5Polymer-chelation approach to high-performance Fe-Nx-C catalyst towards oxygen reduction reaction显示文摘Pyrolyzed Fe-Nx-C with atomically dispersed Fe-Nxsites are hailed as the most promising alternative to the noble metal Pt-based catalysts towards oxygen reduction reaction(ORR). However, the conventional micropore-confinement synthetic approach usually causes the insufficient utilization of active sites and mass transport resistance as the sites are located inside the micropore. We herein report a polymerchelation strategy to directly disperse the Fe-Nxactive sites onto the carbon surface. The N-rich monomer was in-situ polymerized on the carbon support and then chelated with Fe. The strong Fe-N chelating interaction is crucial to suppress Fe aggregation when undergoing the high-temperature pyrolysis. Due to the enriched surface sites, hierarchically porous structure and excellent conductivity of carbon support,the optimal catalyst(denoted as Fe-Nx-C@C-900) exhibits impressive ORR activity of onset and half-wave potential of 1.02 and 0.87 V, respectively, superior to the Pt/C benchmark.Xue Wang Li Zhang Meiling Xiao Junjie Ge Wei Xing Changpeng Liu Jianbing Zhu 2023Chinese Chemical Letters2023,34,4:1
6High coercivity Nd-Ce-Fe-B nanostructured ribbons prepared from melt spinning technique显示文摘The Ce-substituted(Nd1-xCex)12.2 Fe81.6 B6.2(x=0.0, 0.2, 0.4, 0.6) nanocrystalline ribbons were prepared by annealing amorphous ribbons from melt spinning. It is found that all ribbons are in a multiphase state consisting of a-Fe phase, Nd(Ce)-rich phases and RE2 Fe14 B(RE = Nd, Ce) phases. However, the coercivity of all annealed ribbons can reach a considerably high value without doping any heavy rare earth or other coercivity enhanced elements. A strong intergranular exchange coupling appears in these nanocrystalline ribbons. The Nd12.2 Fe81.6 B6.2 ribbons with multiphase have a coercivity of about 11.3 k Oe, and the coercivity decreases slightly with increasing Ce content. A coercivity of 7.5 kOe can be obtained when60 at% of Nd is replaced by Ce(x = 0.6) due to the grain refinement and the strong intergranular exchange coupling. 10Human 8-cell embryos enable efficient induction of disease-preventive mutations without off-target effect by cytosine base editor显示文摘Approximately 140 million people worldwide are homozygous carriers of APOE4(ε4),a strong genetic risk factor for late onset familial and sporadic Alzheimer’s disease(AD),91%of whom will develop AD at earlier age than heterozygous carriers and noncarriers.Susceptibility to AD could be reduced by targeted editing of APOE4,but a technical basis for controlling the off-target effects of base editors is necessary to develop low-risk personalized gene therapies.Here,we first screened eight cytosine base editor variants at four injection stages(from 1-to 8-cell stage),and found that FNLS-YE1 variant in 8-cell embryos achieved the comparable base conversion rate(up to 100%)with the lowest bystander effects.In particular,80%of AD-susceptibleε4 allele copies were converted to the AD-neutralε3 allele in humanε4-carrying embryos.Stringent control measures combined with targeted deep sequencing,whole genome sequencing,and RNA sequencing showed no DNA or RNA off-target events in FNLS-YE1-treated human embryos or their derived stem cells.Furthermore,base editing with FNLS-YE1 showed no effects on embryo development to the blastocyst stage.Finally,we also demonstrated FNLS-YE1 could introduce known protective variants in human embryos to potentially reduce human susceptivity to systemic lupus erythematosus and familial hypercholesterolemia.Our study therefore suggests that base editing with FNLS-YE1 can efficiently and safely introduce known preventive variants in 8-cell human embryos,a potential approach for reducing human susceptibility to AD or other genetic diseases.Yinghui Wei Meiling Zhang Jing Hu Yingsi Zhou Mingxing Xue Jianhang Yin Yuanhua Liu Hu Feng Ling Zhou Zhifang Li Dongshuang Wang Zhiguo Zhang Yin Zhou Hongbin Liu Ning Yao Erwei Zuo Jiazhi Hu Yanzhi Du Wen Li Chunlong Xu Hui Yang 2023Protein & Cell2023,14,6:0
