Generation of inactivated IL2RG and RAG1 monkeys with severe combined immunodeficiency using base editing显示文摘Severe combined immunodeficiency(SCiD)encompasses a range of inherited disorders that lead to a profound deterioration of the immune system.Among the pivotal genes associated with SCID,RAG1 and IL2RG play crucial roles.IL2RG is essential for the development, differentiation, and functioning of T,B,and NK cells, while RAG1 critically contributes to adaptive immunity by facilitating V(D)J recombination during the maturation of lymphocytes.Animal models carrying mutations in these genes exhibit notable deficiencies in their immune systems.Xiao Zheng Chunhui Huang Yingqi Lin Bofeng Han Yizhi Chen Caijuan Li Jiawei Li Yongyan Ding Xichen Song Wei Wang Weien Liang Jianhao Wu Jiaxi Wu Jiale Gao Chengxi Wei Xudong Zhang Zhuchi Tu Sen Yan 2023Signal Transduction and Targeted Therapy2023,8,10:0
Tauopathy promotes spinal cord-dependent production of toxic amyloid-beta in transgenic monkeys显示文摘Tauopathy, characterized by the hyperphosphorylation and accumulation of the microtubule-associated protein tau,and the accumulation of Aβ oligomers, constitute the major pathological hallmarks of Alzheimer's disease. However, the relationship and causal roles of these two pathological changes in neurodegeneration remain to be defined, even though they occur together or independently in several neurodegenerative diseases associated with cognitive and movement impairment.Zhuchi Tu Sen Yan Bofeng Han Cajuan Li Weien Liang Yingqi Lin Yongyan Ding Huiyi Wei Lu Wang Hao Xu Jianmeng Ye Bang Li Shihua Li Xiao-Jiang Li 2023Signal Transduction and Targeted Therapy2023,8,10:0
Expert consensus on prevention and cardiopulmonary resuscitation for cardiac arrest in COVID-19显示文摘Background:Cardiopulmonary resuscitation(CPR)strategies in COVID-19 patients differ from those in patients suffering from cardiogenic cardiac arrest.During CPR,both healthcare and non-healthcare workers who provide resuscitation are at risk of infection. The Working Group for Expert Consensus on Prevention and Cardiopulmonary Resuscitation for Cardiac Arrest in COVID-19 has developed this Chinese Expert Consensus to guide clinical practice of CPR in COVID-19 patients.Main recommendations:1)A medical team should be assigned to evaluate severe and critical COVID-19 for early monitoring of cardiac-arrest warning signs.2)Psychological counseling and treatment are highly recommended, since sympathetic and vagal abnormalities induced by psychological stress from the COVID-19 pandemic can induce cardiac arrest.3)Healthcare workers should wear personal protective equipment(PPE).4)Mouth-to-mouth ventilation should be avoided on patients suspected of having or diagnosed with COVID-19.5)Hands-only chest compression and mechanical chest compression are recommended.6)Tracheal-intubation procedures should be optimized and tracheal-intubation strategies should be implemented early.7)CPR should be provided for 20-30 min.8)Various factors should be taken into consideration such as the interests of patients and family members, ethics, transmission risks, and laws and regulations governing infectious disease control.Changes in management:The following changes or modifications to CPR strategy in COVID-19 patients are proposed:1)Healthcare workers should wear PPE.2)Hands-only chest compression and mechanical chest compression can be implemented to reduce or avoid the spread of viruses by aerosols.3)Both the benefits to patients and the risk of infection should be considered.4)Hhealthcare workers should be fully aware of and trained in CPR strategies and procedures specifically for patients with COVID-19.Wei Song Jie Wei Xiangdong Jian Deren Wang Yanhong Ouyang Yuanshui Liu Xianjin Du Ying Chen Yingqi Zhang Heping Xu Shuming Xianyu Qiong Ning Xiang Li Xiaotong Han Feng Zhan Tao Yu Wenteng Chen Jun Zhang Wenwei Cai Sheng’ang Zhou Shengyang Yi Yu Cao Xiaobei Chen Shunjiang Xu Zong’an Liang Duohu Wu Fen Ai Zhong Wang Qingyi Meng Yuhong Mi Sisen Zhang Rongjia Yang Shouchun Yan Wenbin Han Yong Lin Chuanyun Qian Wenwu Zhang Yan Xiong Jun Lv Baochi Liu Yan Cao Xiaojun He Xuelian Sun Yufang Cao Tian’en Zhou 2021Asian Pacific Journal of Tropical Medicine2021,14,6:0
