1大豆抗胞囊线虫病种质rhg_1和Rhg_4位点的单核苷酸多态性(SNPs)显示文摘研究以 5 7份中美大豆抗胞囊线虫病种质资源为实验材料 ,利用基于检测微珠的单碱基延伸方法 ,对与大豆胞囊线虫病 (SCN)抗性基因rhg1和Rhg4紧密连锁的SNPs进行分析 ,目的是阐明我国大豆抗性种质在这两个位点的SNPs等位变异分布频率 ,为中国大豆种质抗SCN资源的利用奠定基础。分析结果表明 ,SNPs的抗性等位基因与中国大豆种质综合抗性的关系比不同生理小种的抗性关系更为密切。在rhg1和Rhg4位点 ,美国的 9份抗性种质中 ,有 7份抗性种质的SNPs均为纯合抗病基因型 ,而中国 4 8份抗性种质中有 32份 ,分别占鉴定总数的 77 8%和 6 6 7% ,推测大豆抗SCN种质中 ,以rhg1和Rhg4这两个基因协同作用表现出的抗性可能占多数 ,但还存在其他的抗性机制。邱丽娟 常汝镇 王文辉 Perry Cregan Dechun Wang Yiwu Chen 马凤鸣 2003植物遗传资源学报2003,4,2:19
2Fatigue behavior of Ti-6Al-4V cellular structures fabricated by additive manufacturing technique显示文摘Porous titanium and its alloys have been considered as promising replacement for dense implants, as they possess low elastic modulus comparable to that of compact human bones and are capable of providing space for in-growth of bony tissues to achieve a better fixation. Recently, the additive manufacturing(AM) method has been successfully applied to the fabrication of Ti-6 Al-4 V cellular meshes and foams.Comparing to traditional fabrication methods, the AM method offers advantages of accurate control of complex cell shapes and internal pore architectures, thus attracting extensive attention. Considering the long-term safety in the human body, the metallic cellular structures should possess high fatigue strength.In this paper, the recent progress on the fatigue properties of Ti-6 Al-4 V cellular structures fabricated by the AM technique is reviewed. 3Punicalagin ameliorates collagen-induced arthritis by downregulating M1 macrophage and pyroptosis via NF-κB signaling pathway显示文摘Rheumatoid arthritis(RA)is a chronic inflammatory disease that eventually leads to disability.Inflammatory cell infiltration,severe joint breaking and systemic bone loss are the main clinical symptoms.In this study,we established a collagen-induced arthritis(CIA)model and found a large number of M1 macrophages and pyroptosis,which are important sources of proinflammatory cytokines.Punicalagin(PUN)is an active substance extracted from pomegranate peel.We found that it inhibited joint inflammation,cartilage damage and systemic bone destruction in CIA mice.PUN effectively alleviated the high expression of inflammatory cytokines in synovial tissue in vivo.PUN treatment shifted macrophages from the M1 phenotype to the M2 phenotype after stimulation with lipopolysaccharide(LPS)and interferon(IFN)-γ.The expression of inducible nitric oxide synthase(i NOS)and other proinflammatory cytokines released by M1 macrophages was decreased in the PUN treatment group.However,simultaneously,the expression of markers of anti-inflammatory M2 macrophages,such as arginase(Arg)-1 and interleukin(IL)-10,was increased.In addition,PUN treatment attenuated pyroptosis by downregulating the expression of NLRP3 and caspase-1,thereby preventing inflammatory cell death resulting from the release of IL-1βand IL-18.Mechanistically,PUN inhibited the activation of receptor activators of the nuclear factor-κB(NF-κB)signaling pathway,which contributes to M1 polarization and pyroptosis of macrophages.We concluded that PUN ameliorated pathological inflammation by inhibiting M1 phenotype polarization and pyroptosis and has great potential as a therapeutic treatment for human RA.Gaoran Ge Jiaxiang Bai Qing Wang Xiaolong Liang Huaqiang Tao Hao Chen Minggang Wei Junjie Niu Huilin Yang Yaozeng Xu Yuefeng Hao Yi Xue Dechun Geng 2022Science China(Life Sciences)2022,65,3:14
4Assessing the numbers of SNPs needed to establish molecular IDs and characterize the genetic diversityof soybean cultivars derived from Tokachi nagaha显示文摘The development of a core set of SNP molecular markers that could be widely used in soybean genetic research would greatly facilitate research into the genetic diversity of soybean.We conducted an analysis of Tokachi nagaha and 137 of its descendant soybean cultivars using 4044 SNP markers with the goal of determining the appropriate number of single-nucleotide polymorphisms(SNPs)needed to construct unambiguous molecular IDs and characterize genetic diversity based on a genetic distance matrix correlation method.When the number of SNPs was held constant,the number of accession pairs that could be distinguished increased as the polymorphism informative content(PIC)value of the SNPs increased.A core panel of 20 selected SNPs from 11 linkage groups with a mean PIC value of 0.3703 and a range of 0.3640–0.3749 was able to identify almost all of the accession pairs in our study[9445 pairs(99.92%)].The eight accession pairs that could not be identified with this core SNP set all originated from the same province and some of them had the same parental cultivars.The molecular IDs of the 138 accessions were constructed using the core 20 SNPs.It is known that both the number of SNPs and PIC values should be considered when SNPs are selected for use in the analysis of genetic diversity.In this study,when the PIC value was 0.3460,the correlation coefficient between the genetic distance matrices associated with a panel of 200 SNPs and the total population was>0.800,indicating satisfactory correlation.Our high-accuracy,high-resolution core SNP panel for germplasm fingerprinting and our findings about assessing genetic diversity will likely markedly improve the management and utilization efficiency of soybean germplasm resources.Zhangxiong Liu Jun Li Xuhong Fan Nang Myint Phyu Sin Htwe Shuming Wang Wen Huang Jiyu Yang Lili Xing Lijun Chen Yinghui Li Rongxia Guan Ruzhen Chang Dechun Wang Lijuan Qiu 2017The Crop Journal2017,5,4:10
