High quality reference genome of drumstick tree(Moringa oleifera Lam.), a potential perennial crop显示文摘The drumstick tree(Moringa oleifera Lam.) is a perennial crop that has gained popularity in certain developing countries for its high-nutrition content and adaptability to arid and semi-arid environments. Here we report a high-quality draft genome sequence of M. oleifera. This assembly represents 91.78% of the estimated genome size and contains 19,465 protein-coding genes. Comparative genomic analysis between M. oleifera and related woody plant genomes helps clarify the general evolution of this species, while the identification of several species-specific gene families and positively selected genes in M. oleifera may help identify genes related to M. oleifera's high protein content, fast-growth, heat and stress tolerance. This reference genome greatly extends the basic research on M. oleifera, and may further promote applying genomics to enhanced breeding and improvement of M. oleifera. | TIAN Yang ZENG Yan ZHANG Jing YANG Cheng Guang YAN Liang WANG XuanJun SHI ChongYing XIE Jing DAI TianYi PENG Lei ZENG HUAN Yu XU An Ni HUANG YeWei ZHANG JiaJin MA Xiao DONG Yang HAO ShuMei SHENG Jun | 2015 | Science China(Life Sciences)2015,58,7: | 10
Revisiting the role of delta-1-pyrroline-5-carboxylate synthetase in drought-tolerant crop breeding显示文摘The delta-1-pyrroline-5-carboxylate synthetase(P5CS)gene exercises a protective function in stressed plants. However, the relationship between proline accumulation caused by P5CS and abiotic stress tolerance in plants is not always clear,as P5CS overexpression has been reported to repress plant growth under normal conditions in several reports. We re-evaluated the role of P5CS in drought-tolerant rice breeding by expressing the AtP5CS1 and feedback-inhibition-removed AtP5CS1(AtP5CS1^(F128A))genes under the regulation of an ABA-inducible promoter to avoid the potential side effects of P5CS overexpression under normal conditions.ABA-inducible AtP5CS1 and AtP5CS1^(F128A) increased seedling growth in a nutrient solution(under osmotic stress)and grain yield in pot plants. However, the evidently deleterious effects of AtP5CS1 on grain quality, tiller number, and grain yield in the field indicated the unsuitability of P5CS for drought-tolerance breeding. | Xuanjun Feng Yue Hu Weixiao Zhang Rongqian Xie Huarui Guan Hao Xiong Li Jia Xuemei Zhang Hanmei Zhou Dan Zheng Ying Wen Qingjun Wang Fengkai Wu Jie Xu Yanli Lu | 2022 | The Crop Journal2022,10,4: | 2

Strategies to suppress the shuttle effect of redox mediators in lithium-oxygen batteries显示文摘Rechargeable lithium-oxygen(Li-O_(2))batteries are the next generation energy storage devices due to their ultrahigh theoretical capacity.Redox mediators(RMs)are widely used as a homogenous electrocatalyst in non-aqueous Li-O_(2)batteries to enhance their discharge capacity and reduce charge overpotential. However, the shuttle effect of RMs in the electrolyte solution usually leads to corrosion of the Li metal anode and uneven Li deposition on the anode surface, resulting in unwanted consumption of electrocatalysts and deterioration of the cells. It is therefore necessary to take some measures to prevent the shuttle effect of RMs and fully utilize the soluble electrocatalysts.Herein,we summarize the strategies to suppress the RM shuttle effect reported in recent years, including electrolyte additives, protective separators and electrode modification. The mechanisms of these strategies are analyzed and their corresponding requirements are discussed. The electrochemical properties of Li-O_(2)batteries with different strategies are summarized and compared. The challenges and perspectives on preventing the shuttle effect of RMs are described for future study. This review provides guidance for achieving shuttle-free redox mediation and for designing Li-O_(2)cells with a long cycle life, high energy efficiency and highly reversible electrochemical reactions. | Xinbin Wu Wei Yu Kaihua Wen Huanchun Wang Xuanjun Wang Ce-Wen Nan Liangliang Li | 2021 | Journal of Energy Chemistry2021,30,9: | 1

Alkaline-lacustrine deposition and paleoenvironmental evolution in Permian Fengcheng Formation at the Mahu sag, Junggar Basin,NW China显示文摘Alkaline-lacustrine deposition and its evolution model in Permian Fengcheng Formation at the Mahu sag, Junggar Basin were investigated through core and thin-section observation, geochemical and elemental analysis, logging response and lithofacies identification. Six lithofacies are developed in the Fengcheng Formation. The Feng 2 Member(P1 f2) is dominated by lithofacies with alkaline minerals, while the upper part of the Feng 1 Member(P1 f1) and the lower part of the Feng 3 Member(P1 f3) are primarily organic-rich mudstones that are interbedded with dolomite and dolomitic rock. Paleoenvironment evolution of Fengcheng Formation can be divided into 5 stages, which was controlled by volcanic activity and paleoclimate. The first stage(the early phase of P1 f1) was characterized by intensive volcanic activity and arid climate, developing