| 1 | Focal adhesion protein Kindlin-2 regulates bone homeostasis in mice显示文摘Our recent studies demonstrate that the focal adhesion protein Kindlin-2 is critical for chondrogenesis and early skeletal development. Here, we show that deleting Kindlin-2 from osteoblasts using the 2.3-kb mouse Col1 a1-Cre transgene minimally impacts bone mass in mice, but deleting Kindlin-2 using the 10-kb mouse Dmp1-Cre transgene, which targets osteocytes and mature osteoblasts, results in striking osteopenia in mice. Kindlin-2 loss reduces the osteoblastic population but increases the osteoclastic and adipocytic populations in the bone microenvironment. Kindlin-2 loss upregulates sclerostin in osteocytes,downregulates β-catenin in osteoblasts, and inhibits osteoblast formation and differentiation in vitro and in vivo. Upregulation ofβ-catenin in the mutant cells reverses the osteopenia induced by Kindlin-2 deficiency. Kindlin-2 loss additionally increases the expression of RANKL in osteocytes and increases osteoclast formation and bone resorption. Kindlin-2 deletion in osteocytes promotes osteoclast formation in osteocyte/bone marrow monocyte cocultures, which is significantly blocked by an anti-RANKLneutralizing antibody. | 2 | Coupling between the Grain for Green Program and a household level-based agricultural eco-economic system in Ansai,Shaanxi Province of China显示文摘The implementation of the Grain for Green Program(GGP)has changed the development track of the agricultural eco-economic system in China.In response to the results of a lag study that investigated the coupling between the GGP and the agricultural eco-economic system in a loess hilly region,we used a structural equation model to analyze the survey data from 494 households in Ansai,a district of Yan’an City in Shaanxi Province of China in 2015.The model clarified the direction and intensity of the coupling between the GGP and the agricultural eco-economic system.The coupling benefits were derived through linkages between the program and various chains in the agricultural eco-economic system.The GGP,the agroecosystem of Ansai and their potential coupling effects were in a state of general coordination.The agroecosystem directly affected the coupling effect,with the standardized path coefficient of 0.87,indicating that the agroecosystem in Ansai at this stage provided basic material support for the coupling between the GGP and the agricultural eco-economic system.The direct path coefficient of agroeconomic system impacted on the coupling effect was-0.76,indicating that partial contradictions occurred between the agroeconomic system and the coupling effect.Therefore,although the current agroecosystem in Ansai should be provided sufficient agroecological resources for the benign coupling between the program and the agricultural eco-economic system,agricultural development failed to effectively transform agroecological resources into agricultural economic advantages in this region,which resulted in a relative lag in the development of the agricultural economic system.Thus,the coupling between the GGP and the agricultural eco-economic system was poor.To improve the coupling and the sustainable development of the agricultural eco-economic system in cropland retirement areas,the industrial structure needs to be diversified,the agricultural resources(including agroecological resources,agricultural economic resources and agricultural social resources)need to be rationally allocated,and the chain structure of the agricultural eco-economic system needs to be continuously improved. | LI Yue WANG Jijun HAN Xiaojia GUO Mancai CHENG Simin QIAO Mei ZHAO Xiaocui | 2020 | Journal of Arid Land2020,12,2: | 2 |
| 3 | The interaction between polyphyllin I and SQLE protein induces hepatotoxicity through SREBP-2/HMGCR/SQLE/LSS pathway显示文摘Polyphyllin I(PPI)and polyphyllin II(PII)are the main active substances in the Paris polyphylla.However,liver toxicity of these compounds has impeded their clinical application and the potential hepatotoxicity mechanisms remain to be elucidated.In this work,we found that PPI and PII exposure could induce significant hepatotoxicity in human liver cell line L-02 and zebrafish in a dose-dependent manner.The results of the proteomic analysis in L-02 cells and transcriptome in zebrafish indicated that the hepatotoxicity of PPI and PII was associated with the cholesterol biosynthetic pathway disorders,which were alleviated by the cholesterol biosynthesis inhibitor lovastatin.Additionally,3-hydroxy-3-methy-lglutaryl CoA reductase(HMGCR)and squalene epoxidase(SQLE),the two rate-limiting