| 2 | B cell-derived anti-beta 2 glycoprotein I antibody mediates hyperhomocysteinemia-aggravated hypertensive glomerular lesions by triggering ferroptosis显示文摘Hyperhomocysteinemia(HHcy)is a risk factor for chronic kidney diseases(CKDs)that affects about 85%CKD patients.HHcy stimulates B cells to secrete pathological antibodies,although it is unknown whether this pathway mediates kidney injury.In HHcytreated 2-kidney,1-clip(2K1C)hypertensive murine model,HHcy-activated B cells secreted anti-beta 2 glycoprotein I(β2GPI)antibodies that deposited in glomerular endothelial cells(GECs),exacerbating glomerulosclerosis and reducing renal function.Mechanistically,HHcy 2K1C mice increased phosphatidylethanolamine(PE)(18:0/20:4,18:0/22:6,16:0/20:4)in kidney tissue,as determined by lipidomics.GECs oxidative lipidomics validated the increase of oxidized phospholipids upon Hcy-activated B cells culture medium(Hcy-B CM)treatment,including PE(18:0/20:4+3[O],PE(18:0a/22:4+1[O],PE(18:0/22:4+2[O]and PE(18:0/22:4+3[O]).PE synthases ethanolamine kinase 2(etnk2)and ethanolamine-phosphate cytidylyltransferase 2(pcyt2)were increased in the kidney GECs of HHcy 2K1C mice and facilitated polyunsaturated PE synthesis to act as lipid peroxidation substrates.In HHcy 2K1C mice and Hcy-B CM-treated GECs,the oxidative environment induced by iron accumulation and the insufficient clearance of lipid peroxides caused by transferrin receptor(TFR)elevation and down-regulation of SLC7A11/glutathione peroxidase 4(GPX4)contributed to GECs ferroptosis of the kidneys.In vivo,pharmacological depletion of B cells or inhibition of ferroptosis mitigated the HHcy-aggravated hypertensive renal injury.Consequently,our findings uncovered a novel mechanism by which B cell-derived pathogenic anti-β2GPI IgG generated by HHcy exacerbated hypertensive kidney damage by inducing GECs ferroptosis.Targeting B cells or ferroptosis may be viable therapeutic strategies for ameliorating lipid peroxidative renal injury in HHcy patients with hypertensive nephropathy. | Xing Du Xiaolong Ma Ying Tan Fangyu Shao Chun Li Yang Zhao Yutong Miao Lulu Han Guohui Dang Yuwei Song Dongmin Yang Zhenling Deng Yue Wang Changtao Jiang Wei Kong Juan Feng Xian Wang | 2023 | Signal Transduction and Targeted Therapy2023,8,4: | 2 |

| 4 | High-power,electronically controlled source of user-defined vortex and vector light beams based on a few-mode fiber amplifier显示文摘Optical angular momentum(OAM)-based structured light beams provide an additional degree of freedom for practical applications ranging from optical communication to laser-based material processing. Many techniques exist for generating such beams within laser sources and these primarily rely upon the use of specially designed optical components that limit laser power scaling and ready tunability of the topological charge and polarization of the output OAM beams. Here we show that some of these limitations can be overcome by employing a computer controlled reflective phase-only spatial light modulator to adaptively tailor the input(and subsequent output) beam wavefront and polarization in a few-mode fiber amplifier. In this way modal-coupling-induced beam distortion within the fiber amplifier can be mitigated and we are able to generate at will any desired supported spatial mode guided in the fiber, including conventional linearly polarized(LP) modes, scalar OAM modes, and cylindrical vector modes, at average powers >10 W and with a peak power of >11 kW. | 5 | Study of hot electrons generated from intense laser-plasma interaction employing Image Plate显示文摘Image Plate (IP) is convenient to be used and very suitable for radiation detection because of its advantages such as wide dynamic range, high detective quantum efficiency, ultrahigh sensitivity and superior linearity. The function mechanism and characteristics of IP are introduced in this paper. IP was employed in the study of hot electrons generated from intense laser-plasma interaction. The angular distri- bution and energy spectrum of hot electrons were measured with IP in the experi- ments. | 7 | A metabolic intervention strategy to break evolutionary adaptability of tumor for reinforced immunotherapy显示文摘The typical hallmark of tumor evolution is metabolic dysregulation.In addition to secreting immunoregulatory metabolites,tumor cells and various immune cells display different metabolic pathways and plasticity.Harnessing the metabolic differences to reduce the tumor and immunosuppressive cells while enhancing the activity of positive immunoregulatory cells is a promising strategy.We develop a nanoplatform(CLCeMOF)based on