pyroclastics and sedimentary volcaniclastic rocks. The secondary stage(the later phase of P1 f1) had weak volcanic activity and humid climate that contributed to the development of organic-rich mudstone, forming primary source rock in the Fengcheng Formation. The increasing arid climate at the third stage(the early phase of P1 f2) resulted in shrinking of lake basin and increasing of salinity, giving rise to dolomite and dolomitic rocks. The continuous aird climate, low lake level and high salinity at the fourth stage(the later phase of P1 f2) generated special alkaline minerals, e.g., trona, indicating the formation of alkaline-lacustrine. The humid climate made lake level rise and desalted lake water, therefore, the fifth stage(P1 f3) dominated by the deposition of terrigenous clastic rocks and dolomitic rocks. | ZHANG Zhijie YUAN Xuanjun WANG Mengshi ZHOU Chuanmin TANG Yong CHEN Xingyu LIN Minjie CHENG Dawei | 2018 | Petroleum Exploration and Development2018,45,6: | 0
Types, characteristics and geological significance of event deposits of Chang 9 Member of Triassic Yanchang Formation in southwestern Ordos Basin,NW China显示文摘Through core observation, thin section identification, and logging and testing data analysis, the types and characteristics of event deposits in the ninth member of Yanchang Formation of Triassic(Chang 9 Member)in southwestern Ordos Basin,China, are examined. There are 4 types and 9 subtypes of event deposits,i.e.earthquake,gravity flow, volcanic and anoxic deposits,in the Chang 9 Member in the study area.Based on the analysis of the characteristics and distribution of such events deposits,it is proposed that the event deposits are generally symbiotic or associated, with intrinsic genetic relations and distribution laws.Five kinds of sedimentary microfacies with relatively developed event deposits are identified, and the genetic model of event deposits is discussed.Seismites are mainly developed in the lake transgression stage when the basin expands episodically, and commonly affected by liquefaction flow, gravity action and brittle shear deformation.Gravity flow, mainly distributed in the high water level period, sandwiched in the fine-grained sediments of prodelta or semi-deep lake,or creates banded or lobate slump turbidite fan.It is relatively developed above the seismites strata. The volcanic event deposits are only seen in the lower part of the Chang 9 Member, showing abrupt contact at the top and bottom, which reflects the volcanic activity at the same time.Anoxic deposits are mostly formed in the late stage of lake transgression to the highstand stage.Very thick organic-rich shales are developed in the highstand stage of Chang 9 Member, and the event deposits in the depositional period of these shales are conducive to potential reservoirs. | WANG Ziye MAO Zhiguo YUAN Xuanjun DENG Xiuqin HUI Xiao ZHANG Zhongyi CUI Jingweil | 2023 | Petroleum Exploration and Development2023,50,3: | 0
Non-cytotoxic nanoparticles re-educating macrophages achieving both innate and adaptive immune responses for tumor therapy显示文摘Macrophages are important antigen-presenting cells to combat tumor via both innate and adaptive immunity, while they are programmed toM2 phenotype in established tumors and instead promote cancer development and metastasis. Here,we develop a nanomedicine that can re-educate M2 polarized macrophages to restore their anti-tumor activities. The nanomedicine has a core-shell structure to co-load IPI549,a PI3Kγinhibitor,and CpG,a Toll-like receptor 9 agonist.Specifically, the hydrophobic IPI549 is self-assembled into a pure drug nano-core,while MOF shell layer is coated for CpG encapsulation, achieving extra-high total drugs loading of 44%.Such nanosystem could facilitate intracellular delivery of the payloads but without any cytotoxicity, displaying excellent biocompatibility. After entering macrophages, the released IPI549 and CpG exert a synergistic effect to switch macrophages from M2 to M1 phenotype, which enables anti-tumor activities via directly engulfing tumor cells or excreting tumor killing cytokines. Moreover, tumor antigens released from the dying tumor cells could be effectively presented by the re-educated macrophages owing to the up-regulation of various antigen presenting mediators, resulting in infiltration and activation of cytotoxic T lymphocytes. As a result, the nanosystem triggers a robust antitumor immune response in combination with PD-L1 antibody to inhibit tumor growth and metastasis. This work provides a non-cytotoxic nanomedicine to modulate tumor immune microenvironment by reprograming macrophages. | Shengmei Wang Xuanjun Liu Minghua Yang Linqi Ouyang Jinsong Ding Shengfeng Wang Wenhu Zhou | 2022 | Asian Journal of Pharmaceutical Sciences2022,17,4: | 0

