| 3 | A New Indicator for Global Food Security Assessment: Harvested Area Rather Than Cropland Area显示文摘Cropland area has long been used as a key indicator of food security.However,grain yield is not solely controlled by the area of the cropland.Therefore,we proposed a new indicator to assess food security.Results show that from 1992 to 2004,the global cropland area increased by 840200 km^(2)(99.4%),but the grain yield increased only by 310 million t(29.1%);and from 2004 to 2015,the cropland area decreased by 39000 km^(2)(4.64%),but the grain yield increased by 370 million t(70.84%).This result showed that grain yield was not linearly correlated with cropland area,and delimiting the threshold of cropland protection may not guarantee food security.Combined with further correlation analysis,we found that the increase in the global grain yield was more closely related to the harvested area(R^(2)=0.94),which indicated that the harvested area is a more scientific and accurate indicator than cropland area in terms of guaranteeing food security.Therefore,if governments want to ensure the food security,they should choose a new and more accurate indicator:harvested area rather than cropland area. | SONG Fengjiao WANG Shijie BAI Xiaoyong WU Luhua WANG Jinfeng LI Chaojun CHEN Huan LUO Xuling XI Huipeng ZHANG Sirui LUO Guofeng YAN Mengqi ZHEN Qingqing | 2022 | Chinese Geographical Science2022,32,2: | 2 |

| 5 | Research and development of a novel subunit vaccine for the currently circulating pseudorabies virus variant in China显示文摘Pseudorabies(PR)is a devastating viral disease which leads to fatal encephalitis and respiratory disorders in pigs.Commercial gE-deleted live pseudorabies virus(PRV)vaccine has been widely used to control this disease in China.However,the new-emerging variants of PRV compromises the protection provided by current vaccines and lead to the outbreak of PR in vaccinated pig herds.Several killed and live vaccine candidates based on current PRV variants have been reported to be effective to control the disease.A subunit vaccine based on gB protein,one major PRV glycoprotein which elicits strong humoral and cellular immune responses,however,was never evaluated for protection against the current circulating PRV variants.In this study,full-length PRV gB protein was successfully expressed in baculovirus/insect cells in the soluble format and was tested on 3-week-old piglets as a subunit vaccine.Compared with unvaccinated pigs,the gB-vaccinated pigs developed specific antibody-mediated responses and were protected from the virulent PRV HN1201 challenge.All vaccinated pigs survived without showing any PRV-specific respiratory and neurological signs,but all unvaccinated pigs died within 7 days after HN1201 challenge.Hence,this novel gB-based vaccine could be applied as an effective subunit vaccine to control PRV variant in China. | Yuzhou WANG Tongyan WANG He YAN Fanli YANG Linghua GUO Qingyuan YANG Xule HU Feifei TAN Yan XIAO Xiangdong LI Kegong TIAN | 2015 | Frontiers of Agricultural Science and Engineering2015,2,3: | 2 |

| 7 | Sinoatrial node pacemaker cells share dominant biological properties with glutamatergic neurons显示文摘Activation of the heart normally begins in the sinoatrial node(SAN).Electrical impulses spontaneously released by SAN pacemaker cells(SANPCs)trigger the contraction of the heart.However,the cellular nature of SANPCs remains controversial.Here,we report that SANPCs exhibit glutamatergic neuron-like properties.By comparing the single-cell transcriptome of SANPCs with that of cells from primary visual cortex in mouse,we found that SANPCs co-clustered with cortical neurons.Tissue and cellular imaging confirmed that SANPCs contained key elements of glutamatergic neurotransmitter system,expressing genes encoding glutamate synthesis pathway(G/s),ionotropic and metabotropic glutamate receptors(Grina,Gria3,Grm1 and Grm5)t and glutamate transporters(Slc17a7).SANPCs highly expressed cell markers of glutamatergic neurons(Snap25 and S/-c17a7)t whereas Gad1,a marker of GABAergic neurons,was negative.Functional studies revealed that inhibition of glutamate receptors or transporters reduced spontaneous pacing frequency of isolated SAN tissues and spontaneous Ca2+transients frequency in single SANPC.Collectively,our work suggests that SANPCs share dominant biological properties with glutamatergic neurons,and the glutamatergic neurotransmitter system may act as an intrinsic regulation module of heart rhythm,which provides a potential intervention target for pacemaker cell-associated arrhythmias. | Dandan Liang Zhigang Xue Jinfeng Xue Duanyang Xie Ke Xiong Huixing Zhou Fulei Zhang Xuling Su Guanghua Wang Qicheng Zou Yi Liu Jian Yang Honghui Ma Luying Peng Chunyu Zeng Gang Li Li Wang Yi-Han Chen | 2021 | Protein & Cell2021,12,7: | 1 |

