| 1 | Elimination of senescent cells byβ-galactosidase-targeted prodrug attenuates inflammation and restores physical function in aged mice显示文摘Cellular senescence,a persistent state of cell cycle arrest,accumulates in aged organisms,contributes to tissue dysfunction,and drives age-related phenotypes.The clearance of senescent cells is expected to decrease chronic,low-grade inflammation and improve tissue repair capacity,thus attenuating the decline of physical function in aged organisms.However,selective and effective clearance of senescent cells of different cell types has proven challenging.Herein,we developed a prodrug strategy to design a new compound based on the increased activity of lysosomalβ-galactosidase(β-gal),a primary characteristic of senescent cells.Our prodrug SSK1 is specifically activated byβ-gal and eliminates mouse and human senescent cells independently of senescence inducers and cell types.In aged mice,our compound effectively cleared senescent cells in different tissues,decreased the senescence-and age-associated gene signatures,attenuated low-grade local and systemic inflammation,and restored physical function.Our results demonstrate that lysosomalβ-gal can be effectively leveraged to selectively eliminate senescent cells,providing a novel strategy to develop anti-aging interventions. | Yusheng Cai Huanhuan Zhou Yinhua Zhu Qi Sun Yin Ji Anqi Xue Yuting Wang Wenhan Chen Xiaojie Yu Longteng Wang Han Chen Cheng Li Tuoping Luo Hongkui Deng | 2020 | Cell Research2020,30,7: | 12 |
| 2 | Underground ground control monitoring and interpretation,and numerical modeling, and shield capacity design显示文摘Mine or longwall panel layout is a 3D structure with highly non-uniform stress distribution. Recognition of such fact will facilitate underground problem identification/investigation and solving by numerical modeling through proper model construction. Due to its versatility, numerical modeling is the most popular method for ground control design and problem solving. However numerical modeling results require highly experienced professionals to interpret its validity/applicability to actual mining operations due to complicated mining and geological conditions. Underground ground control monitoring is routinely performed to predict roof behavior such as weighting and weighting interval without matching observation of face mining condition while the mining pressures are being monitored, resulting in unrealistic interpretation of the obtained data on mining pressure. The importance of ground control pressure monitoring and simultaneous observation of mining and geological conditions is illustrated by an example of shield leg pressure monitoring and interpretation in an U.S. longwall coal mine: it was found that the roof strata act like a plate, not an individual block of the size of a shield dimension, as commonly assumed by all researchers and shield capacity is not a fixed property for a longwall panel or a mine or a coal seam. | 3 | Confined van der Waals Epitaxial Growth of Two-Dimensional Large Single-Crystal In_(2)Se_(3) for Flexible Broadband Photodetectors显示文摘The controllable growth of two-dimensional(2D)semiconductors with large domain sizes and high quality is much needed in order to reduce the detrimental efect of grain boundaries on device performance but has proven to be challenging.Here,we analyze the precursor concentration on the substrate surface which signifcantly infuences nucleation density in a vapor deposition growth process and design a confned micro-reactor to grow 2D In_(2)Se_(3) with large domain sizes and high quality.Te uniqueness of this confned micro-reactor is that its size is∼102-103 times smaller than that of a conventional reactor.Such a remarkably small reactor causes a very low precursor concentration on the substrate surface,which reduces nucleation density and leads to the growth of 2D In_(2)Se_(3) grains with sizes larger than 200�m.Our experimental results show large domain sizes of the 2D In_(2)Se_(3) with high crystallinity.Te fexible broadband photodetectors based on the as-grown In_(2)Se_(3) show rise and decay times of 140 ms and 25 ms,efcient response(5.6 A/W),excellent detectivity(7×10^(10) Jones),high external quantum efciency(251%),good fexibility,and high stability.Tis study,in principle,provides an efective strategy for the controllable growth of high quality 2D materials with few grain boundaries. | Lei Tang Changjiu Teng Yuting Luo Usman Khan Haiyang Pan Zhengyang Cai Yue Zhao Bilu Liu Hui-Ming Cheng | 2019 | Research2019,,1: | 7 |

| 5 | Domain Adaptation Network Based on Autoencoder显示文摘The domain adaptation uses labeled source domain data to train a classifier to be used in the target domain with no or small amount of labeled data. Usually there exists discrepancy in terms of marginal and conditional distributions for both source and target domains,which is of critical importance to minimize the distribution discrepancy between domains. As a classical model in deep learning, the autoencoder is capable of realizing distribution matching and enhancing classification accuracy by extracting more abstract and effective features from data. A Domain adaptation network based on autoencoder(DANA) is proposed. The DANA structure consists of a couple of encoding layers: a feature extraction layer and a classification layer. For the feature extraction layer,the marginal distributions of source and target domains are matched by using the nonparametric maximum mean discrepancy measurement. For the classification layer, the softmax regression model is applied to encode the label information of source domains meanwhile to match the conditional distribution. | 7 | The Scorpion Venom Peptide Smp76 Inhibits Viral Infection by Regulating Type-Ⅰ Interferon Response显示文摘Dengue virus(DENV) and Zika virus(ZIKV) have spread throughout many countries in the developing world and infect millions of people every year, causing severe harm to human health and the economy. Unfortunately, there are few effective vaccines and therapies available against these viruses. Therefore, the discovery of new antiviral agents is critical.Herein, a scorpion venom peptide(Smp76) characterized from Scorpio maurus palmatus was successfully expressed and purified in Escherichia coli BL21(DE3). The recombinant Smp76(rSmp76) was found to effectively inhibit DENV and ZIKV infections in a dose-dependent manner in both cultured cell lines and primary mouse macrophages. Interestingly,rSmp76 did not inactivate the viral particles directly but suppressed the established viral infection, similar to the effect of interferon(IFN)-b. Mechanistically, rSmp76 was revealed to upregulate the expression of IFN-b by activating interferon regulatory transcription factor 3(IRF3) phosphorylation, enhancing the type-Ⅰ IFN response and inhibiting viral infection.This mechanism is significantly different from traditional virucidal antimicrobial peptides(AMPs).
