4Inhibiting effect of antisense oligonucleotides phosphorthioate on gene expression of TIMP-1 in rat liver fibrosis显示文摘AIM To observe the inhibition of antisenseoligonucleotides (asON) phosphorthioate to thetissue inhibitors metalloproteinase-1 (TIMP-1)gene and protein expression in the liver tissue ofimmunologically induced hepatic fibrosis rats.The possibility of reversing hepatic fibrosisthrough gene therapy was observed.METHODS Human serum albumin (HSA) wasused to attack rats, as hepatic fibrosis model, inwhich asONs were used to block the gene andprotein expressing TIMP-1. According to theanalysis of modulator, structure protein, codingseries of TIMP-1 genome, we designed fourdifferent asONs. These asONs were injected intothe hepatic fibrosis models through coccygealvein. The results was observed by RT-PCR formeasuring TIMP-1 mRNA expression,immunohistochemistry and in situ hybridizationfor collagen Ⅰ, Ⅲ, special staining of collagenfiber, and electron microscopic examination.RESULTS Hepatic fibrosis could last within 363days in our modified model. The expressinglevel of TIMP-1 was high during hepatic fibrosisprocess. It has been proved by theimmunohistochemical and the electronmicroscopic examination that the asONphosphorthioate of TIMP-1 could exactly expressin vivo. The effect of colchicine wasdemonstrated to inhibit the expressing level ofmRNA and the content of collagen Ⅰ, Ⅲ in theliver of experimental hepatic fibrosis rats.However, the electron microscopy research andthe pathologic grading of hepatic fibrosisshowed that there was no significant differencebetween the treatment group and the modelgroup (P>0.05).CONCLUSION The experimental rat model ofhepatic fibrosis is one of the preferable modelsto estimate the curative effect of anti-hepaticfibrosis drugs. The asON phosphorthioate ofTIMP-1 could block the gene and proteinexpression of TIMP-1 in the liver of experimentalhepatic fibrosis rats at the mRNA level. It ispossible to reverse hepatic fibrosis, and it isexpected to study a new drug of anti-hepaticfibrosis on the genetic level. 52018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents显示文摘Syncope belongs to the transient loss of consciousness(TLOC), characterized by a rapid onset, short duration, and spontaneous complete recovery. It is common in children and adolescents, accounting for 1% to 2% of emergency department visits.Recurrent syncope can seriously affect children's physical and mental health, learning ability and quality of life and sometimes cardiac syncope even poses a risk of sudden death. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. Based on the globally recent development and the evidence-based data in China, 2018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents was jointly prepared by the Pediatric Cardiology Society, Chinese Pediatric Society, Chinese Medical Association(CMA)/Committee on Pediatric Syncope, Pediatricians Branch, Chinese Medical Doctor Association(CMDA)/Committee on Pediatric Cardiology, Chinese College of Cardiovascular Physicians, Chinese Medical Doctor Association(CMDA)/Pediatric Cardiology Society, Beijing Pediatric Society, Beijing Medical Association(BMA). 6Methodologic research on TIMP-1,TIMP-2 detection as a new diagnostic index for hepatic fibrosis and its significance显示文摘AIM: To set up a new method to detect tissue inhibitors ofmetalloproteinase1 and -2(TIMP-1 and TIMP-2) in sero ofpatients with hepatic cirrhosis, and to investigate theexpression and location of TIMP-1 and TIMP-2 in liver tissueof patients with hepatic cirrhosis, and the correlationbetween TIMPs in liver and those in sera so as to discusswhether TIMPs can be used ss a diagnosis index of hepaticfibrosisMETHODS: The monoclonal antibodies (McAbs) of TIMP-1and TIMP-2 were used to sensitize erythrocytes, and solid-phase absorption to sensitized erythrocytes (SPASE) wasused to detect TIMP-1 and TIMP-2 in the sera of patients withhepatic cirrhosis. Meanwhile, with the method of in situhybridization and immunohistochemistry, we studied themRNA expression and antigen location of TIMP-1 and TIMP-2in the livers of 40 hepatic cirrhosis patients with pathologicdiagnosis.RESULTS: With SPASE, they were 16.4 % higher in theacute hepatitis group, 33.3 % higher in the chronic hepatitisgroup, and the positive rates were 73.6 % and 61. 2 %respectively in sero of hepatic cirrhosis patients, which wereremarkably higher than those in chronic hepatitis and acutehepatitis group ( P < 0. 