| 3 | 食物与鼻咽癌——广州市居民鼻咽癌病例对照研究显示文摘1983年3月1日至1985年8月31日对经病理确诊,年龄在50岁以下的广州市居民鼻咽癌新发病例进行病例一对照研究。应用Logistic多因素回归分析方法来分析各项数据,结果如下: 1.在各个被调查时期,母亲和对象咸鱼摄入经常度与鼻咽癌发病有显著相关。 2.除咸鱼外,儿童期进食其他五种腌制食品或酱油表明与鼻咽癌发病呈显著相关。这五种腌制食品为鱼露、咸虾酱、腐乳、陈皮梅、嘉应子。而发病前三年进食这些食品显示与鼻咽癌发病无关。 3.在各个被调查时期,病例组较对照组更少进食新鲜蔬菜和水果,显示与鼻咽癌发病有相关。 4.检测咸鱼、鱼露、咸虾酱、腐乳、陈皮梅、嘉应子、西红柿对鼻咽癌发病的作用,有三种食物显示其在鼻咽癌发病上是作为独立的危险因素,它们是咸鱼、腐乳、陈皮梅,而西红柿则为保护因素。 综上述可见:从多个流行病学调查和实验室研究均表明:进食含有亚硝胺类致癌物的咸鱼与鼻咽癌发病密切相关,而且在儿童期越年幼进食,相对危险性越大。 | 黄腾波 Mimi C. | 4 | Substrate stiffness modulates bone marrow-derived macrophage polarization through NF-κB signaling pathway显示文摘The stiffness of the extracellular matrix(ECM)plays an important role in regulating the cellular programming.However,the mechanical characteristics of ECM affecting cell differentiation are still under investigated.Herein,we aimed to study the effect of ECM substrate stiffness on macrophage polarization.We prepared polyacrylamide hydrogels with different substrate stiffness,respectively.After the hydrogels were confirmed to have a good biocompatibility,the bone marrow-derived macrophages(BMMs)from mice were incubated on the hydrogels.With simulated by the low substrate stiffness,BMMs displayed an enhanced expression of CD86 on the cell surface and production of reactive oxygen species(ROS)in cells,and secreted more IL-1βand TNF-αin the supernatant.On the contrary,stressed by the medium stiffness,BMMs expressed more CD206,produced less ROS,and secreted more IL-4 and TGF-β.In vivo study by delivered the hydrogels subcutaneously in mice,more CD68+CD86+cells around the hydrogels with the low substrate stiffness were observed while more CD68+CD206+cells near by the middle stiffness hydrogels.In addition,the expressions of NIK,phosphorylated p65(pi-p65)and phosphorylated IκB(pi-IκB)were significantly increased after stimulation with low stiffness in BMMs.Taken together,these findings demonstrated that substrate stiffness could affect macrophages polarization.Low substrate stiffness promoted BMMs to shift to classically activated macrophages(M1)and the middle one to alternatively activated macrophages(M2),through modulating ROS-initiated NF-κB pathway.Therefore,we anticipated ECM-based substrate stiffness with immune modulation would be under consideration in the clinical applications if necessary. | Mimi Chen Yu Zhang Pinghui Zhou Xingzhi Liu Huan Zhao Xichao Zhou Qiaoli Gu Bin Li Xuesong Zhu Qin Shi | 2020 | Bioactive Materials2020,5,4: | 7 |

| 6 | Archean-Paleoproterozoic Lithospheric Mantle at the Northern Margin of the North China Craton Represented by Tectonically Exhumed Peridotites显示文摘Tectonically emplaced peridotites from North Hebei Province, North China Craton, have retained an original harzburgite mineral assemblage of olivine(54%–58%) + orthopyroxene(40%–46%)+minor clinopyroxene(<1%)+spinel. Samples with boninite-like chemical compositions also coexist with these peridotites. The spinels within the peridotites have high-Al end-members with Al_2O_3 content of 30 wt % –50 wt %, typical of mantle spinels. When compared with experimentally determined melt extraction trajectories, the harzburgites display a high degree of melting and enrichment of SiO_2, which is typical of cratonic mantle peridotites. The peridotites display variably enriched light rare earth elements(REEs), relatively depleted middle REEs and weakly fractionated heavy REEs, which suggest a melt extraction of over 25% in the spinel stability field. The occurrence of arc-and SSZ-type chromian spinels in the peridotites suggests that melt extraction and metasomatism occurred mostly in a subduction-related setting. This is also supported by the geochemical data of the coexisting boninite-like samples. The peridotites have ^(187)Os/^(188)Os ratios ranging from 0.113–0.122, which is typical of cratonic lithospheric mantle. These ^(187)Os/^(188)Os ratios yield model melt extraction ages(TRD) ranging from 981 Ma to 2054 Ma, which may represent the minimum estimation of the melt extraction age. The Al_2O_3-^(187)Os/^(188)Os-proxy isochron ages of 2.4 Ga–2.7 Ga suggest a mantle melt depletion age between the Late Achaean and Early Paleoproterozoic. | 7 | Lactobacillus reuteri-derived extracellular vesicles maintain intestinal immune homeostasis against lipopolysaccharideinduced inflammatory responses in broilers显示文摘Background:Lactobacillus reuteri strains are widely used as probiotics to prevent and treat inflammatory bowel disease by modulating the host’s immune system.However,the underlying mechanisms by which they communicate with the host have not been clearly understood.Bacterial extracellular vesicles(EVs)have been considered as important mediators of host-pathogen interactions,but their potential role in commensals-host crosstalk has not been widely studied.Here,we investigated the regulatory actions of EVs produced by L.reuteri BBC3,a gut-associated commensal bacterium of Black-Bone chicken,in the development of lipopolysaccharide(LPS)-induced intestinal inflammation in a chicken model using both in vivo and in vitro experiments.Results:L.reuteri BBC3 produced nano-scale membrane