1Impact of meteorological conditions on a nine-day particulate matter pollution event observed in December 2013,Shanghai,China显示文摘A severe particulate matter pollution event occurred in Shanghai from 1 to 9 December 2013.The mean hourly mass concentrations of PM_(2.5) and PM_(10) were 211.9 and 249.0 μg/m^3,respectively.Reanalysis data,in situ,and remote-sensing measurements were used to examine the impacts of meteorological conditions on this event.It was found that the synoptic pattern of weak pressure,the reduced planetary boundary layer height,and the passage of two cold fronts were key factors causing the event.Four stages were identified during this event based on the evolution of its PM_(2.5) levels and weather conditions.The highest concentration of PM2.5(602 μg/m^3) was observed in stage 3.High PM2.5 concentrations were closely associated with a low local ventilation index,with an average of 505m^2/s,as well as with the influx of pollutants from upstream,transported by the cold fronts.Jianming Xu Fengxia Yan Ying Xie Fengyun Wang Jianbin Wu Qingyan Fu 2015Particuology2015,13,3:17

3An analytical biomarker for treatment of patients with recurrent B-ALL after remission induced by infusion of anti-CD19 chimeric antigen receptor T(CAR-T) cells显示文摘Anti-CD19 chimeric antigen receptor-modified T(CAR-T-19) cells have emerged as a powerful targeted immunotherapy for B-cell lineage acute lymphoblastic leukemia with a remarkable clinical response in recent trials. Nonetheless, few data are available on the subsequent clinical monitoring and treatment of the patients, especially those with disease recurrence after CAR-T-19 cell infusion. Here, we analyzed three patients who survived after our phase I clinical trial and who were studied by means of biomarkers reflecting persistence of CAR-T-19 cells in vivo and predictive factors directing further treatment. One patient achieved 9-week sustained complete remission and subsequently received an allogeneic hematopoietic stem cell transplant. Another patient who showed relapse after 20 weeks without detectable leukemia in the cerebrospinal fluid after CAR-T-19 cell treatment was able to achieve a morphological remission under the influence of stand-alone low-dose chemotherapeutic agents. The third patient gradually developed extensive extramedullary involvement in tissues with scarce immune-cell infiltration during a long period of hematopoietic remission after CAR-T-19 cell therapy. Long-term and discontinuous increases in serum cytokines(mainly interleukin 6 and C-reactive protein) were identified in two patients(Nos. 1 and 6) even though only a low copy number of CAR molecules could be detected in their peripheral blood. This finding was suggestive of persistent functional activity of CAR-T-19 cells. 4Astragalus injection inhibits c-Jun N terminal kinase mRNA expression following oxygen-glucose deprivation and reintroduction in rat hippocampal neurons显示文摘BACKGROUND:In studies concerning cell injury induced by cerebral ischemia-reperfusion,current experiments have primarily focused on altered protein levels.In addition,the apoptotic proteins Bax and Bcl-2 have been thoroughly studied with regard to initiating neuronal apoptosis. OBJECTIVE:To establish an in vitro model of oxygen-glucose deprivation and reintroduction in the rat hippocampus to simulate cerebral ischemia-reperfusion injury;to observe c-Jun N-terminal kinase 3(JNK3) mRNA expression in hippocampal neurons following Astragalus injection;and thus to determine changes in the signaling and downstream pathways of neuronal apoptosis at the cellular and molecular level. DESIGN,TIME AND SETTING:A randomized,controlled,cellular and molecular experiment was performed at the Department of Central Laboratory,Chengde Medical College from February to June 2008. MATERIALS:Astragalus injection,the main ingredient of astragaloside,was purchased from Chengdu Di'ao Jiuhong Pharmaceutical Manufactory,China.JNK3 mRNA probe and in situ hybridization kit were purchased from Tianjin Haoyang Biological Technology,China,and JNK3 RT-PCR