Hypoglycemic effects of black brick tea with fungal growth in hyperglycemic mice model显示文摘Dark tea(containing Eurotium cristatum)and black tea have hypoglycemic effects. The black brick tea with fungal growth is obtained from black tea by adding E.cristatum, followed by steaming, pressing, fungal growth, and drying. However, the hypoglycemic effects of black brick tea are still unexplored. Here,we used black brick tea with fungal growth and black tea as raw materials to study their hypoglycemic effects in a hyperglycemic mice model. Both these types of black tea could lower the content of blood glucose and increase the content of hepatic glycogen by upregulating the proteins and m RNA expression of phosphatidylinositol-4,5-bisphosphate3-kinase,glycogen synthase, protein kinase B and phosphoinositidedependent protein kinase-1 and downregulating the protein and m RNA expression of glycogen synthase kinase 3β.These regulatory steps were followed by high activities of glutathione peroxidase and superoxide dismutase and low content of malondialdehyde. These teas can promote blood glucose transport and absorption by upregulating the protein and m RNA expression of insulin receptor substrate-1 and glucose transporter-2 and stimulate glycolysis by upregulating the protein and m RNA expression of 6-phosphofructo-2-kinase/fructose-2,6-biphosphatase 2.Our study suggests that the black brick tea was more effective than the black tea in terms of hypoglycemic.Wei Xu Yang Zhou Ling Lin Dongyin Yuan Yingqi Peng Li Li Wenjun Xiao Zhihua Gong 2022Food Science and Human Wellness2022,11,3:0
Bioinformatics exploration of potential common therapeutic targets for systemic and pulmonary arterial hypertension-induced myocardial hypertrophy显示文摘Systemic and pulmonary arterial hypertension(PAH)can induce left and right ventricular hypertrophy, respectively, but common therapeutic targets for both left and right hypertrophy are limited. In this study,we attempt to explore potential common therapeutic targets and screen out potential target drugs for further study.Cardiac mRNA expression profiles in mice with transverse aortic constriction(TAC)and pulmonary arterial constriction(PAC)are obtained from online databases. After bioinformatics analyses,we generate TAC and PAC mouse models to validate the phenotypes of cardiac remodelling as well as the identified hub genes.Bioinformatics analyses show that there are 214 independent differentially expressed genes(DEGs)in GSE136308(TAC related)and 2607 independent DEGs in GSE30922(PAC related),while 547 shared DEGs are associated with the function of the extracellular matrix(ECM)or involved in the PI3K-Akt signaling pathway,cytokine-cytokine receptor interactions, and ECM-receptor interactions. We identifyd Fn1,Il6,Col1a1,Igf1,Col1a2,Timp1,Col3a1,Cd44,Ctgf and Postn as hub genes of the shared DEGs,and most of them are associated with myocardial fibrosis. Those hub genes and phenotypes of cardiac remodelling are validated in our TAC and PAC mouse models.Furthermore,we identify dehydroisoandrosterone(DHEA),iloprost and 4,5-dianilinophthalimide(DAPH)as potential therapeutic drugs targeting both left and right ventricular hypertrophy and validate the effect of DHEA.These findings suggest that DHEA could be an effective drug for pressure overload-induced left or right ventricular hypertrophy by regulating the shared hub differentially expressed genes associated with fibrosis.Lu Chen Mingjue Li Mengjia Shen Yingqi Zhu Kaitong Chen Xiaoxia Huang Cankun Zheng Qiancheng Wang Hairuo Lin Wangjun Liao Jianping Bin Siyuan Ma Yulin Liao 2023Acta Biochimica et Biophysica Sinica2023,55,5:0
One‑Pot Rational Deposition of Coaxial Double‑Layer MnO_(2)/Ni(OH)_(2) Nanosheets on Carbon Nanofibers for High‑Performance Supercapacitors显示文摘Constructing“nanoglue”between inorganic electroactive species and conductive carbon scaffolds is an effective strategy to improve their compatibility and binding interaction, holding a great promise for fabricating high-performance hybrid electrodes for supercapacitors. However, multistep reactions are usually required to obtain these multicomponent systems, thus giving rise to the complicated and time-consuming issues.Herein,we for the first time, demonstrate a green one-pot method to anchor coaxial double-layer MnO_(2)/Ni(OH)_(2)nanosheets on electrospun carbon nanofibers(CNFs)(denoted as MNC),where the intermediate MnO_(2)layer serves as the“nanoglue”to couple the vertically aligned Ni(OH)_(2)nanosheets and conductive CNFs.Benefiting from the unique chemical composition and hierarchical architecture, the resultant electrode delivers outstanding electrochemical performance, including an excellent specific capacitance(1133.3 F g^(-1)at 1 A g^(-1))and an ultrahigh rate capability(844.4 F g^(-1)at 20 A g^(-1)).Moreover,the asymmetric supercapacitor assembled by using the MNC as positive electrode and the CNF as negative electrode can achieve an optimal energy density of 35.1 Wh kg^(-1)and a maximum power density of 8000 W kg^(-1).The one-pot strategy that stabilizes electroactive metal hydroxides on conductive carbons using a MnO_(2)“nanoglue”to design advanced hybrid electrodes is expected to be broadly applicable not only to the supercapacitor technology but also to other electrochemical applications.Guangdi Nie Zhenyuan Zhang Yingqi Liu Jian Wang Chen Fu Haiqi Yin Juan Chen Lin Zhao Zhenghui Pan 2022Advanced Fiber Materials2022,4,5:0