cerium metal-organic framework(CeMOF)by lactate oxidase(LOX)modification and glutaminase inhibitor(CB839)loading.The cascade catalytic reactions induced by CLCeMOF generate reactive oxygen species“storm”to elicit immune responses.Meanwhile,LOX-mediated metabolite lactate exhaustion relieves the immunosuppressive tumor microenvironment,preparing the ground for intracellular regulation.Most noticeably,the immunometabolic checkpoint blockade therapy,as a result of glutamine antagonism,is exploited for overall cell mobilization.It is found that CLCeMOF inhibited glutamine metabolism-dependent cells(tumor cells,immunosuppressive cells,etc.),increased infiltration of dendritic cells,and especially reprogrammed CD8^(+)T lymphocytes with considerable metabolic flexibility toward a highly activated,long-lived,and memory-like phenotype.Such an idea intervenes both metabolite(lactate)and cellular metabolic pathway,which essentially alters overall cell fates toward the desired situation.Collectively,the metabolic intervention strategy is bound to break the evolutionary adaptability of tumors for reinforced immunotherapy. | Qianhua Feng Yutong Hao Shuaiqi Yang Xiaomin Yuan Jing Chen Yuying Mei Lanlan Liu Junbiao Chang Zhenzhong Zhang Lei Wang | 2023 | Acta Pharmaceutica Sinica B2023,13,2: | 1 |

| 12 | Molecular mechanism of lycopene cyclases regulating carotenoids ratio in different branches during tea leaf and flower development显示文摘Carotenoids are essential components in tea quality, contributing to leaf color and aroma. However, little information about carotenoids in different tea cultivars and their biosynthesis regulation mechanism during leaf development is known. Here we analyzed carotenoids by HPLC in the buds and leaves of 113 tea cultivars harvested on the same day. By profile clustering, carotenoids were divided into five groups. Same group cultivars displayed divergence in the total content of carotenoids but a similar molar ratio. To figure out the molecular mechanisms of this phenomenon, we further characterized all functional lycopene cyclases, which are the branch point of the carotenoid biosynthesis pathway. Two β-lycopene cyclases(CsLCYB1 and CsLCYB2) and one ε-lycopene cyclase(CsLCYE1) were cloned. Subcellular localization analysis showed that all cloned CsLCYs were localized in plastids. Enzyme activity assays in E. coli indicated both CsLCYBs catalyzed lycopene into β-carotene, and CsLCYE1 produced δ-carotene and ε-carotene. We found CsLCYB1 and CsLCYE1 predominantly expressed in leaf, while CsLCYB2was mainly expressed during flowering stages. Suppression by antisense oligonucleotides reduced CsLCYB1 and CsLCYE1 transcripts and led to reduction of both β,β-branch and β,ε-branch carotenoids in leaf. The expression levels of CsLCYB1 showed a significant positive correlation withβ,β-branch carotenoids in leaf. Our study provides carotenoid profiles of different tea cultivars, which can assist tea producers in selecting cultivars of interest. | 16 | GDF15 negatively regulates chemosensitivity via TGFBR2-AKT pathway-dependent metabolism in esophageal squamous cell carcinoma显示文摘Treating patients with esophageal squamous cell carcinoma(ESCC)is challenging due to the high chemoresistance.Growth differentiation factor 15(GDF15)is crucial in the development of various types of tumors and negatively related to the prognosis of ESCC patients according to our previous research.In this study,the link between GDF15 and chemotherapy resistance in ESCC was further explored.The relationship between GDF15 and the chemotherapy response was investigated through in vitro and in vivo studies.ESCC patients with high levels of GDF15 expression showed an inferior chemotherapeutic response.GDF15 improved the tolerance of ESCC cell lines to low-dose cisplatin by regulating AKT phosphorylation via TGFBR2.Through an in vivo study,we further validated that the anti-GDF15 antibody improved the tumor inhibition effect of cisplatin.Metabolomics showed that GDF15 could alter cellular metabolism and enhance the expression of UGT1A.AKT and TGFBR2 inhibition resulted in the reversal of the GDF15-induced expression of UGT1A,indicating that TGFBR2-AKT pathway-dependent metabolic pathways were involved in the resistance of ESCC cells to cisplatin.The present investigation suggests that a high level of GDF15 expression leads to ESCC chemoresistance and that GDF15 can be targeted during chemotherapy,resulting in beneficial therapeutic outcomes. | Yingxi Du Yarui Ma Qing Zhu Yong Fu Yutong Li Ying Zhang Mo Li Feiyue Feng Peng Yuan Xiaobing Wang | 2023 | Frontiers of Medicine2023,17,1: | 0 |