vesicles with the size range of 60-250 nm.Biochemical and proteomic analyses showed that L.reuteri BBC3-derived EVs(LrEVs)carried DNA,RNA and several bioactive proteins previously described as mediators of other probiotics’beneficial effects such as glucosyltransferase,serine protease and elongation factor Tu.In vivo broiler experiments showed that administration of LrEVs exerted similar effects as L.reuteri BBC3 in attenuating LPS-induced inflammation by improving growth performance,reducing mortality and decreasing intestinal injury.LrEVs suppressed the LPS-induced expression of pro-inflammatory genes(TNF-α,IL-1β,IL-6,IL-17 and IL-8),and improved the expression of anti-inflammatory genes(IL-10 and TGF-β)in the jejunum.LrEVs could be internalized by chicken macrophages.In vitro pretreatment with LrEVs reduced the gene expression of TNF-α,IL-1βand IL-6 by suppressing the NF-κB activity,and enhanced the gene expression of IL-10 and TGF-βin LPS-activated chicken macrophages.Additionally,LrEVs could inhibit Th1-and Th17-mediated inflammatory responses and enhance the immunoregulatory cells-mediated immunosuppression in splenic lymphocytes of LPS-challenged chickens through the activation of macrophages.Finally,we revealed that the reduced content of both vesicular proteins and nucleic acids attenuated the suppression of LrEVs on LPS-induced inflammatory responses in ex vivo experiments,suggesting that they are essential for the LrEVs-mediated immunoregulation.Conclusions:We revealed that LrEVs participated in maintaining intestinal immune homeostasis against LPSinduced inflammatory responses in a chicken model.Our findings provide mechanistic insight into how commensal and probiotic Lactobacillus species modulate the host’s immune system in pathogens-induced inflammation. | Rujiu Hu Hua Lin Mimi Wang Yuezhen Zhao Haojing Liu Yuna Min Xiaojun Yang Yupeng Gao Mingming Yang | 2021 | Journal of Animal Science and Biotechnology2021,12,3: | 5 |

| 9 | Activation of Piezo1 contributes to matrix stiffness-induced angiogenesis in hepatocellular carcinoma显示文摘Background:Despite integrin being highlighted as a stiffness-sensor molecule in matrix stiffness-driven angiogenesis,other stiffness-sensor molecules and their mechanosensory pathways related to angiogenesis in hepatocellular carcinoma(HCC)remain obscure.Here,we explored the interplay between Piezo1 and integrinβ1 in the mechanosensory pathway and their effects on HCC angiogenesis to better understand matrix stiffness-induced angiogenesis.Methods:The role of Piezo1 in matrix stiffness-induced angiogenesis was investigated using orthotopic liver cancer SD rat models with high liver stiffness background,and its clinical significance was evaluated in human HCC tissues.Matrix stiffness-mediated Piezo1 upregulation and activation were assayed using an in vitro fibronectin(FN)-coated cell culture system with different stiffness,Western blotting and Ca^(2+)probe.The effects of shPiezo1-conditioned medium(CM)on angiogenesis were examined by tube formation assay,wound healing assay and angiogenesis array.The underlying mechanism by which Piezo1 participated in matrix stiffness-induced angiogenesis was analyzed by microRNA quantitative real-time polymerase chain reaction(qRT-PCR),matrix stiffness measurement,dual-luciferase reporter assay,ubiquitination assay and co-immunoprecipitation.Results:Increased matrix stiffness significantly upregulated Piezo1 expression at both cellular and tissue levels,and high expression of Piezo1 indicated an unfavorable prognosis.High matrix stiffness also noticeably enhanced the activation level of Piezo1,similar to its expression level.Piezo1 knockdown significantly suppressed tumor growth,angiogenesis,and lung metastasis of HCC rat models with high liver stiffness background.shPiezo1-CM from HCC cells attenuated tube formation and migration abilities of vascular endothelial cells remarkably,and analysis of differentially expressed pro-angiogenic factors revealed that Piezo1 promoted the expression and secretion of vascular endothelial growth factor(VEGF),CXC chemokine ligand 16(CXCL16)and insulin-like growth factor binding protein 2(IGFBP2).Matrix stiffness-caused Piezo1 upregulation/activation restrained hypoxia inducible factor-1α(HIF-1α)ubiquitination,subsequently enhanced the expression of downstream pro-angiogenic factors to accelerate HCC angiogenesis.Besides,collagen 1(COL1)-reinforced tissue stiffening resulted in more expression of Piezo1 via miR-625-5p.Conclusions:This study unravels a new mechanism by which the inte-grinβ1/Piezo1 activation/Ca2+influx/HIF-1αubiquitination/VEGF,CXCL16 and IGFBP2 pathway participates in matrix stiffness-driven HCC angiogenesis.Simultaneously,a positive feedback regulation loop as stiff matrix/integrinβ1/miR-625-5p/Piezo1 and COL1/stiffer matrix mediates matrix stiffness-caused Piezo1 upregulation. | Miao Li Xi Zhang Mimi Wang Yaohui Wang Jiali Qian Xiaoxia Xing Zhiming Wang Yang You Kun Guo Jie Chen Dongmei Gao Yan Zhao Lan Zhang Rongxin Chen Jiefeng Cui Zhenggang Ren | 2022 | Cancer Communications2022,42,11: | 3 |