primers were designed by Shanghai Bio-engineering,China. METHODS:Primary cultures of hippocampal neurons derived from Sprague Dawley rats,aged 1-2 days,were established.After 8 days,the hippocampal neurons were assigned to the following interventions:model group,Astragalus group,and vehicle control group,cells were subjected to oxygen-glucose reintroduction after oxygen-glucose deprivation for 30 minutes in sugar-free Earle's solution and a hypoxia device,which contained high-purity nitrogen.The normal control group was subjected to primary culture techniques and was not treated using above-mentioned interventions. In addition,the Astragalus and vehicle control groups were treated with Astragalus injection(0.5 g/L raw drug) or sterile,deionized water at 2 hours prior to oxygen-glucose deprivation,respectively. MAIN OUTCOME MEASURES:JNK3 mRNA expression was measured by in situ hybridization and RT-PCR at 0,0.5,2,6,24,72,and 120 hours after oxygen-glucose reintroduction. RESULTS:Hippocampal neuronal morphology was normal in the normal control group. Hippocampal neurons exhibited apparent apoptosis-like pathological changes in the model,as well as the vehicle control,groups.The apoptosis-like pathological changes in the hippocampal neurons were less in the Astragalus group.Results from in situ hybridization and RT-PCR showed that JNK3 mRNA expression significantly increased in hippocampal neurons from model group,as well as the vehicle control group,compared with the normal control group(P<0.05).In addition,JNK3 mRNA expression significantly decreased in hippocampal neurons of the Astragalus group,compared with the model group and vehicle control group(P<0.05). 6Biodegradation of organochlorine pesticide endosulfan by bacterial strain Alcaligenes faecalis JBW4显示文摘The recently discovered endosulfan-degrading bacterial strain Alcaligenes faecalis JBW4 was isolated from activated sludge. This strain is able to use endosulfan as a carbon and energy source. The optimal conditions for the growth of strain JBW4 and for biodegradation by this strain were identified, and the metabolic products of endosulfan degradation were studied in detail. The maximum level of endosulfan biodegradation by strain JBW4 was obtained using broth at an initial pH of 7.0, an incubation temperature of 40°C and an endosulfan concentration of 100 mg/L. The concentration of endosulfan was determined by gas chromatography. Strain JBW4 was able to degrade 87.5% of α-endosulfan and 83.9% of β-endosulfan within 5 days. These degradation rates are much higher than the previously reported bacterial strains. Endosulfan diol and endosulfan lactone were the major metabolites detected by gas chromatography-mass spectrometry; endosulfan sulfate, which is a persistent and toxic metabolite, was not detected. These results suggested that A. faecalis JBW4 degrades endosulfan via a non-oxidative pathway. The biodegradation of endosulfan by A. faecalis is reported for the first time. 7A novel recombinant DNA vaccine encodingMycobacterium tuberculosis ESAT-6 and FL protects againstMycobacterium tuberculosis challenge in mice显示文摘Mycobacterium tuberculosis 6-kDa early secretory antigenic target(ESAT-6)is a dominant target antigen for cell-mediated immunity in the early phase of tuberculosis.The fms-like tyrosine kinase 3 ligand(FL)that induces potent immune response has been used as an adjuvant in vaccine development.In this study,a new recombinant plasmid(pIRES-epitope-peptides-FL)encoding three T cell epitopes of ESAT-6 and FL was constructed,and the immunogenicity of the DNA vaccine was assessed in C57BL/6 mice immunized with the plasmid DNA vaccine.Additionally,a strategy of intramuscular injection with the DNA vaccine(prime)and intranasal administration of the epitope peptides(boost)was employed to induce higher immune reaction of the mice.The results showed that mice vaccinated with the recombinant plasmid DNA vaccine and boosted with the peptides not only increased the levels of Th1 cytokines(IFN-γand IL-12),the number of IFN-γ+T cells and activities of cytotoxic T lymphocytes as well as IgG,but also enhanced protection against Mycobacterium tuberculosis challenge.In conclusion,these data indicate that the novel recombinant pIRES-epitope-peptides-FL plasmid is a useful DNA vaccine for preventing Mycobacterium tuberculosis infection.Qingtao Jiang Jing Zhang Xia Chen Mei Xia Yanlai Lu Wen Qiu Ganzhu Feng Dan Zhao Yan Li Fengxia He Guangyong Peng Yingwei Wang 2013The Journal of Biomedical Research2013,27,5:2