001 ). In 40 samples of hepaticcirrhosis tissues, all of them showed positive expression ofTIMP-1 and TIMP-2 mRNA detected withimmunohistochemistry or in situ hybridization (positive ratewas 100 % ). Expression of TIMPs in different degrees couldbe found in liver tissue with cirrhosis. 7Programmable time-domain digital-coding metasurface for non-linear harmonic manipulation and new wireless communication systems显示文摘Optical non-linear phenomena are typically observed in natural materials interacting with light at high intensities, and they benefit a diverse range of applications from communication to sensing. However,controlling harmonic conversion with high efficiency and flexibility remains a major issue in modern optical and radio-frequency systems. Here, we introduce a dynamic time-domain digital-coding metasurface that enables efficient manipulation of spectral harmonic distribution. By dynamically modulating the local phase of the surface reflectivity, we achieve accurate control of different harmonics in a highly programmable and dynamic fashion, enabling unusual responses, such as velocity illusion. As a relevant application, we propose and realize a novel architecture for wireless communication systems based on the time-domain digital-coding metasurface, which largely simplifies the architecture of modern communication systems, at the same time yielding excellent performance for real-time signal transmission. 8SlMYB75,an MYB-type transcription factor,promotes anthocyanin accumulation and enhances volatile aroma production in tomato fruits显示文摘Genetic manipulation of genes to upregulate specific branches of metabolic pathways is a method that is commonly used to improve fruit quality.However,the use of a single gene to impact several metabolic pathways is difficult.Here,we show that overexpression of the single gene SlMYB75(SlMYB75-OE)is effective at improving multiple fruit quality traits.In these engineered fruits,the anthocyanin content reached 1.86mg g−1 fresh weight at the red-ripe stage,and these SlMYB75-OE tomatoes displayed a series of physiological changes,including delayed ripening and increased ethylene production.In addition to anthocyanin,the total contents of phenolics,flavonoids and soluble solids in SlMYB75-OE fruits were enhanced by 2.6,4,and 1.2 times,respectively,compared to those of wild-type(WT)fruits.Interestingly,a number of aroma volatiles,such as aldehyde,phenylpropanoid-derived and terpene volatiles,were significantly increased in SlMYB75-OE fruits,with some terpene volatiles showing more than 10 times higher levels than those in WT fruits.Consistent with the metabolic assessment,transcriptomic profiling indicated that the genes involved in the ethylene signaling,phenylpropanoid and isoprenoid pathways were greatly upregulated in SlMYB75-OE fruits.Yeast one-hybrid and transactivation assays revealed that SlMYB75 is able to directly bind to the MYBPLANT and MYBPZM cis-regulatory elements and to activate the promoters of the LOXC,AADC2 and TPS genes.The identification of SlMYB75 as a key regulator of fruit quality attributes through the transcriptional regulation of downstream genes involved in several metabolic pathways opens new avenues towards engineering fruits with a higher sensory and nutritional quality.Wei Jian Haohao Cao Shu Yuan Yudong Liu Juanfang Lu Wang Lu Ning Li Jianhui Wang Jian Zou Ning Tang Chan Xu Yulin Cheng Yanqiang Gao Wanpeng Xi Mondher Bouzayen Zhengguo Li 2019Horticulture Research2019,6,1:39
9Rho/ROCK pathway and neural regeneration: a potential therapeutic target for central nervous system and optic nerve damage显示文摘· Rho-associated kinase (ROCK) is a serine/threonine kinase and one of the major downstream effectors of the small GTPase RhoA. The Rho/ROCK pathway is closely related to the pathogenesis of several central nervous system (CNS) disorders, and involved in many aspects of neuronal functions including neurite outgrowth and retraction. In the adult CNS, the damaged neuron regeneration is very difficult due to the presence of myelin-associated axon growth inhibitors such as Nogo, myelin-associated glycoprotein (MAG) and oligodendrocyte-myelin glycoprotein (Omgp), etc. The effects of these axon growth inhibitors are reversed by blocking the Rho/ROCK pathway in vitro, and the inhibition of Rho/ROCK pathway can promote axon regeneration and functional recovery in the injured CNS in vivo. In addition, the therapeutic effects of the Rho/ROCK inhibitors have also been demonstrated in some animal models and the Rho/ROCK pathway becomes an attractive target for the development of drugs for treating CNS disorders. 