11Simultaneously achieving high energy density and responsivity in submicron BaTiO_(3) film capacitors integrated on Si显示文摘In the research field of energy storage dielectrics,the“responsivity”parameter,defined as the recyclable/recoverable energy density per unit electric field,has become critically important for a comprehensive evaluation of the energy storage capability of a dielectric.In this work,high recyclable energy density and responsivity,i.e.,W_(rec)=161.1 J·cm^(-3) and ξ=373.8 J·(kV·m^(2))^(-1),have been simultaneously achieved in a prototype perovskite dielectric,BaTiO_(3),which is integrated on Si at 500℃ in the form of a submicron thick film.This ferroelectric film features a multi-scale polar structure consisting of ferroelectric grains with different orientations and inner-grain ferroelastic domains.A LaNiO_(3) buffer layer is used to induce a{001}textured,columnar nanograin microstructure,while an elevated deposition temperature promotes lateral growth of the nanograins(in-plane diameter increases from~10-20 nm at lower temperatures to~30 nm).These preferably oriented and periodically regulated nanograins have resulted in a small remnant polarization and a delayed polarization saturation in the film’s P-E behavior,leading to a high recyclable energy density.Meanwhile,an improved polarizability/dielectric constant of the BaTiO_(3) film has produced a much larger maximum polarization than those deposited at lower temperatures at the same electric field,leading to a record-breaking responsivity for this simple perovskite.Jun Ouyang Yinxiu Xue Chuanqi Song Meiling Yuan Kun Wang Yuyao Zhao Hongbo Cheng Hanfei Zhu Chao Liu 2024Journal of Advanced Ceramics2024,13,2:0

13Does airway pressure release ventilation offer new hope for treating acute respiratory distress syndrome?显示文摘Mechanical ventilation(MV)is an essential life support method for patients with acute respiratory distress syn-drome(ARDS),which is one of the most common critical illnesses with high mortality in the intensive care unit(ICU).A lung-protective ventilation strategy based on low tidal volume(LTV)has been recommended since a few years;however,as this did not result in a significant decrease of ARDS-related mortality,a more optimal venti-lation mode was required.Airway pressure release ventilation(APRV)is an old method defined as a continuous positive airway pressure(CPAP)with a brief intermittent release phase based on the open lung concept;it also perfectly fits the ARDS treatment principle.Despite this,APRV has not been widely used in the past,rather only as a rescue measure for ARDS patients who are difficult to oxygenate.Over recent years,with an increased under-standing of the pathophysiology of ARDS,APRV has been reproposed to improve patient prognosis.Nevertheless,this mode is still not routinely used in ARDS patients given its vague definition and complexity.Consequently,in this paper,we summarize the studies that used APRV in ARDS,including adults,children,and animals,to illustrate the settings of parameters,effectiveness in the population,safety(especially in children),incidence,and mechanism of ventilator-induced lung injury(VILI)and effects on extrapulmonary organs.Finally,we found that APRV is likely associated with improvement in ARDS outcomes,and does not increase injury to the lungs and other organs,thereby indicating that personalized APRV settings may be the new hope for ARDS treatment.Jiangli Cheng Aijia Ma Meiling Dong Yongfang Zhou Bo Wang Yang Xue Peng Wang Jing Yang Yan Kang 2022Journal of Intensive Medicine2022,2,4:0
14Natural long-chain saturated fatty acids doped LNPs enabling spleen selective mRNA translation and potent cancer immunotherapy显示文摘Rationally tailored lipid nanoparticles(LNPs)with efficient and tunable delivery of mRNA in vivo are crucial for mRNA vaccines.Selective expression of antigenic protein in lymphoid tissues/organs could improve the immunostimulatory efficacy and safety of LNPs-based mRNA vaccines.Inspired by the metabolic behavior that long-chain saturated fatty acids tending to enter lymphoid tissue rather than the liver,we developed fatty acid-doped LNPs capable of mediating differential protein expressions in the liver and spleen when administered intravenously.When the molar ratio of saturated fatty acid located 60%–70%,the doped LNPs achieved the spleen selective mRNA translation.The mechanism could be attributed to the different cellular uptake behaviors of saturated fatty acids in hepatocytes.Immunization with a model antigen(ovalbumin)mRNA-loaded spleen selective LNPs,we observed enhanced antigen-specific T cell immune responses,and potent immunotherapeutic and immunoprophylactic efficacy in the mouse lymphoma model.Our natural long-chain saturated fatty acids metabolic characteristics-inspired design of LNPs for spleen-selective mRNA vaccines delivery will provide references for designing mRNA vaccines with high efficacy and safety for tumor immunotherapy.Fazhan Wang Meng Zhang Meiling Tian Jia Lou Longze Pan Xiaoke Gao Lijing Zhang Xiaohan Lou Linyu Zhu Yuqiao Sheng Ming Wang Rui Xue Wenjing Deng Shuai Shao Zhihai Qin 2024Nano Research2024,17,3:0
