| 1 | Biomonitoring for traditional herbal medicinal products using DNA metabarcoding and single molecule, real-time sequencing显示文摘Global concerns have been paid to the potential hazard of traditional herbal medicinal products(THMPs). Substandard and counterfeit THMPs, including traditional Chinese patent medicine, health foods, dietary supplements, etc. are potential threats to public health. Recent marketplace studies using DNA barcoding have determined that the current quality control methods are not sufficient for ensuring the presence of authentic herbal ingredients and detection of contaminants/adulterants. An efficient biomonitoring method for THMPs is of great needed. Herein, metabarcoding and single-molecule, realtime(SMRT) sequencing were used to detect the multiple ingredients in Jiuwei Qianghuo Wan(JWQHW), a classical herbal prescription widely used in China for the last 800 years. Reference experimental mixtures and commercial JWQHW products from the marketplace were used to confirm the method. Successful SMRT sequencing results recovered 5416 and 4342 circular-consensus sequencing(CCS) reads belonging to the ITS2 and psb A-trn H regions. The results suggest that with the combination of metabarcoding and SMRT sequencing, it is repeatable, reliable, and sensitive enough to detect species in the THMPs, and the error in SMRT sequencing did not affect the ability to identify multiple prescribed species and several adulterants/contaminants. | 2 | An integrated virtual geographic environmental simulation framework:a case study of flood disaster simulation显示文摘Dynamic flood disaster simulation is an emerging and promising technology significantly useful in urban planning,risk assessment,and integrated decision support systems.It is still an important issue to integrate the large assets such as dynamic observational data,numerical flood simulation models,geographic information technologies,and computing resources into a unified framework.For the intended end user,it is also a holistic solution to create computer interpretable representations and gain insightful understanding of the dynamic disaster processes,the complex impacts,and interactions of disaster factors.In particular,it is still difficult to access and join harmonized data,processing algorithms,and models that are provided by different environmental information infrastructures.In this paper,we demonstrate a virtual geographic environments-based integrated environmental simulation framework for flood disaster management based on the notion of interlinked resources,which is capable of automated accumulating and manipulating of sensor data,creating dynamic geo-analysis and three-dimensional visualizations of ongoing geo-process,and updating the contents of simulation models representing the real environment.The prototype system is evaluated by applying it as a proof of concept to integrate in situ weather observations,numerical weather and flood disaster simulation models,visualization,and analysis of the real time flood event.Case applications indicate that the developed framework can be adopted for use by decision-makers for short-term planning and control since the resulting simulation and visualization are completely based on the latest status of environment. | Yulin DING Qing ZHU Hui LIN | 2014 | Geo-Spatial Information Science2014,17,4: | 9 |

| 6 | Improved activity of MC3T3-E1 cells by the exciting piezoelectric BaTiO_(3)/TC4 using low-intensity pulsed ultrasound显示文摘Developing bioactive materials for bone implants to enhance bone healing and bone growth has for years been the focus of clinical research.Barium titanate(BT)is an electroactive material that can generate electrical signals in response to applied mechanical forces.In this study,a BT piezoelectric ceramic coating was synthesized on the surface of a TC4 titanium alloy,forming a BT/TC4 material,and low-intensity pulsed ultrasound(LIPUS)was then applied as a mechanical stimulus.The combined effects on the biological responses of MC3T3-E1 cells were investigated.Results of scanning electron microscopy,energy-dispersive X-ray spectroscopy,and X-ray diffraction showed that an uniform nanospheres-shaped BT coating was formed on TC4 substrate.Piezoelectric behaviors were observed using piezoelectric force microscopy with the piezoelectric coefficient d_(33)of 0.42 pC/N.Electrochemical measures indicated that LIPUS-stimulated BT/TC4 materials could produce a microcurrent of approximately 