11Identification of a Stable Quantitative Trait Locus for Percentage Grains with White Chalkiness in Rice (Oryza sativa)显示文摘High chalkiness is a major problem in many rice-producing areas of the world, especially in hybrid rice (Oryza sativa L.) in China. We previously showed a major quantitative trait locus for the percentage of grains with white chalkiness (QTLqPGWC-8) in the interval G1149-R727 on chromosome 8 using a chromosome segment substitution line (CSSL). Here, we selected the line-CSSL50 harboring the QTLqPGWC-8 allele from the CSSLs derived from a cross between Asominori (as a recurrent parent) and IR24 (as a donor parent), which had higher percentage chalkiness, markedly different from that of Asominori. There were also significant differences in starch granules, appearance of amylose content (AAC) and milling qualities between Asominori and CSSL50, but not in grain size or thousand grain weight (TGW). The BC4F2 and BC4F3 populations from a cross between CSSL50 and Asominori were used for fine mapping of qPGWC-8. We narrowed down the location of this QTL to a 142 kb region between Indel markers 8G-7 and 8G-9. QTLqPGWC-8 accounted for 50.9% of the difference in PGWC between the parents. 15Detection of apoptosis induced by new type gosling viral enteritis virus in vitro through fluorescein annexin V-FITC/PI double labeling显示文摘AIM: To achieve a better understanding of the pathogenesis of new type gosling viral enteritis virus (NGVEV) and the relationship between NGVEV and host cells. METHODS: The apoptosis of duck embryo fibroblasts (DEF) induced by NGVEV was investigated by fluorescence-activated cell sorter (FACS) and fluorescence microscope after the cells were stained with Annexin V-FITC and propidium iodide (PI). RESULTS: By staining cells with a combination of fluorescein annexin V-FITC and PI, it is possible to distinguish and quantitatively analyze non-apoptotic cells (Annexin V-FITC negative/PI negative), early apoptotic cells (Annexin V-FITC positive/PI negative), late apoptotic/necrotic cells (Annexin V-FITC positive/ PI positive) and dead cells (Annexin V-FITC negative/PI positive) through flow cytometry and fluorescence microscope. 17A complex variable meshless method for fracture problems显示文摘Based on the moving least-square (MLS) approximation, the complex variable moving least-square approximation (CVMLS) is discussed in this paper. The complex variable moving least-square approximation cannot form ill-conditioned equations, and has greater precision and computational efficiency. Using the analytical solution near the tip of a crack, the trial functions in the complex variable moving least-square approxi- mation are extended, and the corresponding approximation function is obtained. And from the minimum potential energy principle, a complex variable meshless method for fracture problems is presented, and the formulae of the complex variable meshless method are obtained. 18Distribution of the COVID-19 epidemic and correlation with population emigration from Wuhan,China显示文摘Background:The ongoing new coronavirus pneumonia(Corona Virus Disease 2019,COVID-19)outbreak is spreading in China,but it has not yet reached its peak.Five million people emigrated from Wuhan before lockdown,potentially representing a source of virus infection.Determining case distribution and its correlation with population emigration from Wuhan in the early stage of the epidemic is of great importance for early warning and for the prevention of future outbreaks.Methods:The official case report on the COVID-19 epidemic was collected as of January 30,2020.Time and location information on COVID-19 cases was extracted and analyzed using ArcGIS and WinBUGS software.Data on population migration from Wuhan city and Hubei province were extracted from Baidu Qianxi,and their correlation with the number of cases was analyzed.Results:The COVID-19 confirmed and death cases in Hubei province accounted for 59.91%(5806/9692)and 95.77%(204/213)of the total cases in China,respectively.Hot spot provinces included Sichuan and Yunnan,which are adjacent to Hubei.The time risk of Hubei province on the following day was 1.960 times that on the previous day.The number of cases in some cities was relatively low,but the time risk appeared to be continuously rising.The correlation coefficient between the provincial number of cases and emigration from Wuhan was up to 0.943.The lockdown of 17 cities in Hubei province and the implementation of nationwide control measures efficiently prevented an exponential growth in the number of cases.Conclusions:The population that emigrated from Wuhan was the main infection source in other cities and provinces.Some cities with a low number of cases showed a rapid increase in case load.Owing to the upcoming Spring Festival return wave,understanding the risk trends in different regions is crucial to ensure preparedness at both the individual and organization levels and to prevent new outbreaks.Ze-Liang Chen Qi Zhang Yi Lu Zhong-Min Guo Xi Zhang Wen-Jun Zhang Cheng Guo Cong-Hui Liao Qian-Lin Li Xiao-Hu Han Jia-Hai Lu 2020Chinese Medical Journal2020,,9:15