5Real-time rapid prediction of variations of Earth's rotational rate显示文摘Real-time rapid prediction of variations of the Earth's rotational rate is of great scientific and practical importance. However, due to the complicated time-variable characteristics of variations of the Earth's rotational rate (i.e., length of day, LOD), it is usually difficult to obtain satisfactory predictions by con-ventional linear time series analysis methods. This study employs the nonlinear artificial neural net-works (ANN) to predict the LOD variations. The topology of the ANN model is determined by minimizing the root mean square errors (RMSE) of the predictions. Considering the close relationships between the LOD variations and the atmospheric circulation movement, the operational prediction series of axial atmospheric angular momentum (AAM) is incorporated into the ANN model as an additional input in the real-time rapid prediction of LOD variations with 1-5 days ahead. 6Effects of adeno-associated virus (AAV) of transforming growth factors β_1 and β_3 (TGFβ_(1,3)) on promoting synthesis of glycosaminoglycan and collagen type II of dedifferentiated nucleus pulposus (NP) cells显示文摘The effects of AAV-TGFβ1 and AAV-TGFβ3 on promoting synthesis of glycosaminoglycan and collagen type II of dedifferentiated rabbit lumbar disc NP cells were studied in this work. The rabbit lumbar disc NP cells were isolated and cultured. The earlier and later dedifferentiated NP cells were established by subculture. The AAV transfection efficiency to dedifferentiated NP cells was analyzed with AAV-EGFP in vitro. After dedifferentiated NP cells were transfected by AAV-TGFβ1 or AAV-TGFβ3, their biological effects on promoting synthesis of glycosaminoglycan or collagen type II were detected and compared by the methods of 35S incorporation or immunoblotting. The experimental results showed that AAV could transfect efficiently the earlier dedifferentiated NP cells, but its transfection rate was shown to be at a low level to the later dedifferentiated NP cells. Both AAV-TGFβ1 and AAV-TGFβ3 could promote the earlier dedifferentiated NP cells to synthesize glycosaminoglycan and collagen type II, and the effect of AAV-TGFβ1 was better than that of AAV-TGFβ3. For the later dedifferentiated NP cells, the AAV-TGFβ3 could promote their synthesis, but AAV-TGFβ1 could slightly inhibit their synthesis. 10Changes in compressed neurons from dogs with acute and severe cauda equina constrictions following intrathecal injection of brain-derived neurotrophic factor-conjugated polymer nanoparticles显示文摘This study established a dog model of acute multiple cauda equina constriction by experimental constriction injury (48 hours) of the lumbosacral central processes in dorsal root ganglia neurons. The repair effect of intrathecal injection of brain-derived neurotrophic factor with 15 mg encapsulated biodegradable poly(lactide-co-glycolide) nanoparticles on this injury was then analyzed. Dorsal root ganglion cells (L7) of all experimental dogs were analyzed using hematoxylin-eosin staining and immunohistochemistry at 1, 2 and 4 weeks following model induction. Intrathecal injection of brain-derived neurotrophic factor can relieve degeneration and inflammation, and elevate the expression of brain-derived neurotrophic factor in sensory neurons of compressed dorsal root ganglion. Simultaneously, intrathecal injection of brain-derived neurotrophic factor obviously improved neurological function in the dog model of acute multiple cauda equina constriction. 12A programmed surface on polyetheretherketone for sequentially dictating osteoimmunomodulation and bone regeneration to achieve ameliorative osseointegration under osteoporotic conditions显示文摘Polyetheretherketone(PEEK)is a desirable alternative to conventional biomedical metals for orthopedic implants due to the excellent mechanical properties.However,the inherent bioinertness of PEEK contributes to inferior osseointegration of PEEK implants,especially under pathological conditions of osteoporosis.Herein,a programmed surface is designed and fabricated on PEEK to dictate osteoimmunomodulation and bone regeneration sequentially.A degradable hybrid coating consisting of poly(lactide-co-glycolide)and alendronate(ALN)loaded nano-hydroxyapatite is deposited on PEEK and then interleukin-4(IL-4)is grafted onto the outer surface of the hybrid coating with the aid of N_(2) plasma immersion ion implantation and subsequent immersion in IL-4 solution.Dominant release of IL-4 together with ALN and Ca^(2+) during the first few days synergistically mitigates the early acute inflammatory reactions and creates an osteoimmunomodulatory microenvironment that facilitates bone regeneration.Afterwards,slow and sustained delivery of ALN and Ca^(2+) in the following weeks boosts osteogenesis and suppresses osteoclastogenesis simultaneously,consequently ameliorating bone-implant osseointegration even under osteoporotic conditions.By taking into account the different phases in bone repair,this strategy of constructing advanced bone implants with sequential functions provides customizable and clinically viable therapy to osteoporotic patients.Yanyan Zheng Ang Gao Jiaxiang Bai Qing Liao Yuzheng Wu Wei Zhang Min Guan Liping Tong Dechun Geng Xin Zhao Paul K.Chu Huaiyu Wang 2022Bioactive Materials2022,7,8:2
