2SlMYB75,an MYB-type transcription factor,promotes anthocyanin accumulation and enhances volatile aroma production in tomato fruits显示文摘Genetic manipulation of genes to upregulate specific branches of metabolic pathways is a method that is commonly used to improve fruit quality.However,the use of a single gene to impact several metabolic pathways is difficult.Here,we show that overexpression of the single gene SlMYB75(SlMYB75-OE)is effective at improving multiple fruit quality traits.In these engineered fruits,the anthocyanin content reached 1.86mg g−1 fresh weight at the red-ripe stage,and these SlMYB75-OE tomatoes displayed a series of physiological changes,including delayed ripening and increased ethylene production.In addition to anthocyanin,the total contents of phenolics,flavonoids and soluble solids in SlMYB75-OE fruits were enhanced by 2.6,4,and 1.2 times,respectively,compared to those of wild-type(WT)fruits.Interestingly,a number of aroma volatiles,such as aldehyde,phenylpropanoid-derived and terpene volatiles,were significantly increased in SlMYB75-OE fruits,with some terpene volatiles showing more than 10 times higher levels than those in WT fruits.Consistent with the metabolic assessment,transcriptomic profiling indicated that the genes involved in the ethylene signaling,phenylpropanoid and isoprenoid pathways were greatly upregulated in SlMYB75-OE fruits.Yeast one-hybrid and transactivation assays revealed that SlMYB75 is able to directly bind to the MYBPLANT and MYBPZM cis-regulatory elements and to activate the promoters of the LOXC,AADC2 and TPS genes.The identification of SlMYB75 as a key regulator of fruit quality attributes through the transcriptional regulation of downstream genes involved in several metabolic pathways opens new avenues towards engineering fruits with a higher sensory and nutritional quality.Wei Jian Haohao Cao Shu Yuan Yudong Liu Juanfang Lu Wang Lu Ning Li Jianhui Wang Jian Zou Ning Tang Chan Xu Yulin Cheng Yanqiang Gao Wanpeng Xi Mondher Bouzayen Zhengguo Li 2019Horticulture Research2019,6,1:39
3Distribution characteristic and geological signiflcance of rare earth elements in Lopingian mudstone of Permian,Panxian county,Guizhou province显示文摘In order to discuss the geochemical characteristic of REEs(rare earth elements) and their geological application,we measured the contents of rare earth elements,trace elements and minerals of 29 Lopingian(Late Permian) mudstone samples in Panxian county,carrying out ICP-MS and XRD analysis.The results show that the amount of REEs(185.56-729.46 10à6) is high.The ratios of w(LREE)/w(HREE)(6.84-13.86) and w(La)N/w(Yb)N(1.01-3.02) show clear differentiation of LREEs and HREEs.RREE has a significantly or critically positive correlation with lithophile elements Th,Nb,Ta,Ti,Ga,Sc,Cs,Zr,Hf,Sr,Be and chalcophile element Zn,a critically negative correlation with siderophile element Fe and a slightly positive correlation with illite,illite smectite mixed layers and siderite.REEs originate mainly from terrigenous minerals,in an inorganic phase.Source rocks of our samples consist of Emeishan basalt and a small part of sedimentary rocks,as suggested by the distribution patterns of REEs and w(PREE)-w(La)/w(Yb) diagram.Moreover,abnormal surfaces near the sequence boundaries(SB2,SB3,SB4) are related with the boundaries,identifled by geochemical characteristics of the REEs,such as PREE,w(LREE)/w(HREE),Eu/Eu and Ceanom.Shao Yubao Guo Yinghai Qin Yong Shen Yulin Tian Li 2011Mining Science and Technology2011,21,4:23
4Accumulation of soil organic carbon during natural restoration of desertified grassland in China's Horqin Sandy Land显示文摘China's Horqin Sandy Land,a formerly lush grassland,has experienced extensive desertification that caused considerable carbon(C) losses from the plant-soil system.Natural restoration through grazing exclusion is a widely suggested option to sequester C and to restore degraded land.In a desertified grassland,we investigated the C accumulation in the total and light fractions of the soil organic matter from 2005 to 2013 during natural restoration.To a depth of 20 cm,the light fraction organic carbon(LFOC) storage increased by 221 g C/m2(84%) and the total soil organic carbon(SOC) storage increased by 435 g C/m2(55%).The light fraction dry matter content represented a small proportion of the total soil mass(ranging from 0.74% in 2005 to 1.39% in 2013),but the proportion of total SOC storage accounted for by LFOC was remarkable(ranging from 33% to 40%).The C sequestration averaged 28 g C/(m2·a) for LFOC and 54 g C/(m2·a) for total SOC.The total SOC was strongly and significantly positively linearly related to the light fraction dry matter content and the proportions of fine sand and silt+clay.The light fraction organic matter played a major role in total SOC sequestration.Our results suggest that grazing exclusion can restore desertified grassland and has a high potential for sequestering SOC in the semiarid Horqin Sandy Land.YuQiang LI XueYong ZHAO FengXia ZHANG Tala AWADA ShaoKun WANG HaLin ZHAO TongHui ZHANG YuLin LI 2015Journal of Arid Land2015,7,3:21
