mTORC2 promotes type I insulin-like growth factor receptor and insulin receptor activation through the tyrosine kinase activity of mTOR显示文摘rapamycin (mTOR ) 的哺乳动物的目标是 raptor-mTOR (mTORC1 ) 和控制多样的细胞的进程的 rictor-mTOR (mTORC2 ) 建筑群的一个核心部件。 | YancunYin Hui Hua Minjing Li Shu Liu Qingbin Kong Ting Shao Jiao Wang Yuanming Luo Qian Wang Ting Luo Yangfu Jiang | 2016 | Cell Research2016,26,1: | 11
Signaling pathways and targeted therapy for myocardial infarction显示文摘Although the treatment of myocardial infarction(MI)has improved considerably,it is still a worldwide disease with high morbidity and high mortality.Whilst there is still a long way to go for discovering ideal treatments, therapeutic strategies committed to cardioprotection and cardiac repair following cardiac ischemia are emerging.Evidence of pathological characteristics in MI illustrates cell signaling pathways that participate in the survival, proliferation, apoptosis, autophagy of cardiomyocytes, endothelial cells, fibroblasts, monocytes, and stem cells. These signaling pathways include the key players in inflammation response,e.g.,NLRP3/caspase-1 and TLR4/MyD88/NF-κB;the crucial mediators in oxidative stress and apoptosis, for instance,Notch,Hippo/YAP,RhoA/ROCK,Nrf2/HO-1,and Sonic hedgehog;the controller of myocardial fibrosis such as TGF-β/SMADs and Wnt/β-catenin;and the main regulator of angiogenesis,PI3K/Akt,MAPK,JAK/STAT,Sonic hedgehog, etc.Since signaling pathways play an important role in administering the process of MI,aiming at targeting these aberrant signaling pathways and improving the pathological manifestations in MI is indispensable and promising.Hence, drug therapy, gene therapy, protein therapy, cell therapy, and exosome therapy have been emerging and are known as novel therapies. In this review,we summarize the therapeutic strategies for MI by regulating these associated pathways, which contribute to inhibiting cardiomyocytes death, attenuating inflammation, enhancing angiogenesis, etc.so as to repair and re-functionalize damaged hearts. | Qing Zhang Lu Wang Shiqi Wang Hongxin Cheng Lin Xu Gaiqin Pei Yang Wang Chenying Fu Yangfu Jiang Chengqi He Quan Wei | 2022 | Signal Transduction and Targeted Therapy2022,7,4: | 11

Temperature-dependent optical resonance in a thin-walled tubular oxide microcavity显示文摘This work proposes a temperature-response capability of optical resonance in tubular optical oxide microcavities. The thin wall thickness with a subwavelength scale enables these microcavities to interact with the environment effectively. By optimization of the geometries and materials, the tubular microcavities can be tuned into temperature-inert in vacuum, and the experiments support this design. The experiments prove the idea of utilizing them as temperature-inert microcavities. Contrary wavelength shifts from previous studies were observed, which can be explained with the theoretical model. Furthermore, the theoretical results of the present work suggest that novel rolled-up microtubes could act as an exceptional optical microcavity for the application in temperature response. | Yangfu Fang Xianyun Lin Shiwei Tang Borui Xu Jiao Wang Yongfeng Mei | 2017 | Progress in Natural Science:Materials International2017,27,4: | 1

Chaperone-mediated Autophagy Regulates Cell Growth by Targeting SMAD3 in Glioma显示文摘Previous studies suggest that the reduction of SMAD3(mothers against decapentaplegic homolog 3)has a great impact on tumor development, but its exact pathological function remains unclear. In this study,we found that the protein level of SMAD3 was greatly reduced in human-grade IV glioblastoma tissues,in which LAMP2A(lysosome-associated membrane protein type 2A)was significantly up-regulated.LAMP2A is a key ratelimiting protein of chaperone-mediated autophagy(CMA),a lysosome pathway of protein degradation that is activated in glioma. We carefully analyzed the amino-acid sequence of SMAD3 and found that it contained a pentapeptide motif biochemically related to KFERQ,which has been proposed to be a targeting sequence for CMA.In vitro,we confirmed that SMAD3 was degraded in either serum-free or KFERQ motif deleted condition, which was regulated by LAMP2A and interacted with HSC70(heat shock cognate 71 kDa protein).Using isolated lysosomes,amino-acid residues 75 and 128 of SMAD3 were found to be of importance for this process, which affected the CMA pathway in which SMAD3 was involved.Similarly,down-regulating SMAD3 or up-regulating LAMP2A in cultured glioma cells enhanced their proliferation and invasion.Taken together, these results suggest that excessive activation of CMA regulates glioma cell growth by promoting the degradation of SMAD3.Therefore,targeting the SMAD3-LAMP2Amediated CMA-lysosome pathway may be a promising approach in anti-cancer therapy. | Hanqun Liu Yuxuan Yong Xingjian Li Panghai Ye Kai Tao Guoyou Peng Mingshu Mo Wenyuan Guo Xiang Chen Yangfu Luo Yuwan Lin Jiewen Qiu Ziling Zhang Liuyan Ding Miaomiao Zhou Xinling Yang Lin Lu Qian Yang Pingyi Xu | 2022 | Neuroscience Bulletin2022,38,6: | 0
Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector. Some of JUNO's features make it an excellent location for^8B solar neutrino measurements, such as its low-energy threshold, high energy resolution compared with water Cherenkov detectors, and much larger target mass compared with previous liquid scintillator detectors. In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable, assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition, approximately 60,000 signal and 30,000 background events are expected. This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter, which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle. Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better, depending on the central value, and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND experiment. | 10 | Thermal management for the underwater frontend and readout electronics of JUNO 20‑inch PMTs显示文摘Background The Jiangmen underground neutrino observatory(JUNO)is a new generation of long-term operation neutrino experimental platform under construction in Jiangmen,Guangdong province of southern China. The underwater frontend and readout electronics(F&R electronics)are placed nearby the 20-inch PMTs and readout current signals from three PMTs.The electronics are sealed in a stainless-steel underwater box to keep them dry,and temperature control of the electronics plays a significant role in their reliability. Methods A conductive cooling structure based on the passive cooling method is designed for the 20-inch PMT F&R electronics. Both numerical simulation and experimental tests were performed to evaluate the cooling structures'thermal performance. Results The results show that the case temperature of the respective chip for 20-inch PMT F&R electronics is less than 30℃in ambient still water. Conclusion The case temperature of the respective chip meets the strict temperature requirement for the F&R electronics. The JUNO collaboration has adopted this cooling structure for the 20-inch PMT readout electronics. | Yangfu Wang Xiaoshan Jiang Jun Hu Peiliang Wang Lei Fan Shaojing Hou Zhe Ning Yunhua Sun Xiongbo Yan Zheng Wang Ziyue Yan Peiran Ye Jie Zhang Alberto Garfagnini Antonio Bergnoli Riccardo Brugnera Beatrice Jelmini Filippo Marini Marco Grassi Mariia Redchuk Andrea Serafini Katharina von Sturm | 2023 | Radiation Detection Technology and Methods2023,7,3: | 0
Retrieval-and-alignment based large-scale indoor point cloud semantic segmentation显示文摘Current methods for point cloud semantic segmentation depend on the extraction of descriptive features. However, unlike images, point clouds are irregular and often lack texture information, making it demanding to extract discriminative features. In addition, noise, outliers, and uneven point distribution are commonly present in point clouds, which further complicates the segmentation task.To address these problems,a novel architecture is proposed for direct and accurate large-scale point cloud segmentation based on point cloud retrieval and alignment. The proposed approach involves using a feature-based point cloud retrieval method for searching for reference point clouds with annotations from a dataset. In the following segmentation stage,an overlap-based point cloud registration method has been developed to align the target and reference point clouds. For accurate and robust alignment,an overlap region estimation module is trained to locate the optimal overlap region between two pieces of point clouds in a coarse-to-fine manner. In the detected overlap region, the global and local features of the points are extracted and combined for featuremetric registration to obtain accurate transformation parameters between the target and reference point clouds. After alignment, the annotated segmentation of the reference is transferred to the target point clouds to obtain accurate segmentation results.Extensive experiments are conducted to show that the developed method outperforms the state-of-the-art approaches in terms of both accuracy and robustness against noise and outliers. | Zongyi XU Xiaoshui HUANG Bo YUAN Yangfu WANG Qianni ZHANG Weisheng LI Xinbo GAO | 2024 | Science China(Information Sciences)2024,67,4: | 0
