1Effects of dietary yeast nucleotides on growth, non-specific immunity intestine growth and intestinal microbiota of juvenile hybrid tilapia Oreochromis niloticus ♀ × Oreochromis aureus ♂显示文摘This study investigated the effect of dietary supplementation of yeast nucleotides on the growth, nonspecific immunity, intestine growth and intestinal microbiota of juvenile hybrid tilapia. Tilapia(initial average weight of 8.02 g) was fed test diets supplemented with a yeast-originated nucleotide mixture(0,0.15, 0.30, 0.60, and 1.20 g/100 g diet) for 8 weeks. Fish fed the diet with 0.60% nucleotide had significantly higher weight gain than the control group(P < 0.05). Feed efficiency was improved in the fish fed 0.60 and 1.20% nucleotide compared with that in the control group. The optimal doses of nucleotides supplementation for growth and feed efficiency of fish were determined as 0.63 and 0.81%, respectively.Intestinal growth was improved in the 0.30 and 0.60% groups, as indicated by significant increase in intestine length. The fish fed 0.60 and 1.20% nucleotide showed higher super oxide dismutase(SOD)activity and lower malondialdehyde(MDA) level in the liver than the control fish, indicating enhancement of the anti-oxidant status. Serum lysozyme activity was significantly increased in the 0.15 and 0.3%nucleotide supplementation groups, suggesting an enhancement effect on the non-specific immune response. Lastly, dietary nucleotides supplementation exerted moderate influence on the intestinal microbiota of hybrid tilapia. A reduction in the cumulative abundance of putative butyrate-producing species was observed in the intestinal microbiota of fish fed diets with 0.60% nucleotide compared with the control, implying an interaction between dietary nucleotides and butyrate production. 2Double-composite rectangular truss bridge and its joint analysis显示文摘This paper describes a novel composite tubular truss bridge with concrete slab and concrete-filled rectangular chords. With concrete slab plus truss system and joints reinforced with concrete and Perfobond Leiste rib, double composite truss bridge proved to be a fairly suitable solution in negative moment area. Perfobond Leiste shear connector(PBL) is widely implemented in the composite structure for its outstanding fatigue resistance. In this pilot bridge, Perfobond Leister ribs(PBR) were installed in the truss girder's joints,which played double roles as shear connector and stiffener. An erection method and overall bridge structural analysis were then presented. Typical joints in the pilot bridge were selected to analyze the effect of PBR. Investigation of the effect of PBR in concretefilled tubular joints was elaborated. Comparison has revealed that concrete-filled tubular joints with PBR have much higher constraint capability than joints without PBR. For rectangular tubular truss, the punching shear force of the concrete filled joint with PBR is approximately 43% larger than that of the joint without PBR. Fatigue performance of the joint installed with PBR was improved, which was found through analysis of the stress concentration factor of joint. The PBR installed in the joints mitigated the stress concentration factor in the chord face. 3Expression of Aspergillus niger 9891 Endoinulinase in Pichia pastoris显示文摘An endoinulinase gene from Aspergillus niger 9891 (CGMCC0991) has been expressed in Pichia pastoris GS115 using pPIC9 vector. The recombinant endoinulinase was highly expressed and the optimization of the expression in a 7 liter of fermentor has been investigated. In fermented broth, the concentration of protein secreted is 2.15 mg/ml. 4Bifidobacteria DNA Induces Murine Macrophages Activation in vitro显示文摘Previous studies have shown that oligodeoxynucleotides containing unmethylated CpG motifs were used as adjuvants for immunoregulation and immune response. This study was to explore the activation effects of Bifidobacteria DNA containing unmethylated CpG motifs (CpG DNA) on murine macrophage J774A.1 cells. The genomic DNA of Bifidobacteria was extracted and purified, and the methylation degree of CpG motifs was tested.The phagocytic ability of the macrophages was detected by flow cytometry. The cytokines (IL-1β, IL-6, IL-12p40 and TNF-α) levels in the culture supernatants of Bifidobacteria DNA treated J774A.1 cells were assayed by ELISA. The content of nitric oxide (NO) was detected by Griess reagent. After treated with Bifidobacteria DNA for 24h,Nile Red stain increased in J774A.1 macrophage, which suggested that the lipid metabolism increased in the macrophages. The phagocytic ability and levels of NO and cytokines of IL-1β, IL-6, IL-12p40 and TNF-α were significantly higher than PBS group and CT DNA group. 