| 1 | Signaling pathways and targeted therapy for myocardial infarction显示文摘Although the treatment of myocardial infarction(MI)has improved considerably,it is still a worldwide disease with high morbidity and high mortality.Whilst there is still a long way to go for discovering ideal treatments,therapeutic strategies committed to cardioprotection and cardiac repair following cardiac ischemia are emerging.Evidence of pathological characteristics in MI illustrates cell signaling pathways that participate in the survival,proliferation,apoptosis,autophagy of cardiomyocytes,endothelial cells,fibroblasts,monocytes,and stem cells.These signaling pathways include the key players in inflammation response,e.g.,NLRP3/caspase-1 and TLR4/MyD88/NF-κB;the crucial mediators in oxidative stress and apoptosis,for instance,Notch,Hippo/YAP,RhoA/ROCK,Nrf2/HO-1,and Sonic hedgehog;the controller of myocardial fibrosis such as TGF-β/SMADs and Wnt/β-catenin;and the main regulator of angiogenesis,PI3K/Akt,MAPK,JAK/STAT,Sonic hedgehog,etc.Since signaling pathways play an important role in administering the process of MI,aiming at targeting these aberrant signaling pathways and improving the pathological manifestations in MI is indispensable and promising.Hence,drug therapy,gene therapy,protein therapy,cell therapy,and exosome therapy have been emerging and are known as novel therapies.In this review,we summarize the therapeutic strategies for MI by regulating these associated pathways,which contribute to inhibiting cardiomyocytes death,attenuating inflammation,enhancing angiogenesis,etc.so as to repair and re-functionalize damaged hearts. | Qing Zhang Lu Wang Shiqi Wang Hongxin Cheng Lin Xu Gaiqin Pei Yang Wang Chenying Fu Yangfu Jiang Chengqi He Quan Wei | 2022 | Signal Transduction and Targeted Therapy2022,7,4: | 11 |
| 2 | Synergistic immunoreaction of acupuncture-like dissolving microneedles containing thymopentin at acupoints in immune-suppressed rats显示文摘Dissolving microneedles carried drug molecules can effectively penetrate the stratum corneum of skin to improve the transdermal drug delivery. The traditional Chinese medicine acupuncture is based on the needle stimulation at a specific location(acupoint) to generate and transmit biochemical and physiological signals which alter the pathophysiological state of patients. However, the pain associated with conventional acupuncture needles and the requirement of highly trained professionals limit the development of acupuncture in non-Asian countries. The purpose of this study is to investigate whether the dissolving microneedles can be utilized as a self-administered painless replacement for acupuncture and locally released drug molecules can achieve expected therapeutic outcomes. Immunosuppressive rats were treated with acupuncture at Zusanli(ST36) acupoint using microneedles containing thymopentin.The immune functions and psychological mood of the immunosuppressed animals were examined. The proliferation of splenocytes was examined by CCK-8 assay. CD4 and CD8 expression patterns in spleen cells were detected by flow cytometry. The current study showed that use of either microneedles containing thymopentin or conventional acupuncture both resulted in immune cell proliferation, which was confirmed by flow cytometry. | 3 | Influence of physical properties of carrier on the performance of dry powder inhalers显示文摘Dry powder inhalers(DPIs) offer distinct advantages as a means of pulmonary drug delivery and have attracted much attention in the field of pharmaceutical science. DPIs commonly contain micronized drug particles which, because of their cohesiveness and strong propensity to aggregate, have poor aerosolization performance. Thus carriers with a larger particle size are added to address this problem. However, the performance of DPIs is profoundly influenced by the physical properties of the carrier, particularly their particle size, morphology/shape and surface roughness. | 4 | Triple Medium Physical Model of Post Fracturing High-Rank Coal Reservoir in Southern Qinshui Basin显示文摘In this paper, influences on the reservoir permeability, the reservoir architecture and the fluid flow pattern caused by hydraulic fracturing are analyzed. Based on the structure and production fluid flow model of post fracturing high-rank coal reservoir, Warren-Root Model is improved. A new physical model that is more suitable for post fracturing high-rank coal reservoir is established. The results show that the width, the flow conductivity and the permeability of hydraulic fractures are much larger than natural fractures in coal bed reservoir. Hydraulic fracture changes the flow pattern of gas and flow channel to wellbore, thus should be treated as an independent medium. Warrant-Root Model has some limitations and can't give a comprehensive interpretation of seepage mechanism in post fracturing high-rank coal reservoir. Modified Warrant-Root Model simplifies coal bed reservoir to an ideal system with hydraulic fracture, orthogonal macroscopic fracture and cuboid matrix. Hydraulic fracture is double wing, vertical and symmetric to wellbore. Coal bed reservoir is divided into cuboids by hydraulic fracture and further by macroscopic fractures. Flow behaviors in coal bed reservoir are simplified to three step flows of gas and two step flows of water. The swap mode of methane between coal matrix and macroscopic