2The transcription factor OpWRKY2 positively regulates the biosynthesis of the anticancer drug camptothecin in Ophiorrhiza pumila显示文摘The limited bioavailability of plant-derived natural products with anticancer activity poses major challenges to the pharmaceutical industry.An example of this is camptothecin,a monoterpene indole alkaloid with potent anticancer activity that is extracted at very low concentrations from woody plants.Recently,camptothecin biosynthesis has been shown to become biotechnologically amenable in hairy-root systems of the natural producer Ophiorrhiza pumila.Here,time-course expression and metabolite analyses were performed to identify novel transcriptional regulators of camptothecin biosynthesis in O.pumila.It is shown here that camptothecin production increased over cultivation time and that the expression pattern of the WRKY transcription factor encoding gene OpWRKY2 is closely correlated with camptothecin accumulation.Overexpression of OpWRKY2 led to a more than three-fold increase in camptothecin levels.Accordingly,silencing of OpWRKY2 correlated with decreased camptothecin levels in the plant.Further detailed molecular characterization by electrophoretic mobility shift,yeast one-hybrid and dual-luciferase assays showed that OpWRKY2 directly binds and activates the central camptothecin pathway gene OpTDC.Taken together,the results of this study demonstrate that OpWRKY2 acts as a direct positive regulator of camptothecin biosynthesis.As such,a feasible strategy for the over-accumulation of camptothecin in a biotechnologically amenable system is presented.Xiaolong Hao Chenhong Xie Qingyan Ruan Xichen Zhang Chao Wu Bing Han Jun Qian Wei Zhou Hans-Wilhelm Nutzmann Guoyin Kai 2021Horticulture Research2021,8,1:7
3Foraging strategies in the greater horseshoe bat(Rhinolophus ferrumequinum)on Lepidoptera in summer显示文摘The diet of the bat Rhinolophus ferrumequinum was studied at the Zhi’an Village of Ji’an City in China, from June to August 2004. The bats were trained in a laboratory (volume: 9×4×4 m3). Foraging strategies of the bat were ob- served at night and prey remains were collected and identi- fied. The results showed that the diet consisted mainly of Lepidoptera in summer, including 11 families, more than 30 species of moths, such as Noctuidae (36.6% by number), Sphingidae (24.1%), Geometridae (13.4%) and Limacodidae (9.5%). The length of culled wings ranged from 10―40 mm (97.7%). Pearson correlation analysis showed that the bat R. ferrumequinum foraged their prey selectively, but not op- portunistically. From field studies, two ways were observed in which the bats retrieved their prey including aerial hawk- ing during peak active period of the insects and flycatching during the insects’ non-peak activity period. 6Numerical Study of the Effect of Imaginary Circle Diameter and Initial Flow Field on the Aerodynamic Field in a Tangentially Fired Furnace显示文摘In this paper, the effect of the imaginary circle diameter opi and the initial flow field on the aerodynamic field in a tangentially fired furnace was studied by numerical simulation and experiments in the cold model. Results show that merely reducing the imaginary circle diameter rki can not significantly reduce the rotatiollal diameter op in the range considered. The flow still rotates coullter-clockwise stably and does not change rotation direction when the direction of all jet axes are defiected suddenly to the opposite rotation direction by up to 5.4' in a countereclockwise llow field. It is the first time that the numerical sAnulation results were obtained which agreed quite well with this experimental phenomena qualitatively. The experimental data, i.e., the rotational diameter & and the maximum velocity on the syrnmetric central line of fUrnace Vm, are only a bit larger than the simulation results. It is shown that the initial flow field has an important influence on the aerodynamic field in the furnace. Other measures have to be taken as wel1 in order to reduce & to resist slagging and high temperature corrosion of furnace tubes. 