| 10 | simple and effective aerosol pathogen disinfection test for a flowing air disinfector显示文摘Aerosol transmission is an important disease transmission route and has been especially pertinent to hospital and biosafety laboratories during the SARS-CoV-2 pandemic.The thermal resistance of airborne SARS-CoV-2 is lower than that of Bacillus subtilis spores,which are often used to test the effectiveness of SARS-CoV-2 and other pathogen disinfection methods.Herein,we propose a new method to test the disinfection ability of a flowing air disinfector(a digital electromagnetic induction air heater)using B.subtilis spores.The study provides an alternative air disinfection test method.The new test system combined an aerosol generator and a respiratory filter designed in-house and could effectively recover spores on the filter membrane at the air outlet after passing through the flowing air disinfector.The total number of bacterial spores used in the test was within the range of 5×10^(5)–5×10^(6)colony-forming units(CFUs)specified in the technical standard for disinfection.The calculation was based on the calculation method in Air Disinfection Effect Appraisal Test in Technical Standard for Disinfection(2002 Edition).At an air speed of 3.5 m/s,we used a digital electromagnetic induction air heater to disinfect flowing air containing 4.100×10^(6)CFUs of B.subtilis spores and determined that the minimum disinfection temperature was 350℃for a killing rate of 99.99%.At 400℃,additional experiments using higher spore concentrations(4.700×10^(6)±1.871×10^(5)CFU)and a higher airspeed(4 m/s)showed that the killing rate remained>99.99%.B.subtilis spores,as a biological indicator for testing the efficiency of dry-heat sterilization,were killed by the high temperatures used in this system.The proposed method used to test the flowing air disinfector is simple,stable,and effective.This study provides a reference for the development of test systems that can assess the disinfection ability of flowing air disinfectors. | Xuling Liu Zhiran Qin Linqing Wang Xiaoting Xie Yifang Fu Jianhai Yu Zuxin Liang Xiaoen He Jingshu Li Hong Dai Jinxiu Yao Qinghua Wu Weiwei Xiao Li Zhu Chengsong Wan Bao Zhang Wei Zhao | 2023 | Journal of Biosafety and Biosecurity2023,5,1: | 0 |

| 13 | Long noncoding RNA lnc_217 regulates hepatic lipid metabolism by modulating lipogenesis and fatty acid oxidation显示文摘Nonalcoholic fatty liver disease (NAFLD) is considered a major health epidemic with an estimated 32.4% worldwide prevalence. No drugs have yet been approved and therapeutic nodes remain a major unmet need. Long noncoding RNAs are emerging as an important class of novel regulators influencing multiple biological processes and the pathogenesis of NAFLD. Herein, we described a novel long noncoding RNA, lnc_217, which was liver enriched and upregulated in high-fat diet-fed mice, and a genetic animal model of NAFLD. We found that liver specific knockdown of lnc_217 was resistant to high-fat diet-induced hepatic lipid accumulation and decreased serum lipid in mice. Mechanistically, we demonstrated that knockdown of lnc_217 not only decreased de novo lipogenesis by inhibiting sterol regulatory element binding protein-1c cleavage but also increased fatty acid β- oxidation through activation of peroxisome proliferator-activated receptor α and carnitine palmitoyltransferase-1α. It is necessary to decrease this unexpected immunological response.A novel 4-arm poly(ethylene glycol-5000)4-cholesteryl methyl amide(4-arm PEG5000-CHMA)has been synthesized. The structure of 4-arm PEG5000-CHMA was confirmed by IR and 1H-NMR spectrum. The pharmacokinetics of the tocopheryl nicotinate(TN)-loaded nanoemulsions modified with 4-arm PEG5000-CHMA or/and 1, 2-distearoyl-Sn-glycero-3-phosphoethanolamine-n-[methoxy(poly-ethyleneglycol)-2000](mPEG2000-DSPE) have been studied. Furthermore, the ABC phenomenon has been detailed investigated in rats by TN-loaded nanoemulsions modified with 4-arm PEG5000-CHMA and mPEG2000-DSPE(CPNE). The plasma levels of TN and anti-PEG IgM antibody were determined by HPLC and ELISA, respectively.The circulation time of the CPNEs were comparable to the mPEG2000-DSPE coated nanoemulsions. Moreover, the ABC phenomenon can be decreased by CPNEs. | 16 | AtMYBS1 negatively regulates heat tolerance by directly repressing the expression of MAX1 required for strigolactone biosynthesis in Arabidopsis显示文摘Heat stress caused by global warming requires the development of thermotolerant crops to sustain yield.It is necessary to understand the molecular mechanisms that underlie heat tolerance in plants.Strigolactones(SLs)are a class of carotenoid-derived phytohormones that regulate plant development and responses to abiotic or biotic stresses.Although SL biosynthesis and signaling processes are well established,genes that directly regulate SL biosynthesis have rarely been reported.Here,we report that the MYB-like transcription factor AtMYBS1/AtMYBL,whose gene expression is repressed by heat stress,functions as a negative regulator of heat tolerance by directly inhibiting SL biosynthesis in Arabidopsis.Overexpression of AtMYBS1 led to heat hypersensitivity,whereas atmybs1 mutants displayed increased heat tolerance.Expression of MAX1,a critical enzyme in SL biosynthesis,was induced by heat stress and downregulated in AtMYBS1-overexpression(OE)plants but upregulated in atmybs1 mutants.Overexpression of MAX1 in the AtMYBS1-OE background reversed the heat hypersensitivity of AtMYBS1-OE plants.Loss of MAX1 function in the atmyb1 background reversed the heat-tolerant phenotypes of atmyb1 mutants.Yeast one-hybrid assays,chromatin immunoprecipitation‒qPCR,and transgenic analyses demonstrated that AtMYBS1 directly represses MAX1 expression through the MYB binding site in the MAX1 promoter in vivo.The atmybs1d14 double mutant,like d14 mutants,exhibited hypersensitivity to heat stress,indicating the necessary role of SL signaling in AtMYBS1-regulated heat tolerance.Our findings provide new insights into the regulatory network of SL biosynthesis,facilitating the breeding of heat-tolerant crops to improve crop production in a warming world. | Xiang Li Jianhua Lu Xuling Zhu Yanqi Dong Yanli Liu Shanshan Chu Erhui Xiong Xu Zheng Yongqing Jiao | 2023 | Plant Communications2023,4,6: | 0 |