14Effect of intracranial hypertension on cerebral hemorrhage induced autonomic nerve imbalance显示文摘BACKGROUND: Cerebral hemorrhage can cause the imbalance of nerve function, whereas its mechanism and main impact factors are still not quite clear. OBJECTIVE: To explore the rules about the changes of intracranial pressure in brainstem hemorrhage and internal capsule hemorrhage, and analyze the role of intracranial hypertension in the changes of nerve function caused by cerebral hemorrhage. DESIGN: A self-controlled trial. SETTING: Department of Physiology, Tianjin Medical University. MATERIALS: Sixty-five healthy male Japanese white rabbits with long ears (1.5–1.8 kg) were supplied and fed by the Department of Animal Experiment of Tianjin Medical University. The RM6240B biological signal collecting and processing system was used. METHODS: The experiments were conducted in the Department of Physiology, Tianjin Medical University from August 2001 to May 2006. ① The rabbits were anesthetized, then fixed onto the brain stereotaxic apparatus, and afterwards fenestration on skull and intubation to lateral ventricle were performed.The dynamic changes of intracranial pressure were monitored continuously. Rabbits were infused with autologous arterial blood (0.3 mL) into midbrain corpora quadrigemina inferior colliculus to induce model of acute brainstem hemorrhage; models of internal capsule hemorrhage were established by infusing autologous arterial blood into internal capsule. ② The dynamic intracranial pressures under the above conditions were recorded continuously with the RM6240B biological signal collecting and processing system. ③ An animal model of persistent intracranial hypertension was established by infusion of physiologic saline into lateral ventricle. ④ The changes of the intensity of autonomic nerve discharge were analyzed, using the biological signal collecting and processing system before and after hemorrhage and under persistent intracranial hypertension. ⑤ Ten animal models of internal capsule hemorrhage and 10 of brainstem hemorrhage were selected respectively, then gross pathological samples were cut open, and the accuracy of hemorrhage models was affirmed. Histological sections in hemorrhage point and around this point were prepared for with hematoxylin and eosin staining, and the pathological changes were observed under light microscope. MAIN OUTCOME MEASURES: ① Changes of intracranial pressures before and after internal capsule hemorrhage and brainstem hemorrhage; ② Changes of the discharge intensity of cervical vagus nerve trunk in animal models of internal capsule hemorrhage, brainstem hemorrhage and persistent intracranial hypertension without hemorrhage; ③ Accuracy of location of internal capsule hemorrhage and brainstem hemorrhage confirmed by gross pathological samples and sections. RESULTS: Totally 65 rabbits were involved in the analysis of results. ① Dynamic state of intracranial pressure: Intracranial pressure increased obviously at 45 minutes after internal capsule hemorrhage and brainstem hemorrhage, the intracranial pressures were (1.31±0.30), (1.82±0.45) kPa, which were obviously higher than those before hemorrhage [(1.04±0.18), ( 1.05±0.19) kPa, P < 0.01]. ② Discharge of vagus nerve: Under intracranial hypertension, the discharge of cervical vagus nerve trunk was enhanced, and the discharge intensity of vagus nerve trunk was significantly different before and after persistent intracranial hypertension [(364.28±78.55), (1252.19±151.75) μV·s, P < 0.01]. The discharges of cervical vagus nerve trunk were significantly enhanced after internal capsule hemorrhage and brainstem hemorrhage (P < 0.01). ③ Validation of hemorrhage sites: The hemorrhage sites were internal capsule and brainstem on histopathological sections. 