9Antidepressants for bipolar disorder A meta-analysis of randomized, double-blind, controlled trials显示文摘OBJECTIVE:To examine the efficacy and safety of short-term and long-term use of antidepressants in the treatment of bipolar disorder.DATA SOURCES:A literature search of randomized,double-blind,controlled trials published until December 2012 was performed using the PubMed,ISI Web of Science,Medline and Cochrane Central Register of Controlled Trials databases.The keywords'bipolar disorder,bipolar I disorder,bipolar II disorder,bipolar mania,bipolar depression,cyclothymia,mixed mania and depression,rapid cycling and bipolar disorder',AND'antidepressant agent,antidepressive agents secondgeneration,antidepressive agents tricyclic,monoamine oxidase inhibitor,noradrenaline uptake inhibitor,serotonin uptake inhibitor,and tricyclic antidepressant agent'were used.The studies that were listed in the reference list of the published papers but were not retrieved in the above-mentioned databases were supplemented.STUDY SELECTION:Studies selected were double-blind randomized controlled trials assessing the efficacy and safety of antidepressants in patients with bipolar disorder.All participants were aged 18 years or older,and were diagnosed as having primary bipolar disorder.Antidepressants or antidepressants combined with mood stabilizers were used in experimental interventions.Placebos,mood stabilizers,antipsychotics and other antide pressants were used in the control interventions.Studies that were quasi-randomized studies,or used antidepressants in combination with antipsychotics in the experimental group were excluded.All analyses were conducted using Review Manager 5.1 provided by the Cochrane Collaboration.MAIN OUTCOME MEASURES:The primary outcome was the response and switching to mania.The secondary outcomes included remission,discontinuation rate,and suicidality.RESULTS:Among 5 001 treatment studies published,14 double-blind randomized controlled trials involving 1 244 patients were included in the meta-analysis.Eleven short-term studies and three maintenance studies were included.Studies suggested that patients treated with antidepressants were not significantly more likely to achieve higher response and remission rates in the short-term or long-term treatment than patients treated with placebo and other medications.Antidepressants were not associated with an increased risk of discontinuation,relapse or suicidality.When one antidepressant was compared with another,no significant difference in efficacy and tolerability was found.CONCLUSION:Existing evidence of efficacy does not support the short-term or long-term application of antidepressant therapy in patients with bipolar disorder,although the tolerability and safety of antidepressants have been generally acknowledged.There is a need for large-sample,double-blind,randomized controlled trials to elucidate the role of antidepressants in patients with different subcategories of bipolar disorder.Yingli Zhang Huan Yang Shichang Yang Wei Liang Ping Dai Changhong Wang Yalin Zhang 2013Neural Regeneration Research2013,8,31:3

11The direct and gut microbiota-mediated effects of dietary bile acids on the improvement of gut barriers in largemouth bass(Micropterus salmoides)显示文摘Fish gut barrier damage under intensive culture model is a significant concern for aquaculture industry.This study aimed to investigate the effects of bile acids(BAs)on gut barriers in Micropterus salmoides.A germ-free(GF)zebrafish model was employed to elucidate the effects of the direct stimulation of BAs and the indirect regulations mediated by the gut microbiota on gut barrier functions.Four diets were formulated with BAs supplemented at 0,150,300 and 450 mg/kg,and these 4 diets were defined as control,BA150,BA300 and BA450,respectively.After 5 weeks of feeding experiment,the survival rate of fish fed with BA300 diet was increased(P<0.05).Histological analysis revealed an improvement of gut structural integrity in