10μA/cm^(2).In vitro,the greatest osteogenesis(cell adhesion,proliferation,and osteogenic differentiation)was found in MC3T3-E1 cells when BT/TC4 was stimulated using LIPUS.Furthermore,the intracellular calcium ion concentration increased in these cells,possibly because opening of the L-type calcium ion channels was promoted and expression of the Ca_(V)1.2 protein was increased.Therefore,the piezoelectric BT/TC4 material with LIPUS loading synergistically promoted osteogenesis,rending it a potential treatment for early stage formation of reliable bone-implant contact. | Kunzhan Cai Yilai Jiao Quan Quan Yulin Hao Jie Liu Lin Wu | 2021 | Bioactive Materials2021,6,11: | 6 |
| 7 | Facile one-step synthesis of graphitic carbon nitride-modified biochar for the removal of reactive red 120 through adsorption and photocatalytic degradation显示文摘Converting waste biomass into value-added biochar has been considered as a green and sustainable strategy for resource management and pollution control.In this study,graphitic carbon nitride(g-C_(3)N_(4))modified biochars(BCs)were produced through one-pot pyrolysis of urea and hickory chips in differential ratios at 520℃.The resulting BC/g-C_(3)N_(4)composites were evaluated in laboratory for their physicochemical,adsorptive,and photocatalytic properties.The characterization tests showed the successful synthesis of the BC/g-C_(3)N_(4)composites that introduced g-C_(3)N_(4)structure,N-containing surface func-tional groups,reduced surface area,and better thermal stability to the biochar.After modification,the BC/g-C_(3)N_(4)composites showed better adsorption ability to reactive red 120(RR120)than the pristine BC,due to the strong electrostatic attrition between N-containing functional groups of g-C_(3)N_(4)on biochar surface and anionic RR120.The BC/g-C_(3)N_(4)composites also inherited g-C_(3)N_(4)’s photocatalytic activity,which is visible light responsive to generate free radicals for RR120 degradation.In addition,the composites with higher urea modification ratios were more effective in the degradation of RR120.Overall,this study demonstrates the feasibility and promising potential of combining biochar and photocatalyst for the removal of aqueous dye.Because of the synergistic adsorption and photodegradation ability,BC/g-C_(3)N_(4)composites present a novel and cost-effective solution for the removal of aqueous dye and other photodegradable contaminants under natural conditions. | Yulin Zheng Yichen Yang Yue Zhang Weixin Zou Yidan Luo Lin Dong Bin Gao | 2019 | Biochar2019,1,1: | 5 |

| 9 | Simplified full-depth precast concrete deck panel systems for accelerated bridge construction显示文摘A simplified full-depth precast concrete deck panel system for accelerating bridge construction(ABC) is introduced and a finite element analysis(FEA) is conducted to investigate the static and dynamic responses of this conceptual deck system. The FEA results are compared to those of the traditional full-depth precast concrete deck panel system. The comparison results show that the mechanical behavior of the new deck system is different from that of the traditional deck system. The concrete decks in the new system act as two-way slabs, instead of the one-way slab in the traditional system. Meanwhile, the connections in both the longitudinal and transverse directions may need to accommodate the negative moments.Compared to those in the traditional system, the longitudinal nominal stress at middle span increases a lot in the new deck system and the effective flange width varies significantly. | 11 | The LTR of endogenous retrovirus ev21 retains promoter activity and exhibits tissue specific transcription in chicken显示文摘Endogenous viruses integrate into the host genome and influence the expression of neighboring genes through their long terminal repeats (LTRs). In this study, we analyzed the promoter, enhancer and transcriptional activities of the chicken endogenous retrovirus ev21 LTR. The LTR was cloned into pGL3-basic and pGL3-promoter luciferase vectors in forward and reverse orientation separately. The luciferase activities were detected respectively in chicken embryonic fibroblast cell, human embryonic kidney cell line, human lung carcinoma cell line, Chinese hamster ovary cell line, and murine melanoma cell line. Relative luciferase activity analysis indicated that the ev21 LTR retained bi-directional promoter activity but no detectable enhancer activity in these cells. The constructs