5Determining "abnormal" levator hiatus distensibility using three-dimensional transperineal ultrasound in Chinese women显示文摘The dimension of the levator hiatus is a possible predictor of pelvic organ prolapse (POP). Thisretrospective study investigated 360 women who went to urogynecological clinic for pelvic floor discomfort.Levator hiatus dimensions were obtained by three-dimensional transperineal ultrasound and results werecompared between women with and without significantly objective prolapse (International Continence SocietyPOP quantification, grade 2 or higher). Receiver operating characteristic (ROC) curve analyses were performed todetermine valid screening index for detecting abnormal levator hiatus distensibility. Women with significantlyobjective prolapse had significantly higher levator hiatus dimensions than those without (all P 〈 0.001). ROCcurve analyses confirmed that hiatal area (HA) of 19.5 cm2 during Valsalva maneuver can be used as single-screening index for abnormal levator hiatus distensibility with sensitivity of 0.80 and specificity of 0.70. In thisstudy, we used a two-step method and achieved higher sensibility (0.80 vs. 0.87) without reducing specificity (0.70vs. 0.71) compared with a single-screening index method. 6Fatigue behavior of Ti-6Al-4V cellular structures fabricated by additive manufacturing technique显示文摘Porous titanium and its alloys have been considered as promising replacement for dense implants, as they possess low elastic modulus comparable to that of compact human bones and are capable of providing space for in-growth of bony tissues to achieve a better fixation. Recently, the additive manufacturing(AM) method has been successfully applied to the fabrication of Ti-6 Al-4 V cellular meshes and foams.Comparing to traditional fabrication methods, the AM method offers advantages of accurate control of complex cell shapes and internal pore architectures, thus attracting extensive attention. Considering the long-term safety in the human body, the metallic cellular structures should possess high fatigue strength.In this paper, the recent progress on the fatigue properties of Ti-6 Al-4 V cellular structures fabricated by the AM technique is reviewed. 7Biofunctional magnesium coated Ti6Al4V scaffold enhances osteogenesis and angiogenesis in vitro and in vivo for orthopedic application显示文摘The insufficient osteogenesis and osseointegration of porous titanium based scaffold limit its further application.Early angiogenesis is important for scaffold survival.It is necessary to develop a multifunctional surface on titanium scaffold with both osteogenic and angiogenic properties.In this study,a biofunctional magnesium coating is deposited on porous Ti6Al4V scaffold.For osseointegration and osteogenesis analysis,in vitro studies reveal that magnesium-coated Ti6Al4V co-culture with MC3T3-E1 cells can improve cell proliferation,adhesion,extracellular matrix(ECM)mineralization and ALP activity compared with bare Ti6Al4V cocultivation.Additionally,MC3T3-E1 cells cultured with magnesium-coated Ti6Al4V show significantly higher osteogenesisrelated genes expression.In vivo studies including fluorochrome labeling,micro-computerized tomography and histological examination of magnesium-coated Ti6Al4V scaffold reveal that new bone regeneration is significantly increased in rabbits after implantation.For angiogenesis studies,magnesium-coated Ti6Al4V improve HUVECs proliferation,adhesion,tube formation,wound-healing and Transwell abilities.HUVECs cultured with magnesium-coated Ti6Al4V display significantly higher angiogenesis-related genes(HIF-1αand VEGF)expression.Microangiography analysis reveal that magnesium-coated Ti6Al4V scaffold can significantly enhance the blood vessel formation.This study enlarges the application scope of magnesium and provides an optional choice to the conventional porous Ti6Al4V scaffold with enhanced osteogenesis and angiogenesis for further orthopedic applications.Peng Gao Bo Fan Xiaoming Yu Wenwen Liu Jie Wu Lei Shi Di Yang Lili Tan Peng Wan Yulin Hao Shujun Li Wentao Hou Ke Yang Xiaokang Li Zheng Guo 2020Bioactive Materials2020,5,3:15