fractures is pseudo steady fluid channeling. The flow behaviors of methane to wellbore no longer follow Darcy's Law and are mainly affected by inertia force. The flow pattern of water follows Darcy's Law. | 5 | Ripple-associated high-firing interneurons in the hippocampal CA1 region显示文摘By simultaneously recording the activity of individual neurons and field potentials in freely behaving mice, we found two types of interneurons firing at high frequency in the hippocampal CA1 region, which had high correlations with characteristic sharp wave-associated ripple oscillations (100―250 Hz) during slow-wave sleep. The firing of these two types of interneurons highly synchronized with ripple oscillations during slow-wave sleep, with strongly increased firing rates corresponding to individual ripple episodes. Interneuron type I had at most one spike in each sub-ripple cycle of ripple episodes and the peak firing rate was 310±33.17 Hz. Interneuron type II had one or two spikes in each sub-ripple cycle and the peak firing rate was 410±47.61 Hz. During active exploration, their firing was phase locked to theta oscillations with the highest probability at the trough of theta wave. Both two types of interneurons increased transiently their firing rates responding to the startling shake stimuli. | 9 | Room temperature compressive properties and strengthening mechanism of Mg96.17Zn3.15Y0.50Zr0.18 alloy solidified under high pressure显示文摘The microstructures of Mg96.17Zn3.15Y0.50Zr0.18 alloys solidified under 2-6 GPa high pressure were investigated by employing SEM(EDS) and TEM.The strengthening mechanism of experimental alloy solidified under high pressure is also discussed by analyzing the compressive properties and compression fracture morphology.The results show that the microstructure of experimental alloy becomes significantly fine-grained with increasing GPa level high pressure during solidification process,and the secondary dendrite arm spacing reduces from 40 μm at atmospheric pressure to 10 μm at 6 GPa pressure.The morphology of the second phases changes from the net structure by the lamellar-type eutectic structure at atmospheric pressure to discontinuous thin rods or particles at 6 GPa pressure.Besides,the solid solubility of Zn in the Mg matrix is improved with the increase of the solidification pressure.Compared with atmospheric-pressure solidification,high-pressure solidification can improve the strength of the experimental alloy.The compressive stre ngth is improved from 263 to 437 MPa at 6 GPa.The fracture mechanism of the experimental alloy changes from cleavage fracture at atmospheric pressure to quasi-cleavage fracture at high pressure.The main mechanism of the strength improvement of the experimental alloy includes the grain refinement strengthening caused by the refinement of the solidification microstructure,the second phase strengthening caused by the improvement of the morphology and distribution of the second phases,and solid solution strengthening caused by the increase of the solid solubility of Zn in the Mg matrix. | Xiaoping Lin Zhibin Fan Chang Xu Kunyu Guo Chang Xu Shiqi Guo | 2020 | Journal of Rare Earths2020,38,6: | 2 |

| 12 | Chip-based SALDI-MS for rapid determination of intracellular ratios of glutathione to glutathione disulfide显示文摘Alterations in the ratio of glutathione(GSH) to glutathione disulfide(GSSG) reveal the cell living state and are associated with a variety of diseases. In this study, an Au NPs grafted nanoporous silicon chip was used for surface assisted laser desorption ionization-mass spectrometry(SALDI-MS) detection of GSH. Due to the bond interaction between thiol of GSH and Au NPs modified on the chip surfaces, GSH could be captured from the complex cellular lysate. Meanwhile, the composite nanostructures of Au NPs grafted porous silicon surface presented good desorption/ionization efficiency for GSH detection. The GSH levels in different tumor cells were successfully detected. Chip-based SALDI-MS was optimized for quantification of intracellular GSH/GSSG ratio changing under drug stimulation in liver tumor cells, GSSG was reduced to GSH by reductant of tris(2-carboxyethyl)phosphine(TCEP) and isotope-labeling GSH was as an internal standard. It was found that the increasing concentration of drug irinotecan and hypoxia culture condition caused the rapid consumption of GSH and a decrease of GSH/GSSG ratio in liver tumor cells. Advanced microfluidic technologies enable to create more realistic in vitro microenvironment by spatial and temporal control of cell growth and co-culture. In this work, we design a microfluidic device to achieve the co-culture of PC12 cells and 293 cells, and study in vitro cell-cell interaction via cell metabolic analysis by mass spectrometry. The membraneintegrated microfluidic device was firstly used for cell co-culture, and the cellular metabolite was further investigated by mass spectrometer(MS). Our results showed that the differentiation of PC12 cells could be successfully induced by m NGF and also greatly influenced by the microchannel treatment of fetal bovine serum(FBS) solution. The identification of cell morphology, microtubule-associated protein 2(MAP-2) expression and viability of differentiated PC12 cells were conducted before 293 cells being introduced into the top microfluidic channels and stimulated to secrete cell metabolism products.