15Mechanism of interactions betweenα-conotoxin RegIIA and carbohydrates at the humanα3β4 nicotinic acetylcholine receptor显示文摘Conotoxins are marine peptide toxins from marine cone snails.Theα-conotoxin RegIIA can selectively act on human(h)α3β4 nicotinic acetylcholine receptor(nAChR),and is an important lead for drug development.The high-resolution cryo-electron microscopy structure of theα3β4 nAChR demonstrates several carbohydrates are located near the orthosteric binding sites,which may affectα-conotoxin binding.Oligosaccharide chains can modify the physical and chemical properties of proteins by changing the conformation,hydrophobicity,quality and size of the protein.The purpose of this study is to explore the effect of oligosaccharide chains on the binding modes and activities of RegIIA and its derivatives at hα3β4 nAChRs.Through computational simulations,we designed and synthesized RegIIA mutants at position 14 to explore the importance of residue H14 to the activity of the peptide.Molecular dynamics simulations suggest that the oligosaccharide chains affect the binding of RegIIA at the hα3β4 nAChR through direct interactions with H14 and by affecting the C-loop conformation of the binding sites.Electrophysiology studies on H14 analogues suggest that in addition to forming direct interactions with the carbohydrates,the residue might play an important role in maintaining the conformation of the peptide.Overall,this study further clarifies the structure–activity relationship ofα-conotoxin RegIIA at the hα3β4 nAChR and,also provides important experimental and theoretical basis for the development of new peptide drugs.Meiling Zheng Han-Shen Tae Liang Xue Tao Jiang Rilei Yu 2022Marine Life Science & Technology2022,4,1:0
16Sialoglycoproteins Isolated from the Eggs of Gadus morhua Inhibit Bone Resorption in Ovariectomized Rats by Suppressing the MAPK and NF-κB Pathways显示文摘The present study investigated the anti-osteoporosis function and the mechanism of sialoglycoproteins isolated from the eggs of Gadus morhua(Gm-SGP)on ovariectomized(OVX)rats.After 3 months of Gm-SGP treatment,OVX-induced bone loss was suppressed and uncoupling bone turnover was balanced,as indicated by systemic biomarkers of bone metabolism;no uterine estrogenicity was observed.Moreover,rats administered with Gm-SGP exhibited increased bone mineral density and biomechanical strength and significant restoration of the trabecular microarchitecture compared with rats in the control group.Gm-SGP significantly decreased bone resorption-related indicators in serum.Investigation of the associated mechanisms revealed that Gm-SGP significantly increases the OPG/RANKL ratio at the mRNA and protein levels.Further research suggested that Gm-SGP inhibits the mRNA and protein expressions of important transcription factors of the MAPK and NF-κB signaling pathways.It also attenuates the activation of related transduction signaling pathways by inhibiting phosphorylation of JNK,ERK,p38,and NF-κB,and ultimately suppresses the induction of c-Fos and NFATc1.Overall,these results demonstrate that Gm-SGP inhibits bone resorption by suppressing osteoclastogenesis-related MAPK and NF-κB pathways,thereby improving osteoporosis.MAO Lei WANG Yiming WANG Meiling DAI Yufeng WANG Jingfeng XUE Changhu 2019Journal of Ocean University of China2019,18,5:0
17Afterglow Carbon Dots:From Fundamentals to Applications显示文摘The ability of carbon dots(CDs)to emit afterglow emission in addition to fluorescence in response to UV-to-visible excitation allows them to be a new class of luminescent materials.When compared with traditional organic or inorganic afterglow materials,CDs have a set of advantages,including small size,ease of synthesis,and absence of highly toxic metal ions.In addition,high dependence of their afterglow color output on temperature,excitation wavelength,and aggregation degrees adds remarkable flexibility in the creation of multimode luminescence of CDs without the need for changing their intrinsic attributes.These characteristics make CDs particularly attractive in the fields of sensing,anticounterfeiting,and data encryption.In this review,we first describe the general attributes of afterglow CDs and their fundamental afterglow mechanism.We then highlight recent strategic advances in the generation or activation of the afterglow luminescence of CDs.Considerable emphasis is placed on the summarization of their emergent afterglow properties in response to external stimulation.We further highlight the emerging applications of afterglow CDs on the basis of their unique optical features and present the key challenges needed to be addressed before the realization of their full practical utility.Chenxi Peng Xue Chen Meiling Chen Shenci Lu Yu Wang Suli Wu Xiaowang Liu Wei Huang 2021Research2021,,1:0
