| 3 | NIR-triggered thermo-responsive biodegradable hydrogel with combination of photothermal and thermodynamic therapy for hypoxic tumor显示文摘Hypoxia is a typical feature of solid tumors,which highly limits the application of the oxygen-dependent therapy.Also,the dense and hyperbaric tumor tissues impede the penetration of nanoparticles into the deep tumor.Thereby,we designed a novel localized injectable hydrogel combining the photothermal therapy(PTT)and the thermodynamic therapy(TDT),which is based on the generation of free radicals even in the absence of oxygen for hypoxic tumor therapy.In our study,gold nanorods(Au NRs)and 2,2-Azobis[2-(2-imidazalin-2-yl)propane]dihydrochlaride(AIPH)were incorporated into the hydrogel networks,which were formed by the copolymerization of hydrophobic N-isopropyl acrylamide(NIPAM)and hydrophilic glycidyl methacrylate modified hyaluronic acid(HAGMA)to fabricate an injectable and near-infrared(NIR)responsive hydrogel.The crosslinked in situ forming hydrogel could not only realize PTT upon the NIR laser irradiation,but also generate free radicals even in hypoxic condition.Meanwhile the shrink of hydrogels upon thermal could accelerate the generation of free radicals to further damage the tumors,achieving the controlled drug release on demand.The designed hydrogel with a sufficient loading capacity,excellent biocompatibility and negligible systemic toxicity could serve as a long-acting implant for NIR-triggered thermo-responsive free radical generation.The in vitro cytotoxicity result and the in vivo antitumor activity illustrated the excellent therapeutic effect of hydrogels even in the absence of oxygen.Therefore,this innovative oxygen-independent platform combining the antitumor effects of PTT and TDT would bring a new insight into hypoxic tumor therapy by the application of alkyl free radical. | Xiaoqi Sun Di Liu Xiaoyu Xu Yifeng Shen Yanjuan Huang Zishan Zeng Meng Xia Chunshun Zhao | 2020 | Asian Journal of Pharmaceutical Sciences2020,15,6: | 5 |
| 4 | Deficient Rnf43 potentiates hyperactive Kras-mediated pancreatic preneoplasia initiation and malignant transformation显示文摘Background:Largely due to incidental detection,asymptomatic pancreatic cystic le-sions(PCLs)have become prevalent in recent years.Among them,intraductal papillary mucinous neoplasm(IPMN)infrequently advances to pancreatic ductal adenocarcinoma(PDAC).Conservative surveillance versus surgical intervention is a difficult clinical decision for both caregivers and PCL patients.Because RNF43 loss-of-function mutations and KRAS gain-of-function mutations concur in a subset of IPMN and PDAC,their biological significance and therapeutic potential should be elucidated.Methods:Pancreatic Rnf43 knockout and Kras activated mice(Rnf43^(−/−);Kras^(G12D))were generated to evaluate their clinical significance in pancreatic pre-neoplastic initiation and malignant transformation.Results:Loss of Rnf43 potentiated the occurrence and severity of IPMN and PDAC in oncogenic Kras mice.The Wnt/β-catenin signaling pathway was activated in pan-creatic Kras^(G12D)and Rnf43 knockout mice and the PORCN inhibitor LGK974 blocked pancreatic IPMN initiation and progression to PDAC accordingly.Conclusions:Rnf43 is a tumor suppressor in the prevention of pancreatic malignant transformation.This genetically reconstituted autochthonous pancreatic Rnf43^(−/−);Kras^(G12D)preclinical cancer model recapitulates the pathological process from pancreatic cyst to cancer in humans and can be treated with inhibitors of Wnt/β-catenin signaling.Since the presence of RNF43 and KRAS mutations in IPMNs predicts future development of advanced neoplasia from PCLs,patients with these genetic anomalies warrant surveillance,surgery,and/or targeted therapeutics such as Wnt/β-catenin inhibitors. | Xian Zhou Zhichao Sun Mengdi Zhang Xiaoyu Qu Shuhui Yang Lianmei Wang Yanling Jing Li Li Weiwei Deng Fangming Liu Jin Di Jie Chen Jian Wu Hongbing Zhang | 2022 | Animal Models and Experimental Medicine2022,5,1: | 3 |

| 6 | Is there an optimal time to initiate adjuvant chemotherapy to predict benefit of survival in non-small cell lung cancer?显示文摘Objective: Adjuvant chemotherapy(AC) after curative resection is known to improve the survival of patients with non-small cell lung cancer(NSCLC); however, few studies have reported the correlation between the time to initiation of AC(TTAC) and survival in NSCLC patients.Methods: The clinical data of 925 NSCLC patients who received curative resection and post-operative AC at the Cancer Hospital of Chinese Academy of Medical Sciences between 2003 and 2013 were retrospectively analyzed.TTAC was measured from the date of surgery to the initiation of AC. Disease-free survival(DFS) was defined as the duration from surgery to the time of tumor recurrence or last follow-up evaluation. The optimal cut-off value of TTAC was determined by maximally selected log-rank statistics. The DFS curve was estimated using the Kaplan-Meier method, and the Cox proportional hazards regression model was used to identify risk factors independently associated with DFS. Propensity score matching(PSM) was performed for survival analysis using the match data.Results: The optimal discriminating cut-off value of TTAC was set at d 35 after curative resection based on which the patients were assigned into two groups: group A(≤35 d) and group B(>35 d). There was no significant difference in the DFS between the two groups(P=0.246), indicating that the TTAC is not an independent prognostic factor for DFS. A further comparison continued to show no significant difference in the DFS among 258 PSM pairs(P=0.283).Conclusions: There was no significant correlation between the TTAC and DFS in NSCLC patients. | 10 | Cooperative Privacy Provisioning for Energy Harvesting Based Cognitive Multi-Relay Networks显示文摘In order to provide privacy provisioning for the secondary information,we propose an energy harvesting based secure transmission scheme for the cognitive multi-relay networks.In the proposed scheme,two secondary relays harvest energy to power the secondary transmitter and assist the secondary secure transmission without interfere the secondary transmission.Specifically,the proposed secure transmission policy is implemented into two phases.In the first phase,the secondary transmitter transmits the secrecy information and jamming signal through the power split method.After harvesting energy from a fraction of received radio-frequency signals,one secondary relay adopts the amplify-and-forward relay protocol to assist the secondary secure transmission and the other secondary relay just forwards the new designed jamming signal to protect the secondary privacy information and degrade the jamming interference at the secondary receiver.For the proposed scheme,we first analyze the average secrecy rate,the secondary secrecy outage probability,and the ergodic secrecy rate,and derive their closed-form expressions.Following the above results,we optimally allocate the transmission power such that the secrecy rate is maximized under the secrecy outage probability constraint.For the optimization problem,an AI based simulated annealing algorithm is proposed to allocate the transmit power.Numerical results are presented to validate the performance analytical results and show the performance superiority of the proposed scheme in terms of the average secrecy rate. | Dawei Wang Wei Liang Xiaoyu Hu Daosen Zhai Di Zhang | 2020 | China Communications2020,17,2: | 1 |

| 14 | Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND experiment. The tightest statistical constraints on the dense matter equation of state will come from pulse profile modelling of accretion-powered pulsars, burst oscillation sources, and rotation-powered pulsars. Additional constraints will derive from spin measurements, burst spectra, and properties of the accretion flows in the vicinity of the neutron star.