A platinum@polymer-catechol nanobraker enables radio-immunotherapy for crippling melanoma tumorigenesis, angiogenesis, and radioresistance显示文摘Malignant melanoma cell-intrinsic PD-1:PD-L1 interaction thrusts tumorigenesis, angiogenesis, and radioresistance via mTOR hyperactivation to aggravate circumjacent aggression.Interdicting melanoma intrinsic growth signals, including the blockade of PD-L1 and mTOR signaling concurrently, cooperative with radiotherapy may provide a vigorous repertoire to alleviate the tumor encumbrance.Thence,we design a three-pronged platinum@polymer-catechol nanobraker to deliver mTOR inhibitor TAK228 and anti-PD-L1 antibody(aPD-L1)for impeding the melanoma-PD-1-driven aggression and maximizing the melanoma eradication. The aPD-L1 collaborated with TAK228 restrains melanoma cell-intrinsic PD-1:PD-L1 tumorigenic interaction via blocking melanoma-PD-L1 ligand and the melanoma-PD-1 receptor-driven mTOR signaling;corresponding downregulation of mTOR downstream protumorigenic cellular MYC and proangiogenic hypoxia-inducible factor 1-alpha is conducive to preventing tumorigenesis and angiogenesis,respectively.Further,high-Z metal platinum sensitizing TAK228-enhanced radiotherapy confers the nanobraker on remarkable tumoricidal efficacy.Hereto, the customized three-pronged nanobrakers efficiently suppress melanoma tumorigenesis and angiogenesis concomitant with the amplification of radiotherapeutic efficacy.Such an ingenious tactic may provide substantial benefits to clinical melanoma patients. | Wenxi Li Jie Yan Hao Tian Bei Li Guohao Wang Wei Sang Zhan Zhang Xuanjun Zhang Yunlu Dai | 2023 | Bioactive Materials2023,,4: | 0
Investigation of hydroxyl-terminated polybutadiene propellant breaking characteristics and mechanism impacted by submerged cavitation water jet显示文摘A submerged cavitation water jet(SCWJ)is an effective method to recycle solid propellant from obsolete solid engines by the breaking method.Solid propellant's breaking modes and mechanical process under SCWJ impact are unclear. This study aims to understand those impact breaking mechanisms. The hydroxyl-terminated polybutadiene(HTPB)propellant was chosen as the research material, and a self-designed test system was used to conduct impact tests at four different working pressures. The high-speed camera characterized crack propagation, and the DIC method calculated strain change during the impact process.Besides, micro and macro fracture morphologies were characterized by scanning electron microscope(SEM)and computed tomography(CT)scanning. The results reveal that the compressive strain concentration region locates right below the nozzle, and the shear strain region distributes symmetrically with the jet axis, which increases to 4% at first 16th ms,the compressive strain rises to 2% and 6% in the axial and transverse direction, respectively. The two tensile cracks formed first at the compression strain concentrate region, and there generate many shear cracks around the tensile cracks, and those shear cracks that develop and aggregate cause the cracks to become wider and cut through the tensile cracks, forming the tensile-shear cracks and the impact parts eventually fail. The HTPB propellant forms a breaking hole shaped conical after impact 10 s.The mass loss increases by 17 times at maximum, with the working pressure increasing by three times.Meanwhile, the damage value of the breaking hole remaining on the surface increases by 7.8 times while 2.9 times in the depth of the breaking hole. The breaking efficiency is closely affected by working pressures. The failure modes of HTPB impacted by SCWJ are classified as tensile crack-dominated and tensile-shear crack-dominated damage mechanisms. | Wenjun Zhou Meng Zhao Bo Liu Youzhi Ma Youzhi Zhang Xuanjun Wang | 2024 | Defence Technology(防务技术)2024,33,3: | 0

A molecular sieve-containing protective separator to suppress the shuttle effect of redox mediators in lithium-oxygen batteries显示文摘Lithium-oxygen(Li-O_(2))batteries have a great potential in energy storage and conversion due to their ultra-high theoretical specific energy, but their applications are hindered by sluggish redox reaction kinetics in the charge/discharge processes.Redox mediators(RMs),as soluble catalysts, are widely used to facilitate the electrochemical processes in the Li-O_(2)batteries.A drawback of RMs is the shuttle effect due to their solubility and mobility, which leads to the corrosion of a Li metal anode and the degradation of the electrochemical performance of the batteries.Herein,we synthesize a polymer-based composite protective separator containing molecular sieves. The nanopores with a diameter of 4Åin the zeolite powder(4A zeolite)are able to physically block the migration of 2,2,6,6-tetramethylpiperidinyloxy(TEMPO)molecules with a larger size;therefore, the shuttle effect of TEMPO is restrained. With the assistance of the zeolite molecular sieves, the cycle life of the Li-O_(2)batteries is significantly extended from~20 to 170 cycles at a current density of 250 mA·g^(-1)and a limited capacity of 500 mAh·g^(-1).Our work provides a highly effective approach to suppress the shuttle effects of RMs and boost the electrochemical performance of Li-O_(2)batteries. | Xinbin Wu Huiping Wu Shundong Guan Ying Liang Kaihua Wen Huanchun Wang Xuanjun Wang Ce-Wen Nan Liangliang Li | 2023 | Nano Research2023,16,7: | 0