9Progress and records in the study of endogenetic mineralization during collisional orogenesis显示文摘To develop and perfect the theory of plate tectonics and regional metallogeny, metal-logenesis during collisional orogenesis should be thoroughly studied and will attract increasing at-tention of more and more scientists. This paper presents the main aspects of research and discus-sions on metallogenesis during collisional orogenesis after the development of plate tectonics, and accordingly divides the study history into two stages, i.e. the junior stage during 1971-1990 and the senior stage after 1990. Beginning with the negation of mineralization in the collision regime by Guild (1971), the focus of study was put on whether there occurred any mineralization during collisional orogenesis at the junior stage. 10Metamodel-based Global Optimization Using Fuzzy Clustering for Design Space Reduction显示文摘High fidelity analysis are utilized in modern engineering design optimization problems which involve expensive black-box models.For computation-intensive engineering design problems,efficient global optimization methods must be developed to relieve the computational burden.A new metamodel-based global optimization method using fuzzy clustering for design space reduction(MGO-FCR) is presented.The uniformly distributed initial sample points are generated by Latin hypercube design to construct the radial basis function metamodel,whose accuracy is improved with increasing number of sample points gradually.Fuzzy c-mean method and Gath-Geva clustering method are applied to divide the design space into several small interesting cluster spaces for low and high dimensional problems respectively.Modeling efficiency and accuracy are directly related to the design space,so unconcerned spaces are eliminated by the proposed reduction principle and two pseudo reduction algorithms.The reduction principle is developed to determine whether the current design space should be reduced and which space is eliminated.The first pseudo reduction algorithm improves the speed of clustering,while the second pseudo reduction algorithm ensures the design space to be reduced.Through several numerical benchmark functions,comparative studies with adaptive response surface method,approximated unimodal region elimination method and mode-pursuing sampling are carried out.The optimization results reveal that this method captures the real global optimum for all the numerical benchmark functions.And the number of function evaluations show that the efficiency of this method is favorable especially for high dimensional problems.Based on this global design optimization method,a design optimization of a lifting surface in high speed flow is carried out and this method saves about 10 h compared with genetic algorithms.This method possesses favorable performance on efficiency,robustness and capability of global convergence and gives a new optimization strategy for engineering design optimization problems involving expensive black box models.LI Yulin LIU Li LONG Teng DONG Weili 2013Chinese Journal of Mechanical Engineering2013,26,5:13
11N and P resorption in a pioneer shrub(Artemisia halodendron) inhabiting severely desertified lands of Northern China显示文摘Nutrient resorption is an important conservation mechanism for plants to overcome nutrient limitation in the less fertile area of desertified land.In the semi-arid Horqin Sandy Land of Northern China,the shrub Artemisia halodendron usually colonizes into the bare ground of severely desertified land as a pioneer species.It is,therefore,expected that A.halodendron will be less dependent on current nutrient uptake through efficient and proficient resorption of nutrients.In this study,we found that averaged nitrogen(N)and phosphorus(P)concentrations in senesced leaves significantly varied from 12.3 and 1.2 mg/g in the shifting sand dune to 15.9 and 1.9 mg/g in the fixed sand dune,respectively,suggesting that foliar N and P resorption of A.halodendron were more proficient in the shifting sand dune.In particular,positive relationships between nutrient concentrations in senesced leaves and soil nutrient availability indicate that A.halodendron in infertile habitats is more likely to manage with a low level of nutrients in senesced leaves,giving this species an advantage in infertile soil.Moreover,foliar N-and P-resorption efficiencies and proficiencies showed limited inter-annual variability although annual precipitation varied greatly among 2007–2009.However,N and P resorption of A.halodendron were not more efficient and proficient than those previously reported for other shrubs,indicating that the pioneer shrub in sand dune environments does not rely more heavily than other plants on the process of resorption to conserve nutrients.Incomplete resorption of nutrients in A.halodendron suggests that senesced-leaf fall would return litter with high quality to the soil,and thereby would indirectly improve soil nutrient availability.The restoration of desertified land,therefore,may be accelerated after A.halodendron pioneers into shifting sand dunes.YuLin LI Chen JING Wei MAO Duo CUI XinYuan WANG XueYong ZHAO 2014Journal of Arid Land2014,6,2:11