the BA150 and BA300 groups.Compared with the control group,the expression of genes related to chemical barrier(mucin,lysozyme and complement 1)and physical barrier(occludin and claudin-4)was increased in the BA150 and BA300 groups(P<0.05),and the expression of genes related to immunological barrier(interleukin[IL]-6,tumor growth factorβ,IL-10,macrophage galactosetype lectin and immunoglobulin M[Ig M])was significantly increased in the BA300 group(P<0.05),but the expression of genes related to chemical barrier(hepcidin)and immunological barrier(IL-1β,tumor necrosis factor-a,IL-6 and arginase)was significantly decreased in the BA450 group(P<0.05).Gut microbiota composition analysis revealed that the abundance of Firmicutes was augmented prominently in the BA150 and BA300 groups(P<0.05),while that of Actinobacteriota and Proteobacteria showed a downward trend in the BA150 and BA300 groups(P>0.05).The results of the gut microbiota transferring experiment demonstrated an upregulation of gut barrier-related genes,including immunoglobulin Z/T(Ig Z/T),IL-6,IL-1βand IL-10,by the gut microbiota transferred from the BA300 group compared with the control(P<0.05).Feeding the BA300 diet directly to GF zebrafish resulted in enhanced expression of Ig M,Ig Z/T,lysozyme,occludin-2,IL-6 and IL-10(P<0.05).In conclusion,BAs can improve the gut barriers of fish through both direct and indirect effects mediated by the gut microbiota.Rui Xia Qingshuang Zhang Dongmei Xia Qiang Hao Qianwen Ding Chao Ran Yalin Yang Aizhi Cao Zhen Zhang Zhigang Zhou 2023Animal Nutrition2023,,3:2

16High prevalence of pathogenic Leptospira in wild and domesticated animals in an endemic area of China显示文摘Objective:To assess the prevalence of Leptospira detected in wildlife and domesticated animals in Jiangxi Province,China,in 2009.Methods:Urine samples from 28 buffaloes and kidney samples from 50 pigs,50 dogs and 38 rats were collected from Fuliang and Shangrao County,Jiangxi Province,China,in October 2009.Polymerase chain reaction(PCR) and culture analyses were used to detect Leptospira.The cultured isolates were typed using the microscopic agglutination lest(MAT).Results:The results showed that rats potentially serve as the main reservoir of leptospiral infection,followed by dogs.Although 16%of rats(6/38) were positive using culture analysis,PCR analysis using the diagnostic primers G1/G2 and B64Ⅰ/B64Ⅱor lipLS2 showed identification as 50%and 24%,respectively,of the rat samples as positive for the presence of leptospiral DNA.Conclusions:PCR-based detection of leptospiral DNA in infected kidney tissues of reservoirs is more efficient when using G1/G2 primers than lipL32 primers. 18Internal Loads and Bioavailability of Phosphorus and Nitrogen in Dianchi Lake,China显示文摘Sediments have a significant influence on the cycling of nutrient elements in lake environments. In order to assess the distribution characteristics and estimate the bioavailability of phosphorus and nitrogen in Dianchi Lake, organic and inorganic phosphorus and nitrogen forms were analysed. The 210 Pb radiometric dating method was employed to study temporal changes in the phosphorus and nitrogen pools in Dianchi Lake. The result show that the total phosphorus(TP) and total nitrogen(TN) were both at high concentrations, ranging from 697.5–3210.0 mg/kg and 1263.7–7155.2 mg/kg, respectively. Inorganic phosphorus(IP) and total organic nitrogen(TON) were the main constituents, at percentages of 59%–78% and 74%–95%, respectively, in the sediments. Spatially, there was a decreasing trend in phosphorus and nitrogen contents from the south and north to the lake centre, which is related to the distribution pattern of local economic production. The burial rates of the various phosphorus and nitrogen forms increased in same spatially and over time. Particularly in the past two decades, the burial rates doubled, with that TN reached to 1.287 mg/(cm^2·yr) in 2014. As the most reactive forms, nitrate nitrogen(NO_3-N) and ammonia nitrogen(NH_4-N) were buried more rapidly in the south region, implying that the potential for releasing sedimentary nitrogen increased from north to south. Based on their concentrations and burial rates, the internal loads of phosphorus and nitrogen were analysed for the last century. A TP pool of 71597.6 t and a TN pool of 81191.7 t were estimated for Dianchi Lake.