15Analysis of anticancer compound,indole-3-carbinol,in broccoli using a new ultrasound-assisted dispersive-filter extraction method based on poly(deep eutectic solvent)-graphene oxide nanocomposite显示文摘Indole-3-carbinol(I3C),an important anticancer compound found in broccoli,has attracted considerable attention.The rapid extraction and accurate analysis of I3C in the pharmaceutical industry in broccoli is challenging as I3C is unstable at low pH and high temperature.In this study,a rapid,accurate,and lowcost ultrasound-assisted dispersive-filter extraction(UADFE)technique based on poly(deep eutectic solvent)-graphene oxide(PDES-GO)adsorbent was developed for the isolation and analysis of I3C in broccoli for the first time.PDES-GO with multiple adsorption interactions and a fast mass transfer rate was synthesized to accelerate adsorption and desorption.UADFE was developed by combining dispersive solid-phase extraction(DSPE)and filter solid-phase extraction(FSPE)to realize rapid extraction and separation.Based on the above two strategies,the proposed PDES-GO-UADFE method coupled with high-performance liquid chromatography(HPLC)allowed the rapid(15-16 min),accurate(84.3%-96.4%),and low-cost(adsorbent:3.00 mg)analysis of I3C in broccoli and was superior to solid-phase extraction,DSPE,and FSPE methods.The proposed method showed remarkable linearity(r=0.9998;range:0.0840-48.0 mg/g),low limit of quantification(0.0840 mg/g),and high precision(relative standard deviation<5.6%).Therefore,the PDES-GO-UADFE-HPLC method shows significant potential in the field of pharmaceutical analysis for the separation and analysis of anti-cancer compounds in complex plant samples.Yanan Yuan Huanhuan Chen Yehong Han Fengxia Qiao Hongyuan Yan 2022Journal of Pharmaceutical Analysis2022,12,2:0

18Dppa3 is critical for Lin28a-regulated ES cells na?ve–primed state conversion显示文摘Lin28a is a pluripotent factor that promotes somatic cell reprogramming. Unlike other pluripotent factors, Lin28a expression is transient and accumulated in primed embryonic stem (ES) cells, but its exact function and mechanism in the conversion of ES cells from naive to primed state remain unclear. Here, we present evidence for Dppa3, a protein originally known for its role in germ cell development, as a downstream target of Lin28a in naive–primed conversion. Using rescue experiment, we demonstrate that Dppa3 functions predominantly downstream of Lin28a during naive–primed state conversion. Higher level of Lin28a prevents let-7 maturation and results in Dnmt3a/b (target of let-7) upregulation, which in turn induces hypermethylation of the Dppa3 promoter. Dppa3 demarcates naive versus primed pluripotency states. 20Study on Air-Purifying Performance of Semi-Flexible Asphalt Samples Coated with Titanium Dioxide Using Different Methods显示文摘Titanium dioxide(TiO_(2))was recently employed to apply onto road surfaces to degrade the harmful compounds from vehicle emissions.However,it remains a challenging task to find a highly compatible pavement type for TiO_(2)application to achieve durable and efficient air-purifying performance.This study proposed to coat TiO_(2)particles onto semi-flexible pavement surface and tried to investigate an optimum coating method.Three coating methods,including direct mixing TiO_(2)(MT)with asphalt mixture,spraying dry TiO_(2)(ST)coating and watersolution-based TiO_(2)(WT)coating on semi-flexible pavement surface.To achieve this objective,semi-flexible samples were prepared to evaluate and compare the performances of three coating methods by employing resistance to wearing,NO removal efficiency tests and residual texture depth tests.It was found that the ST method not only provided better NO degrading efficiency but also improved the resistance to wearing than the other two methods.Shoujing Yan Yangyang Wang Fengxia Chi Xue Luo Tianjie Zhang Chenchen Xi 2022Journal of Renewable Materials2022,10,5:0