7China’s Recent Progresses in Polar Climate Change and Its Interactions with the Global Climate System显示文摘During the recent four decades since 1980,a series of modern climate satellites were launched,allowing for the measurement and record-keeping of multiple climate parameters,especially over the polar regions where traditional observations are difficult to obtain.China has been actively engaging in polar expeditions.Many observations were conducted during this period,accompanied by improved Earth climate models,leading to a series of insightful understandings concerning Arctic and Antarctic climate changes.Here,we review the recent progress China has made concerning Arctic and Antarctic climate change research over the past decade.The Arctic temperature increase is much higher than the global-mean warming rate,associated with a rapid decline in sea ice,a phenomenon called the Arctic Amplification.The Antarctic climate changes showed a zonally asymmetric pattern over the past four decades,with most of the fastest changes occurring over West Antarctica and the Antarctic Peninsula.The Arctic and Antarctic climate changes were driven by anthropogenic greenhouse gas emissions and ozone loss,while tropical-polar teleconnections play important roles in driving the regional climate changes and extreme events over the polar regions.Polar climate changes may also feedback to the entire Earth climate system.The adjustment of the circulation in both the troposphere and the stratosphere contributed to the interactions between the polar climate changes and lower latitudes.Climate change has also driven rapid Arctic and Southern ocean acidification.Chinese researchers have made a series of advances in understanding these processes,as reviewed in this paper.Xichen LI Xianyao CHEN Bingyi WU Xiao CHENG Minghu DING Ruibo LEI Di QI Qizhen SUN Xiaoyu WANG Wenli ZHONG Lei ZHENG Meijiao XIN Xiaocen SHEN Chentao SONG Yurong HOU 2023Advances in Atmospheric Sciences2023,40,8:1

The TiC/γ interface in as-cast sample is of coherent feature. Its orientation relationship is (020)γ//(220)TiC, [001]γ||[001]TiC. 11Delayed Antarctic melt season reduces albedo feedback显示文摘Antarctica’s response to climate change varies greatly both spatially and temporally.Surface melting impacts mass balance and also lowers surface albedo.We use a 43-year record(from 1978 to 2020)of Antarctic snow melt seasons from space-borne microwave radiometers with a machine-learning algorithm to show that both the onset and the end of the melt season are being delayed.Granger-causality analysis shows that melt end is delayed due to increased heat flux from the ocean to the atmosphere at minimum sea-ice extent from warming oceans.Melt onset is Granger-caused primarily by the turbulent heat flux from ocean to atmosphere that is in turn driven by sea-ice variability.Delayed snowmelt season leads to a net decrease in the absorption of solar irradiance,as a delayed summer means that higher albedo occurs after the period of maximum solar radiation,which changes Antarctica’s radiation balance more than sea-ice cover.Lei Liang Huadong Guo Shuang Liang Xichen Li John C.Moore Xinwu Li Xiao Cheng Wenjin Wu Yan Liu Annette Rinke Gensuo Jia Feifei Pan Chen Gong 2023National Science Review2023,10,9:0
12Viral RNA-based transfection and expression of enhanced green fluorescent protein in the parasitic protozoan Eimeria stiedae显示文摘Eimeria stiedae(E.stiedae),the most common protozoan pathogen of rabbits,causes coccidiosis.This disease is problematic for rabbit breeders [1].There is little information about the cellular and molecular biology of this pathogen and a lack of genetic tools.Genetic manipulation is necessary to advance our understanding of parasite biology and ultimately develop vaccines and treat diseases.DNA-mediated and viral RNA-mediated transfection systems have successfully been established in many protozoan parasites [2,3].It was discovered that E.stiedae possesses a dsRNA virus(EsRV1),and more recently,the genome of this virus has been sequenced.To date,however,there is no report on the use of viral RNA-mediated transfection for E.stiedae.In this study,we report a transient viral RNA-mediated transfection of E.stiedae using enhanced green fluorescent protein(EGFP)as a reporter gene.Our study provides a valuable genetic tool for studying the biology of E.stiedae.Caiyan Xin Jianhua Li Pengtao Gong Bin Wu Nan Zhang Ju Yang Xuepeng Cai Xichen Zhang 2019Acta Biochimica et Biophysica Sinica2019,51,2:0