13Rational integration of defense and repair synergy on PEEK osteoimplants via biomimetic peptide clicking strategy显示文摘Polyetheretherketone(PEEK)has been widely used as orthopedic and dental materials due to excellent mechanical and physicochemical tolerance.However,its biological inertness,poor osteoinduction,and weak antibacterial activity make the clinical applications in a dilemma.Inspired by the mussel adhesion mechanism,here we reported a biomimetic surface strategy for rational integration and optimization of anti-infectivity and osteo-inductivity onto PEEK surfaces using a mussel foot proteins(Mfps)-mimic peptide with clickable azido terminal.The peptide enables mussel-like adhesion on PEEK biomaterial surfaces,leaving azido groups for the further steps of biofunctionalizations.In this study,antimicrobial peptide(AMP)and osteogenic growth peptide(OGP)were bioorthogonally clicked on the azido-modified PEEK biomaterials to obtain a dual-effect of host defense and tissue repair.Since bioorthogonal clicking allows precise collocation between AMP and OGP through changing their feeding molar ratios,an optimal PEEK surface was finally obtained in this research,which could long-term inhibit bacterial growth,stabilize bone homeostasis and facilitate interfacial bone regeneration.In a word,this upgraded mussel surface strategy proposed in this study is promising for the surface bioengineering of inert medical implants,in particular,achieving rational integration of multiple biofunctions to match clinical requirements.Meng Li Jiaxiang Bai Huaqiang Tao Li Hao Weiling Yin Xiaoxue Ren Ang Gao Ning Li Miao Wang Shiyuan Fang Yaozeng Xu Liang Chen Huilin Yang Huaiyu Wang Guoqing Pan Dechun Geng 2022Bioactive Materials2022,7,2:2
14Targeting strategies for bone diseases:signaling pathways and clinical studies显示文摘Since the proposal of Paul Ehrlich’s magic bullet concept over 100 years ago,tremendous advances have occurred in targeted therapy.From the initial selective antibody,antitoxin to targeted drug delivery that emerged in the past decades,more precise therapeutic efficacy is realized in specific pathological sites of clinical diseases.As a highly pyknotic mineralized tissue with lessened blood flow,bone is characterized by a complex remodeling and homeostatic regulation mechanism,which makes drug therapy for skeletal diseases more challenging than other tissues.Bone-targeted therapy has been considered a promising therapeutic approach for handling such drawbacks.With the deepening understanding of bone biology,improvements in some established bone-targeted drugs and novel therapeutic targets for drugs and deliveries have emerged on the horizon.In this review,we provide a panoramic summary of recent advances in therapeutic strategies based on bone targeting.We highlight targeting strategies based on bone structure and remodeling biology.For bone-targeted therapeutic agents,in addition to improvements of the classic denosumab,romosozumab,and PTH1R ligands,potential regulation of the remodeling process targeting other key membrane expressions,cellular crosstalk,and gene expression,of all bone cells has been exploited.For bone-targeted drug delivery,different delivery strategies targeting bone matrix,bone marrow,and specific bone cells are summarized with a comparison between different targeting ligands.Ultimately,this review will summarize recent advances in the clinical translation of bone-targeted therapies and provide a perspective on the challenges for the application of bone-targeted therapy in the clinic and future trends in this area.Hao Xu Wentao Wang Xin Liu Wei Huang Chen Zhu Yaozeng Xu Huilin Yang Jiaxiang Bai Dechun Geng 2023Signal Transduction and Targeted Therapy2023,8,6:2

19Research on the photoluminescence of spectral broadening by rapid thermal annealing on InAs/GaAs quantum dots显示文摘Photoluminescence (PL) test was conducted to investigate the effect of rapid thermal annealing (RTA) on the opticalperformance of self-assembled InAs/GaAs quantum dots (QDs) at the temperatures of 16 and 300 K. It was found that after RTAtreatment, the PL spectrum of the QDs sample had a large blue-shift and significantly broadened at 300 K. Compared with theas-grown InAs QDs sample, the PL spectral width has increased by 44.68 meV in the InAs QDs sample RTA-treated at800 ℃.