19SSR Analysis of Genetic Relationship and Classification in Chrysanthemum Germplasm Collection显示文摘To further test the accuracy and rationality of the Chinese classification system for chrysanthemum(Chrysanthemum morifolium), 10 simple sequence repeat(SSR) markers were used to identify a collection of 88 chrysanthemum and its related genera accessions. In total, 42 effective alleles across 88 accessions were detected; 3 429 bands were obtained by PCR amplification, including 2 630 polymorphic bands. The similarity coefficient ranged from 0.53 to 0.88. Cluster analysis based on UPGMA illustrated that the wild species and large-flower cultivars were first divided into two clusters, then the large-flower cultivars formed five distinct groups according to petal type, indicated that petal type can be a classification criterion. In the wild species cluster, C. vestitum and C. zawadaskii grouped with A. trilobata, suggested that the Ajania genera was closely related to the Chrysanthemum genera. ‘Hangbaiju’, ‘Gongju’ and ‘Chuju’ were grouped together, and ‘Boju’, O. longilobus and C. mongolicum constituted another branch, showed a correlation with geographic region of origin. Population structure analysis was subsequently performed with K values ranging from 2 to 10, and the most likely estimate for the population structure is five subpopulations, which is nearly consistent with the clustering results. Principle component analysis was further performed to verify the classification results. 20Relation among p130Cas,E-cadherin and β-catenin expression,clinicopathologic significance and prognosis in human hepatocellular carcinoma显示文摘BACKGROUND:p130Cas(p130Crk-associated substance) is a junction protein that is important to the adhesion between cytoskeleton and extracellular matrix.Also the adhesion molecules E-cadherin andβ-catenin play important roles in the invasiveness of carcinoma.This study was undertaken to investigate the effects of p130Cas E-cadherin andβ-catenin on the invasion,metastasis and prognosis of hepatocellular carcinoma(HCC). METHODS:Immunohistochemistry was used to evaluate the expression of p130Cas,E-cadherin,andβ-catenin in 40 patients with HCC.All patients were followed up postoperatively,and the relationship between expression and clinicopathological prognostic parameters was analyzed RESULTS:The positive expression rates of p130Cas and E-cadherin in HCC tissue(n=40)were 62.50%and 55.00% but in normal liver tissue 10%,and 100%,respectively (P<0.05).The abnormal expression rate ofβ-catenin in HCC tissue was 70%,while in normal liver tissue it was 13.33%(P<0.05).The positive rate of p130Cas was correlated with lymph node invasion,pathological stage TNM stage,and a worse prognosis,but not with gender age,HBV infection,hepatic cirrhosis,alpha-fetoprotein (AFP)level before operation,and tumor diameter Similarly,the expression of E-cadherin andβ-catenin was correlated with lymph node invasion,pathological stage,TNM stage,and worse prognosis,but not with gender,age,HBV infection,hepatic cirrhosis,AFP level before operation,and tumor size.Correlations were found between p130Cas and abnormal E-cadherin/β-catenin expression(P<0.001 and<0.05,respectively).CONCLUSIONS:In HCC,there is a negative correlation between the positive expression of p130Cas and the normal expression of the adhesion molecules E-cadherin/β-catenin, and p130Cas plays important roles in the invasion, metastasis and prognosis of HCC.p130Cas may be involved in alterating the structure and function of E-cadherin/ β-catenin,by regulating tyrosine phosphorylation via the p130Cas-Src signal pathway.Cheng Guo,Qing-Guang Liu,Wei Yang,Zheng-Liang Zhang and Ying-Min Yao Department of Hepatobiliary Surgery,First Affiliated Hospital,School of Medicine,Xi’an Jiaotong University,Xi’an 710061, China 2008Hepatobiliary & Pancreatic Diseases International2008,7,5:15