12Discovery of mantle and lower crust xenoliths from early Cretaceous volcanic rocks of southwestern Tianshan,Xinjiang显示文摘In Tuoyun area of southwestern Tianshan, mantle and lower crust xenoliths are present In the volcanic rocks with ages of 101-123 Ma. Mantle xenoliths include mineral megacrysts such as kaersutite and pargasite, feldspar, biotite, and rare pyroxene and rock fragments such as perodotite, pyroxenite, amphibolite, and rare glimmerite. Lower crust xenoliths are mainly banded and massive granulite. The volcanic rocks were produced by within-plate magmatism. Occurrence of hydrous and volatile mineral megacrysts, amphibolite, and some pyroxenite containing hydrous and volatile minerals indicates that mantle metasomatism was intense. 13Cotton bollworm resistance to Bt transgenic cotton: A case analysis显示文摘Cotton bollworm (Helicoverpa armigera) is one of the most serious insect pests of cotton. Transgenic cotton expressing Cry toxins derived from a soil bacterium, Bacillus thuringiensis (Bt), has been produced to target this pest. Bt cotton has been widely planted around the world, and this has resulted in efficient control of bollworm populations with reduced use of synthetic insecticides. However, evolution of resistance by this pest threatens the continued success of Bt cotton. To date, no field populations of bollworm have evolved significant levels of resistance; however, several laboratory-selected Cry-resistant strains of H. armigera have been obtained, which suggests that bollworm has the capacity to evolve resistance to Bt. The development of resistance to Bt is of great concern, and there is a vast body of research in this area aimed at ensuring the continued success of Bt cotton. Here, we review studies on the evolution of Bt resistance in H. armigera, focusing on the biochemical and molecular basis of Bt resistance. 16Establishment of intestinal organoid cultures modeling injury-associated epithelial regeneration显示文摘The capacity of 3D organoids to mimic physiological tissue organization and functionality has provided an invaluable tool to model development and disease in vitro.However,conventional organoid cultures primarily represent the homeostasis of self-organizing stem cells and their derivatives.Here,we established a novel intestinal organoid culture system composed of 8 components,mainly including VPA,EPZ6438,LDN193189,and R-Spondin 1 conditioned medium,which mimics the gut epithelium regeneration that produces hyperplastic crypts following injury;therefore,these organoids were designated hyperplastic intestinal organoids(Hyper-organoids).Single-cell RNA sequencing identified different regenerative stem cell populations in our Hyper-organoids that shared molecular features with in vivo injury-responsive Lgr5+stem cells or Clu+revival stem cells.Further analysis revealed that VPA and EPZ6438 were indispensable for epigenome reprogramming and regeneration in Hyper-organoids,which functioned through epigenetically regulating YAP signaling.Furthermore,VPA and EPZ6438 synergistically promoted regenerative response in gut upon damage in vivo.In summary,our results demonstrated a new in vitro organoid model to study epithelial regeneration,highlighting the importance of epigenetic reprogramming that pioneers tissue repair.Molong Qu Liang Xiong Yulin Lyu Xiannian Zhang Jie Shen Jingyang Guan Peiyuan Chai Zhongqing Lin Boyao Nie Cheng Li Jun Xu Hongkui Deng 2021Cell Research2021,31,3:9
17Genome-wide Dissection of Co-selected UV-B Responsive Pathways in the UV-B Adaptation of Qingke显示文摘Qingke (Tibetan hulless barley) has long been cultivated and exposed to long-term and strong UV-B radiation on the Tibetan Plateau, which renders it an ideal species for elucidating novel UV-B responsive mechanisms in plants. Here we report a comprehensive metabolite profiling and metabolite-based genome-wide association study (mGWAS) using 196 diverse qingke and barley accessions. Our results demonstrated both constitutive and induced accumulation, and common genetic regulation, of metabolites from different branches of the phenylpropanoid pathway that are involved in UV-B protection. A total of 90 significant mGWAS loci for these metabolites were identified in barley-qingke differentiation regions, and a number of high-level metabolite trait alleles were found to be significantly enriched in qingke, suggesting co-selection of various phenylpropanoids. Upon dissecting the entire phenylpropanoid pathway, we identified some key determinants controlling natural variation of phenylpropanoid content, including three novel proteins, a flavone C-pentosyltransferase, a tyramine hydroxycinnamoyl acyltransferase, and a MYB transcription factor. 