containing the LTR and F1 region show stronger promoter activity than the constructs containing only LTR. The transcriptional pattern of ev21 LTR varied in tissues of late feathering White Leghorn chicken at post-hatch day one. Skin exhibits the highest expression in the tissues examed. | 15 | The interaction between polyphyllin I and SQLE protein induces hepatotoxicity through SREBP-2/HMGCR/SQLE/LSS pathway显示文摘Polyphyllin I(PPI)and polyphyllin II(PII)are the main active substances in the Paris polyphylla.However,liver toxicity of these compounds has impeded their clinical application and the potential hepatotoxicity mechanisms remain to be elucidated.In this work,we found that PPI and PII exposure could induce significant hepatotoxicity in human liver cell line L-02 and zebrafish in a dose-dependent manner.The results of the proteomic analysis in L-02 cells and transcriptome in zebrafish indicated that the hepatotoxicity of PPI and PII was associated with the cholesterol biosynthetic pathway disorders,which were alleviated by the cholesterol biosynthesis inhibitor lovastatin.Additionally,3-hydroxy-3-methy-lglutaryl CoA reductase(HMGCR)and squalene epoxidase(SQLE),the two rate-limiting enzymes in the cholesterol synthesis,selected as the potential targets,were confirmed by the molecular docking,the overexpression,and knockdown of HMGCR or SQLE with siRNA.Finally,the pull-down and surface plasmon resonance technology revealed that PPI could directly bind with SQLE but not with HMGCR.Collectively,these data demonstrated that PPI-induced hepatotoxicity resulted from the direct binding with SQLE protein and impaired the sterol-regulatory element binding protein 2/HMGCR/SQLE/lanosterol synthase pathways,thus disturbing the cholesterol biosynthesis pathway.The findings of this research can contribute to a better understanding of the key role of SQLE as a potential target in drug-induced hepatotoxicity and provide a therapeutic strategy for the prevention of drug toxic effects with similar structures in the future. | Zhiqi Li Qiqi Fan Meilin Chen Ying Dong Farong Li Mingshuang Wang Yulin Gu Simin Guo Xianwen Ye Jiarui Wu Shengyun Dai Ruichao Lin Chongjun Zhao | 2023 | Journal of Pharmaceutical Analysis2023,13,1: | 2 |

| 19 | Astrocyte reactivity in related brain regions in a mouse model of MPTP-induced Parkinson's disease显示文摘BACKGROUND:Severe injury to dopaminergic neuronal cell bodies and their axon terminals in the substantia nigra pars compacta(SNC) has been observed in both Parkinson's disease(PD) patients or in 1-methy-4-phenyl-1,2,3,6-tetrahydropyrindine(MPTP)-induced PD animal models, but only slight injury occurs in the adjacent ventral tegmental area(VTA).The mechanisms underlying this selective injury remain poorly understood. OBJECTIVE:To comparatively observe astrocyte reactivity in the SNC,caudate putamen(CPu), VTA,and frontal association cortex(FrA). DESIGN,TIME AND SETTING:A cellular and molecular biology,randomized,controlled experiment was performed at the Institute of Neurobiology,Department of Human Anatomy, Medical School of Nantong University,between December 2006 and September 2008. MATERIALS:A total of 80 healthy adult male C57BL/6 mice were included in this study.MPTP was purchased from Sigma,USA. METHODS:Mice were randomly divided into a model group(n=64) and a sham-operated group (n=16).PD was induced in the mice from the model group by intraperitoneal injection of 20 mg/kg MPTP,once every three hours,for a total of 4 times. MAIN OUTCOME MEASURES:Tyrosine hydroxylase(TH)-immunoreactive neurons and glial fibrillary acidic protein(GFAP)-immunoreactive astrocytes were examined by dual immunofluorescence labeling.GFAP-immunoreactive astrocytes in the CPu and FrA were determined by immunofluorescent staining.GFAP mRNA expression in the SNC,CPu,VTA,and FrA was detected using real-time polymerase chain reaction.TH protein levels in the TH-immunoreactive axon terminals of the CPu and FrA were detected by Western blotting. RESULTS:Numbers of TH-immunoreactive neurons in the SNC,and TH protein level in the CPu, markedly decreased(by approximately 68%) 1 day after MPTP injection,and gradually increased at 3 days.Simultaneously,astrocyte reactivity was strengthened,in particular at 7 days.However, after MPTP injection,decreases in the numbers of TH-immunoreactive neurons in the VTA,and TH protein levels in the FrA,were less apparent(approximately 15%).Also,no obvious astrocyte reactivity was observed.