enzymes in the cholesterol synthesis,selected as the potential targets,were confirmed by the molecular docking,the overexpression,and knockdown of HMGCR or SQLE with siRNA.Finally,the pull-down and surface plasmon resonance technology revealed that PPI could directly bind with SQLE but not with HMGCR.Collectively,these data demonstrated that PPI-induced hepatotoxicity resulted from the direct binding with SQLE protein and impaired the sterol-regulatory element binding protein 2/HMGCR/SQLE/lanosterol synthase pathways,thus disturbing the cholesterol biosynthesis pathway.The findings of this research can contribute to a better understanding of the key role of SQLE as a potential target in drug-induced hepatotoxicity and provide a therapeutic strategy for the prevention of drug toxic effects with similar structures in the future. | Zhiqi Li Qiqi Fan Meilin Chen Ying Dong Farong Li Mingshuang Wang Yulin Gu Simin Guo Xianwen Ye Jiarui Wu Shengyun Dai Ruichao Lin Chongjun Zhao | 2023 | Journal of Pharmaceutical Analysis2023,13,1: | 2 |

| 9 | Flexible feeding patterns of copepod Centropages tenuiremis in fluctuating conditions: a possible survival strategy to cope with disturbance显示文摘Centropages tenuiremis is a species with a wide distribution range in disturbed coastal waters.However,due to a lack of dietary information,it remains unclear as to how they maintain such dominance in fluctuating conditions.In this study,C.tenuiremis was collected from the Daya Bay Nuclear Power Plant both in inlet and outfall regions at 06:00,12:00 and 18:00 on April 27,2011 and their in situ diet was analyzed using a PCR protocol targeting 18S ribosomal genes.Thirty-four species of prey organisms were identified totally,including Dinophyta,Baciliariophyta,Viridiplantae,Rhizaria,Apicomplexa,Chordata,Mollusca,Arthropoda and Fungi,indicating an obvious omnivorous feeding habit of C.tenuiremis.Centropages tenuiremis obviously exhibited spatial and temporal variations in diet composition.More plant prey(land plants and phytoplankton)were consumed in the morning(~50%),while more animal prey(metazoans and protozoans)were ingested at midday and night(60%–70%).Furthermore,a more diverse diet was detected in the outfall region(10–11 taxa),where the temperatures were relatively higher and more fluctuating,than in the control region(5–10 taxa).This finding indicated that C.tenuiremis could potentially expand its food spectrum under stressful condition.Specifically,C.tenuiremis exhibited phytoplankton preference(58.62%–67.64%)in the outfall region with a lower omnivory index(0.27–0.35)than in the control region(0.51–0.95).However,phytoplankton density was lower than that in the control region,suggesting a possible herbivorous tendency of C.tenuiremis under elevated temperatures to balance the energy acquirement and feeding effort.The flexible food choices of C.tenuiremis observed here could effectively buffer environmental fluctuations and might be an important survival strategy in coastal ecosystems. | Cuilian Xu Simin Hu Zhiling Guo Tao Li Hui Huang Leo Lai Chan Sheng Liu | 2020 | Acta Oceanologica Sinica2020,39,2: | 0 |
| 10 | Improving current and mitigating energy loss in ternary organic photovoltaics enabled by two well-compatible small molecule acceptors显示文摘Ternary organic photovoltaic(OPV)strategy is an effective but facile approach to enhance the photovoltaic performance for single-junction devices.Herein,a series of ternary OPVs were fabricated by employing a wide bandgap donor(PBDB-TF)and two acceptor-donor-acceptor(A-D-A)-type nonfullerene small molecule acceptors(NF-SMAs,called F-2 Cl and 3 TT-OCIC).As the third component,the near-infrared SMA,3 TT-OCIC,has complementary absorption spectrum,narrow bandgap and wellcompatible crystallization property to the host acceptor(F-2 Cl)for efficient ternary OPVs.With these,the optimal ternary devices yield significantly enhanced power conversion efficiency of 15.23%,one of the very few examples with PCE higher than15%other than Y6 systems.This is mainly attributed to the increased short-circuit current density of 24.92 m A cm^(-2) and dramatically decreased energy loss of 0.53 e V.This work presents a successful example for simultaneously improving current,minimizing energy loss and together with modifying the morphology of active layers in OPVs,which will contribute to the further construction of high performance ternary OPVs. | Yanna Sun Huan-Huan Gao Simin Wu Lingxian Meng Xiangjian Wan Mengyang Li Zaifei Ma Ziqi Guo Shitong Li Hongtao Zhang Chenxi Li Yongsheng Chen | 2021 | Science China Chemistry2021,64,4: | 0 |