18Impending HRT wave precursors to the Wenchuan M_s8.0 earthquake and methods of earthquake impending prediction by using HRT wave显示文摘We deployed four geo-electric monitoring stations in Sichuan and Yunnan provinces from 2004, using the new generation of equipment (PS-100) and technologies to capture the HRT wave earthquake precursor. Before the Wenchuan Ms8.0 earthquake, we recorded the HRT wave precursor at the only operating station in Hongge (HG, Δ=465 km) and found that significant impending signal had been recorded at the station in the early morning ( 0―5 am) of 12th of May, 2008. The precursor for this earthquake is consistent with precursors recorded for other strong earthquakes. The measured physical properties (geo-resistivity and telluric-current) show tidal wave period oscillations from several days to several months before the earthquakes and the amplitude of such HT oscillation increases significantly towards the occurrence of an earthquake. These HT and RT waves from the epicenter have a causal relationship with the earthquakes that happened several days later. The arrival time of two RT waves is proportional to the distance from the station to the epicenter. The estimated natural decay of the amplitude is correlated with the natural period (T0) of the earthquake fault, which is proportional to the fault length. From this relationship, we can predict the earthquake magnitude. For magnitude 6―9 earthquakes, the natural period is about 1―6 hours. Such oscillation comes from the epicenter area and they can propagate several thousand kilometers in the Earth's crust. Before a strong earthquake in the shallow crust, the conductive pore fluid will experience major changes before the fault rapture. Such fluid change will emit an oscillation in the pore fluid pressure. This is the mechanism for the HRT wave generation. Since the China Earthquake Administration funded the HRT wave short-term earth-quake prediction project in 2003, the first record of HRT precursor wave has been recorded from the 2004-12-26 Sumatra Mw9.0 earthquake with the largest epicentre distance Δ=2900 km. Thereafter, we have captured HRT waves from more than twenty strong earthquakes, which are well-matched and show repeatability, consistency and regularity. All our observation with the HRT waves demonstrate that HRT wave precursors to earthquakes indeed exist. Strong earthquakes can be predicted and short-term and impending earthquake prediction is achievable in the very near future. 19Neural substrates of data-driven scientific discovery:An fMRI study during performance of number series completion task显示文摘Although much has been known about how humans psychologically perform data-driven scientific discovery,less has been known about its brain mechanism.The number series completion is a typical data-driven scientific discovery task,and has been demonstrated to possess the priming effect,which is attributed to the regularity identification and its subsequent extrapolation.In order to reduce the heterogeneities and make the experimental task proper for a brain imaging study,the number magnitude and arithmetic operation involved in number series completion tasks are further restricted.Behavioral performance in Experiment 1 shows the reliable priming effect for targets as expected.Then,a factorial design (the priming effect:prime vs.target;the period length:simple vs.complex) of event-related functional magnetic resonance imaging (fMRI) is used in Experiment 2 to examine the neural basis of data-driven scientific discovery.The fMRI results reveal a double dissociation of the left DLPFC (dorsolateral prefrontal cortex) and the left APFC (anterior prefrontal cortex) between the simple (period length=1) and the complex (period length=2) number series completion task.The priming effect in the left DLPFC is more significant for the simple task than for the complex task,while the priming effect in the left APFC is more significant for the complex task than for the simple task.The reliable double dissociation may suggest the different roles of the left DLPFC and left APFC in data-driven scientific discovery.The left DLPFC (BA 46) may play a crucial role in rule identification,while the left APFC (BA 10) may be related to mental set maintenance needed during rule identification and extrapolation.ZHONG Ning LIANG PeiPeng QIN YuLin LU ShengFu YANG YanHui LI KunCheng 2011Science China(Life Sciences)2011,54,5:8
20Ultrasonic vibrations and coal permeability: Laboratory experimental investigations and numerical simulations显示文摘Ultrasonic vibrations in coal lead to cavitation bubble oscillation, growth, shrinkage, and collapse, and the strong vibration of cavitation bubbles not only makes coal pores break and cracks propagate, but plays an important role in enhancing the permeability of coal. In this paper, the influence of ultrasonic cavitation on coal and the effects of the sonic waves on crack generation, propagation, connection, as well as the effect of cracks on the coal permeability, are studied. The experimental results show that cracks in coal are generated even connected rapidly after ultrasonic cavitation. Under the effect of ultrasonic cavitation,the permeability increases between 30% and 60%, and the number of cracks in coal also significantly increased. Numerical experiments show that the effective sound pressure is beneficial to fracture propagation and connection, and it is closely related to the permeability.
