrostatic assembly of NPs with oppositely charged 2D nanosheets to afford sandwich-like 0D/2D/0D heterostructures. The barely covered surfaces and the advantageous architectures of such sandwich-like 0D/2D/0D heterostructures induce the desired synergistic effect, making them particularly suitable for electrochemical energy storage and conversion. We demonstrate this by employing MXene/NiFe_(2)O_(4) and MXene/Fe3O4 heterostructures for high-performance electrocatalytic oxygen evolution and supercapacitors, respectively. | Shuqing Xue Guanhong Wu Mingzhong Li Zihan Liu Yuwei Deng Wenqian Han Xuanyu Lv Siyu Wan Xiangyun Xi Dong Yang Angang Dong | 2022 | Nano Research2022,15,1: | 0
Information space of sensor networks:Lagrangian,energy-momentum tensor, and applications显示文摘It is a challenge to investigate the interrelationship between the geometric structure and performance of sensor networks due to the increasingly complex and diverse architecture of them. This paper presents two new formulations for the information space of sensor networks, including Lagrangian and energy–momentum tensor, which are expected to integrate sensor networks target tracking and performance evaluation from a unified perspective. The proposed method presents two geometric objects to represent the dynamic state and manifold structure of the information space of sensor networks.Based on that, the authors conduct the property analysis and target tracking of sensor networks. To the best of our knowledge,it is the first time to investigate and analyze the information energy-momentum tensor of sensor networks and evaluate the performance of sensor networks in the context of target tracking.Simulations and examples confirm the competitive performance of the proposed method. | Mo TAO Shaoping WANG Hong CHEN Han PAN Jian SHI Yuwei ZHANG | 2023 | Chinese Journal of Aeronautics2023,36,3: | 0
Bacteria-mediated tumor-targeted delivery of tumstatin(54-132)significantly suppresses tumor growth in mouse model by inhibiting angiogenesis and promoting apoptosis显示文摘Tumor growth is an angiogenesis-dependent process and accompanied by the formation of hypoxic areas.Tumstatin is a tumor-specific angiogenesis inhibitor that suppresses the proliferation and induces the apoptosis of tumorous vascular endothelial cells.VNP20009,an attenuated Salmonella typhimurium strain, preferentially accumulates in the hypoxic areas of solid tumors. In this study,a novel Salmonella-mediated targeted expression system of tumstatin(VNP-Tum5)was developed under the control of the hypoxia-induced J23100 promoter to obtain anti-tumor efficacy in mice.Treatment with VNP-Tum5 effectively suppressed tumor growth and prolonged survival in the mouse model of B16F10 melanoma.VNP-Tum5 exhibited a higher efficacy in inhibiting the proliferation and inducing the necrosis and apoptosis of B16F10 cells in vitro and in vivo compared with VNP(control).VNP-Tum5 significantly inhibited the proliferation and migration of mouse umbilical vascular endothelial cells to impede angiogenesis.VNP-Tum5 downregulated the expression of anti-vascular endothelial growth factor A,platelet endothelial cell adhesion molecule-1,phosphorylated phosphoinositide 3 kinase, and phosphorylated protein kinase B and upregulated the expression of cleaved-caspase 3 in tumor tissues. This study is the first to use tumstatin-transformed VNP20009 as a tumor-targeted system for treatment of melanoma by combining anti-tumor and anti-angiogenic effects. | Feifei Bao Mengjie Liu Wenhua Gai Yuwei Hua Jing Li Chao Han Ziyu Zai Jiahuang Li Zichun Hua | 2022 | Frontiers of Medicine2022,16,6: | 0
A cell marker-based clustering strategy(cmCluster)for precise cell type identification of scRNA-seq data显示文摘Background:The precise and efficient analysis of single-cell transcriptome data provides powerful support for studying the diversity of cell functions at the single-cell level. The most important and challenging steps are cell clustering and recognition of cell populations. While the precision of clustering and annotation are considered separately in most current studies,it is worth attempting to develop an extensive and flexible strategy to balance clustering accuracy and biological explanation comprehensively. Methods:The cell marker-based clustering strategy(cmCluster),which is a modified Louvain clustering method, aims to search the optimal clusters through genetic algorithm(GA)and grid search based on the cell type annotation results. Results:By applying cmCluster on a set of single-cell transcriptome data, the results showed that it was beneficial for the recognition of cell populations and explanation of biological function even on the occasion of incomplete cell type information or multiple data resources. In addition,cmCluster also produced clear boundaries and appropriate subtypes with potential marker genes. The relevant code is available in GitHub website(huangyuwei301/cmCluster).Conclusions:We speculate that cmCluster provides researchers effective screening strategies to improve the accuracy of subsequent biological analysis, reduce artificial bias, and facilitate the comparison and analysis of multiple studies. | Yuwei Huang Huidan Chang Xiaoyi Chen Jiayue Meng Mengyao Han Tao Huang Liyun Yuan Guoqing Zhang | 2023 | Quantitative Biology2023,11,2: | 0
He-Wei Granule enhances anti-tumor activity of cyclophosphamide by changing tumor microenvironment显示文摘Objective:He-Wei Granule(HWKL)is a modern product derived from the modified formulation of traditional Chinese medicine Banxia Xiexin Decoction(BXD),which remarkedly enhanced the anti-proliferation activity of cyclophosphamide(CTX)on HepG2 and SGC-7901 cell lines in vitro in our previous research. The aim of the study was to investigate the synergistic effects of HWKL and CTX using a transplanted H22 hepatocellular carcinoma mouse model. Methods:The CTX-toxic-reducing efficacy of HWKL was evaluated by hematology indexes, organ indexes and marrow DNA detection. To investigate the underlying mechanisms, histopathology test, immunohistochemistry test and TUNEL staining were conducted. The efficacy of HWKL on the micro-vessel density(MVD)in tumor tissue was also evaluated by measuring CD34 level. Results:High dose HWKL(6.75 g/kg)markedly attenuated CTX-induced hepatotoxicity and myelosuppression while significantly enhanced CTX anticancer efficacy in vivo.Further mechanism investigation suggested that high dose HWKL significantly increased cleaved Caspase 3 level and promoted apoptosis in tumor tissue by up-regulating Bax expression and down-regulating Bcl-2 and FasL expressions.Compared with CTX alone group, the decrease in LC-3B and Beclin 1 levels suggested that the autophagy in H22 carcinoma was significantly inhibited with addition of high dose HWKL.ELISA assay results indicated that the autophagy inhibition was achieved by decreasing p53 expression, blocking PI3K/AKT/mTOR pathway and recovering Th1/Th2 cytokine balance. In addition,CD34 and EGFR immunohistochemistry assay suggest that high dose HWKL could significantly decrease micro-vessel density(MVD)and inhibit angiogenesis in H22 carcinoma. Conclusion:It can be concluded that high-dose HWKL enhanced CTX efficacy by promoting apoptosis, inhibiting autophagy and angiogenesis in tumor tissue while significantly alleviated CTX-induced toxicity, and could be applied along with CTX in clinical treatment as a supplement agent. | Jianxiu Zhai Zehai Song Hang Chang Yuwei Wang Na Han Zhihui Liu Jun Yin | 2022 | Chinese Herbal Medicines2022,14,1: | 0
